Reduction of Nitrotoluenes and Simultaneous Removal of Hydrogen Sulfide and Nitrotoluenes by Co3+-centered Hematoporphyrin (포피린의 촉매작용에 의한 니트로톨루엔의 환원 및 니트로톨루엔과 황화수소의 동시 제거)

  • Mononitrotoluenes were reduced to aminotoluenes using porphyrin as a catalyst in the presence of several types of reductants including hydrogen sulfide and 1, 4-dithiothreitol(DTT). Intermediates and final products of porphyrin-catalyzed reduction of mononitrotoluenes were identified and a pathway for the reduction of the nitro group to the corresponding amino group was proposed. The optimum pH for the reduction was determined. The catalytic activity of the porphyrin was confirmed by UV/VIS absorption spectra and basic kinetics of porphyrin-catalyzed reduction were studied. Of several types of reductants tested, DTT sodium hydrosulfite, and hydrogen sulfide were seen to give significant reduction of nitrobodies. When hydrogen sulfide was used as a reductant hydrogen sulfide and nitrotoluenes were removed simultaneously.

Purification and Characterization of Cholesterol Oxidase Produced by Streptomyces sp. No.4 (방선균 Streptomyces sp. No.4가 생산하는 Cholesterol Oxidase의 정제 및 특성)

  • The cholesterol oxidase(EC.1.1.3.6) produced from Streptomyces sp. No.4 which isolated from soil was purified and investigated for the enzymatic properties. The enzyme was purified specifically by cholesterol affinity column chromatography with a yield of 28.3%. The purified enzyme showed a single polypeptide on SDS-PAGE and the molecular weight was estimated to be 60,000 daltons. The enzyme activity was strongly inhibited by metal ions such as $HgCl_2$ and $CuSO_4$. Dithiothreitol and mercaptoethanol inhibited the enzyme activity at concentration of 1mM. The Michaelis constant(Km) for cholesterol was found to be 1.38mM by Lineweaver-Burk plot analysis. Amino acid analysis showed that the enzyme protein was composed of 416 amino acid residues including 52moles of glycine and 19moles of tryptophane.

  • The synthetic oligomers called nos right palindrome (RP) element and left palindrome (LP) element were inserted into nos.minimal promoter nos 5'-101 deletion mutant The activity of nos promoter was measured by studying the expression pattern of gene fusion between nos promoter and reporter genes such as chloramphenicol acetyltransferase and $\beta$-glucuconidase. Analysis of transgenic tobacco plane carrying transgene showed that the activity of nos minimal promoter activity was recovered by insertion of synthetic nos RP element. Nos RP element insertion of nos minimal promoter was induced by auxin, dithiothreitol, salicylic acid and methyl jasmonate.

한국원자력연구소 방사선방어기술 개발 및 연구 현황

  • 1959년 한국원자력연구소가 창립됨과 동시에 &Health Physics&, 즉 보건물리라고 하는 명칭과 조직이 탄생되어, 방사선안전관리의 실무와 보건물리의 연구가 시작되었다. 최초 10년간은 선진제국의 보건물리분야의 연구와 기술을 추적하여 우리나라의 방사선안전관리 기술의 기초를 다지는 시기로서 개인방사선모니터링기술, 환경방사선(능) 모니터링기술 및 방사선방어용계측기기의 교정기술 개발에 중점을 두고 연구개발이 추진되었으며, TRIGA Mark-II 연구용원자로의 가동에 따라 원자로 생체차폐체의 건전성 검증에 관한 유익한 방사선량 측정자료도 얻게 되었다. 즉 이 기간은 방사선안전관리의 체제정비 및 기초기술 확립에 노력한 기간이었다. 1970년대는 원자력 연구개발에 대한 기본방향과 정책의 변경등으로 보건물리 연구조직은 방사선안전관리, 환경연구 그리고 방사화학분야로 분산되었으며, 그로인하여 연구개발활동은 거의 정체되어 겨우 방사선안전관리 실무만이 그 명맥을 유지하였다. 그 결과 우리나라 방사선안전관리 및 그와 관련된 연구개발의 기반이 흔들리게 되었으나, 그러한 환경하에서도 방사선량측정평가기술, 방사선차폐설계기술 및 원자로사고시 피폭선량평가기술의 선진화에 필요한 지식을 얻었으며, 방사선 안전관리에 유익한 실무경험도 축적하게 되었다. 1980년대는 통합된 원자력 연구개발체제의 구축으로 방사선작업종사자 및 일반공중의 피폭저감화 기술개발에 필요한 각종 최신기술을 도입하였고, 관리업무에 있어서도 측정의 정확도와 신뢰성향상 및 새로운 관리기술의 개발에 많은 노력을 한 결과, 유익한성과를 얻게된 기간이다. 특히, 이 기간은 방사선안전관리기술의 선진화를 위한 지식이 축적되어 90년대의 방사선안전관리기술자립화를 위한 전환기로서, 이와같이 축적된 기술은 원자력의 평화적 이용에 크게 기여할 것으로 기대된다.서 dithiothreitol를 투여한 군에서는 우라늄단독투여군에 비해 cretinine의 배설이 상당히 증가하였다(P<0.05). 6. 우라늄오염에 의한 신장의 소견에 있어 우라늄단독투여군은 근위곡세뇨관상피의 공포화 및 종창, microvilli와 brush border의 손실, 세뇨관 상피의 괴사가 관찰되었으며, 간장의 충혈, 중심성 괴사 및 모세관 확장증도 관찰되었다. 그리고 sodium bicarbonate와 생리적 식염수를 병행투여한 군과 우라늄을 투여하고 30분이 지나서 dithiothreitol를 투여한 군에서는 우라늄 단독투여군에 비해 높은 방호효과가 관찰되었으나 다른 실험군에서는 큰 효과가 없는 것으로 나타났다. 결론적으로 우라늄의 체내오염시에는 sodium bicarbonate와 생리적 식염수를 가능한 빨리 병행투여하거나 dithiothreitol을 체내오염후 30분이 지나서 투여하는 방법이 우라늄오염에 대한 제염에 매우 유효할 것으로 생각되며, 특히 우라늄에 의한 인체장해를 유의하게 경감시켜줄 것으로 사료되었다.내의 어떤 부위와도 관계가 되는 것으로 간주되는데 이것이 $(^3H)$ QNB가 $(^3H)$ NMS보다 높은 최대 결합능력 $(B_{max})$을 나타낼 이유이다.   • Hemolytic property is a specific feature of bacteria to obtain iron which is essential for its survival in host tissues. Therefore, it is thought to be one of several factors of virulence. The purpose of this study was to investigate the hemolytic properties of Prevotella nigrescens isolated from the teeth diagnosed as pulp necrosis and apical periodontitis under the presence of hemolysin inhibitors such as $NaN_3$ and dithiothreitol. heat, various pH and cultural conditions. The results were as follows; 1. Clinically isolated P. nigrescens strains and standard P. nigrscens ATCC 33563 showed hemolytic activity. 2. P. nigrescens showed higher hemolytic activity against human erythrocytes than sheep or horse erythrocytes. 3. $NaN_3$ and dithiothreitol (DTT) reduced the hemolytic activity of P. nigrescens in a dose dependent manner (p<0.05). 4. Optimal pH for the maximum hemolytic activity of P. nigrescens was 4.0 and the hemolysin was stable under the $50^{\circ}C$, but the hemolytic activity was significantly decreased at $95^{\circ}C$. 5. P. nigrescens cultured in $10\%\;CO_2$ condition showed higher hemolytic activity than the bacteria cultured in the anaerobic condition.

