Pfiesteriaand Pfiesteria-like organisms were reported to be linked to major fish kills(involving well over a billion fish) in North Carolina and Maryland estuaries on the U.S. east coast during the 1990s. Occurrences of these species have been recently reported from Korean waters including Chinhae Bay and the coast of Yeosu. In this study, the life cycle of Cryptoperidiniopsis brodyi and Pfiesteria piscicida were examined using DAPI staining. Their excystment and growth were stimulated directly by the addition of prey cells such as Rhodiminas salina. Amoeboid stages in C. brodyi and P. piscicida were never observed in culture, even after addition of filter-sterile fish mucus and tissue. The dominant life cycle stages consisted of motile flagellated zoospores and cysts. A typical dinoflagellate life cycle was demonstrated by direct observation and DAPI staining.

The phototrophic dinoflagellate genus Scrippsiella is known to have a worldwide distribution. Here, we report for the first time, the occurrence of Scrippsiella lachrymosa in Korean waters. Unlike the other stains of S. lachrymosa whose cultures had been established from cysts in the sediments, the clonal culture of the Korean strain of S. lachrymosa was established from motile cells. When the sulcal plates of S. lachrymosa, which have not been fully described to date, were carefully examined using scanning electron microscopy, the Korean strain of S. lachrymosa clearly exhibited the anterior sulcal plate (s.a.), right sulcal plate (s.d.), left sulcal plate (s.s.), median sulcal plate (s.m.), and posterior sulcal plate (s.p.). When properly aligned, the large subunit (LSU) rDNA sequence of the Korean strain of S. lachrymosa was ca. 1% different from those of two Norwegian strains of S. lachrymosa, the only strains for which LSU sequences have been reported. The internal transcribed spacer (ITS) rDNA sequence of the Korean strain of S. lachrymosa was also ca. 1% different from those of the Scottish and Chinese strains and 3% different from those of the Canadian, German, Greek, and Portuguese strains. Thus, the Korean S. lachrymosa strain has unique LSU and ITS sequences. The abundances of S. lachrymosa in the waters of 28 stations, located in the East, West, and South Sea of Korea, were quantified in four seasons from January 2016 to October 2017, using quantitative real-time polymerase chain reaction method and newly designed specific primer-probe sets. Its abundances were >$0.1cells\;mL^{-1}$ at eight stations in January and March 2016 and March 2017, and its highest abundance in Korean waters was $26cells\;mL^{-1}$. Thus, S. lachrymosa has a nationwide distribution in Korean waters as motile cells.
On the outbreak of paralytic shellfish poisoning in April 1993 in most of shellfish harvesting areas in Jinhae Bay, Korea, to clarify the toxin production of causative organism Alexandrium species, 19 axenic clonal isolates established from the benthic resting cysts in three different stations of those culture grounds were subjected to PSP toxin analysis by HPLC. Individual toxin content per cell was highly variable among the strains isolated from a sampling area and originated from an individual cyst. Average toxin contents in those areas revealed higher values of 54-70 fmol/cell. Toxin profiles included C1/C2(epiGTX8/GTX8), GTX1/GTX4 and neoSTX as the major components, and GTX2/GTX3, GTX5, C4, dcSTX and STX as the minor or sporadic ones. neoSTX on the dominant toxins showed not only most diverse compositional changes comprising $5-54 mol\%$ ranges but also no detection on the half of the strains examined, which were implicated in arising of heterogeneity with a genetic trait within a geographical region. When average toxin composition was compared, carbamate toxins comprised large proportions of $57\%,\;54\%\;and\;67\%$ as total toxin in St. 1, St. 2 and St. 4, respectively. These results suggested that an extensive paralytic shellfish toxification in Jinhae Bay could be largely due to the production of highly potent carbamate toxins in the causative dinoflagellate Alexandrium species.
To investigate the occurrences of resting cysts of Diplopsalis and Protoperidinium species, sediment trap samples were collected from Gamak Bay, Korea. Based on the germination experiments, nine species of order Peridiniales were identified; cyst of Diplopsalis lebourae, cyst of Protoperidinium excentricum, cyst of Protoperidinium sp. cf. nudum, cyst of Protoperidinium obtusum, cyst of Protoperidinium ventricum, cyst of Protoperidinium sp.1, cyst of Protoperidinium sp.2, Protoperidinium sinuosum (Brigantedinium majusculum) and unidentified Peridiniales.
