"Leak Current" correction for critical current measurement of no-insulation HTS coil

  • Discrepancy between a power supply current and an actual "spiral" coil current makes the conventional 4-probe measurement of a critical current ($I_c$) of a no-insulation (NI) high temperature superconductor (HTS) coil inaccurate and time-consuming. This paper presents a fast and accurate approach for $I_c$ measurement of NI HTS coils. With an NI HTS coil energized at a constant ramping rate, a complete analytic expression for the spiral coil current was obtained from a first-order partial differential equation that derived from an equivalent circuit model of the NI coil. From the analytic solution, both spiral coil current and radial leak current can be obtained simultaneously, which enables fast and accurate measurement of the NI coil $I_c$. To verify the proposed approach, an NI double-pancake (DP) coil, wound with GdBCO tapes of $6mm{\times}0.1mm$, was constructed and its $I_c$ was repeatedly measured with various ramping rates in a bath of liquid nitrogen at 77 K. The measured results agreed well with the calculated ones, which validates the proposed approach to measure $I_c$ of an NI HTS coil.

A Genome-wide Approach for Functional Analysis Using Rice Mutant

  • Rapid extension of genomic database leads to the remarkable advance of functional genomics. This study proposes a novel methodology of functional analysis using 5-methyltrytophan (5 MT) mutant together with their 2-DE analysis and public microarray database. A total of 24 proteins was changed in 5 MT mutant and four remarkably different expressed proteins were identified. Among them, three spots were converted to Affymetrix probe. A total of 155 microarray samples from Gene Expression Omnibus (GEO) in NCBI was retrieved and followed by constructing gene co-expression networks over a broad range of biological issues through Self-Organising Tree Algorithm. Three co-expressing gene clusters were retrieved and each functional categorization with differential expression pattern was exhibited from 5 MT resistance mutant rice. It was indicated new co-expression networks in the mutant. This study suggests that on investigating possibility which correspond 2-DE to microarray database with their full potential.

$Cu(In,Ga)Se_2$ 박막 태양전지 제작을 위한 폴리이미드 기판의 열분석 및 후면전극 특성 분석

  • CIGS 박막 태양전지는 일반적으로 soda-lime glass(SLG)를 기판으로 사용하여 SLG/Mo/CIGS/CdS/ZnO/ITO/Grid의 구조로 제작된다. 하지만 SLG를 기판으로 사용할 경우, 유리의 특성상 무게가 무겁고, 유연성이 없기 때문에 건축물 적용에 적합하지 않다. 이러한 문제점을 극복하기 위해 가볍고 유연한 금속 및 폴리이미드 기판을 이용한 CIGS 태양전지가 널리 연구되고 있다. 그러나, 폴리이미드 기판의 경우, 특성이 우수한 CIGS 박막을 얻기 위한 고온 공정을 사용할 수 없기 때문에 이에 대한 고려가 필요하다. 본 논문에서는 CIGS 박막 태양전지 제작을 위한 폴리이미드 기판의 특성과 그 위에 형성한 후면 전극의 특성을 논의하고자 한다. 4종류의 폴리이미드 기판에 대한 열 특성을 시차주사열량계(differential scanning calorimeter)와 열중량분석기(thermogravimetric analysis), 열기계분석기(thermo mechanical anaylsis)를 이용해 분석하였다. 또한 Mo 후면 전극을 DC-sputter를 이용해 형성한 후, XRD와 AFM, 4-point probe를 이용하여 결정성 및 표면 거칠기, 면저항을 분석하였다. 결정성과 거칠기는 SLG에 증착했을 때와 동일한 결과를 보였으며, 면저항은 폴리이미드 필름에 증착 할 경우 더 크게 측정되었다. 본 연구는 중소기업청 산연기술개발사업(SL122689) 및 과학기술연합대학원대학교(UST)의 지원을 받아 수행된 "공동연구 지원사업"의 연구결과입니다.

