Gardnerella vaginalis에 대한 목단피, 소목, 어성초, 연교, 하고초, 현삼의 항균효과 (in vitro activities of Moutan Cortex Radicis, Caesalpiniae Lignum, Houttuyniae Herba, Forsythiae Fructus, Prunellae Herba, Scrophuiariae Radix against Gardnerella vaginalis)

  • The purpose of this thesis is to dedicate to get the objectivity of herbal medicine treatments by choosing herbs likely to work as antibacterial agents, Moutan Cortex Radicis, Caesalpiniae Lignum, Houttuyniae Herba, Forsythiae Fructus, Prunellae Herba, Scrophulariae Radix against Gardnerella vaginalis which is associated with Bacterial vaginosis, making experiments on them and getting the significant results. Each herb's efficacy on control the number of Bacterial vaginosis is noticed by using Disk Susceptibility test with six herbs medicine and Broth dilution assay of the culture. Disk Suseptibility Test : The efficiency strength is as follows in a row : Caesaipiniae Lignum, Moutan Cortex Radicis, Prunellae Herba, Forsythiae Fructus, Houttuyniae Herba, Scrophulariae Radix. MIC Prunellae Herba is 1.5 mg/ml, Caesalpiniae Lignum is 5 mg/ml, Forsythiae Fructus and Moutan Corex Radicis is 10mg/ml for G. vaginalis. As a results, herbmed which tested in this study have an antibacterial effects against G. vaginalis and that may be used for treament of Bacterial vaginosis. Assistant use of external medicine to p.o medicine is expected to bring the good effect of treatment which is used for the origin of this disease.

Salmonella typhimurium 에 대한 E-ball 및 상용화된 소독약품의 살균 및 소독 효능에 대한 연구 (Study on the sterilizing and disinfective effects of E-ball and commercialized disinfectants against Salmonella typhimurium)

  • Many chemical disinfectants are using to protect the foot and mouth disease (FMD) and avian influenza (AI) in Korea since 2000. This study was performed to confirm disinfective ability of commercialized chemical disinfectants and to investigate the sterilizing ability of E-ball as alterative to chemical disinfectants. 4 kinds of acidulant, 3 kinds of aldehyde, 1 kind of oxidizer and 300 g of E-ball were used in this study. Dilution rate of disinfective power of all chemical disinfectants were to 200 times. The sterilizing ability of aldehydes were better than the acidulant and oxidizer with Salmonella typhimurium. The sterilizing ability of E-ball treated solution was guessed due to the friction of E-ball deads. In the case of the friction of 2 beads of E-ball, Salmonella typhimurium was sterilizted on $1{\times}10^6/mL$ CFU in the E-ball treated solution. The E-ball treated solution had superior sterizing power compared with the chemical disinfectants in the bacteria of soil for antibacterial examination. E-ball treated solution has a possibility as the substitute of chemical disinfectants to protective the animal diseases contains FMD, AI.

백강균(Beauveria bassiana)을 이용한 식물병원성세균 생장억제효능 검증 (Effects of the growth inhibition against plant pathogenic bacteria using Beauveria bassiana)

  • 뽕나무병원성세균에 대한 Beaveria bassiana 6균주의 항균활성을 paper disc법으로 조사하였고 최소성장저해농도(MIC)를 액체배지희석법으로 조사하였다. 항균활성은 B. bassiana J200에서만 나타났으며, Erwinia rhapontici KACC 10407에서는 13mm, Pseudomonas syringae KACC 10390와 Xanthomonas campestris KACC 12134에서는 각각 17 mm의 항균활성이 나타났다. MIC 측정 결과 E. rhapontici KACC 10407, P. syringae KACC 10390 및 X. campestris KACC 12134 모두 4.0%에서 MIC를 보였다. 이상의 결과로 B. bassiana의 뽕나무병원성세균에 대한 생물학적 방재제로서의 이용 가능성을 확인하였다.

Effects of Contrast Agent Concentration on the Signal Intensity and Turbo Factor of TSE and Slice-selective IR in T1-weighted Contrast Imaging

  • The present study analyzes T1 TSE and T1 slice sel. IR (dark_fluid) signal strength according to the degree of gadolinium contrast agent dilution and analyzes the turbo factors with regard to changes in the maximum and overall signal strength to study correlations between changes and signal-to-noise ratios (SNRs) and compare peak-to-peak SNR (PSNR) enhancement in order to improve the quality of T1-weighted images. Enhancement TR (600 msec) evaluated to determine the T1 TSE turbo factor and obtain the maximum signal strength, T1WI were used sequentially to experiment with turbo factors_1-4. T1 slice sel. IR (dark-fluid) was used to sequentially test turbo factors_2-5 but not turbo factor_1 at a TR (1500 msec) and compare data at an increase in T1 of 900 msec. The T1 TSE was reduced according to the contrast agent concentration. Phantom signal strength increased, whereas turbo factors_1-4 exhibited maximum signal strength at a concentration of 3 mmol, followed by a gradual decrease. In the turbo factors_2-5, the signal strength increased sharply to maximum signal strength at 0.7 mmol, followed by a reduction. T1 TSE had a greater maximum signal strength than did T1 slice sel. IR (dark_fluid). A comparison of SNR found that T1 TSE imaging was superior (33.3 dB) in turbo factor_1 and T1 slice sel. IR (dark_fluid) was highest (33.9 dB) at turbo factor_5. A PSNR comparison analysis was not sufficient to distinguish between the images obtained with both techniques at 30 dB or higher under all experimental conditions.

