Effect of $\gamma$-Irradiation of Volatile Organic Compounds of Houttuynia cordata Thunb. (방사선 조사에 의한 어성초의 휘발성 유기성분 변화)

  • This study was performed to examine the effect of $\gamma$-irradiation on the volatile organic compounds in Houttuynia cordata Thunb.(H. cordata). 1be volatile compounds of non-irradiated and 10 kGy $\gamma$-irradiated H. cordata were isolated using SDE apparatus and analyzed by GC/MS. For each treatment the number of volatile compounds were detected at 83 and 85, respectively, 1be predominant functional groups of volatile organic compounds from H. cordata were identified as alcohols and ketones. H. cordata was mainly composed of hexahydrofarnesyl acetone (12.81 %), phytol, decanoic acid, dodecanoic acid, octadecanol, caryophyllene oxide, 2-undecanone and menthol. Houttuynum which is characteristic compound of H. cordata was found in all samples and there was no significant difference with irradiation doses. Consequently, $\gamma$-irradiation of H. cordata would be an effective process for sanitation and to increase extraction efficiency.

Profiling of Volatile Components Using Gas Chromatography-Mass Spectrometry in Commercial Pine Needle (Pinus densiflora S. and Z.) Powder

  • Volatile components in commercial pine needle (Pinus densiflora S. and Z.) powder were extracted using simultaneous steam distillation and a solvent extraction (SDE) apparatus, and were analyzed by gas chromatography-mass spectrometry (GC-MS). A total of 230 compounds divided into 13 groups were identified, which included alcohols (42), ketones (39), aldehydes (32), terpenes (30), alkenes (17), esters (14), furans (14), benzenes (10), alkanes (8), napthalenes (7), acids (6), miscellaneous compounds (6), and phenols (5). Among the 230 compounds identified, 96 compounds were positively confirmed and quantified, and the rest of the compounds were tentatively identified. The major volatile components identified at relatively high levels were dodecanoic acid, hexadecanoic acid, hexanal, benzaldehyde, (Z)-3-hexen-1-ol, 1-penten-3-one, limonene, and $\beta$-caryophyllene oxide. Among the groups, terpenes accounted for 60.18% of the total concentration of all the volatile components. Some volatile components might account for the unique aroma and the biological activity of the sample.

  • In this research, the enhancement of electron-transfer activity of hemoglobin (Hb) in dodecanoic acid film was investigated for the first time. This type of composite film was made on glassy carbon electrode by casting method. Cyclic voltammetric result of the modified electrode displays a well defined redox peaks which was attributed to the direct electrochemical response of Rb. Our results illustrate that Rb exchange electrons directly with electrode and exhibits the characteristics of peroxidase. When we apply this modified electrode as a biosensor, it gives excellent performances in the electrocatalytic reduction of hydrogen peroxide ($H_{2}O_{2}$). Through the optimal conditions, the proposed biosensor shows the linear range for H2O2 determination was from $1{\times}10^{-5}$ to $1.25{\times}10^{-4}mol/L$ with a detection limit of $1{\times}10^{-7}mol/L$. The biosensor retained more than 90% of the initial response after 14 days.

  • Soluble epoxide hydrolase (sEH) is a metabolic regulator of epoxyeicosatrienoic acids (EETs). EETs have many beneficial effects, vasodilation, anti-diabetes, anti-inflammation, cardiovascular protection, renal protection. Therefore, selective sEH inhibitors have a potential for treating these diseases. In the present study, screening methods for sEH inhibitors using PHOME ((3-phenyl-oxiranyl)-acetic acid cyano-(6-methoxynaphthalen-2-yl)-methyl ester) and 14-15-EET as substrates were established. To determine selectivity, microsomal epoxide hydrolase (mEH) inhibition assay was also developed using styrene oxide as a substrate of microsomal epoxide hydrolase. Our results obtained from 12-[[(tricyclo[3.3.1.13,7]dec-1-ylamino)carbonyl]amino]-dodecanoic acid (AUDA) used as a positive sEH inhibitor and valpromide used as a positive mEH inhibitor showed that these methods are useful for discovery of drug candidates.