뇌조직으로부터 정제한 Glutamate decarboxylase의 활성부위 구조 연구

  • 돼지 뇌조직으로부터 순수 분리 정제된 Glutamate decarboxylase (GAD)는 효소 dimer당 0.8mole 보조 인자인 pyridoxal-5-phosphate(PLP)가 강하게 binding되어 있었다. 이러한 부분적으로 resolved된 효소에 외부로부터 PLP를 넣어주면 효소의 활성도는 최대값으로 증가하였다. 정제된 GAD는 sulfydryl시약에 의한 화학변형에 의하여 효소의 활성도를 상실하였으며 환원제인 dithiothreitol이나 2-mercaptoethanol의 첨가에 의하여 효소의 활성도가 복구되는 것으로 보아 효소의 활성부위의 활성에 직접 관여하는 중요한 cysteinyl잔기가 존재하고 있는 것을 알 수 있다.

  • PpsR from the facultative photohetrotroph Rhodobacter sphaeroides is involved in repression of photo system gene expression. SDS-PAGE analysis showed that some portion of PpsR is oxidized so that intra- or inter-disulfide bond is formed between the two cysteins in each subunit. The disulfide bond was reduced by dithiothreitol and the binding activity to puc promoter region was increased.(omitted)

  • A technique for detection of Bacillus in soil and waste water treatment system was developed. Mixed col- tured solutions of 22 Bacillus strains were applied for selection of probe and pretreatment method for FISHAmong the three probes known as useful tool for FISH method,S-G-Bacill-0597-a-A-22 was best for detec-tion of Bacillus. Ninety five percent of DAPl count was observed with FISH method with S-G-Bacill-0597-a-A-22 probe. For increasing the permeability to Bacillus cell walls, pretreatment with lysozyme was betterthan that with lysozyme and SDS/DTT (dithiothreitol). Bacillus spore was not detected with FISH. So, Bacil-lus detection in ecosystem requires FISH with pretreatment of lysozyme and spore staining.

Purification and Enzymatic Properties of Myrosinase in Korean Mustard Seed(Brassica juncea) (한국산 겨자중 Myrosinase의 정제 및 효소학적 특성)

  • Myrosinase was purified from Korean mustard seed(Brassica juncea) by a sequential process of DEAE-cellulose, concanavalin A-sepharose, and Superose 6 chromatography. The molecular weight of puri-fied myrosinase(II-2) determined by SDS-polyacrylamide electrophoresis was 67KD. About a 248-fold purification for myrosinase II-2 was obtained after Superose 6 chromatography. Optimum pH of the myrosinase was 7.0 and optimum temperature of the enzyme was $3^{\circ}C.$ The enzyme was stable at pH 7.0, and below $30^{\circ}C.$ Cu, Hg and Fe ion significantly inhibited the enzyme activity, but ascorbic acid enhanced, resulting in a maximum activity by 1mM ascorbic acid. Among tile ascorbic acid ana-logues, dehydroascorbic acid inhibited the enzyme activity, whereas others showed a little effect. Reducing agents such as 2-mercaptoethanol and dithiothreitol inhibited the enzyme activity, but the reducing agents with ascorbic acid was enhanced enzyme activity.