전기점화 엔진에서 개질가스 첨가에 의한 희박연소특성 연구 (Lean Operation Characteristics of a Spark Ignition Engine with Reformed Gas Addition)

  • Hydrogen can extend the lean misfire limit to a large extent when it is mixed with conventional fuels for a spark ignition engine. In this study, hydrogen-enriched gaseous fuels by reforming process were simulated according to their proportions of $H_2$, CO, $CO_2$ and $N_2$ gases. Pure hydrogen and two different hydrogen-enriched gaseous mixtures(A-, B-composition) were tested for their basic effects on the engine performances and emissions in a single cylinder research engine. A- and B-composition showed different results from 100% $H_2$ addition because air/fuel mixtures were more diluted by their additions. Even though the energy fraction of reformed gases was increased, combustion stabilities and lean misfire limits were not sensitively improved. It means that combustion augmentation by $H_2$ addition was offset by the charge dilution of $N_2$ and $CO_2$. In addition, the low flammability of CO gas deteriorated thermal efficiencies. CO emission was drastically increased with B-composition which included higher CO component. However, $NO_x$ was reduced as energy fraction($X_e$) rised except for the case of 100% $H_2$ addition at $\lambda=1.2$ and was, for A-composition, lowered to a factor of ten when compared with that of $H_2$ addition. HC emissions were largely influenced by $COV_{imep}$ due to misfire and partial burns.

Antimicrobial and Anti-Biofilm Activities of the Methanol Extracts of Medicinal Plants against Dental Pathogens Streptococcus mutans and Candida albicans

  • Several medicinal plants are ethnomedically used in Korea as agents for treating infection, anti-inflammation, and pain relief. However, beyond typical inhibitory effects on cell growth, little is known about the potential anti-biofilm activity of these herbs, which may help to prevent cavities and maintain good oral health. This study aimed to investigate the antimicrobial and anti-biofilm activities of the methanol extracts of 37 Korean medicinal plants against dental pathogens Streptococcus mutans and Candida albicans, which synergize their virulence so as to induce the formation of plaque biofilms in the oral cavity. The antimicrobial activities were investigated by broth dilution and disk diffusion assay. The anti-biofilm and antioxidant activities were evaluated based on the inhibitory effect against glucosyltransferase (GTase) and the DPPH assay, respectively. Among 37 herbs, eight plant extracts presented growth and biofilm inhibitory activities against both etiologic bacteria. Among them, the methanol extracts (1.0 mg/ml) from Camellia japonica and Thuja orientalis significantly inhibited the growth of both bacteria by over 76% and over 83% in liquid media, respectively. Minimum inhibitory concentration (MIC) values of these methanol extracts were determined to be 0.5 mg/ml using a disk diffusion assay on solid agar media. Biofilm formation was inhibited by more than 92.4% and 98.0%, respectively, using the same concentration of each extract. The present results demonstrate that the medicinal plants C. japonica and T. orientalis are potentially useful as antimicrobial and anti-biofilm agents in preventing dental diseases.

Effects of four novel root-end filling materials on the viability of periodontal ligament fibroblasts

  • Objectives: The aim of this in vitro study was to evaluate the biocompatibility of newly proposed root-end filling materials, Biodentine, Micro-Mega mineral trioxide aggregate (MM-MTA), polymethylmethacrylate (PMMA) bone cement, and Smart Dentin Replacement (SDR), in comparison with contemporary root-end filling materials, intermediate restorative material (IRM), Dyract compomer, ProRoot MTA (PMTA), and Vitrebond, using human periodontal ligament (hPDL) fibroblasts. Materials and Methods: Ten discs from each material were fabricated in sterile Teflon molds and 24-hour eluates were obtained from each root-end filling material in cell culture media after 1- or 3-day setting. hPDL fibroblasts were plated at a density of $5{\times}10^3/well$, and were incubated for 24 hours with 1:1, 1:2, 1:4, and 1:8 dilutions of eluates. Cell viability was evaluated by XTT assay. Data was statistically analysed. Apoptotic/necrotic activity of PDL cells exposed to material eluates was established by flow cytometry. Results: The Vitrebond and IRM were significantly more cytotoxic than the other root-end filling materials (p < 0.05). Those cells exposed to the Biodentine and Dyract compomer eluates showed the highest survival rates (p < 0.05), while the PMTA, MM-MTA, SDR, and PMMA groups exhibited similar cell viabilities. Three-day samples were more cytotoxic than 1-day samples (p < 0.05). Eluates from the cements at 1:1 dilution were significantly more cytotoxic (p < 0.05). Vitrebond induced cell necrosis as indicated by flow cytometry. Conclusions: This in vitro study demonstrated that Biodentine and Compomer were more biocompatible than the other root-end filling materials. Vitrebond eluate caused necrotic cell death.