Cashew Nut Oil: Extraction, Chromatographic and Rheological Characterisation.

  • Oil was extracted from cashew nuts. The physicochemical parameters of the oil were determined. A chromatographic assay of the oil was carried out using Gas Chromatography-Mass Spectrometry. Seventeen compounds were detected: Phenol, Phenol 2-methyl-, Cyclohexene 4, 4-dimethyl-, m-Fluoro-2-diazoacetophenone 4-dimethyl-, Tetradecanoic acid, Phenol 4-octyl-, n-Hexadecanoic acid. Others are 9, 12-Octadecadienoic acid (Z, Z) - methyl ester, Hexadecanoic acid methyl ester, Methyl stearate, Dodecanoic acid methyl ester, 9, 12, 15-Octadecatrienoic acid methyl ester, 9, 12, 15-Octadecatrienoic acid (Z, Z, Z)-, Oleic acid, Octadecanoic acid, Tetracosanoic acid and 9-Octadecenoic acid methyl ester. Among the components are omega three and omega six essential free fatty acids. The rheological profiling and flow properties of cashew nut oil were determined using a Programmable Rheometer. Cashew nut oil exhibits slight dilatant behaviour at the low end of shear rate. The long chain and high molecular weight of its constituents controlled its rheology. Long-chained 9-Octadecenoic acid methyl ester, 9, 12-Octadecadienoic acid (Z, Z) - methyl ester, Tetracosanoic acid and methyl stearate, coupled with their high molecular weights are responsible for the shear thickening effect observed. Two models, Carreau-Yasuda and Ostwald-de Waele Power Law were employed to fit the rheological data. The Carreau-Yasuda model followed well the data.

Changes of Aroma and Volatile Components of Korean Leaf Tobaccos from a Different Stalk Positions (국산 원료잎담배의 착엽위치에 따른 향기성분 및 휘발성 성분의 변화)

  • This study was conducted to determine the aroma and volatile component changes from a different stalk positions of Korean flue-cured tobacco. Eight different stalk positions of flue-cured leaf tobaccos harvested in 2001 were used for this study. Thermal extraction method at two different treatment temperature($50\;amp;\;80^{\circ}C$) was applied for this experiment. Forty eight kinds of aroma and volatile components such as 2,4-heptadienal, hexadecane, 1-methyl-1H- pyridine, 2,5-dimethyl-1H-pyrrole were analyzed by using thermal extraction method. All of aroma and volatile components of leaf tobaccos were changed from a different stalk positions and treatment temperature. Leaf tobaccos in middle stalk position have a higher concentration of aroma and volatile components such as norsolanidione, 4-pyridinecarboxaldehyde, 4-methyl-4-OH-2-pentanone, acetic acid, propylene glycol, 1-methyl-2-pyrrolidinone, 2,5-dimethyl-1H-pyrrole. Also, Megastigmatrienone 1, 3-oxo-[alpha]­ionol, 6,10,14-trimethyl-2-pentadecanone, heptadecane, 6-methyl-2-isohexyl-l-heptene concentration were low in the middle stalk position and high in both bottom and upper position. Treatment temperature affected on the changes of many aroma and volatile components in leaf tobacco. Most of aroma and volatile components such as, 2,4-Heptadienal, dodecanoic methylester, famesol isomer and 3-acetylpyridine were sharply increased as increasing treatment temperature. This results can be used to estimate the aroma characteristics of cigarette blend using a different stalk position of leaf tobacco.

Volatile flavor components of Dioscorea japonica (참마의 휘발성 풍미성분)

  • An attempt was made to derermine the volatile flavor components of Dioscorea japonica. Essential oils from roots of the samples were isolated by simultaneous steam distillation-extraction(SDE) method using diethyl ether as solvent. Concentrated samples were analyzed by gas chromatography(GC) and combined gas chromatography-mass spectrometry(GC-MS). Fifty nine volatile flavor components, including 35 hydrocarbons, 5 aldehydes, 1 ketone, 9 alcohols, 2 esters, 3 acids and 4 miscellaneous ones were confirmed in the young roots of Dioscorea japonica. Forty two components, including 23 hydrocarbons, 2 aldehydes, 7 alcohols, 1 ester and 8 acids and 1 miscellaneous one were confirmed in the roots of mature stage. ${\sigma}-3-Carene$ and dodecanoic acid were regarded as the most abundant components in young and mature roots repectively. The profile of volatile flavor components was markedly different in young and mature roots of Dioscorea japonica.

