PAF Regulate Blastocyst Development to Hatching Stage through PKC Activity in the Mouse

  • The developmental regulation of the preimplantation mammalian embryos is a fundamental step for preparing the implantation and it may be regulated by several autocrine and paracrine factors including platelet-activating factor. PAF improved the embryonic survival and implantation but its role during blastocyst development is still largely unknown. In this study, the effects and the possible pathway of PAF on developmental regulation of blastocyst to hatching stage were investigated. Developmental pattern in hatching embryo was a concentration-response curve showing maximal activity at 1 nM PAF, with decreasing activity at higher concentrations. $50{\mu}M$ 1-(5-isoquinolimnesulfonyl)-2-methylpiperazinme dihydrochloride (H-7), a PKC inhibitor, inhibited the progression of blastocyst to hatching embryo. In addition H-7 blocked the PAF effects on the blastocyst development. Besides tetradecanoylphorbol acetate (TPA), a PKC activator stimulated development of blastocyst to the hatching stage. These finding revealed that PAF support the blastocyst development to the hatching embryo. Also it is suggested that PAF action pathways in hatching supporting include the PKC signaling pathway.

Potentiometric Sensor for the Determination of Dibucaine in Pharmaceutical Preparations and Electrochemical Study of the Drug with BSA

  • Plasticized poly(vinyl chloride), PVCs, with different membrane compositions tested for use in the construction of an ion-selective sensor for the determination dibucaine. A prepared membrane with dioctyl phthalate-PVC and ion-pair of N-(1-naphthyl)ethylenediamine dihydrochloride-tetraphenyl borate had a good potential to acts as a potentiometric sensor for the analysis of dibucaine. A linear relationship was obtained between potential and logC varying between $1.0{\times}10^{-6}$ and $1.0{\times}10^{-2}$ M dibucaine with a good repeatability and reproducibility. The sensor was applied for the determination of the drug in pharmaceuticals and biological fluids such as plasma and urine samples with satisfactory results. The drug electrode has also been used to study the interaction of bovine serum albumin (BSA) with dibucaine. The saturated quantities of dibucaine binding were 13.04, 5.30 and 9.70 mol/mol in 0.01, 0.02 and 0.1% of protein, respectively.

  • Hexamethylenediamine dihydroiodide is monoclinic, with cell dimensions $a=4.85{\AA}$, $b=12.77{\AA}$, $c=9.73{\AA}$, ${\beta}=91.5^{\circ}$ The space group is $P2_1/c$, with two molecules per unit cell. It has a center of symetry in the molecule. All atomic positions are determined by means of a two-dimensional patterson synthesis and fourier synthesis. The C-N bond distance is $1.48{\AA}$ and the C-C bond distances are lying between $1.55{\AA}$, and $1.59{\AA}$. The iodine atom is bonded by hydrogen bridges of $3.59{\AA}{\pm}0.1{\AA}$ to nitrogen atoms and surrounded by three nitrogen atoms. The hexamethylenediamine chain is zigzag in the hexamethylenediamine dihydrochloride molecules though, it is not zigzag in the hexamethylenediamine dihydroiodide.

Crystal Form of Olmutinib

  • Olmutinib, N-[3-({2-[4-(4-methylpiperazine-1-yl)aniline]thieno[3,2-d]Pyrimidin-4-yl}oxy)phenyl]prop-2-enamide dihydrochloride monohydrate, $Olita^{TM}$ is an oral, third-generation epidermal growth factor receptor tyrosine kinase inhibitor (EGFR TKI) that was developed by Boehringer Ingelheim and Hanmi Pharmaceutical Co. Ltd for the treatment of non-small cell lung cancer (NSCLC). The aim of this work was to investigate the existence of polymorphs and pseudopolymorphs of olmutinib. Three crystal forms of olmutinib have been isolated by recrystallization and characterized by differential scanning calorimetry (DSC), thermogravimetric (TG) analysis and powder X-ray diffractometry (PXRD). From the DSC and TG data it was confirmed that Form 1 is monohydrate, Form 2 is dihydrate, Form 3 is 1.5 hydrate. The PXRD patterns of three crystal forms were different respectively. After storage of 1 month at $2^{\circ}C$, 24% RH (Relative Humidity), Form 1, Form 2, and Form 3 were not transformed.

