Fluorescent Crack Sensor in a Polymeric Material (고분자 재료의 형광 크랙센서)

  • A styrylpyrylium salt (DHSP) was synthesized and then photodimerized to obtain a cyclobutane-type dimer (DHSP dimer). Methacryloyl group was incorporated into DHSP dimer to obtain DMSP dimer. Based on FT-IR analysis of a crosslinked DMSP dimer, it was considered that the cyclobutane structure reversed to styryl C=C bonds upon crack formation. Fluorescence measurement of DHSP in solid state (excitation at 385 nm) showed emission centered at 626 nm, while DHSP dimer revealed very weak emission. Fluorescent emission from microcracks in a film of crosslinked DMSP dimer was observed upon exposure to $330{\sim}385\;nm$ light.

Study on the Structure of DNA Containing a Thymine Dimer and $T_4$ Endonuclense V * DNA Complex (Thymine Dimer를 포함한 DNA와 $T_4$ Endonuclease V * DNA 복합체의 구조에 관한 연구)

  • In order to obtain insight into the repair mechanism of DNA containing thymine photo-dimer, the conformation of the duplex d(GCGGTTGGCG).d(CGCCAACCGC) with a thymine dimer incorporated has been studied by proton NMR. NOE data show that, although the local environment around the thymine dimer is altered, the gross structural changes are relatively small. T$_4$endonuclease V exhibited a conformational change on complex formation with DNA. This conformational change occurred around histidine 16 which was close to tyrosine 129 located in the aromatic segment (WYKYY) near the C-terminus. It is likely that the interaction between T$_4$endonuclease V and DNA is strong since the complex was not dissociated up to 1.6 M NaCl.

Preparation and characterization of fullerene dimer [C120] by trichloroperbenzoic acid oxidation method

  • In this study, we present the preparation and characterization of oxidized fullerene and fullerene dimer [$C_{120}$]. From the XRD data, other weak peaks with pristine fullerene [$C_{60}$] peaks were observed in the X-ray diffraction patterns for fullerene dimer [$C_{120}$]. SEM micrographs for the fullerene dimer [$C_{120}$] indicated that practically all the surface state was shown the drastic morphology changes and its outer surface is clearly visible and resulted in clogging and frost-like formation. From the MALDI-TOF mass spectra, the differences in the spectra recorded on two kinds of fullerene are due to the oxidation including chemical bonding and bridging between the $C_{60}$ molecules. We also obtained additional information from FT-IR spectra on functional component on the chemically modified surface of oxidized fullerene and fullerene dimer [$C_{120}$].

NMR study of the interaction of T$_4$ Endonuclease V with DNA

  • In order to obtain insight into the mechanism by which DNA containing a thymine photo-dimer is recognized by the excision repair enzyme, T$_4$ endonuclease V, we have taken NMR study of this protein and its complex with oligonucleotides. The conformations of five different DNA duplexes DNA I : d(GCGGATGGCG).d(CGCCTACCGC), DNA II d(GCGGTTGGCG) .d(CGCCAACCGC), DNA III : d(GCGGT ^ TGGCG) .d(CGCCAACCGC), DNA IV d(GCGGGCGGCG).d(CGCCCGCCGC) and DNA V d(GCGGCCGGCG) . d(CGCCGGCCGC) were studied by $^1$H NMR. The NMR spectra of these five DNA duplexes in the absence of the enzyme clearly show that the formation of a thymine dimer within the DNA induces only a minor distortion in the structure, and that the overall structure of B type DNA is retained. The photo-dimer formation is found to cause a large change in chemical shifts at the GC7 base pair, which is located at the 3'-side of the thymine dimer, accompanied by the major conformational change at the thymine dimer site. The binding of a mutant T$_4$ endonuclease V (E23Q), which is unable to digest DNA containing a thymine dimer, to the DNA duplex d(GCGGT ^ TGGCG)ㆍd(CGCCAACCGC) causes a large down-field shift in the imino proton resonance of GC7. Therefore, this position is thought to be either the crucial point of the interaction wi th T$_4$ endonuclease V, or the si to of a conformational change in the DNA caused by the binding of T$_4$ endonuclease V. Usually, it is very difficult to assign NMR peaks in DNA * protein complex because of severe peak overlaps. In order to overcome these peak overlaps, we used a method of deuterium incorporation.

