Objective : This study was designed to determine whether manual acupuncture therapy is effective for symptoms of patients with idiopathic Parkinson's disease. Methods : Nineteen patients consecutively enrolled in this study. The subjects were divided into two groups : the first group (n=12) discontinued antiparkinsonian drug treatment, the second group (n=7) continued antiparkinsonian drug treatment Acupuncture therapy was performed twice a week for 8 weeks. In the first group, the acupuncture was performed after 2 weeks of drug wash-out period. The patient's symptoms were assessed at pre-treatment, 4 weeks and 8 weeks after the acupuncture by unified Parkinson's disease rating scale (UPDRS), modified Hoehn-Yahr (H-Y) stage, Schwab & England activity of daily living and Freezing of Gait Questionnaire(FOGQ). Results : In the first group that discontinued. antiparkinsonian drug treatment, the total UPDRS scores were significantly improved after 4 weeks (p=0.0l4) and after 8 weeks (p=0.049) compared to the pre-treatment. Particularly, the scores of UPDRS III were significantly improved after 8 weeks (p=0.032). In the second group that continued antiparkinsonian drug treatment, the total UPDRS scores were significantly improved after 4 weeks (p=0.027) and after 8 weeks (p=0.018). Particularly, the scores of UPDRS II were significantly improved after 4 weeks (p=0.042) and after 8 weeks (p=0.043), and UPDRS IV were significantly improved after 8 weeks (p=0.043). The scores of ADL were significantly improved after 8 weeks (p=0.046). Conclusion : This study suggests that manual acupuncture therapy is effective for the improvement of certain symptoms in patients with idiopathic Parkinson's disease.
The efficacy of anticancer drugs depends on a variety of signaling pathways, which can be positively or negatively regulated. In this study, we show that SETDB1 HMTase is down-regulated at the transcriptional level by several anticancer drugs, due to its inherent instability. Using RNA sequence analysis, we identified FosB as being regulated by SETDB1 during anticancer drug therapy. FosB expression was increased by treatment with doxorubicin, taxol and siSETDB1. Moreover, FosB was associated with an increased rate of proliferation. Combinatory transfection of siFosB and siSETDB1 was slightly increased compared to transfection of siFosB. Furthermore, FosB was regulated by multiple kinase pathways. ChIP analysis showed that SETDB1 and H3K9me3 interact with a specific region of the FosB promoter. These results suggest that SETDB1-mediated FosB expression is a common molecular phenomenon, and might be a novel pathway responsible for the increase in cell proliferation that frequently occurs during anticancer drug therapy.

Background and Objectives: Human induced pluripotent stem cell (hiPSC)-derived cardiomyocyte (CM) hold great promise as a cellular source of CM for cardiac function restoration in ischemic heart disease. However, the use of animal-derived xenogeneic substances during the biomanufacturing of hiPSC-CM can induce inadvertent immune responses or chronic inflammation, followed by tumorigenicity. In this study, we aimed to reveal the effects of xenogeneic substances on the functional properties and potential immunogenicity of hiPSC-CM during differentiation, demonstrating the quality and safety of hiPSC-based cell therapy. Methods and Results: We successfully generated hiPSC-CM in the presence and absence of xenogeneic substances (xeno-containing (XC) and xeno-free (XF) conditions, respectively), and compared their characteristics, including the contractile functions and glycan profiles. Compared to XC-hiPSC-CM, XF-hiPSC-CM showed early onset of myocyte contractile beating and maturation, with a high expression of cardiac lineage-specific genes (ACTC1, TNNT2, and RYR2) by using MEA and RT-qPCR. We quantified N-glycolylneuraminic acid (Neu5Gc), a xenogeneic sialic acid, in hiPSC-CM using an indirect enzyme-linked immunosorbent assay and liquid chromatography-multiple reaction monitoring-mass spectrometry. Neu5Gc was incorporated into the glycans of hiPSC-CM during xeno-containing differentiation, whereas it was barely detected in XF-hiPSC-CM. Conclusions: To the best of our knowledge, this is the first study to show that the electrophysiological function and glycan profiles of hiPSC-CM can be affected by the presence of xenogeneic substances during their differentiation and maturation. To ensure quality control and safety in hiPSC-based cell therapy, xenogeneic substances should be excluded from the biomanufacturing process.

