Salinity Effects on the Survival of the Metazooplankton in the Coastal Waters off the Seamankeum Areas

  • A huge freshwater reservoir (ca. 12,000 ha) will be created when the construction of a 33­km dike on a huge mud flat of the Saemankeum areas is established. A large quantity of freshwater will emerge to the adjacent sea from the reservoir through two big gates. Marine organisms outside the dike are expected to frequently experience low salinity waters. To investigate the salinity effects on the dominant metazooplankton in the coastal waters off Saemankeum areas, we measured the survival (Survival 1H and Survival 24H) of 11 different taxa (the copepods Acartia omorii, A. pacifica, Calanus sinicus, Centropages abdominalis, Paracalanus indicus, Pseudodiaptomus inopinus, Tortanus forcipatus, and a hydromedusa, and barnacle nauplius, polychaeta larva, and a chaetognath Sagitta sp.) at salinities of 0, 5, 10, 15, 20, 25, 30, 35, and 40 psu when the organisms were exposed for 1 and 24 h, respectively. Survival 1Hs of P. inopinus and barnacle nauplius were 100% between 5 and 35 psu, while they were 0% at salinities of 0 and 40 psu. Survival 1Hs of A. omorii and A. pacifica, P. indicus, T. forcipatus, and polychaeta larva were 100% at $salinities\;\geq\;10$ psu, while they were 0% at lower salinities. Survival 1Hs of a hydromedusa and Sagitta sp. were 100% at $salinities\;\geq\;15$ psu, while they were 0% at lower salinities. Survival 1H of C. abdominalis and C. sinicus was 100% at $salinities\;\geq\;20$ psu, while they were 0% at lower salinities. Survival 24Hs of A. omorii, A. pacifica, C. abdominalis, barnacle nauplius, and polychaeta larva were the same as Survival 1 Hs at the same salinity, while those of the other metazooplankton were lower than Survival 1Hs. The results of the present study suggest that low salinity water emerging from big gates may cause the death of the metazooplankton, but the salinities at which death of the metazooplankton occurs may differ by species.

  • This study aimed to determine the appropriate salinity condition for optimal cultivation of the juvenile hybrid grouper Epinephleus akaara♀×E. lanceolatus♂ in conditions of decreasing salinity. The survival, growth, metabolic rate, stress, and histological response were investigated in juvenile hybrid grouper exposed to different salinities for 30 days. At 0 psu, the survival rate of the juvenile hybrid grouper was 60% on the 2nd day of exposure and all individuals died on the 3rd day of exposure. At salinities above 3 psu, all animals survived throughout the exposure period. Growth rate for body weight of the juvenile hybrid grouper was the highest in the control and the lowest at 10 psu. Plasma osmolality of the juvenile hybrid grouper exposed to different salinities was 313-355 mg Osmol/kg at salinities above 10 psu, and then decreased to 225-264 mg Osmol/kg at salinities below 5 psu. The oxygen consumption rate tended to decrease as the salinity decreased. Stress responses of the juvenile hybrid grouper were analyzed with decreasing salinity. Therefore, it is considered that a salinity of more than 20 psu is suitable for the culture of the juvenile hybrid grouper.

Effects of Salinity Changes on Hematological Responses in Juvenile Ship Sturgeon Acipenser nudiventris

  • A 10-day trial was conducted to evaluate the salinity tolerance and hematological responses of juvenile ship sturgeon Acipenser nudiventris under different salinity levels. A total of 240 fish averaging $6.2{\pm}0.13g$ (mean${\pm}$SD) were randomly distributed into 12 tanks in groups of 20, and each tank was then randomly assigned to one of three replicates of four different salinities at 0, 4, 8 and 12‰. At the end of 10 days of experimental period, plasma protein value of fish exposed to 0‰ and 4‰ salinities was significantly higher than those of fish exposed to 8‰ and 12‰ salinities (P < 0.05). Significantly higher values for plasma cortisol and glucose were obtained in fish exposed to 12‰ salinity compared to the fish exposed to 0‰ and 4‰ salinities (P < 0.05). Plasma potassium and sodium values increased significantly by increment of salinity levels (P < 0.05). Therefore, these results indicated that in natural environments, juvenile ship sturgeon, A. nudiventris in migratory populations could be able to migrate and adjust successfully into brackish water with a salinity of up to 8‰ without any short-term hematological stress responses.

