The molecular understanding of resistance and susceptibility of host plants to scab, a most threatful disease to pome fruit production worldwide, is very limited. Comparing resistant line '93-3-98' to susceptible one 'Sweet Skin' at seven time points of 0, 0.5, 1, 2, 3, 4, 8 days post inoculation, RNA-sequencing data derived from infected and mock-inoculated young leaves were analyzed to evaluate the tolerant response and to mine candidate genes of pear to the scab pathogen Venturia nashicola. Analysis of the mapped reads showed that the infection of V. nashicola led to significant differential expression of 17,827 transcripts with more than 3-fold change in the seven pairs of libraries, of which 9,672 (54%) are up- and 8,155(46%) are down-regulated. These included mainly receptor (NB-ARC domains-containing, CC-NBS-LRR, TIR-NBS-LRR, seven transmembrane MLO family protein) and transcription factor (ethylene responsive element binding, WRKY DNA-binding protein) related gene. An arsenal of defense response of highly resistant pear accessions derived from European pear was probably supposed no sooner had V. nashicola infected its host than host genes related to disease suppression like Polyketide cyclase/dehydrase and lipid transport protein, WRKY family transcription factor, lectin protein kinase, cystein-rich RLK, calcium-dependent phospholipid-binding copine protein were greatly boosted and eradicated cascade reaction induced by pathogen within 24 hours. To identify transcripts specifically expressed in response to V. nashicola, RT-PCRs were conducted and compare to the expression patterns of seven cultivars with a range of highly resistant to highly susceptible symptom. A DEG belonging to the PR protein family genes that were higher expressed in response to V. nashicola suggesting extraordinary role in the resistance response were led to the identification. This study provides the first transcriptional profile by RNA-seq of the host plant during scab disease and insights into the response of tolerant pear plants to V. nashicola.
Cognitive reserve (CR) is a concept that can explain the discrepancies between the pathologic burden of the disease and clinical manifestations. It refers to the individual susceptibility to age-related brain changes and pathologies related to Alzheimer's disease, thus recognized as a factor affecting the trajectories of the disease. The purpose of this study was to explore the current states of clinical studies on neural substrates of CR in Alzheimer's disease using functional magnetic resonance imaging. We searched for clinical studies on CR using fMRI in the Pubmed, Cochrane library, RISS, KISS and ScienceON on August 14, 2023. Once the online search was finished, studies were selected manually by the inclusion criteria. Finally, we analyzed the characteristics of selected articles and reviewed the neural substrates of CR. Total thirty-four studies were included in this study. As surrogate markers of CR, not only education and occupational complexity, but also composite score and questionnaire-based method, which cover various areas of life, were mainly used. The most utilized methods in resting-state fMRI were independent component analysis, seed-based analysis, and graph theory analysis. Through the analysis, we demonstrated that neuroimaging techniques could capture the neural substrates associated with cognitive reserve. Moreover, functional connectivity of brain regions centered on prefrontal and parietal cortex and network areas such as default mode network showed a significant correlation with CR, which indicated a significant association with cognitive performance. CR may induce differential effects according to the disease status. We hope that this perspective on cognitive reserve would be helpful when conducting clinical researches on the mechanisms of traditional Korean medicine for Alzheimer's disease in the future.
목 적 : Shiga-like toxin (SLT)을 생산하는 Esherichia coli에 의한 감염은 설사, 출혈성 대장염(hemorrhagic colitis) 및 용혈성 요독 증후군(hemolytic uremic syndrome)을 특징으로 하며, 특히 5세 이하의 소아에게서 심각한 결과를 초래한다. SLT-I의 병인으로는 다양한 숙주 매개인자들이 알려져 있다. 본 연구에서는 E. coli 0157 : H7 (ATCC 43890)로부터 정제한 SLT-I이 포유동물 세포들에 대한 세포독성과 종양괴사인자(tumor necrosis factor-${\alpha}$; TNF-${\alpha}$)의 생산에 미치는 효과를 측정하였으며, SLT-I의 수용체인 glycolipid globotriaosylceramide (Gb3)의 발현과 SLT-I의 세포독성의 관계를 규명하고자 하였다. 방 법 : SLT-I과 SLT-I B를 순수분리 정제하고 SLT-I B-FLTC 접합체를 제조하여 vero 세포, 대식세포 및 수지상세포를 대상으로 세포독성능을 측정하고 세포독성능의 차이가 SLT-I의 수용체인 Gb3의 발현과 상관관계가 있는지를 Flow cyotmetry로 분석하였다. 또한 대식세포의 종양괴사인자 생산능은 ELISA법으로 시행하였다. 결 과 : SLT-I은 대식세포(Raw264.7)로부터 TNF-${\alpha}$의 생산을 증가시켰다. 연구 대상 세포 중 SLT-I에 감수성을 나타낸 Vero 세포와 수지상세포(dendritic cells)는 Gb3 발현이 각각 83%와 68%로 높았으며, 29%의 낮은 Gb3 발현을 보인 Raw264.7 세포는 감수성을 보이지 않았다. 따라서 위의 결과로부터 SLT-I에 감수성을 보이지 않은 Raw264.7 세포를 대상으로 Gb3 발현 정도와 SLT-I의 세포독성의 관계를 규명하고자 Gb3의 발현을 증가 시킨 후 SLT-I의 세포독성을 재차 평가하였다. 이 결과 TNF-${\alpha}$의 처리에 의하여 6 h에 Gb3의 발현이 정점(43.5%)에 이르렀으며 36 h에 정상 수준(25.0%)으로 환원되었다. 그러나, Gb3의 발현이 증가함에도 불구하고 SLT-I의 세포독성에는 변화가 관찰되지 않았다. 따라서, SLT-I에 의한 세포독성은 세포의 종류에 따라서 다르며 또한, Gb3의 발현정도에만 의존적이지는 않을 것으로 생각된다. 결 론 : 이와 같은 결과는 E. coli 0157의 감염증 병인 연구에 있어 SLT-I과 Gb3의 발현의 상관관계에 대한 보다 심도 있는 연구가 필요함을 시사한다.