We observed a treatment with constant drug dosage whose efficacy is less than full treatment may lead HIV-infected patient to LTNP. It turns out that the treatment whose efficacy is less than full treatment is better in the point of performance on controllability.
Objectives: The objective of this study was to report a clinical case that suggests potential beneficial effects of Korean medicine therapy for the treatment of levosulpiride-induced acute dyskinesia. Methods: A patient having drug-induced acute dyskinesia was given a series of Korean medicine therapy treatments, including administration of the herbal medicine Chengsimyeonja-tang-gamibang, acupuncture, electroacupuncture, pharmacopuncture, and moxibustion over the 17 days of the hospitalization period. During the therapy, the progression of the disease was measured by the Abnormal Involuntary Movement Scale (AIMS). Results: During the therapy, the remission of orofacial dyskinesia and improvement of AIMS scores were observed. Conclusion: Korean medicine therapy has potential benefits for the treatment of drug-induced acute dyskinesia.
Statins are competitive inhibitors of hydroxy-3-methyl glutaryl coenzyme A (HMG-CoA) reductase and used most frequently to reduce plasma cholesterol levels and to decrease cardiovascular events. However, statins also have been reported to have undesirable side effects such as myotoxicity and hepatotoxicity associated with their intrinsic efficacy mechanisms. Clinical studies recurrently reported that statin therapy elevated the level of liver enzymes such as ALT and AST in patients suggesting possible liver toxicity due to statins. This observation has been drawn great attention since statins are the most prescribed drugs and statin-therapy was extended to a larger number of high-risk patients. Here we employed rat primary hepatocytes and microarray technique to understand underlying mechanism responsible for statin-induced liver toxicity on cell level. We isolated genes whose expressions were commonly modulated by statin treatments and examined their biological functions. It is of interest that those genes have function related to response to stress in particular immunity and defense in cells. Our study provided the basic information on cellular mechanism of statin-induced cytotoxicity and may serve for finding indicator genes of statin -induced toxicity in rat primary hepatocytes.

In the past decade, significant progress has been achieved in the battle against hepatitis B virus. In addition to the immunomodulating agents such as interferon-.alpha., and thymosin, many novel antiviral agents have been discovered, among which nucleoside analogues are the mainstay. New-generation compounds such as 3TC and famciclovir have shown promise in the treatment of patients chronically infected by this virus, and are on the line for approval. However, viral rebound after cessation of therapy still remains a major problem. Additionally, the reports on the drug resistance to these antiviral agents suggest that combination therapy will be the eventual strategy (Bartholomew et al., 1997; Tipples et al., 1996). Therefore, developments of safe and effective antiviral agents which do not cross-resist with currently available antiviral drugs are still much needed.

Development of dual functional liposome has been studied for cancer theragnostics. Therefore, we focused on ultrasound-sensitive liposomes with doxorubicin (DOX) and gadolinium (Gd) as a theragnostic carrier having a potential for cancer therapy and diagnosis. In this study, Gd(III)-DOTA-modified sonosensitive liposomes (GL) was developed using chemically synthesized Gd(III)-DOTA-DPPE lipid. Sonosensitivity of GL to 1 MHz ultrasound induced 25% of DOX release. The relaxivities ($r_1$) of GL were $7.33-10.34\;mM^{-1}s^{-1}$, which was higher than that of MR-bester$^{(R)}$. Intracellular delivery of DOX from GL by ultrasound irradiation was evaluated according to ultrasound intensity, resulting in increase of uptake of DOX released from ultrasound-triggered GLs compared to GL3 or Doxil$^{(R)}$ without ultrasound. Taken together, this study shows that the paramagnetic and sonosensitive liposomes, GL, is a novel and highly effective delivery system for drug with the potential for broad applications in human disease.
