Selection of PCR Markers and Its Application for Distinguishing Dried Root of Three Species of Angelica

  • An analysis of RAPD-PCR (random amplified polymorphic DNA-polymerase chain reaction) was performed with three Angelica species (A. gigas Nakai, A. sinensis (Olive.) Diels and A. acutiloba Kitag) in an effort to distinguish between members of these three species. Two arbitrary primers (OPC02, OPD11) out of80 primers tested, produced 17 species-specific fragments among the three species. Eight fragments were specific for A. sinensis, four fragments specific for A. gigas, five specific for A. acutiloba. When primers OPC02 and OPD11 were used in the polymerase chain reaction, RAPD-PCR fragments that were specific for each of the three species were generated simultaneously. Primer OPC02 produced eight species-specific fragments: four were specific for A. sinensis, one for A. gigas, and three for A. acutiloba. Primer OPD11 produced nine speciesspecific fragments: four for A. sinensis, three for A. gigas, and two for A. acutiloba. The RAPD-PCR markers that were generated with these two primers should rapidly identify members of the three Angelica species. The consistency of the identifications made with these species-specific RAPD-PCR markers was demonstrated by the observation that each respective marker was generated from three accessions of each species, all with different origins. We also performed the RAPD-PCR analysis with the dried Angelica root samples that randomly collected from marketed and from the OPC02 primer, obtained a A. gigasspecific band and the band were cloned and sequenced.

  • Essential oil of the root of Angelica decursiva(Miq.) Fr. et Sav. (Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain ten monoterpenes such as ${\alpha}-pinene$ (7.0%) etc. Three hydrocarbons, two aldehydes, three sesquiterpenes, two sesquiterpene alcohols, one aromatic compounds, one ketone, isobonyl acetate and two lactones were tentatively identified.

  • In continuation of our studies on essential oils of Angelica genus(Umbelliferae), We report on the components of essential oils obtained from the root of Angelica tenuissima Nakai(藁子). Oils were obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. Essential oils of the root of A. tenuissima(Gaoben) were found to contain $\alpha-pinene,\;camphene,\;\beta-pinene,\;myrcene,\;\alpha-phellandrene,\;\Delta-3-carene,\;p-cymene,\;limonene,\;\gamma-terpinene,\;terpinolene,\;4-vinylguauacol,\;\gamma-elemene$, one aromatic compound, three unidentified sesquiterpene alcohols, butylidenephthalide, senkyunolide and Z-ligustilide which was the most abundant compound comprising 75% of the whole oil. Also butylphthalide and hydroxybutylidenephalide were tentatively identified.

  • This study was conducted to find out the variation modes of the absorbtion contents of N, P and K and the contents of decursin, decursinol and sugar in Angelica gigas Nakai at different growth stages. The results of field study were as follows. The dried weights of root and shoot was increased by the application of nitrogen and potassium fertilizers during the growing seasons. But, the effect of phosphorus application was observed only the increasing of the dried weight of shoot. During the growing seasons, the contents of nitrogen and phosphorus in root was decreased, but in shoot was increased. The potassium contents of root was increased and that of shoot was decreased. The decursin and total sugar contents of root were continually increased, and the decursinol and reducing sugar contents were increased until the middle growth stage and after that were decreased.

  • The ether extract of Dang-Gui, the dried root of Angelica gigas N$_{AKAI}$, was placed into a column with aluminium oxide gel. The elution of it with a mixture of n-hexane and ethyl acetate afforded Nodakenetin (I) and the elution with a mixture of n-hexane, ethyl acetate, acetone and methyl alcohol yielded Umbelliferon (II) and Nodakenin (III). Three coumarin derivatives were identified by comparing with authentic samples. The existence of sucrose and glucose was also proved.

  • Dried parts of the herb medicines are difficult to distinguish morphologically. Heracleum moellendorffii cordata has often been sold instead of Aralia cordata in herbal medicine markets. Therefore, this study was conducted to develop the key for discrimination between them using the RAPD analysis and morphological characteristics. Thirty decarmer oligonucleotide primers were screened for the RAPD analysis, and four primers generated distinct RAPD markers specific to Aralia cordata, Angelica pubescens maxim f. biserrata, and Heracleum moellendorffii. The specific RAPD patterns generated by the selected primers were reproducible from dried materials. In comparison of morphological characteristics, Aralia cordata seems to be entirely developed in xylem fiber, but not developed in pith.

  • Essential oil of the root of Angelica koreana Max. (Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain nine monoterpenes such as ${\alpha}-pinene$(7.0%), camphene, ${\beta}-pinene$, myrcene, ${\alpha}-phellandrene$, ${\Delta}-3-carene$, p-cymene, limonene(2.8%), terpinolene and also found to contain m-cresol(11.6%), citronellol, citral b, methylcinnamate, eudesmol and osthol. 2-Hydroxy-5-methylacetophenone and twelve compounds were tentatively identified.

  • Essential oil of the root of Angelica dahurica Benth et Hook(Umbelliferae) was investigated. Essential oil was obtained from the dried roots by steam distillation and fractionated by column chromatography. Each isolate or fraction was identified by GC, GC-MS and spectral analysis. It was found to contain eleven monoterpenes such as ${\alpha}-pinene(4.74%),\;campben, {\beta}-pinene,\;myrcene,\;{\alpha}-phellandrene,\;{\delta}-3-carene(39.4%),\;{\alpha}-terpinene,\;{\rho}-cymene,\;{\beta}-phellandrene,\;{\alpha}-terpinene,\;terpinolene\;and\;also\;found\;to\;contain\;4-vinylguaiacol,\;iso-elemicin,\;{\beta}-elemene$, caryophyllene, ligustilide, osthol and seven tentatively identified sesquiterpenes.

  • Angelicae gigantis Radix (dried root of Angelica gigas) including major bioactives such as decursin and decursinol angelate provides rich flavors and several healthy benefits. Recent studies have shown that ultrafine powders of herbal medicines provide better physical properties and biological activities. Thus, ultrafine Angelica powder was added into the oligosaccharide syrup to provide flavors and healthy benefits in this study. Angelicae gigantis Radix was pulverized into d(0.1) = 3.220, d(0.5) = 7.822, and d(0.9) = 7.817 $\mu$m respectively using an air-flow mill. The ultrafine Angelica powder was added into the oligosaccharide syrup process with different ratios of water to oligosaccharide syrup at 1:5, 1:8, 1:11, and 1:14. The physicochemical properties such as viscosity and bulk density were measured. The Stokes' law was applied to predict the sedimentation velocity of the added Angelica powder in the syrup. The Angelica syrup prepared in this experiment showed good stability since the Angelica particles precipitated down slowly. The ratio of water to oligosaccharide syrup at 1:11 showed the optimal preparation in terms of the stability and the viscosity. The ultrafine-sized herbal powders such as Angelicae gigantis Radix have potentials for various food and pharmaceutical applications.
