To determine the origin and genetic diversity of Red Chittagong (RC) cattle in Bangladesh, we analyzed mitochondrial DNA displacement loop (D-loop) sequences of 48 samples along with 22 previously published sequences from Bos indicus and Bos taurus breeds. Twenty five haplotypes were identified in RC cattle that were defined by 44 polymorphic sites and nucleotide diversity was $0.0055{\pm}0.0026$. The estimated sequence divergence times between RC and other zebu cattle breeds studied ranged between 22,700-26,900 years before present (YBP) which, it is suggested, predate domestication of RC cattle. Furthermore, it is assumed that introgressions have occurred in this breed mainly from Indian zebu breeds in the recent millennia. The phylogenetic studies showed RC cattle clustered with Bos indicus lineage with two distinct haplogroups representing high genetic variability of this breed. These findings can be used for designing proper breeding and conservation strategies for RC cattle in Bangladesh.
The study attempts to assess the status, distribution, conservation and use value of Medicinal and Aromatic Plant (MAPs) in the Sagarmatha National Park. Altogether 62 species of MAPs belonging to 47 genera and 33 families have been recorded in the study area. 10 species, belonging to 9 families are categorized as the potential species. Out of the these species, most potential in local but threatened species are Allium hypsistum Stearn, Cordyceps sinensis Sacc, Dactylorhiza hatagirea Soo, Nardostachys grandiflora DC, Aconitum orochryseum, Ephedra gerardiana Wall. Ex. Stapf, Swertia multicaulis D. Don, Picrorhiza scrophulariflora Penne, Rheum australe. D. Don, Malva verticillataL and Swertia pedicallata Benerji. By analysis of data using Simpson's diversity index (SI) and Shannon weaver function (H'), there was high diversity (more heterogeneous) MAPs species composition in the Manjo Gate to Large Dobhan. (0.98349). Less diversity (less heterogeneous) MAPs species composition was in Tyanboche to Pangoche (0.90419). Similarly, the Shannon weaver function shows that in plots laying out in Mongla to Phorche are evenly distributed than others However, higher MAPs species (i.e., 31) was found in the way of Manjo Gate to Larja Dobhan than others.

Although there have been many studies of native Korean cattle, Hanwoo, there have been no selective sweep studies in these animals. This study was performed to characterize genetic variation and identify selective signatures. We sequenced the genomes of 12 cattle, and identified 15125420 SNPs, 1768114 INDELs, and 3445 CNVs. The SNPs, INDELs, and CNVs were similarly distributed throughout the genome, and highly variable regions were shown to contain the BoLA family and GPR180, which are related to adaptive immunity. We also identified the domestication footprints of the Hanwoo population by searching for selective sweep signatures, which revealed the RCN2 gene related to BPV resistance. The results of this study may contribute to genetic improvement of the Hanwoo population in Korea.
Asia hosts more than half of the world's 1 billion goats and is also where domestication of wild goats began. Goats, including dairy goats, are adapted to a wide variety of harsh environments and thus play key roles as providers of nutrition, food security and socio-economic status to their human owners in many low-income Asian countries. In many countries in Southeast and East Asia, medium and large scale commercial dairy goat farming can be profitable enterprises because of the high price of goat milk, and good demand due to its health and medicinal properties. In some Asian countries, dairy goats play important roles in non-commercial activities, including use as educational animals in elementary schools in Japan and show animals in Indonesia. Dairy goat farmers in Asia are faced with numerous challenges, such as a shortage of high producing animals adapted to the local environment, lack of quality feeds during a prolonged dry season, many diseases and difficulty getting their product to market, however, the increasing demand for goat milk in the newly developed and developed economies in Asia provides an optimistic future for dairy goat production in this region.
Diospyros mespiliformis is a highly valued and threatened tree species within the Sahelo-Sudanian zone of Africa, but its seed germination requirements under cultivation are not well researched. In a first experiment which aimed at determining germination response of seeds to dehydration, fresh seeds were dried at room temperature for 26 days during which their moisture content, their germinability, and their viability were monitored at two-day intervals. In the second experiment, 14 pre-germination treatments were tested for their effect on the germination of dried seeds. Results showed that fresh seeds had 52.7% moisture and achieved 97.7% germination. As seeds were dried, percentage germination gradually decreased with decreasing moisture content and reached 0% when moisture content had dropped to 18%. Meanwhile, seed viability remained at 100% over drying duration. Seeds that were not germinated after air dry also recorded 100% viability. The most effective treatment for inducing germination of dried seeds was scarification using 98% sulfuric acid for 30 min which resulted in 96.6% germination. This study reports for the first time in D. mespiliformis seeds a desiccation-induced dormancy which can be efficiently alleviated by acid scarification. This study provides useful information that will contribute to efficient management of D. mespiliformis seed resources for propagation.