Improved Procedure for Large-scale Isolation of Mitochondrial DNA from Mammalian Tissues

  • Although there are several methods for the preparation of mitochondrial DNA (mtDNA) from mammalian tissues, most are relatively long ultracentrifugation or manipulations by a small-scale method. We escribed a rapid method for large-scale extraction of mtDNA from human placental and horse liver tissues. The method is based on the preparation and homogenization of tissues, urification of crude mitochondria by differential centrifugations and isolation of mtDNA by alkaline Iysis. It was improved from Pre-existing methods by replacing some steps with simpler ones and discarding many others. This method gives a high yield of pure mtDNA(approximately 1-5mg from one placenta; ca. 400-600 g wet weight), depending on its sources (fresh tissue gave better results than frozen one). The resulting mtDNA indicated that this method can yield mtDNA in sufficient purity and quantity to identify the direct restriction analysis on agarose gel, random-primed labeling as a probe, and end labeling. Therefore, the method is ideal for obtaining good mtDNA samples to conduct routine restriction fragment length polymorphism (RFLP) analyses of natural populations for genetic studies.

The Effects of Two - Phase Swirling Flow on Void Distribution and Pressure Drop in a Vertical Tube (수직관에서 2상선회유동이 보이드분포와 압력강하에 미치는 영향)

  • This experimental investigation has been conducted to determine the effects of swirling angle and flow patterns on distributions of void fraction, bubble velocity and two-phase pressure drop in a vertical straight tube. Swirling angles of $0^{\circ}$ (non swirling), $30^{\circ}$, and $45^{\circ}$ were tested with air-water two components over a range of superficial air velocities. A transparent lucite tube of 38mm in internal diameter was used for the test section. The void fraction and bubble velocities were measured by means of a optical fiber probe at the upper part of the swirler in the test section. Pressure drops which seem to be closely related with flow patterns and swirling angle were measured by a differential pressure transducer. It is shown that the probability density functions of pressure drop demonstrate peculiar features for both swirling angles and flow patterns, whereas the distributions of void fraction and bubble velocities are parabolic and flat shape in the vicinity of tube center, respectively except bubbly flow in any swirling angle cases, and the void fraction increases with increasing swirling angle around the center of tube.

Electrical and Morphology Properties of Self-Assembled Dendrimer on Au (111) Substrate (Au (111)기판에 자기조립된 덴드리머 분자의 형태와 전기적 특성에 관한 연구)

  • To investigate the characteristics of the single dendrimer molecule, we attempt to measure morphology and electrical properties of the self-assembled dendrimer on Au (111)substrate with SPM(scanning probe microscopy). The same self-assembly procedure was used for two different concentrations, $10{\mu}mol/ml$ and $100{\mu}mol/ml$. The case of lower concentration, we can measure the diameter and the height of the single molecule with the tapping mode AFM image. The imaged single molecules were dome shaped and the average diameter and height were 15.6 nm, 1.2 nm respectively. From these sizes, we can calculate the volume of the single molecule. The volume of the single molecule was estimated about $116nm^3$. However, that of higher concentration, it is difficult to obtain obvious image of the single molecule. To add to, I-V characteristics were investigated using STM, on which the phenomenon of negative differential resistance (NDR)was observed between 0.14 V and 0.24 V reproductively.

Mitochondria-Specific Monoclonal Antibodies in Eggs and Embryos of the Ascidian Halocynthia roretzi

  • Ascidian embryos have become an important model for embryological studies, offering a simple example for mechanisms of cytoplasmic components segregation. It is a well-known example that the asymmetric segregation of mitochondria into muscle lineage cells occurs during ascidian embryogenesis. However, it is still unclear which signaling pathway is involved in this process. To obtain molecular markers for studying mechanisms involved in the asymmetric distribution of mitochondria, we have produced monoclonal antibodies, Mito-1, Mito-2 and Mito-3, that specifically recognize mitochondria-rich cytoplasm in cells of the ascidian Halocynthia roretzi embryos. These antibodies stained cytoplasm like reticular structure in epidermis cells, except for nuclei, at the early tailbud stage. Similar pattern was observed in vital staining of mitochondria with DiOC2, a fluorescent probe of mitochondria. Immunostaining with these antibodies showed that mitochondria are evenly distributed in the animal hemisphere blastomeres at cleavage stages, whereas not in the vegetal hemisphere blastomeres. Mitochondria were transferred to the presumptive muscle and nerve cord lineage cells of the marginal zone in the vegetal hemisphere more than to the presumptive mesenchyme, notochord and endoderm lineage of the central zone. Therefore, it is suggested that these antibodies will be useful markers for studying mechanisms involved in the polarized distribution of mitochondria during ascidian embryogenesis.

