Major diseases and insect pests in nursery season of strawberry were anthracnose, powdery mildew, Fusarium wilt, two-spotted spider mite, and aphids. Environmental-friendly and chemical application schedules can improve diseases and insect pests control with relatively fewer organic and chemical materials inputs compared with spray programs when it's occurred. Field experiments were performed in 2012 to 2013 according to calendar-based spray programs with environmental-friendly spray calendar (EFSC) and conventional chemical spray calendar (CSC) for controlling diseases and insect pests of strawberry plants cv, Seolhyang during the two nursery seasons. EFSC did reduce the incidence of diseases and insect pests as compared to the non-treated control. Incidence of anthracnose and powdery mildew by EFSC and CSC was similar in 2012 and 2013 seasons. In addition, occurrence of two-spotted spider mite in EFSC in 2013 was similar to those of CSC and was shown highly in early and mid-June both 2012 and 2013 seasons. Occurrence of aphid in EFSC was shown highly in early and mid-June both 2012 and 2013 seasons. These results suggest that EFSC program may be effective for controlling strawberry diseases and insect pests by using environmental-friendly organic materials.

Beetles Protaetia brevitarsis seulensis Kolbe (Coleoptera: Cetoniidae) and Allomyrina dichotoma Linn. (Coleoptera: Scarabaeidae) are widely used in traditional medicine, and the number of insect-rearing farms is increasing in South Korea. The purpose of this study was to establish a multiplex PCR-based assay for rapid simultaneous detection of multiple pathogens causing insect diseases. Six insect parasites such as fungi Beauveria bassiana (Bals.-Criv.) Vuill. (Hypocreales: Cordycipitaceae) and Metarhizium anisopliae (Metschn.) Sorokin (Hypocreales: Clavicipitaceae), bacteria Bacillus thuringiensis Berliner (Bacillales: Bacillaceae), Pseudomonas aeruginosa Migula (Pseudomonadales: Pseudomonadaceae), and Serratia marcescens Bizio (Enterobacteriales: Enterobacteriaceae), and Oryctes rhinoceros nudivirus were chosen based on the severity and incidence rate of insect diseases in South Korea. Pathogen-specific primers were designed and successfully applied for simultaneous detection of multiple infectious agents in farm-bred insects P. b. seulensis and A. dichotoma using multiplex PCR and high resolution capillary electrophoresis. Our results indicate that multiplex PCR is an effective and time-saving method for simultaneous detection of multiple infections in insects, and the QIAxcel capillary electrophoresis system is useful for quantitative evaluation of the individual impact of each infectious agent on the severity of insect disease. The approach designed in this study can be utilized for rapid and accurate diagnostics of infection in insect farms.
Three chalcones and a stilbenoid have been isolated from the stem bark of Lindera erythrocarpa. They were elucidated as 2'-hydroxy-3',4',5',6'- tetramethoxychalcone, 2',4'-dihydroxy-3',6'-dimethoxy chalcone, 2',4',5',6'- tetrahydroxy-3'-methoxychalcone, and 5,6-dihydroxy-2,3,4-trimethoxystilbene. Chemical structures were elucidated on the basis of MS, $^1H,\;^{13}C$ and 2D-NMR spectroscopic data. This is the first report on the isolation of these compounds from the L. erythrocarpa.
To elucidate the role of silicon in biotic stress such as pests and diseases, standard chrysanthemum was grown in pots filled with soil without application of pesticide and fungicide. Si treatment was largely composed of three groups: $K_2SiO_3$ (50, 100, and $200mg{\cdot}L^{-1}$), three brands of silicate fertilizer (SiF1, SiF2, and SiF3) and tap water as a control. Si sources were constantly drenched into pots for 14 weeks. Application high concentration $K_2SiO_3$ ($200mg{\cdot}L^{-1}$) and three commercial Si fertilizers for 14 weeks improved growth parameters such as plant height and the number of leaves. In the assessment of disease after 4 weeks of Si treatment, percentage of infected leaves was not significantly different from that of control. After 14 weeks of Si treatment, however, the infected leaves were significantly reduced with a 20-50% decrease in high concentration ($200mg{\cdot}L^{-1}$) of potassium silicate and all commercial silicate fertilizers. Colonies of aphid insect (Macrosiphoniellas anborni) were also reduced in Si-treated chrysanthemum, showing 40-57% lower than those of control plants. Accumulation of silicon (approximately $5.4-7.1mg{\cdot}g^{-1}$ dry weight) in shoots of the plants was higher in Si-supplemented chrysanthemum compared to control plants ($3.3mg{\cdot}g^{-1}$ dry weight). These results indicate that using potassium silicate or silicate fertilizer may be a useful for management of disease and aphid insect in soil-cultivated chrysanthemum.