어린 숭어(Mugil cephalus)의 염분농도별 성장과 생존을 비교 (Comparison of Growth and Survival Rates of Juvenile Grey Mullets (Mugil cephalus) in Different Salinities)

  • 어린 숭어, Mugil cephalus에 대한 저염분 해수 및 담수 사육의 가능성을 알아보기 위하여, 전장 $4.8{\pm}0.6$ cm, 체중 $1.0{\pm}0.3$ g 및 전장 $13.6{\pm}0.2$ cm, 체중 $20.0{\pm}0.5$ g의 2군을 대상으로 염분농도별 성장과 생존율을 조사하고, 산소소비량 및 피부의 두께 등을 측정하여 생리활성을 서로 비교하였다. 1, 2차 실험 모두에서 $25\~75\%SW$의 저염 분구가 성장과 생존에 있어 $100\%SW$보다 좋은 편이었다. 그러나, 단계적 순치과정을 거치지 않고 $100\%SW$로부터 $0\%SW$로의 급격한 염분변화는 어체의 성장 및 생존율을 감소시키는 요인이 되었다. 일간사료섭식률은 $25\%SW$에서 $3.6{\pm}0.8\%$로 가장 높았다. 식욕에 있어서는 $50\%SW$에서 가장 높았다. 일간성장률은 $25\%\;SW$에서 가장 좋았고 ($2.7{\pm}0.9\%$), 다음으로 $50\%SW,\;100\%SW,\;0\%SW$이 순으로 나타났다. 염분에 따른 각 실험구별 어체의 표피와 진피의 두께에는 유의차가 없었으나, 지방층은 $0\%SW$의 실험어에서 $50\%SW$$100\%SW$의 숭어 보다 유의하게 두꺼웠다. 숭어의 산소소비량은 염분농도가 감소함에 따라 유의하게 낮아지는 경향을 나타냈다. 염분농도별 숭어의 저산소 내성은 $0\%SW$에서 비교적 약한 것으로 나타났다.

Comparison Between Phylogenetic Relationships Based on 18S rDNA Sequences and Growth by Salinity of Chlorella-like Species (Chlorophyta)

  • This study was carried out to understand the correlation between phylogenetic relationships based on 18S rDNA sequences and growth by salinity of Chlorella-like species. The 18S rDNA sequences of 71 Chlorella-like species which were mainly collected from Korean waters were analyzed. The 18S rDNA sequences of Chlorella-like species were divided into three groups (group A, B and C) and group B was further divided into three subgroups (subgroup B-1, B-2 and B-3). Thirty-seven Chlorella-like species in group A grew well at high salinity (32 psu) but the other groups grew well in freshwater. The sequence identities of the species in group A and B were 97.2-99.5%, but those of 6 species in group C ("Chlorella" saccharophila), which contained group I intron sequences region were 75.0-75.4%. Two representative species of each group were cultured at different salinities (0, 16 and 32 psu) to examine the correlation between the molecular phylogenetic groups and the phenotypic characteristics on cell growth and size by different salinities. The size of cell cultured at different salinities varied according to the species of each molecular phylogenetic group. The size of "Chlorella" saccharophila in group C was bigger and more obviously elliptical rather than that of the other Chlorella-like species. Considering the results on molecular and phenotypic characteristics, the group A and B belonged to Chlorellaceae, but group C was distinctly different from them.

  • This study examined the preferred salinity levels of soups by different demographic groups in order to establish desirable serving conditions for soups with respect to customer satisfaction and health. The salinity levels were measured in August and November to determine seasonal variations using a digital salinity measuring device. Four demographic groups totaling 1,723 individuals were surveyed from institutional foodservice establishments (IFEs) in elementary schools (ES: male 202/female 194), middle-high schools (MHS: 268/216), universities (UNIV: 208/238), and companies (COM: 182/215) in Seoul. Various thin or thick soups typically offered in foodservice establishments were served salty (1.2%) and less salty (0.4%) at $55{\pm}2^{\circ}C$ in a 50 mL portion. The preferred salinities of the soups were found to be 0.71%, 0.79%, 0.76%, and 0.74% for ES, MHS, UNIV, and COM, respectively. Men had 0.02~0.08% higher preferences than women in ES, MHS and UNIV. The customers also preferred higher salinity levels in November than in August, and in thick soups than in thin soups. The salinities preferred by school children were significantly (p<0.01) lower than those of soups by the IFEs. A similar observation was made for MHS, in which each IFE served the soups with significantly (p<0.01) higher salinities than those preferred by the students. Again, at two COM IFEs, the salinities of the soups served were significantly higher than those preferred by the customers. However, for one of UNIV, the salinity of the served soup was significantly (p<0.05) higher than that preferred by the students, while in the two other UNIV no significant differences were noted between the salinity of the served soup and the level preferred by customers. The surveys showed that 9 out of 12 IFEs served soups having salinities that were significantly higher than those preferred by customers. In particular, regardless of preference, the MHS group was served soups with higher salinity levels than the other demographic groups. The salinities of the soups provided by the IFEs were unnecessarily high for the levels preferred by the customers. Therefore, IFEs should make an effort to serve soups that are less salty soups to enhance public health.