  • The ${\pi}-{\pi}$ interactions of the peptide-dimer and peptide-trimer complexes were investigated in the (VQIVYK + LYQLEN) and (VQIVYK + NNQQNY) mixing solutions. The results showed that tyrosine (Y) residues were critical in the formation of hetero peptide-dimers and -trimers during the early oligomerization process. We used collision-induced dissociation (CID) along with electrospray ionization mass spectroscopy (ESI-MS) to obtain the structural information of the hetero-dimers and -trimers. We chose three amyloidogenic peptides-VQIVYK, NNQQNY, and LYQLEN-from tau protein, yeast prion-like protein Sup35, and insulin chain A, respectively. Hetero-dimer, -trimer, -tetramer, and -pentamer complexes were observed in the mass spectra. The tandem mass spectrum of the hetero-dimer and hetero-trimer showed two different fragmentation patterns (covalent and non-covalent bond dissociation). Y-Y interaction structures were also proposed for the hetero-dimer and -trimer complexes.

Influence of Surface Morphology and Substrate on Thermal Stability and Desorption Behavior of Octanethiol Self-Assembled Monolayers

  • The formation and thermal desorption behaviors of octanethiol (OT) SAMs on single crystalline Au (111) and polycrystalline Au, Ag, and Cu substrates were examined by X-ray photoelectron microscopy (XPS), thermal desorption spectroscopy (TDS), and contact angle (CA) measurements. XPS and CA measurements revealed that the adsorption of octanethiol (OT) molecules on these metals led to the formation of chemisorbed self-assembled monolayers (SAMs). Three main desorption fragments for dioctyl disulfide (C8SSC8+, dimer), octanethiolate (C8S+), and octanethiol (C8SH+) were monitored using TDS to understand the effects of surface morphology and the nature of metal substrates on the thermal desorption behavior of alkanethiols. TDS measurements showed that a sharp dimer peak with a very strong intensity on single crystalline Au (111) surface was dominantly observed at 370 K, whereas a broad peak on the polycrystalline Au surface was observed at 405 K. On the other hand, desorption behaviors of octanethiolates and octanethiols were quite similar. We concluded that substrate morphology strongly affects the dimerization process of alkanethiolates on Au surfaces. We also found that desorption intensity of the dimer is in the order of Au>>Ag>Cu, suggesting that the dimerization process occurs efficiently when the sulfur-metal bond has a more covalent character (Au) rather than an ionic character (Ag and Cu).

A Spectroscopic Study of Hydrogen Bonding between Riboflavin and Salicylic Acid Derivatives

  • Specific association phenomena of riboflayin-2',3',4',5',- tetraacetate and salicylic acid derivatives, such as salicylic acid, aspirin and salicylamide have been measured by infrared and fluorescence spectroscopy. Salicylic acid and riboflavin tetraacetate oxyl group of the former. Asprin and riboflavin tetraacetate form the 1:1 cyclic hydrogen bonded dimer by the same mode. Salicylamide froms the 1:1 cyclic hydrogen bonded dimer with riboflavin tetraacetate by using its amide group and carbonyl group. Salicylic acid derivatives are effective quenchers of the fluorescence of riboflavin tetraacetate. It is appeared that salifylamide is the strongest quencher among them. The quenching effect is attributed to the formation of association dimer.

Reaction of NO on Vanadium Oxide Surfaces: Observation of the NO Dimer Formation

  • The adsorption and surface reactions of NO on a VO/V(110) surface have been investigated using X-ray photoelectron spectroscopy (XPS), near-edge X-ray absorption fine structure, and temperature programmed desorption (TPD) technique. NO is molecularly adsorbed on VO/V(110) at 80 K. As the surface coverage of NO increases, the NO dimer is formed on the surface at 80 K. Both NO and (NO)2 are adsorbed on the surface with the N-O bond perpendicular to the surface. (NO)2 decomposes at ~100 K and the reaction product is desorbed as N2O. Decomposition of NO takes place when the surface temperature is higher than 273 K.

Synthesis of Asymmetric Dimer of Quinolone Derivatives Using p-TSA (p-TSA를 이용한 퀴놀론 유도체의 비대칭 이합체 합성)

  • New asymmetric dimers, N,N'-dialkyl-4'-hydroxy-4-oxo-2,2',3,3'-tetrahydro-2,2'-diphenyl-4,4'-quinolones 3a-f were synthesized through the dehydration and dea1coholation of N-alkylanilines and ethyl benzoylacetate. Dimers 3a-f were identified by NMR, IR and GC-MS. A series of dimer 3a-f has been synthesized using acid-catalyzed one-pot reaction that involved the condensation, cyc1ization and dimerization. Similarly, the 6,6'-methoxy (or 7,7'-methoxy) substituted dimers were prepared from N-alkyl-meta-(or para)-anisidines. Formation of dimers was undertaken with p-toluenesulfonic acid (p-TSA) at 90∼11$0^{\circ}C$ in toluene for 2∼6 hours over the Dean-Stark apparatus.