Analytical Techniques for Measurement of Crosslink Densities of Rubber Vulcanizates

  • It is important to analyze crosslink densities of rubber articles because the physical properties are dependent on the crosslink densities. In this paper, analytical techniques for the measurement of crosslink densities of rubber vulcanizates are described. The most widely used method to measure the crosslink density is a swelling method combined with the Flory-Rehner equation. Application of the interaction parameter (${\chi}$) of rubber and swelling solvent is critical because the crosslink density is absolutely dependent on the ${\chi}$ value. Methods for obtaining ${\chi}$ employ not only solubility parameters of the polymer and swelling solvent but also inverse gas chromatography (IGC). The solubilities of rubbers can be obtained using micro differential scanning calorimetry (${\mu}DSC$), intrinsic viscosity measurement, and UV-visible spectroscopy. Nuclear magnetic resonance (NMR) spectroscopy has been also used for the measurement of the crosslink density using the $T_2$ relaxation time, which is determined by spin-spin relaxation in solid-state NMR. For sulfur-cured rubber vulcanizates, crosslink densities according to the crosslink types of mono-, di-, and polysulfides are measured by treating the rubber samples with a chemical probe composed of thiol and amine compounds. Measurement methods of physical crosslinking by filler, crystallization, and ionic bonding have also been introduced.

The Molecular Gas Kinematics of HI Monsters

  • Our HI monster sample is a set of local HI-rich galaxies identified by the ALFALFA survey (Arecibo Legacy Fast Survey ALFA) at z<0.08. Intriguingly, they are also found with a relatively large molecular gas reservoir compared to the galaxies with similar stellar mass and color, yet their star formation rate is quite comparable to normal spirals. This makes our HI monsters good candidates of galaxies in the process of gas accretion which may lead to the stellar mass growth. One feasible explanation for their relatively low star formation activity for a given high cool gas fraction is the gas in monsters being too turbulent to form stars as normal spirals. In order to verify this hypothesis, we probe the molecular gas kinematics of 10 HI monsters which we observed using the Atacama Large Millimeter/sub-millimeter Array (ALMA). We utilize the tilted ring model to investigate what fraction of the molecular gas in the sample is regularly and smoothly rotating. In addition, we model the molecular gas disk using the GALMOD package of the Groningen Image Processing System (GIPSY) and compare with the observations to identify the gas which is offset from the 'co-planar differential rotation'. Based on the results, we discuss the possibility of gas accretion in the sample, and the potential origin of non-regularly rotating gas and the inefficient star formation.

Effects of Local Anesthetics on the Fluidity of Synaptosomal Plasma Membrane Vesicles Isolated from Bovine Brain (국소마취제가 Synaptosomal Plasma Membrane Vesicles의 유동성에 미치는 영향)

  • To elucidate the mechanism of action of local anesthetics, the effects of local anethetics on the microenvironment of the lipid bilayers of synaptosomal plasma membrane vesicles (SPMV) isolated from bovine brain and dimyristoylphosphatidylcholine (DMPC) multilamellar liposomes were investigated employing the intermolecular excimer fluorescence technique and differential scanning calorimetry (DSC). The relative intensities of excimer and monomer fluorescence of pyrene are a simple linear function of the viscosity of a homologous series of solvents. The microviscosity(${\eta}$)of the hydrocarbon region of SPMV was measured by this method and the value was $57.3{\pm}5.3\;cP$ at $37^{\circ}C$. In the presence of lidocaine-HCl and procaine-HCl, the values decreased to $46.5{\pm}5.1\;cP$ and $54.7{\pm}4.8\;cP$, respectvely. The differential scanning thermograms of DMPC multilamellar liposomes showed that local anesthetics significantly lowered the phase transition temperature, broadened the thermogram peaks, and reduced the size of the cooperative unit. These results indicate that local anesthetics have significant fluidizing effects on biomembranes and perturbation of membrane lipids may produce some, but not all, of their pharmacological actions.