Trapping efficiency of various sex pheromone blends of the peach pyralid moth, Dichocrocis punctiferalis was compared by field study to develop monitoring system with its sex pheromone at chestnut orchards in Korea. Five candidates of the sex pheromone blends used for the field trapping of D. punctiferalis males were 70:30, 75:25, 80:20, 85:15 and 90:10 mixture of (E)-10-hexadecenyl aldehyde (E10-16:Al) and (Z)-10-hexadecenyl aldehyde (Z10-16:Al). All lures were treated with 1 or 2 mg of each blends. During 2 years of field survey, the 75:25 blend was usually the most effective in attracting males among 5 blends tested. For the 2nd generation, the best capturing activity for D. punctiferalis male was observed by lure with 75:25 blend. Both 90:10 and 75:25 blends showed highest efficiency for the 3rd generation. In most cases. lures treated with 1 mg of blend caught more male moths than these treated with 2 mg of blend.
This research was carried out to selection of desirable cultivar for organic cultivation of carrot in Korea. The assay of disease resistance, insect tolerance and rate of bolting were investigated on 32 cultivars of genetic resources of carrot. This experiment was carried out at experimental field and greenhouse of Gangneung-Wonju National University. 'PI 223360' was resistant against powdery mildew. Ten cultivars including 'Oxheart Carrot Heirloom' were moderately resistant, 22 cultivars including 'Long Impeator #58' were susceptible to powdery mildew. Although there was no resistant cultivar against black rot and leaf blight, our results showed that several cultivars have moderate resistance. Thirteen cultivars including 'Oxheart Carrot Heirloom' were having insect (Erythroplusia pyropia) tolerance and 'SA 102' and 'Scarlet Keeper Carrot Rare' were susceptible. According to the investigation of bolting, 6 cultivars including 'Hongsim Ouchon Carrot' were early bolting and 4 cultivars including 'Japanese Imperial Long Carrot' were moderate bolting. Other cultivars were identified as late bolting. From above results, we confirmed that 'Oxheart Carrot Heirloom' was suitable cultivar for organic cultivation among 32 genetic resources of carrot.
Accurate identification of pine wood nematode, Bursaphelenchus xylophilus is a prerequisite to diagnose the pine wilt disease. However, a fungivorous nematode, B. mucronatus is highly similar to B. xylophilus and it is difficult to differentiate these two species by morphological features. A molecular diagnosis method, ITSRFLP was applied for the identification of B. xylophilus and B. mucronatus from Korea. Genomic DNA was extracted from a single individual nematode and ITS DNA was amplified by PCR. The size of PCR product was approximately 900bp and the sequence data were obtained after cloning. Amplified ITS was digested by 5 different restriction enzymes (Rsa I, Hae III, Msp I, Hinf I, and Alu I) and provided a discriminatory profile for B. xylophilus and B. mucronatus. Besides, B. mucro- natus was determined to have 2 different genotypes, East Asian type and European type also clearly separated by Rsa I and Hae III digestion. European type of B. mucronatus is recently collected from Pinus koraiensis and has not been reported before. ITS sequnce data were analyzed by Restriction Mapper program and the result supported ITS-RFLP pattern. These data indicated that PCRRFLP method is an accurate and simple way for identification of Bursaphelenchus species.

Rhizina undulata is the fungus, which causes Rhizina root rot on coniferous trees. Nested-PCR using ITS-specific primer was applied to detect R. undulata from the soils of Japanese black pine (Pinus thunbergil) forests infested with the disease in Seocheon, Chungnam Province, South Korea. Soil samples were collected from four different sites, both dead trees and fruit bodies of R. undulata were present, dead trees only present, fruit bodies only present, and both were absent. Nested-PCR products specific to R. undulata ITS-region were amplified. Positive reactions were found in some samples from the sites, where dead trees and fruit bodies of R. undulata were absent as well as where both of those were present. R. undulata was mainly detected in the soil samples from the depth of 5~20 cm under the soil surface. These results show that the nested-PCR could be used to diagnose the presence or potential infestation of R. undulata in the soils of pine forests.
- Turfgrass insect pests and natura.l enemies for biological control were investigated to develop pest management effectively in golf courses at several golf clubs. Twenty eight insect pest species of 10 families in 6 orders were collected from golf courses. The zoysiagrass mite, Eriophyes zoysiae and root-knot nematode, Meloidogyne incognita were also collected from zoysiagrass. White grubs of several scarab beetles and cutworms (Agrotis spp.) damaged seriously at most surveyed golf clubs. In addition, bluegrass webworm (Crambus sp.), Japanese lawngrass cutworm (Spodoptera depravata), scale insects, Tipula sp., and ants (Camponitus japonicus, Formica japonica, and Lasins japonicus) damaged turfgrasses directly or indirectly in golf courses. The entomopathogenic nematodes, Heterorhabditis spp., Steinernema glaseri, and S. longicaudum, entomopathogenic fungi, Beauveria bassiana and Metarhizium anisopliae, and milky disease, Paenibacil/us popil/iae were isolated from white grubs or turfgrass soil as microbial control agents. Besides, dipteran predators, Cophinopoda chinensis, Philonicus albiceps, and Promachus yesonicus and hymenopteran parasitoid, Tiphia sp. were also collected. The P. yesonicus was the most active in golf courses. The root-knot nematode, M. incognita was found from Zoysia japonica, Z. matrella. and Cynodon dactylon.
Ambrosia beetle, Platypus koryoensis, is a vector of oak wilt disease caused by Raffaelea sp. in Korea. The degree of damage by oak wilt disease was dependent on the density of the beetles in the oak trunk, a relationship between the degree of damage by oak wilt disease and the density of beetle on the basis of the number of entry hole was studied. Entry hole distribution within tree trunk was analyzed by the nearest neighbor method. Thirty four oak trees (Quercus mongolica) located in survey area were selected and then degree of damage, the number of attack hole/$623cm^2$ in upper (50cm from the surface) and lower (surface) trunk and the nearest neighbor distance between the holes were measured. The number of hole and the nearest neighbor distance in upper and lower part were positively correlated with each other. As the degree of damage was severer, the number of the holes increased, whereas the nearest neighbor distance decreased. The distribution pattern of the hole was changed from clumped one to uniform as the severity of damage increased. These results suggested that Platypus koryoensis attacked the oak tree in concentrative manner at initial stage of attack but at final stage, it distributed uniformly to reduce intraspecific competition between the beetles.