Polyhydroxyalkanoate (PHA) Production Using Waste Vegetable Oil by Pseudomonas sp. Strain DR2

  • To produce polyhydroxyalkanoate (PHA) from inexpensive substrates by bacteria, vegetable-oil-degrading bacteria were isolated from a rice field using enrichment cultivation. The isolated Pseudomonas sp. strain DR2 showed clear orange or red spots of accumulated PHA granules when grown on phosphate and nitrogen limited medium containing vegetable oil as the sole carbon source and stained with Nile blue A. Up to 37.34% (w/w) of intracellular PHA was produced from corn oil, which consisted of three major 3-hydroxyalkanoates; octanoic (C8:0, 37.75% of the total 3-hydroxyalkanoate content of PHA), decanoic (C10:0, 36.74%), and dodecanoic (C12:0, 11.36%). Pseudomonas sp. strain DR2 accumulated up to 23.52% (w/w) of $PHA_{MCL}$ from waste vegetable oil. The proportion of 3-hydroxyalkanoate of the waste vegetable-oil-derived PHA [hexanoic (5.86%), octanoic (45.67%), decanoic (34.88%), tetradecanoic (8.35%), and hexadecanoic (5.24%)] showed a composition ratio different from that of the corn-oil-derived PHA. Strain DR2 used three major fatty acids in the same ratio, and linoleic acid was the major source of PHA production. Interestingly, the production of PHA in Pseudomonas sp. strain DR2 could not occur in either acetate- or butyrate-amended media. Pseudomonas sp. strain DR2 accumulated a greater amount of PHA than other well-studied strains (Chromobacterium violaceum and Ralstonia eutropha H16) when grown on vegetable oil. The data showed that Pseudomonas sp. strain DR2 was capable of producing PHA from waste vegetable oil.

  • Anionic oligomer surfactants, dodecyl polyoxyethylene ${\alpha}-sulfa$ alkanoates, had been synthesized through the esterification of dodecyl polyoxyethylene glycol and ${\alpha}-sulfa$ alkanoic acid with straight chain alkyl group having from 10 to 18 carbon atoms to good yield. ${\alpha}-sulfa$ alkanoic acids were obtained by reaction with long chain alkanoic acids and sulfur trioxide-dioxane complex, and dodecyl polyoxyethylene glycols, by addition reaction with dodecyl alcohol and ethylene oxide(addition, 5, 10, 20mol) respectively. All the synthetic products could be separated by means of the thin layer and column chromatography, and their structure has characterized with IR, $^1HNMR$ and elemental analysis, respectively.

Comparison of Chemical Compositions of Houttuynia cordata Thunb Cultivated from Different Local Area (재배지역이 다른 어성초의 부위별 화학성분 비교)

  • Proximate composition , volatile compounds, free amino acids, fatty acids and inorganic compounds in leaf, stem and root of Houttyunia cordata Thunb cultivated from two different area, Bosung and Sunchon , were analyzed. Each part of Houttuynia cordata Thunb from both local area showed moisture contents of 80-84% and crude ash contents of 2.1 ∼2.8%. Crude fat and protein contents were less than 3 % showing slightly higher contents in leaf than in both stem and root . Twenty six volatile compunds were identified from the parts of the plant, the volatile contents were high in the oder of leaf, root and stem. Major volatile compounds were mostly derivatives of decanoic acid ; decanoic acid, 20 tridecanoie, decanal and dodecanoic acid. Of free amino acids in leaf asparagine was the highest, while in stem hydroxyproline, proline and arginine were the major free amino acids. Linoleic acid was the highest in the stem and root, and linolenic acid was highest in leaf. the major minerals of all parts were K, Ca. Mg, P, Fe, Zn and Cu, showing highest with K.