목적: 히스톤탈아세틸화효소 억제제는 그 자체의 항암효과뿐만 아니라 방사선 감작제로서의 효과가 점차분명해져가고 있다. 최근 Class I 특이적인 히스톤탈아세틸화효소 억제제의 개발로 계층 특이적인(Class specific) 연구가 가능해짐에 따라, 본 연구에서는 서로 다른 히스톤탈아세틸화효소억제제의 방사선감작효과를 비교함과 동시에 p53 발현도의 차이가 히스톤탈아세틸화효소억제제의 방사선 감수성에 미치는 영향을 알아보고자 하였다. 대상 및 방법: 이를 위해 p53 발현도가 매우 낮은 HeLa 세포에 p53의 핵 외 수송을 억제하여 세포질 내 분해를 차단하는 Leptomycin B를 처리하여 p53의 발현도를 현저하게 높인 후, Trichostatin와 SK7041의 방사선 민감도를 비교 관찰하였다. 결과: 세포생존곡선, SER 및 SF2를 비교 분석 시, p53의 발현이 높은 Leptomycin B 처리군에서 Trichostatin A가 Class I HDAC만을 억제하는 SK7041에 비해 유의하게 높은 방사선 감작효과를 나타내었다. 이는 p53이 Class I 특이적 억제제인 SK7041과 Class I과 II를 모두 억제하는 TSA의 방사선감작효과에 미치는 영향의 차이에 기전적으로 관여함을 시사한다. 결론: Leptomycln B에 의해 유도된 p53의 발현증가는 Class I과 Class I과 II를 모두 억제하는 TSA의 방사선 감작효과를 증강시킨다.
전국 최대 규모의 고추(Capsicum annuum L.) 재배지는 안동이다. 이 지역을 중심으로 본 연구는 시설재배지 고추를 가해하는 총채벌레의 연중(2021년 3월 31일~10월25일) 발생을 보고한다. 황색트랩을 이용하여 포획한 총채벌레를 유관으로 동정한 결과 꽃노랑총채벌레(Frankliniella occidentalis)와 대만총채벌레(F. intonsa)가 우점종으로 나타났다. 전체 포획충은 107,873마리였으며 이 가운데 꽃노랑총채벌레가 약 82%, 대만총채벌레가 약 17% 그리고 기타 총채벌레가 약 0.3%를 차지하였다. 연중 전체적으로 2회 총채벌레 발생 피크를 보였다. 첫 번째 발생피크는 5-6월에 나타났고, 두 번째 발생 피크는 9월 이후에 일어났다. 발생규모는 첫번째보다는 두 번째 발생 피크에서 높았으며 대부분은 꽃노랑총채벌레가 차지하였다. 흥미롭게 7-8월에 이들 총채벌레의 발생이 매우 낮았는데 이들 주요 총채벌레가 고온에 대한 높은 감수성으로 기인되었다. 실내 고온 노출실험은 총채벌레들이 35℃ 이상에서 고온 감수성을 보여 45℃에서는 1시간 노출을 견디지 못하였다. 실제로 안동지역 7-8월 시설재배지는 최고온도가 45℃ 이상을 기록하였다. 이 가운데 꽃노랑총채벌레가 대만총채벌레에 비해 낮은 고온 감수성을 보였다. 반면에 안동에 비해 기온이 낮은 강원지역의 시설 고추 재배지에서는 7-8월에 오히려 최대 총채벌레 발생피크를 나타냈으며, 이 시기 최고온도는 45℃ 이하를 기록하였다. 조사한 안동지역 고추에서는 TSWV (tomato spotted wilt virus)에 의해 발병되는 바이러스병이 발생하였다. 다중 PCR 검정법으로 이 바이러스 보독 유무 및 대상 총채벌레를 동시에 분석하였다. 이 결과 바이러스 보독충 비율은 최대 30%를 기록하며 연중 지속적으로 검출되었는데 이들 모두는 대만총채벌레로 판명되었다. 이상의 결과는 시설 고추재배지에서 꽃노랑총채벌레는 고추 수확기에 최성기를 이루며 고추에 직접피해를 줄 가능성이 높으며, 대만총채벌레는 높은 바이러스 보독율로 고추의 간접피해를 유발할 위험성이 있다는 것을 각각 제시하고 있다.
