Soybean cultivars susceptible to Phytophthora root and stem rot are vulnerable to seed rot and damping-off of seedlings and young plants following an infection by Phytophthora sojae. In this study, the disease responses of Japanese soybean cultivars including currently grown main cultivars during the early growth stages were investigated following infections by multiple P. sojae isolates from Japanese fields. The extent of the resistance to 17 P. sojae isolates after inoculations at 14, 21, and 28 days after seeding varied significantly among 18 Japanese and two US soybean cultivars. Moreover, the disease responses of each cultivar differed significantly depending on the P. sojae isolate and the plant age at inoculation. Additionally, the treatment of 'Nattosyo-ryu' seeds with three fungicidal agrochemicals provided significant protection from P. sojae when plants were inoculated at 14-28 days after seeding. These results indicate that none of the Japanese soybean cultivars are completely resistant to all tested P. sojae isolates during the first month after sowing. However, the severity of the disease was limited when plants were inoculated during the later growth stages. Furthermore, the protective effects of the tested agrochemicals were maintained for at least 28 days after the seed treatment. Japanese soybean cultivars susceptible to Phytophthora root and stem rot that are grown under environmental conditions favorable for P. sojae infections require the implementation of certain practices, such as seed treatments with appropriate agrochemicals, to ensure they are protected from P. sojae during the early part of the soybean growing season.
Identification of host genes involved in disease progresses and/or defense responses is one of the most critical steps leading to the elucidation of disease resistance mechanisms in plants. Soybean mosaic virus (SMV) is one of the most prevalent pathogen of soybean (Glycine max). Although the soybeans are placed one of many important crops, relatively little is known about defense mechanism. In order to obtain host genes involved in SMV disease progress and host defense especially for virus resistance, two different cloning strategies (DD RT-PCR and Subtractive hybridization) were employed to identify pathogenesis- and defenserelated genes (PRs and DRs) from susceptible (Geumjeong 1) and resistant (Geumjeong 2) cultivars against SMV strain G7H. Using these approaches, we obtained 570 genes that expressed differentially during SMV infection processes. Based upon sequence analyses, differentially expressed host genes were classified into five groups, i.e. metabolism, genetic information processing, environmental information processing, cellular processes and unclassified group. A total of 11 differentially expressed genes including protein kinase, transcription factor, other potential signaling components and resistant-like gene involved in host defense response were selected to further characterize and determine expression profiles of each selected gene. Functional characterization of these genes will likely facilitate the elucidation of defense signal transduction and biological function in SMV-infected soybean plants.
Susceptibility of soybean cultivars to Calonectria illicicola was evaluated in a greenhouse by inoculating seedlings with mycelium in agar discs placed on the stems at the soil line. A range of responses was detected among cultivars following inoculation with a virulent isolate of C.ilicicola. Rankings of cultivars between greenhouse tests 1 and 2 were similar for disease severity and areas under the disease progress curves (AUDPC). In addition, rankings of cultivars for Final disease severity were highly correlated with AUDPC in test 1 ($r_s$ =0.88, t =5.48, p<0.001), test 2 ($r_s$ =0.99, t =22.10, p<0.001), and when tests were combined ($r_s$=0.89, t=5.82, p<0. 001). Final disease severity and AUDPC consistently identified Asgrow 7986, Braxton, Cajun, and Forrest as soybean cultivars least susceptible to red crown rot. In 1993 and 1994 field tests, a range in disease susceptibility was observed for tested cultivars but none was completely resistant. Soybean cultivars Braxton, Cajun, and Forrest, which were least susceptible to red crown rot in greenhouse tests, also ranked among cultivars with the lowest disease incidence and AUDPC in field tests. Comparisons .between rankings of the eight cultivars common to greenhouse and field tests showed a correlation between final disease severity from combined greenhouse tests and both final disease incidence ($r_s$=0.63, t =1.99, p<0.1) and AUDPC ($r_s$=0.60, t =1.82, p < 0.2) from the combined field tests. However, AUDPC from greenhouse tests did not correlate with either final disease incidence or AUDPC from field tests. The green-house screening method provided consistent results between greenhouse and field tests and successfully identified the least susceptible cultivars Braxton, Cajun, and Forrest.