어린 숭어 (Mugil cephalus)의 염분별 삼투조절 능력 (Osmoregulation Capability of Juvenile Grey Mullets (Mugil cephalus) with the Different Salinities)

  • 숭어, Mugil cephalus의 염분별 삼투조절 능력을 알아보기 위하여, 전장 $13.6{\pm}0.2cm$, 체중 $20.0{\pm}0.5g$ 크기의 재료를 해수 (SW)에 담수를 첨가하여 만든 $0\%SW\~100\%SW$에 수용한 다음, 60일간 사육하면서 염분별 혈액의 이온 농도와 삼투질 농도 변화를 조사하고, 아가미와 염세포의 조직학적 변화 및 근육의 수분함량을 비교하였다. $0\%SW\~25\%SW$에서 숭어 혈장의 $Na^+$$K^+$ 농도는 실험개시후 8일째부터 안정되었다. $0\%SW$에서의 혈장 $Cl^-$ 농도는 실험개시후 0.5일째 가장 낮았으며 2일 이후부터 안정되었다. 실험종료시에 있어서 숭어 혈장의 삼투질 농도는 저염분 일수록 낮아지는 경향을 보여, $100\%SW$부터 $0\%SW$까지 각각 340, 334, 322 및 316 mOsm/kg 수준으로 안정됨으로써, 저염분에서는 저삼투 조절을 하는 것으로 나타났다. 이와 함께 hematocrit도 염분이 낮아짐에 따라 감소하는 경향이었다. $0\%SW\~50\%SW$에 10일째 노출된 숭어의 아가미에서는 2차새변이 손상되었으며, 특히 $0\%SW$에서는 2차새변의 박리, 유착 및 괴사 등이 관찰되었다. 그러나 손상된 아가미는 이후부터 회복되기 시작하여 사육 15일째에 정상적인 조직상을 나타냈다. 염분에 따른 각 염분구별 어체 근육의 수분함량에는 차이가 없었다.

염분농도에 따른 치어기 은대구(Anoplopoma fimbria)의 혈액학적 성상, 혈장성분 및 항산화반응의 변화 (Alterations of Hematological Parameters, Plasma Constituents and Antioxidant Responses in the Sablefish Anoplopoma fimbria Depending on Salinity)

  • Juvenile Anoplopoma fimbria (mean length $15.6{\pm}1.4cm$, mean weight $68.7{\pm}4.3g$) were exposed to 4 months with the different levels of salinity [100 (35.0), 90 (31.5), 80 (28.0), 70 (24.5), 60 (21.0), 50 (17.5), and 40 (14.0) % (psu)] for 4 months. Hematological parameters such as red blood cell (RBC) counts, hematocrit (Ht), and hemoglobin (Hb) concentrations were substantially decreased under salinities of 50% psu or lower. Of the measured inorganic plasma constituents, magnesium was notably decreased, whereas there was no effect on calcium. Among organic plasma components, glucose and cholesterol were significantly increased, and total protein was decreased. Among enzyme plasma components, glutamic oxalate transaminase (GOT), glutamic pyruvate transaminase (GPT), and alkaline phosphatase (ALP) were significantly increased under salinities of 50% psu or lower. Antioxidant responses such as glutathione S-transferase (GST) and glutathione (GSH) were significantly decreased at salinities of 50% psu or lower. The results of this study indicate that salinity affects the blood parameters, plasma constituents, and antioxidant responses of A. fimbria.

Occurrence and pathogenicity of Pythium (Oomycota) on Ulva species (Chlorophyta) at different salinities

  • Pythium species are ubiquitous organisms known to be pathogens to terrestrial plants and marine algae. While several Pythium species (hereafter, Pythium) are described as pathogens to marine red algae, little is known about the pathogenicity of Pythium on marine green algae. A strain of a Pythium was isolated from a taxonomically unresolved filamentous Ulva collected in an intertidal area of Oslo fjord. Its pathogenicity to a euryhaline Ulva intestinalis collected in the same area was subsequently tested under salinities of 0, 15, and 30 parts per thousand (ppt). The Pythium isolate readily infected U. intestinalis and decimated the filaments at 0 ppt. Mycelium survived on U. intestinalis filaments for at least 2 weeks at 15 and 30 ppt, but the infection did not progress. Sporulation was not observed in the infected algal filaments at any salinity. Conversely, Pythium sporulated on infected grass pieces at 0, 15, and 30 ppt. High salinity retarded sporulation, but did not prevent it. Our Pythium isolate produced filamentous non-inflated sporangia. The sexual stage was never observed and phylogenetic analysis using internal transcribed spacer suggest this isolate belongs to the clade B2. We conclude that the Pythium found in the Oslo fjord was a pathogen of U. intestinalis under low salinity.

담수순화 감성돔 Acanthopagrus schlegelii 동결보존 정자의 수정능력 (Fertilization Ability of Cryoperserved Sperms of Black Porgy Acanthopagrus schlegelii Acclimated in Freshwater)

  • Fertilization rates were over 90% when sperm from black porgy Acanthopagrus schlegelii acclimated and raised in freshwater and black porgy raised in seawater were fertilized with eggs from female black porgy raised in seawater, although cryopreserved sperm were significantly lower in fertilization rates than non-cryopreserved sperm. From the observations of embryos development at different salinities (0, 10, 20 and 32 psu) within 3 hours upon fertilization (16 cell stage), all were dead at 0 psu. However, the development process and speed at 10 and 20 psu were the same as at 32 psu. But, many developing embryos were egg envelope injury at low-salinities of 10 and 20 psu unlike at 32 psu. This is attributable to osmotic shock. Hatching rate of embryos at 32 psu was 60% or so, whereas low as 5% or less were successfully hatched at 10 and 20 psu, implying that osmotic shock could be responsible for the failure of embryo development.