Sanitary and Nutritional Characterization of Commercial Kwamegi from Pacific Herring Clupea pallasii (시판 청어(Clupea pallasii ) 과메기의 위생 및 영양학적 특성)

  • The objective of this study was to investigate the sanitary and nutritional requirements for the industrialization of commercial kwamegi from Pacific herring Clupea pallasii (CK-PH). The proximate composition of CK-PH was 46.4-47.2% moisture, 24.7-25.6% crude protein, 23.6-25.2% crude lipid, and 2.5-2.9% ash, which differed significantly from that of commercial kwamegi from the Pacific saury Cololabis saira. According to the volatile basic nitrogen content, heavy metal content, and viable cell and coliform group counts, products K and F (prepared by a general process) maintained their freshness, whereas product T (treated with green tea extract) did not. Products K and F contained five types of biogenic amine: agmatine sulfate (2,596 and 2,067 mg/kg, respectively), putrescine dihydrochloride (8.5 and 8.0 mg/kg, respectively), cadaverine (3.7 and 3.9 mg/kg, respectively), histamine (17.0 and 12.4 mg/kg, respectively), and spermidine (8.7 and 8.0 mg/kg, respectively). Product T contained six amine types: tyramine (12.5 mg/kg), agmatine sulfate (2,723 mg/kg), putrescine dihydrochloride (29.4 mg/kg), cadaverine (321.6 mg/kg), histamine (45.3 mg/kg), and spermidine (13.6 mg/kg). The total amino acid content of product K (22.16/100g) was 5.8% lower than that of product F. The major amino acids of products K and F were aspartic acid, glutamic acid, leucine, and lysine. No difference was found in the fatty acid composition of products K and F. The major fatty acids in products K and F were 18:1n-9, 20:5n-3, and 22:6n-3. Based on the recommended daily dietary allowances for Koreans, the significant minerals in products K and F were calcium, phosphorus, and magnesium.

Reinforcement of Antioxidative Potentials by Korean Traditional Prescriptions on Mouse Plasma and Liver (전통 한약 탕제 투여에 의한 혈장 및 간 조직의 항산화력 증강 효과)

  • Yungmijihwgang-Won, Yollyunggobon-Dan and Palmi-Hwan, Korea traditional prescriptions composed of oriental medical herbs, have been used successfully to improve human health and regimen. This study was designed to examine the mechanism of healthful effects of the Korea traditional prescriptions through its antioxidative potentials. Using in vitro antioxidative activity assay system such as DPPH radical quenching assay, superoxide anion radical scavenging assay and inhibition of TBARS production, three Korea traditional prescriptions were observed to have nearly the same antioxidative potentials as ascorbic acid, a well-known strong water-soluble antioxidant. Moreover, we observed reinforced antioxidative effects of these drugs in liver from mouse fed these drugs with 4 weeks. When liver homogenate was incubated with 2.2'-azobis(amidinopropane) dihydrochloride(AAPH), as a free radical initiator, we observed that oxidative damages were decreased and antioxidative potentials were increased in liver homogenate treated these drugs. However, enzymatic antioxidative system as superoxide dismutase, catalase, glutathione peroxidase and glutathione reductase was not affected by drug administration.

Culture Conditions of Garlic Resistant Lactic Acid Bacteria for Feed Additives (사료첨가용 생균제 개발을 위한 마늘 내성 유산균의 배양 조건)

  • Culture conditions of L. plantarum TJ-LP-002, the garlic resistant strain isolated from pakimchi (green onion kimchi), were investigated for the use of feed additives. Acetic acid, citric acid, lactic acid, and tartaric acid were detected in the culture supernatant, and especially the concentrations of lactic acid and acetic acid significantly increased during cultivation. The antimicrobial activity of L. plantarum TJ-LP-002 was not affected by proteases, calatase or cellulase, which showed that the antimicrobial activity might be due to the production of acids rather than proteinaceous antimicrobial substances. L. plantarum TJ-LP-002 was resistant to neomycin sulfate, spectinomycin dihydrochloride, and lincomycin hydrochloride, sensitive to streptomycin sulfate, and intermediate resistant to ampicillin trihydrate, chloramphenicol, erythromycin, tetracycline hydrochloride, and kanamycin sulfate. The optimum initial pH of medium, fermentation temperature and time for the cell growth and antibacterial activity were pH 7.0, 30${^{\circ}C}$ and 24hr, respectively. The optimal composition of culture medium for the cell growth and antimicrobial activity was 3%(w/v) glucose as a carbon source, 3%(w/v) yeast extract as a nitrogen source, and manganese sulfate and ammonium citrate as inorganic salts. The combinatorial supplementation of these inorganic salts, rather than sole addition as an inorganic salt, resulted in better antibacterial activity.