Soybean (Glycine max (L) Merr.) provides plant-derived proteins, soy vegetable oils, and various beneficial metabolites to humans and livestock. The importance of soybean is highly underlined, especially when carbon-negative sustainable agriculture is noticeable. However, many diseases by pests and pathogens threaten sustainable soybean production. Therefore, understanding molecular interaction between diverse cultivated varieties and pathogens is essential to developing disease-resistant soybean plants. Here, we established a pathosystem of the Korean domestic cultivar Kwangan against Pseudomonas syringae pv. syringae B728a. This bacterial strain caused apparent disease symptoms and grew well in trifoliate leaves of soybean plants. To examine the disease susceptibility of the cultivar, we analyzed transcriptional changes in soybean leaves on day 5 after P. syringae pv. syringae B728a infection. About 8,900 and 7,780 differentially expressed genes (DEGs) were identified in this study, and significant proportions of DEGs were engaged in various primary and secondary metabolisms. On the other hand, soybean orthologs to well-known plant immune-related genes, especially in plant hormone signal transduction, mitogen-activated protein kinase signaling, and plant-pathogen interaction, were mainly reduced in transcript levels at 5 days post inoculation. These findings present the feature of the compatible interaction between cultivar Kwangan and P. syringae pv. syringae B728a, as a hemibiotroph, at the late infection phase. Collectively, we propose that P. syringae pv. syringae B728a successfully inhibits plant immune response in susceptible plants and deregulates host metabolic processes for their colonization and proliferation, whereas host plants employ diverse metabolites to protect themselves against infection with the hemibiotrophic pathogen at the late infection phase.
In Korea, a local soybean (Glycine max) genotype 56l. was found to be strongly resistant to a virulent bacterial strain of a Pseudomonas sp. SN239. Specific genes involved in the resistance of the soybean genotype 561 were identified and the pattern of gene expression against the Pseudomonas infection was analyzed using differential-display reverse transcription PCR (DDRT-PCR). More than 126 cDNA fragments representing mRNAs were induced within 48 hours of bacteria inoculation. Among them, 28 cDNA fragments were cloned and sequenced. Twelve differentially displayed clones with open reading frames had unknown functions. Sixteen selected cDNA clones were homologous to known genes of other organisms. Some of the identified cDNAs were pathogenesis-related (PR) genes and PR-like genes. These cDNAs included a putative calmodulin-binding protein; an endo-l,3-1,4-$\bate$-D-glucanase; a $\bate$-1,3-endoglucanase; a $\bate$-1,3-exoglucanase; a phytochelatin synthetase-like gene; a thiol protease; a cycloartenol synthase; and a putative receptor-like serine/threonine protein kinase. Among them, four genes were found to be putative PR genes induced significantly by the Pseudomonas infection. These included a calmodulin-binding protein gene, a $\bate$-1,3-endoglucanase gene, a receptor-like serine/threonine protein kinase gene, and pS321 (unknown function). These results suggest that the differentially expressed genes may mediate the strong resistance of soybean 561 to the strain SN239 of Pseudomonas sp.
Typical susceptible symptoms of the bacterial pustule disease caused by Xanthomonas axonopodis pv. glycines are pustule and chlorotic haloes that usually occur in leaves of Glycine max plants. The soybean genotype 'PI96188' showed an atypical response to all tested races X. axonopodis pv. glycines, accompanied with necrosis without chlorotic haloes on the underside of the necrotic symptoms. X. axonopodis pv. glycines 8ra grew to levels from 10 to 100 fold lower on PI96188 than on susceptible cultivar Jinjoo1, but 10-fold higher than on the resistant cultivar CNS. The chlorophyll content in PI96188 leaves remained unchanged until 12 days after bacterial infection. Ultrastructural observation showed that the infected leaf cells of PI96188 had intact normal chloroplasts compared to those of the susceptible cultivar Jinjoo1. Chloroplast degradation or the absence of chloroplasts in cells of the infected tissues was observed in Jinjoo1. Senescence-related ACS7 gene was significantly induced in PI96188 compared to those in Jinjoo1 at 2 days after inoculation. While photosynthesis-related rbcS gene showed the dramatic change in Jinjoo1, this gene was constitutively expressed in PI96188. However, expression of the defense-related genes, such as peroxidase and isoflavone synthase in the infected PI96188 leaves was similar to that in Jinjoo1. Together, these results suggest that the novel necrotic symptom in PI96188 is a kind of resistant response different from a typical hypersensitive response in the resistant genotypes.
Bacterial pustule of soybean caused by Xanthomonas axonopodis pv. glycines is one of the most prevalent bacterial diseases in many areas where soybeans are grown. This study was carried out to evaluate the effect of cultivars, sowing date and cropping system on the suppression of soybean bacterial pustule in the field. One hundred soybean cultivars were screened for disease resistance against bacterial pustule in naturally infested field. Among them, fourteen cultivars including 'Pureun' were found to be high resistant. And thirty cultivars showed to be moderate resistant(less than 3% of diseased leaf area). When Soybean cultivar 'Taekwang' were sown in four different dates, May 25, June 5, June 15, and June 25, at 10 day-interval in Milyang, the diseased leaf area of bacterial pustule was 23.3%-25.7%, 14.7%-18.0%, 10.7%-12.8%, and 1.0%-2.7%, respectively. The lowest percentage of diseased leaf area was recorded in the plots sown on June 25, whereas the highest percentage of diseased leaf area was recorded in the plot sown on May 25. As sowing time was delayed, incidence of soybean bacterial pustule found to be comparatively reduced. From December in 2006 to June in 2007, we surveyed the pathogen population of soybean bacterial pustule in five cropping upland soils where soybean was cultivated. The survey result showed the bacterial pustule pathogens were detected from the all cropping soils. The pathogen populations of soybean bacterial pustule in soybean-barley and soybean-garlic cropping soil were significantly lower than that of the other cropping soils. In addition, the incidence of soybean bacterial pustule was decreased under the two cropping systems.