Scrippsiella trochoidea is a dinoflagellate responsible for red tide in early spring in southern coastal water. Marine bacteria appear to exert critical roles on the development and decay of phytoplankton bloom in marine ecosystem. It is likely that marine bacteria, Pseudomonas spp., share some metabolic processes with S. trochoidea. To investigate interactions between S. trochoidea and Pseudomonas spp. directly, cysts of S. trochoidea isolated from the bottom mud in Masan Bay have been germinated and cultured. From the S. trochoidea cultured medium, we have isolated Pseudomonas spp., a dominant and cultured. From the S. trochoidea cultured medium, we have isolated Pseudomonas spp., a dominant species. Both of Pseudomonas spp. and S trochoidea have been simultaneously inoculated into the sterilized sea water and cultured to examine the change of fatty acids. The major fatty acids that showed increases in composition during the dispecific culture were $C_{18:0/},{\;}C_{20:5}{\;}and{\;}C_{22:5}$ in S. trochoidea, and in Pseudomonas spp. Especially, $C_{20:5}{\;}and{\;}C_{18:0}$ were increased in S. trochoidea but decreased in Pseudomonas spp. These results strongly suggest that two species share some processes in their fatty acid metabolism.
Feeding behaviour of the manila clam Ruditapes philippinarum was qualitatively and quantitatively characterized by comparing the contents composition in digestive tract of the clam and the phytoplankton community in surface sea water in Gomso Bay, west coast of Korea. The contents of digestive tract comprised diatoms (71.5%), dinoflagellates (13.1%), nannoplankton (6.6%), and detritus including mesozooplankton. The abundance of food organisms in digestive tract of the clam was high in winter and spring, while low in summer and autumn. But The biomass of phytoplankton in surface sea water revealed the highest value in autumn. Also, the larger the clam size increases the abundance of food organisms in digestive tract. The dominant species in digestive tract were Paralia sulcata and Navicula arenaria such as benthic diatoms and dinoflagellate cysts, whereas the dominant species in surface sea water were Chaetoceros, Skeletonema, Asterionellopsis such as pelagic diatoms in genus, cryptomonads, and P. sulcata. Analyses of digestive tract revealed that benthic diatoms especially represent an important constituent of food organisms in the malina clam and different of phytoplankton size and morphology explain preference for food selectivity.
Biostratigraphic and paleoenvironmental analyses are carried out on cutting samples from the Dolgorae wells drilled in the Ulleung basin. The clayey, silty, and sandy sediments of the wells yield various microfossil assemblages of relatively good preservation, among which five fossil groups are reported; a total of 97 foraminiferal species of 66 genera, 19 nannofossil species of 12 genera, 86 ostracod species of 41 genera, 107 diatom species of 44 genera, and 124 dinoflagellate cysts species of 45 genera. Based on microfossils the geologic ages of the Dolgorae wells are dated to be from late Early Miocene to Early Pleistocene. Several biohorizons are defined in Neogene successions by the LOD (Last Ocurrence Datum) and FOD (First Ocurrence Datum) of marker species including G. truncatulinoides (LOD: 1.9 Ma) of foraminifera; C. macintyeri (LOD: 1.64-1.60), G. oceanica (FOD: 1.65 Ma), G. caribbeanica (1.72 Ma), D. brouweri (LOD: 2 Ma), R. pseudoumbilica (LOD: 3.66 Ma), P. lacunosa (FOD: 4.2 Ma) of nannofossils; S. ellipsoideus (LOD: 4 Ma), S. palcacantha (LOD: 10.2), C. giusepei (LOD: 14 Ma) of dinocysts; D. seminae v. fossilis (FOD: 3.7 Ma), T. antiqua (LOD: 1.7 Ma), T. convexa (LOD: 2.4 Ma), N. kamtschatica (LOD: 2.58 Ma), T. oestrupii(FOD: 5.1 Ma) of diatoms. Abundance patterns of microfossils throughout the wells reflect changes in paleoenvironmental and sedimentological settings of the basin in relation to sea-level variations. According to these data the large-cycle and small-cycle changes of transgression and regression phases are observed in terrestrial to marine sediments. This high-resolution sequence biostratigraphy established by various fossil groups enabled more reliable correlation between strata and refined interpretation on deposition systems of the basin. It also proved to provide fundamental and precise informations regarding stratigraphic correlation, tectonic events, basin, and depositional history for hydrocarbon explorations, especially in collaboration with seismic-stratigrahic analyses.