Objective: To understand the domestication and spread of horses in history, genetic information is essential. However, mitogenetic traits of ancient or medieval horses have yet to be comprehensively revealed, especially for East Asia. This study thus set out to reveal the maternal lineage of skeletal horse remains retrieved from a 15th century archaeological site (Gongpyeongdong) at Old Seoul City in South Korea. Methods: We extracted DNA from the femur of Equus caballus (SNU-A001) from Joseon period Gongpyeongdong site. Mitochondrial (mt) DNA (HRS 15128-16116) of E. caballus was amplified by polymerase chain reaction. Cloning and sequencing were conducted for the mtDNA amplicons. The sequencing results were analyzed by NCBI/BLAST and phylogenetic tool of MEGA7 software. Results: By means of mtDNA cytochrome b and D-loop analysis, we found that the 15th century Korean horse belonged to haplogroup Q representing those horses that have historically been raised widely in East Asia. Conclusion: The horse is unique among domesticated animals for the remarkable impact it has on human civilization in terms of transportation and trade. Utilizing the Joseon-period horse remains, we can obtain clues to reveal the genetic traits of Korean horse that existed before the introduction of Western horses.
연근해 어선 등 소형선박에 설치되는 항해통신어로기자재는 대외의존도가 높아 외산제품에 의존하고 있다. 국산화 관점에서 해외 제품수준의 국산화 기술개발이 진행되고 있으며, Track Record 확보 차원에서 전용 테스트베드를 제작하여 실증하고자 한다. 본 논문에서는 테스트베드 설계를 위한 기초연구를 통하여 테스트베드 형태 및 설계방안을 도출하고자 하였다.
Disproportionating Enzyme 1 (DPE1) is an a-1,4-D-glucanotransferase that cleavages the a-1,4-glucosidic bonds and transfers glucosyl groups. In rice endosperm, it participates in starch synthesis by transferring maltooligosyl groups from amylose and amylopectin to amylopectin. Here, we investigated the haplotype variations and evolutionary indices (e.g., genetic diversity and population structure) for the DPE1 gene in 374 rice accessions representing seven subgroups (wild, indica, temperate japonica, tropical japonica, aus, aromatic, and admixture). Variant calling analysis of DPE1 coding regions leads to the identification of six functional haplotypes representing/occupying 8 nonsynonymous SNPs. Nucleotide diversity analysis revealed the highest pi-value in wild group (0.0556) compared to other cultivated groups, of which temperate japonica showed the most reduction of genetic diversity value (0.003). A significant positive Tajima's D value (1.6330) of admixture highlights sudden population contraction under balancing selection, while temperate japonica with the lowest Tajima's D value (-1.3523) showed a selection signature of DPE1 domestication which might be the cause of excess of rare alleles. Moreover, these two subpopulations exhibits a greater differentiation (FST=0.0148), indicating a higher genetic diversity. Our findings on functional DPE1 haplotypes will be useful in future breeding programs, and the evolutionary indices can also be applicable in functional studies of the DPE1 gene.
Starch synthase proteins (SSI, SSII and SSIII) in rice are mainly involved in amylopectin synthesis mediating its chain elongation, and the functional loss of SSII can increase amylose accumulation through decreasing of amylopectin chain proportions. For purposes of identifying functional haplotypes and evolutionary analyses of this gene, SSII-1, we investigated 374 rice accessions belonging to different subgroups of origins. We subsequently performed bioinformatic analyses on their variations through haplotyping, resequencing and structuring based on different classified populations. Haplotyping of cultivated rice accessions using genetic variations within SSII-1 genomic region of chromosome 10 revealed a total of 8 haplotypes, representing 6 functional haplotypes by 4 non-synonymous SNPs of three different exons (1, 4 and 10), which effect on protein structure. Higher nucleotide diversity value was found in wild group (0.0055) compared to any of cultivated subpopulations, of which aus showed the most reduction of diversity value (0.0003). Tajima's D analysis exhibits the most Tajima's D value only in admixture group (0.3600) which appears to be the cause of a sudden population contraction by rare alleles scarcity. A clear separation of some wild accessions from the admixed cultivated subpopulations was observed in PCA and phylogenetic analysis. Similar admixed pattern of population structure was estimated with an increased K values of 2 to 8 where genetic components of almost all cultivated subpopulations were shared with the wild which can also be subsequently estimated by very low FST-values by -0.011 (wild-aromatic) and -0.003 (wild-admixture).
Pullulanase (PUL), a debranching enzyme, has been utilized in hydrolyzing the a-1,6 glucosidic linkages in starch, amylopectin, pullulan, as well as related oligosaccharides. It has also been indicated that PUL is a novel indicator of inherent RS (Resistant Starch) formation in rice. In this study, we performed haplotype analysis on 320 bred rice accessions, and additional 54 wild accessions were added to study genetic diversity along with other population-based analyses of the PUL gene. Through these investigations, we summarized a total of 10 functional (non-synonymous) SNPs from 7 different exons on chromosome 4. There were 10 haplotypes, of which only six haplotypes were functional, implicating different subpopulations. Diversity reduction was noticed in temperate japonica (0.0005) compared to the highest one (aus, 0.0154), illustrating their higher genetic differentiation by FST-value (0.926). The highest Tajima^ D value was observed in indica (3.6613), indicating PUL gene domestication signature under balancing selection, while the lowest Tajima's D value was found in temperate japonica (-2.2191) which might have undergone under positive selection and purified due to the excess of rare alleles. PCA, population structure, and phylogenetic analyses provide information on the genetic relatedness between and or among the cultivated subpopulations and the wild based on PUL genomic region.
