고도 불포화지방산으로 산화스트레스가 유도된 흰쥐의 뇌에서 비타민 E의 항산화효소 활성 및 CYP2E1 발현에 미치는 효과 (Vitamin E in vivo Studies on the Activity of Antioxidant Enzymes and CYP2E1 Expression in High PUFA-treated Brains)

  • 본 연구는 식이 불포화지방산으로 산화 스트레스가 유도된 쥐에게서 비타민 E의 항산화 효과를 알아보기 위해 4주령의 쥐 90마리를 1주일간 적응시킨 후, 정상식이를 섭취한 대조군(C), 식이 불포화지방산(올레산, 리놀레산, 리놀렌산, DHA)군, 식이 불포화지방산에 비타민 E를 첨가한 군으로 나누어 8주간 실시하였다. 체중변화는 대조군에 비하여 모든 군에서 증가하였고, 비타민 E 첨가군에서는 LA+비타민 E군을 제외한 모든 군에서 유의적인 체중증가가 나타났다. 뇌 마이크로좀의 지질과산화물 생성은 대조군에 비해 특히 LA, DHA 군에서 유의적으로 높은 경향을 보였다. 비타민 E 첨가 시 모든 군에서 첨가하지 않은 군에 비하여 지질과산화물 농도가 감소하였다. 뇌 세포질의 항산화 효소 SOD, GPx는 대조군에 비하여 모든 지방산 군에서 활성이 증가하였고 비타민 E 첨가 시 활성이 유의하게 감소하였다. 뇌 마이크로좀의 CYP2E1의 활성은 LNA군에서만 유의적으로 증가하고 비타민 E 첨가 시 LNAE군만이 유의하게 감소됨을 확인하였다. 이상의 실험결과, 식이 불포화지방산으로 유도된 뇌의 산화 스트레스는 비타민 E를 식이 불포화지방산과 같이 섭취함으로써 산화 스트레스 감소에 긍정적 영향을 미치는 것으로 나타났다.

Effects of dietary n-6:n-3 polyunsaturated fatty acid ratio on growth performance, blood indexes, tissue fatty acid composition and the expression of peroxisome proliferator-activated receptor gamma signaling related genes in finishing pigs

  • Objective: This study investigated the effects of dietary n-6:n-3 polyunsaturated fatty acid (PUFA) ratio on growth performance, blood indexes, tissue fatty acid composition and the gene expression in finishing pigs. Methods: Seventy-two crossbred ([Duroc×Landrace]×Yorkshire) barrows (68.5±1.8 kg) were fed one of four isoenergetic and isonitrogenous diets with n-6:n-3 PUFA ratios of 2:1, 3:1, 5:1, and 8:1. Results: Average daily gain, average daily feed intake and gain-to-feed ratio had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The concentrations of serum triglyceride, total cholesterol and interleukin 6 were linearly increased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio, while that of high-density lipoprotein cholesterol tended to decrease (p = 0.062), and high-density lipoprotein cholesterol:low-density lipoprotein cholesterol ratio and leptin concentration were linearly decreased (p<0.05). The concentration of serum adiponectin had a quadratic response but the measurement was decreased and then increased (quadratic, p<0.05). The proportion of C18:3n-3 was linearly decreased (p<0.05) in the longissimus thoracis (LT) and subcutaneous adipose tissue (SCAT) as dietary n-6:n-3 PUFA ratio increasing, while the proportion of C18:2n-6 and n-6:n-3 PUFA ratio were linearly increased (p<0.05). In addition, the expression levels of peroxisome proliferator-activated receptor gamma (PPARγ) and lipoprotein lipase in the LT and SCAT, and adipocyte fatty acid binding protein and hormone-sensitive lipase (HSL) in the SCAT had quadratic responses but the measurements were increased and then decreased (quadratic, p<0.05). The expression of HSL in the LT was linearly decreased (p<0.05) with increasing of dietary n-6:n-3 PUFA ratio. Conclusion: Dietary n-6:n-3 PUFA ratio could regulate lipid and fatty acid metabolism in blood and tissue. Reducing dietary n-6:n-3 PUFA ratio (3:1) could appropriately suppress expression of related genes in PPARγ signaling, and result in improved growth performance and n-3 PUFA deposition in muscle and adipose tissue in finishing pigs.