Controlling Size and Distribution for Nano-sized Polystyrene Spheres

  • Highly monodisperse polystyrene (PS) nanospheres were fabricated by surfactant-free emulsion polymerization in water using styrene, 2,2'-azobis(2-methyl propionamidine) dihydrochloride (AIBA), and poly(vinyl pyrrolidone) (PVP). The size and distribution of the PS nanospheres were systematically investigated in terms of initiator concentration, stabilizer concentration, reaction temperature, reaction time, and reactant concentration. With increasing AIBA initiator concentration, PS particle sizes are raised proportionally, and can be controlled from 120 to 380 nm. Particle sizes were reduced with increasing PVP concentration. This decrease occurs because a high PVP concentration leads to a large number of primary nuclei in the early stage of polymerization. When the reaction temperature increased, the sizes of the PS particles decrease slightly. The particles grew quickly during the initial reaction stage (1-3 h) and the growth rate became steady-state after 6 h. The PS sizes approximately doubled when the reactant (styrene, PVP, azo-initiator) concentrations were increased by a factor of eight.

Free radical scavenging effect and protective activity from oxidative stress of broccoli flowers and sprouts (Broccoli flower와 Broccoli sprout의 라디칼 소거능 및 산화적 스트레스 개선 효과)

  • In this study, the antioxidative effect and protective potential against oxidative damage of extract and fractions from broccoli were investigated under in vitro and cellular system. The methanol (MeOH) extracts of broccoli flowers and sprouts were partitioned as dichloromethane, n-butanol (BuOH) and aqueous fractions. The comparison of antioxidative effect of broccoli flowers and sprouts showed that broccoli sprouts exerted the more effective protective activity from 2,2'-azobis (2-aminopropane) dihydrochloride (AAPH)-induced oxidative stress in LLC-$PK_1$ porcine renal epithelial cell. In addition, the extract and fractions from broccoli sprouts showed strong scavenging effect of 1,1-diphenyl-2-picrylhydrazyl radical and the BuOH fraction exerted the strongest activity. Therefore, the BuOH fraction was evaluated as the most active fraction with strong radical scavenging activity among the fractions of broccoli flowers and sprouts. The present study suggests the antioxidative potential against free radical-induced oxidative damage of flowers and sprouts of broccoli. In addition, the BuOH fraction of broccoli is considered as the active fraction with antioxidative effect.

Fabrication of Uniform Hollow Silica Nanospheres using a Cationic Polystyrene Core

  • Uniform, hollow nanosilica spheres were prepared by the chemical coating of cationic polystyrene (cPS) with tetraethylorthosilicate (TEOS), followed by calcination at 600 $^{\circ}C$ under air. cPS was synthesized by surfactant-free emulsion polymerization using 2,2'-azobis (2-methyl propionamidine) dihydrochloride as the cationic initiator, and poly(vinyl pyrrolidone) as a stabilizer. The resulting cPS spheres were 280 nm in diameter, and showed monodispersion. After coating, the hollow silica product was spherically shaped, and 330 nm in diameter, with a narrow distribution of sizes. Dispersion was uniform. Wall thickness was 25 nm, and surface area was 96.4 $m^2/g$, as determined by BET. The uniformity of the wall thickness was strongly dependent upon the cPS surface charge. The effects of TEOS and ammonia concentrations on shape, size, wall thickness, and surface roughness of hollow $SiO_2$ spheres were investigated. We observed that the wall thicknesses of hollow $SiO_2$ spheres increased and that silica size was simultaneously enhanced with increases in TEOS concentrations. When ammonia concentrations were increased, the irregularity of rough surfaces and aggregation of spherical particles were more severe because higher concentrations of ammonia result in faster hydrolysis and condensation of TEOS. These changes caused the silica to grow faster, resulting in hollow $SiO_2$ spheres with irregular, rough surfaces.
