  • The present study was conducted to compare antioxidant activity of Cuscutae Semen, Psoraleae Fructus, Cnidii Fructus and Epimedii Herba by DPPH radical scavenging activity. The extract was studied using diphenyl-picryl-hydrazyl (DPPH) for DPPH method. DPPH radical scavenging activity was measured after. .10, 20 and 30 minutes. The extract was tested by 1, 5, 10, 50, 100, 500 and $1000{\mu}g/ml$ concentrations. The results showed that the extract scavenged DPPH radical with time-dependent manner. Also, the extract showed dose-dependent DPPH radical scavenging activity. The extract of Cuscutae Semen, Psoraleae Fructus, Cnidii Fructus and Epimedii Herba scavenged DPPH radical with the IC50 being 2.7, 3.2, 2.9 and 1.1 mg/ml, respectively. In conclusion, the extract of Epimedii Herba, Cuscutae Semen, Cnidii Fructus and Psoraleae Fructus have antioxidant activity for the treatment of male sterility.

  • The antioxidant activities of methanol extracts of sixteen samples were tested using 1.1-diphenyl-2-picryl hydrazyl(DPPH) reactivity and TBARS substances assay in vitro. The methanol extracts of the rice brans from three wild rice -O. minuta, O. rufipogon, and O. barthii-were found to be the most effective in DPPH radical scavenging activity. The next effective ones were the rice brans of Heugjinjubyeo and leaves of Tapgolbori. When tested on lipid peroxidation using a lipid peroxidation generation system mediated by $H_2O$$_2$/Fe$^{2+}$ in rat liver homogenates, the brans and hull of wild rice (O. minuta, O. rufipogon, and O. barthii) and rice bran of Heugjinjubyeo exhibited protective activities against lipid peroxidation in the order of effectiveness.s.

  • The antioxidant activities of the bread made from waxy black rice flour(WBRF) and wheat flour were investigated. The waxy black rice bread was manufactured with 10, 20, 30, or 40% waxy black rice flour(w/w). As the additive amount of WBRF were increased, DPPH(1,1-diphenyl-2-picryl-hydrazyl) radical scavenging activity and TEAC(trolox equivalent antioxidant capacity) were also increased. WBRF group showed the higher activity than the control group in DPPH radical scavenging activity and TEAC. The group with 10% WBRF added was not significantly different from the control group in metal chelating activity, but over 20% of addition, it had the higher metal chelating activity than that of the control group.

Antioxidant Effects of Raw Ginseng, Soft Red Ginseng, and Red Ginseng Sap (수삼, 연질 홍삼, 수액 홍삼의 항산화 효과)

  • Korean ginseng (Panax ginseng) generally has a good safety profile and contains many bioactive substances, such as ginsenosides or panaxosides. Korean red ginseng might help to stabilize the sympathetic nervous system and improve cognition in individuals. Soft red ginseng is produced by new processing technology. This study focused on investigating whether soft red ginseng produced under the new processing technology reduces or improves the existing antioxidant effects. No significant difference in 2,2'-azino-bis (3-ethylbenzthiazoline-6-sulphonic acid) (ABTS) scavenging activity was found between soft red ginseng and ready-made red ginseng (p<0.05). Soft red ginseng extract showed higher 2,2-diphenyl-1-picryl-hydrazyl-hydrate (DPPH) and hydroxyl radical (OH) scavenging activity than other ginseng extracts. OH scavenging activity was significantly different across three groups (raw ginseng, soft red ginseng, and red ginseng sap) (p<0.05). Nitric oxide (NO) scavenging activity was also significantly different among raw ginseng, soft red ginseng, and purchased red ginseng liquid products (p<0.05). Many calcium crystals appeared on the electron microscope in soft red ginseng. Magnesium and potassium showed no significant difference between soft red ginseng and hard red ginseng. The extract of soft red ginseng scavenged different free radicals efficiently due to the presence of DPPH and OH and may help treat free radical-induced diseases.

Compositions of Astragali Radix and Angelicae Radix by DPPH Radical Scavenging Activity (당귀(當歸)와 황기(黃芪)의 배합 변화가 DPPH 자유기 소거에 미치는 영향 연구)

  • Objectives : This study was conducted to compare antioxidant activity of ratio-related Angelicae Radix and Astragali Radix compositions by DPPH radical scavenging activity. Methods : The species of Angelicae Radix was compared as Angelica gigas, A. sinensis and A. acutiloba. Then various compositions of Angelicae Radix with Astragali Radix were prepared with different ratio. Diphenyl-picryl-hydrazyl (DPPH) radical scavenging activity was measured after 10, 20 and 30 minutes. Each extracts were tested by 1, 5, 10, 50, 100, 500 and 1,000 ${\mu}g/ml$ concentrations. Results : The results showed that Angelica gigas, A. sinensis and A. acutiloba scavenged DPPH radical with 66.8, 61.7 and 56.7% in 1,000 ${\mu}g/ml$ concentration, respectively. According to this result, Angelicae Radix and Astragali Hadix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio were scavenged DPPH radical with 56.3, 49.9, 61.2, 55.7, 54.7, 55.0, 52.8 ,md 50.2% in 1,000 ${\mu}g/ml$ concentration, respectively. The extract of Angelicae Radix and Astragali Radix compositions with 100:0, 10:1, 5:1, 3:1, 1:1, 1:3, 1:5, 1:10 and 0:100 ratio scavenged DPPH radical with the $IC_{50}$ being 85,21, 1016.26, 656.51, 785.67 and 956.74 ${\mu}g/ml$ respectively. Conclusion : In conclusion, the extract of Angelicae Radix and Astragali Radix compositions with 1:1 ratio showed dose-dependent DPPH radical scavenging activity.