냉장중 사슴육 증탕액의 이화학적 특성의 변화 (Changes of the Physico-chemical Characteristics of Venison Extracts during Chilling Storage)

  • This experiment was carried out to investigate the changes in pH, proximate composition, pH, VBN, TBA, minerals, and fatty acid of venison extracts, from three slaughtered deers with 180~210 kg live weight (♀, Elk deer, 28~30 months of age) at 4$^{\circ}C$. Proximate composition was not affected by storage periods. The pH, VBN, and TBA ranged from 4.60~4.62, 13.52~15.75 mg%, and 0.20~0/81mg/kg. respectively. The pH, VBN, and TBA gradually increased during storage period (p<0.050. Among minerals, K, P, Na, Mg, and Ca were major mineral contents and the Ca, mg, Na contents significantly decreased (p<0.05) according to the storage period. The major fatty acid found in venison extracts were oleic acid, palmitic acid, stearic acid, linoleic acid. Oleic acid, linolenic acid, and arachidonic acid decreased during storage, but palmitic acid, heptadeanonic acid, and stearic acid increased during the storage. U/S (unsalturated fatty acid/saturated fatty acid) ratio tended to decrease during the storage.

사료내 Conjugated Linoleic Acid 첨가수준이 육계의 생산성, 도체특성 및 근내 지방산 조성에 미치는 영향 (Effect of Dietary Conjugated Linoleic Acid on Growth Performance, Carcass Characteristics and Muscular Fatty Acid Composition in Broiler)

  • 본 연구는 CLA를 육계사료에 첨가하여 그 급여수준(0, 0.5, 1.0, 1.5, 및 2.0%)에 따라 육계의 생산성과 CLA를 급여한 계육의 도체특성, 혈청콜레스테롤 및 지방산조성의 변화를 검토하고자 육계 200수(Arbor Acre Broiler, male)를 5주 동안 공시하였다 육계의 생산성을 나타내는 증체량, 사료섭취량 및 사료요구율은 CLA급여에 따른 유의성은 없었다. 도체특성중 도체율뿐만 아니라 도체중에 대한 간, 대퇴부위, 흉심 및 북채의 비율은 CLA를 급여함으로서 유의한 변화가 없었지만, 복강지방 비율은 CLA 급여량이 많아짐에 따라 유의한 감소 현상을 확인할 수 있었다(p<0.05). 혈청콜레스테롤은 CLA급여량이 증가할 수록 총콜레스테롤과 LDL-C은 감소하고, HDL-C은 증가하였다(p<0.05). 그리고 지방산 조성 변화는 CLA를 급여함에 따라 palmitic acid는 증가하고, oleic acid, linoleic acid 및 arachidonic acid는 감소하여 포화지방산은 증가하고 불포화지방산은 감소하는 현상을 보였다. 또한, CLA의 이성체함량도 CLA급여량이 증가함에 따라 직선적으로 증가하였다(p<0.05). 결론적으로 계육에 CLA 축적을 극대화하기 위해서는 2%까지 급여도 가능하지만. 지방산조성의 변화가 심하므로 CLA도 축적시키고 지방산 변화도 최소화할 수 있는 1%의 급여수준이 적합할 것으로 판단된다.

식이지방과 Tocopherol 첨가가 혈액의 지질 및 조직의 Tocopherol 함량과 지방산조성에 미치는 영향 (Effect of Dietary Fat and Marginal Tocopherol Supplement on Plasma Lipid, Tocopherol Contet and Fatty acid Composition of Rat Tissues)

  • To observe the effect of the different level of PUFA and marginal tocopherol supplement on HDL-chol, tissue tocopherol content and fatty acid composition, the rats were supplied either safflower oil or conconut oil with or without tocopherol supplement to the experimental diet. Plasma tocopherol level was not greatly influenced by the different dietary fat and similar effect was observed in the liver but not in the adipose tissue. HDL-chol level was reduced in the high PUFA diet regardless of tocopherol content. No effect by tocopherol supplement was observed in the fatty acid composition of liver and adipose tissue lipid in both dietary PUFA levels . There was also no increase in the content of tissue polyenoid acid by tocopherol in the high PUFA diet . Fatty acid composition of tissue lipid was rather more influenced by dietary fat. Lauric and myristic acid contents were higher in the low PUFA diet and linoleic acd and total polynoic acid content were higher in the high PUFA diet. With tocopherol supplement tocopherol /PUFA ratio of tissue was increased but the ratio of high PUFA diet was significantly lower than that of low PUFA diet. Marginal tocopherol supplement could not reduce the peroxidizability index of high PUFA diet.