Bacterial pustule (BP) is a leaf disease of soybean that is most common in Korea. Inoculation of 8ra, pathogen strain, to resistant and susceptible cultivars for finding the BP resistance gene (rxp) was much tried but the sequence of the exact gene is not found. This research performed in order to confirm the rxp gene near molecular marker by using the resistant and susceptible cultivars. Soybean BP resistance gene which related to region of near molecular marker could select the resistant cultivar. For the near molecular marker of rxp, reference genomics data available at sequenced Phytozome was used for designing molecular markers. The rxp was mapped between Satt372 and Satt486 on chromosome 17. According to previous study, rxp released in find mapping 7.2 Mbp to 7.3 Mbp on chromosome 17. In this study, we developed 3 random markers near from 6.6 Mbp to 7.3 Mbp on chromosome 17 identified to increase the genetic resolution of the rxp gene region using resistant and susceptible cultivars. Particularly, Rxp17-700 marker was mostly coincided resistance and susceptible genotype to rxp. This result suggests that Rxp17-700 marker will be more tightly linked to rxp gene.

Xanthomonas axonopodis pv. glycines(Xag) is a pathogen that causes bacterial leaf pustule(BLP) disease in soybeans grown in Korea and the southern United States. Typical and early symptoms of the disease are small, yellow to brown lesions with raised pustules that develop into large necrotic lesions leading to a substantial loss in yield due to premature defoliation. After Xag infects PI 96188, only pustules without chlorotic haloes were observed, indicating the different response to Xag. To identify differentially expressed genes prior to and 24 hr after Xag inoculation to PI 96188 and BLP-resistant SS2-2, an oligonucleotide macroarray was constructed with 100 genes related to disease resistance and metabolism from soybean and Arabidopsis. After cDNAs from each genotype were applied on the oligonucleotide macroarrays with three replicates and dye swapping, 36 and 81 genes were expressed as significantly different between 0 hr and 24 hr in PI 96188 and SS2-2, respectively. Six UniGenes, such as the leucine-rich repeat protein precursor or 14-3-3-like protein, were selected because they down-regulated in PI 96188 and up-regulated in SS2-2 after Xag infection, simultaneously. Using tubulin and cDNA of Jangyeobkong(BLP-susceptible) as controls, the oligonucleotide macroarray data concurred with quantitative real-time RT-PCR(QRT RT-PCR) results in most cases, supporting the accuracy of the oligonucleotide macroarray experiments. Also, QRT RT-PCR data suggested six candidate genes that might be involved in a necrotic response to Xag in PI 96188.
Bacterial pustule of soybean (Glycines max) caused by Xanthomonas axonopodis pv. glycines is one of the most prevalent bacterial diseases of soybean. This bacterium shows strong pathogenicity to the plants and distributes throughout Korea. However, no good control measures including bactericides and resistant cultivars are available to control the disease in Korea. Therefore, this study was conducted to develop chemical control method against soybean bacterial pustule. The present study was undertaken to find out the growth inhibitory effect bactericides (8 antibiotics, 2 copper compounds, quinoline, 18 agro-chemicals) on bacterial pustule pathogen. Antibiotics test showed that tetracycline and streptomycin sulfate significantly suppressed the growth of bacterial pustule pathogen. Also, application of oxolinic acid was found to be effective for pathogen inhibition. However, vancomycin, polymyxin B sulfate and copper compounds did not show the positive suppressive effect on growth of the pathogen. Among the eighteen agro-chemicals, streptomycin sulfate + oxytetracyclin (18.8 + 1.5%) WP, oxytetracycline (17%) WP and oxolinic acid (20%) WP were found to be effective for the inhibition of the pathogen in vitro. The selected 5 agro-chemicals were also applied on soybean in field and their control effects against the soybean bacterial pustule were tested. The foliar application of streptomycin sulfate + oytetracyclin WP and oxytetracycline WP on the naturally infected soybean (Taekwangkong) showed high control value (above 70%). Therefore, it is concluded that the bactericides used in this study showed strong inhibitory effect to soybean bacterial pustule and they can be recommend to farmers to control the disease.