Quantitative Structure-Activity Relationships for Radical Scavenging Activities of Flavonoid Compounds by GA-MLR Technique

  • The quantitative structure-activity relationship (QSAR) of a set of 35 flavonoid compounds presenting antioxidant activity was established by means of Genetic Algorithm-Multiple Linear Regression (GA-MLR) technique. Four-parametric models for two sets of data, the 1,1-diphenyl-2-picryl hydrazyl (DPPH) radical scavenging activity $(R^2=0.788,\;Q^2_{cv}=0.699\;and\;Q^2_{ext}=0.577)$ and scavenging activity of reactive oxgen species (ROS) induced by $H_2O_2 (R^=0.829,\;Q^2_{cv}=0.754\;and\;Q^2_{ext}=0.573)$ were obtained with low external predictive ability on a mass basis, respectively. Each model gave some different mechanistic aspects of the flavonoid compounds tested in terms of the radical scavenging activity. Topological charge, H-bonding complex and deprotonation processes were likely to be involved in the radical scavenging activity.

Jacaranone and Related Compounds from the Fresh Fruits of Ternstroemia japonica and their Antioxidative Activity

  • Jacaranone and related compounds (1-3) were isolated, along with three triterpenes (4-6), from the fresh fruits of Ternstroemia japonica. The compounds were identified as jacaranone (1), 3-hydroxy-2,3-dihydrojacaranone (2), 3-methoxy-2,3-dihydrojacaranone (3), 3-O-acetyl­oleanolic acid (4), 3-O-acetylursolic acid (5), and ursolic acid (6). Jacaranone and its derivatives were isolated for the first time from Theaceae. Of the isolated compounds, compound 3 is a new compound. Jacaranone (1) exhibited weak antioxidative effect on 1, 1-diphenyl-2-picryl-hydrazyl (DPPH) radical.

Antioxidant and Anti-wrinkling Effects of Extracts from Vitex trifolia L. (만형자 추출물의 항산화 및 주름개선 효과)

  • In this study, the antioxidant and anti-wrinkling effects of fraction from Vitex trifolia L. were investigated. Among the fractions, ethyl acetate fraction showed the highest antioxidant activities in 1-1-diphenyl-2-picryl-hydrazyl, 2,2'-azino-bis(3-ethylbenzothiazoine-6-sulfonic acid) radical scavenging and elastase inhibition with 76, 89, and 74%, respectively, at a concentration of $1,000{\mu}g/mL$. This fraction, at the concentration of $25{\mu}g/mL$, inhibited 70% fibroblast cell viability and 86% the matrix metalloprotease (MMP)-1. In addition, the results from Western blot assay showed that this fraction ($25{\mu}g/mL$) expressed the MMP-1 protein level by decreasing 50%. The findings suggest that the ethyl acetate fraction from V. trifolia has great potential as a cosmeceutical ingredient with antioxidant and anti-wrinkling effects.

Antioxidant Activity of Honeydew Honey Produced by Apis mellifera L. (양봉꿀벌이 생산한 감로꿀의 항산화 활성)

  • To evaluate biological activity of honeydew honey produced by Apis mellifera L. in Korea, we measured antioxidant activity by using DPPH (1,1-diphenyl-2-picryl hydrazyl), ABTS [2,2-azobis(3-ethylbenzothiazoline-6-sulfonate)], FRAP (Ferric reducing antioxidant power), and total polyphenol content assays. Korean honeydew honey respectively scavenged 26% and 86% of DPPH and ABTS radicals at the highest concentration of 10 mg/mL. In the result of FRAP assay, Korean honeydew honey showed activity (126 µM of FRAP value) to reduce Fe3+ to Fe2+. Total polyphenol content was 73.41 mg GAE/kg. Korean honeydew honey exhibited excellent antioxidant activity due to having high radicals scavenging ability and reducing power of ferric ion as well as the presence of phenolic compounds. These findings suggest that Korean honeydew honey has great potential as a functional food material.

Study on the Effects of the Extracts from Sophora Japonica L. Flowers, Fruits and Branches on Improvements in Skin Condition (회화나무 꽃, 열매, 가지 추출물의 피부개선 효과에 관한 연구)

  • The purpose of this research was to examine the effect of the ehanol extracts of Sophora japonica L. (S. japonica). flowers, fruits and branches on skin enhancement with assessing anti-oxidative, whitening, and wrinkle enhancement effects. Results showed that l,1-diphenyl-2-picryl-hydrazyl (DPPH) radical scavenging activities were $17.68{\pm}1.59{\sim}51.40{\pm}1.04$, $27.48{\pm}0.22{\sim}50.89{\pm}0.13$ and $30.79{\pm}0.55{\sim}45.17{\pm}0.83%$, respectively, in 50~1,000 mg/L of concentrations. The capacities of inhibiting tyrosinase of ethanol extracts from S. japonica. flowers, fruits and branches were $0.27{\pm}0.12{\sim}11.38{\pm}0.57$, $0.27{\pm}0.02{\sim}0.82{\pm}0.27$ and $0.09{\pm}0.16{\sim}0.55{\pm}0.27%$, respectively. The capacities of preventing porcine pancreatic elastase (PPE) were $3.70{\pm}1.23{\sim}7.28{\pm}1.01$, $3.06{\pm}2.13{\sim}13.03{\pm}2.99$ and $6.00{\pm}0.96{\sim}9.71{\pm}0.44%$, respectively, in the case of 50~1,000 mg/L of concentrations. It is concluded that the effects of S. japonica. flowers, fruits and branches on skin improvement are varied significantly.