정상인과 녹내장 환자의 혈청 인지질 지방산 농도 (Fatty Acid Concentration of Serum Phospholipids in Normal and Glaucoma Patients)

  • Glaucoma is one of the leading causes of blindness worldwide and is characterized by degeneration of the optic nerve. Elevated intraocular pressure (IOP) is the major contributing factor to optic nerve damage. However, some patients develop glaucoma even with normal IOP. Other factors, including age, race, myopia, and nutrition, can affect glaucoma risk. Dietary fat intake and serum fatty acid composition are closely related with the fatty acid profile of the retina and thus can modulate glaucoma risk. In this study, we collected serum samples from 34 glaucoma patients (26 primary open angle glaucoma (POAG) and 8 normal tension glaucoma (NTG)) and 45 healthy controls and analyzed their serum phospholipid fatty acid concentrations. The results show that the ratio of oleic acid to stearic acid (OA/SA), which is an indicator of stearoyl-CoA desaturase activity, was higher in POAG patients when compared to normal controls. The ratio of eicosapentaenoic acid to arachidonic acid (EPA/AA), which is an indirect marker of eicosanoid biosynthesis, was also higher in glaucoma patients (p=0.048). These results imply that dietary fatty acid intake and serum fatty acid profile can influence glaucoma risk. Additional study is necessary to identify the relationship between fatty acids and glaucoma.

식이 Linolenic Acid와 Linoleic Acid 함량변화가 흰주의 연령에 따른 지방대사 및 항혈전효과에 미치는 영향 (The Effect of Diet Varing Linolenic Acid and Linoleic Acid Content on Lipid Metabolism and Antithrombosis in Different Aged Rats)

  • This study was done to investigate the effect of age and dietary linolenic acid content and the linolenic acid/linoleic acid(LNA/LA) ratio on the lipid metabolism and formation of PGI2 and TXA2. The male Sprague-Dawley rats were fed 6 different with 0.2, 0.4, 0.6 of LNA/LA ratio within either 8% LNA(high LNA) or 4% LNA(low LNA) of fatty acid content for different feeding period(1, 4, 12 month). The dietary fat used were sesame oil, perilla oil, soybean oil and beef tallow. The concentration of serum total lipid, total cholesterol and HDL-C were increased with aging. Triglyceride concentration was decreased in 0.2 ratio of LNA/LA. The lipid content of liver showed similar tendency to that of serum. The ratio of PGI2/TXA2 was increased in 1 month rats and antithrombotic effect was reduced significantly with increasing age. The PGI2/TXA2 ratio was tended to be higher in diet of 0.2 and 0.4 LNA/LA ratio at high LNA level and in diet of 0.6 LNA/LA ratio at low LNA level. Especially PGI2/TXA2 ratio was increased linearly with rising LNA/LA ratio at low LNA level. It seemed that the LNA content and LNA/LA ratio had interaction to increase the antithrombotic effect bychanging TXA2 synthesis. And the dietary fatty acid related effect lowering the serum and liver lipid content, excepting triglyceride, was increased when dietary n3/n6 ratio was high(0.6) at both high and low n3 level. Therefore, it could not be recommended to consume large amount of n3 fatty acid or high ratio of n3/n6 to prevent cardiovascular diseases. These results suggested that the dietary fatty acid ratio of n3/n6 could be determined based on the n-3 content of dietary fat to reduce the risk of cardiovascular disease.