X線螢光分析에 依한 珪酸鹽鑛物의 分析 (The X-Ray Fluorescent Spectrographic Analysis of Silicate Minerals)

  • 螢光X線을 利用하여 珪酸鹽鑛物中의 主成分인 $SiO_2$, $Al_2O_3$, $Fe_2O_3$, CaO, MgO 및 $K_2O$를 迅速히 分析할 目的으로 試料의 處理 測定 및 各條件에 對하여 檢討하였다. 試料를 Lithium Tetraborate로 용융하여 300Mesh 以上의 微粉末로 한後 40,000Lb의 壓力으로 成型하여 Tungsten과 Chromium 對陰極의 X-線管과 LiF, EDDT, ADP의 分光結晶을 使用하여 測定하였다. 各成分에 對한 檢量曲線은 Matrix Effect를 고려하여 N.B.S 및 International Rock Standard를 選定 使用하였고 Lanthanum Oxide 및 Binder로서 Borie Acid를 첨가하여 얻었다. 各成分에 對하여 本法의 再現性 및 誤差를 檢討하기를 爲하여 I.R.S T-1을 使用하여 測定한 結果 0.47($SiO_2$), 0.85($Al_2O_3$), 0.05($Fe_2O_3$), 0.07(caO), 0.02($K_2O$), 0.13(MgO)의 標準偏差를 얻었다. 또한 化學分析植에 對한 偏差를 求하고져 Clay, Kaoline, Alunite, Wallastonite 및 Zeolite 等의 珪酸鹽鑛物을 選定하여 化學分析 및 本法에 依한 分析結果를 비교하였다.

Antimicrobial activity of Gynura segetum's leaf extracts and its active fractions

  • $Gynura$ $segetum$ (Lour.) Merr. ($Compositae$) is a well recognized medicinal plant in Indonesia and Malaysia. It is believed to have an anticoagulant effect and is used in treating snake-bites, inflammations and other skin afflictions. This study was carried out to evaluate the antimicrobial effects of $Gynura$ $segetum$ leaves extracts and its fractions. The chemical compositions of the active extracts were also determined. The antimicrobial activities of different solvent extracts of leaves of $Gynura$ $segetum$ were evaluated using the agar well-diffusion method. The Minimum Inhibitory Concentration (MIC) of the active subfractions was determined by the tube dilution method. Gas Chromatography-mass spectrometry (GC-MS) analysis was carried out to identify the chemical compositions of the active extracts. The ethyl acetate fraction and its subfraction E4 performed potent antimicrobial activities and fifteen known chemical constituents were identified by GCMS analysis as 4-vinylphenol, 1-tetradecene, phenol, 2,4-bis(1,1-dimethylethyl), 1-hexadecene, E-15-heptadecenal, hexadecanoic acid, 1,2-benzenedicarboxylic acid, dibutyl ester, 1-docosene, octadecanoic acid, 1-eicosene, cyclotetracosane, 1,2-benzenedicarboxylic acid, bis(2-ethylhexyl)ester, butanedioic acid, monomethyl ester, niacin and 4-hydroxy-benzoic acid. The results of this study suggested a connection between the antimicrobial activities and the chemical structures. The plant may be used as a potential source for antimicrobial agents.

An Effective Antidandruff Agent - IPBC

  • The purpose of this study was to determine the effect of IPBC(3-lodo-2-propynylbutyl carbamate) on dandruff caused by the anthropophilic fungus Maiassezia furfur. The effects of IPBC on dandruff were examined by evaluating (a) the MIC value of IPBC using broth dilution method : (b) the remnant antimicrobial activity of IPBC containing shampoo on skin disc ; (c) the antidandruff efficacy of 1.0 % IPBC containing shampoo in double blind clinical trial. To investigate the remnant antimicrobial activity of IPBC against Maiassezia furfur, guinea pig-skin disc was washed with antidandruff shampoo and then the diameter of inhibition zone per disc was measured. For clinical trial, thirty healthy volunteers, aged 25-35, participated in 4 week study. At 0,2,4 weeks, examinations of scaling, itching on scalp were carried out. The MIC(Minimun Inhibition Concentration) values of IPBC range from 0.10 to 1.00$\mu$ g/ml and it seems that IPBC is more effective in the MIC values than zinc pyrithione, selenium disulphide, piroctone olamine and comparable to ketoconazole, climbazole. When the rimnant antimicrobial activity of IPBC shampoo on skin disc was determined, 0.5% IPBC shampoo and 2.0% Ketoconazole shampoo resulted in similar antimicrobial effect. In addition, 1.0%,2.0% IPBC shampoo was more effective than 2.0% ketoconazole shampoo. After two and four-weeks of 1.0% IPBC shampoo treatment, there was significant reduction of scaling, itching in test group compared to control group. On the basis of these results, it can be concluded that 1.0% IPBC is more effective than 2.0% Ketoconazole in reducing dandruff. It seems that strong capacity of drug binding to the stratum corneum plays a role in its antidandruff effect since adsorption of active ingredients on scalp is very important factor in reducing dandruff.