Dietary Fiber and Large Bowel Cancer

  • Large bowel cancer correlates tightly to dietary factors such as dietary fiber and fat. Dietary fiber prevents the large bowel cancer in different modes of action which depend upon physicochemical and fermentable properties. Water-soluble fiber is fermented easily by intestinal microbes producing short chain fatty acids ; in contrast, water-insoluble fiber occurs effectively more rapid transit time due to greater bulk of gut content, though it is unfermentable. Not only short chain fatty acid is utilized in the proximal and distal colon as primary energy source, but also it lowers pH in the colon to normalize cellular differentiation and helps to stimulate peri staltic movement by acting as an osmotic laxative. In particular, butyric acid may also regulate gene expression and cell growth, though it is an important respiratory fuel for the colonocyte. Since dietary fiber and non-digestible oligosaccharides are the major source of butyric acid, this provides a possible link between dietary fiber and oligosaccharide and prevention of large bowel cancer. But, as with many links between dietary fiber and large bowel cancer, a direct casual association has not been established. In addition, RDA of dietary fiber which is 20~25g/day for adult Japanese, appears to be reasonable for the defecation once daily and the prevention of large bowel cancer.

Conjugated Linoleic Acid 급여기간과 첨가수준이 저장기간 중 육계의 부위별 품질특성에 미치는 영향 (Effects of Dietary Conjugated Linoleic Acid Levels and Periods on Meat Quality in Breast and Thigh Muscles of Broiler)

  • CLA를 이용한 고품질 기능성 육계의 생산 가능성을 알아보기 위해 첨가수준과 급여기간에 따른 부위별 CLA 축적율과 지방산 조성 및 지방산패도를 분석하였다. CLA 축적율은 첨가수준이 증가할수록 유의적으로(P<0.05) 증가하였으며, 급여기간에 따라서도 다소 증가하는 경향을 보였다. 그러나 급여 1주와 급여 2주 및 3주의 비교에서 큰 차이는 나타나지 않았다. CLA 첨가수준에 따른 포화지방산은 증가하는 경향을 보였지만, 불포화 지방산은 감소하는 경향을 보였으며, 특히 CLA와 가장 비슷한 구조를 가지고 있는 linoleic acid(C18:2)는 CLA 함량이 증가한 만큼 감소하는 경향을 나타내었다. CLA 첨가수준에 따른 지방산패도의 변화는 첨가수준이 증가함에 따라 낮게 나타났으며, 대조구에 비해 처리구가 낮게 나타났다. 이상의 결론은 근육내 CLA 축적을 극대화 시키기 위한 최적의 급여기간은 1주이며, 지방산패도의 효과적인 억제를 위해서 첨가수준 4.8%가 가장 적합하다는 것을 나타낸다.

Biosynthesis of Conjugated Linoleic Acid and Its Incorporation into Ruminant's Products

  • Bio-hydrogenation of $C_{18}$-unsaturated fatty acids released from the hydrolysis of dietary lipids in the rumen, in general, occurs rapidly but the range of hydrogenation is quite large, depending on the degree of unsaturation of fatty acids, the configuration of unsaturated fatty acids, microbial type and the experimental condition. Conjugated linoleic acid (CLA) is incompletely hydrogenated products by rumen microorganisms in ruminant animals. It has been shown to have numerous potential benefits for human health and the richest dietary sources of CLA are bovine milk and milk products. The cis-9, trans-11 is the predominant CLA isomer in bovine products and other isomers can be formed with double bonds in positions 8/10, 10/12, or 11/13. The term CLA refers to this whole group of 18 carbon conjugated fatty acids. Alpha-linolenic acid goes through a similar bio-hydrogenation process producing trans-11 $C_{18:1}$ and $C_{18:0}$, but may not appear to produce CLA as an intermediate. Although the CLA has been mostly derived from the dietary $C_{18:2}$ alternative pathway may be existed due to the extreme microbial diversity in the reticulo-rumen. Regardless of the origin of CLA, manipulation of the bio-hydrogenation process remains the key to increasing CLA in milk and beef by dietary means, by increasing rumen production of CLA. Although the effect of oil supplementation on changes in fatty acid composition in milk seems to be clear its effect on beef is still controversial. Thus further studies are required to enrich the CLA in beef under various dietary and feeding conditions.
