This study was to investigate the effects of dietary linoleic acid(18:2\omega6, LA) and aipha-linolenic acid(18:3\omega3. \alpha-LNA) levels on brain development and fatty acid compositions of various lipid classes in the chicken embryo brain tissues. Thirty two ISA Brown layers, 52 weeks-old, were divided into four groups. Birds of each group were given corn-soybean meal based diets added with 1) safflower oil 8%, 2) safflower oil 6% + perilla oil 2%, 3) safflower oil 2% + perilla oil 6%, or 4) perilla oil 8%. Mter 15 days fed the diets. the layers were artificially inseminated to obtain fertile eggs. During the incubation. embryonic brains were sampled at 15th and 21st days. Fatty acid contents were quantitated by using heptadecanoic acid (17:0) as an internal standard. No significant differences in brain weight and in contents of various lipids such as phospholipid. triglyceride, cholesterol. cholesterol ester and free fatty acid in the tissues were found among the dietary groups (P<0.05). The ratios of AA/LA in the brain lipid classes were lowered as the dietary levels of perilla oil were increased. Higher LA was found in phosphatidylcholine(PC) than arachidonic acid (20:4\omega6. AA), meanwhile the level of LA was less than AA in phosphatidylethanolamine(PE). Docosahexaenoic acid(22:6\omega3, DHA) was the* major fatty acid in the tissue and its content in PE was 2.5~3 times higher than in PC. DHA level in the phospholipid reached at a peak (1.7~1.8 mg/brain) in dietary groups added with 6% or 8% perilla oil. suggesting that no more increase in that fatty acid level in the brain tissue could be obtained by consuming more \alpha-LNA, the major precursor of DHA.
The prevalence of atopic dermatitis (AD) continues to rise in industrialized countries related to Western lifestyle, including dietary habits, especially imbalance of intake of dietary fatty acids. The purpose of this study was to evaluate the dietary fatty acids and the assess the blood fatty acid composition and immune parameters in AD patients. AD (n = 50) patients and gender ${\cdot}$ age matched healthy controls (HC) were studied in case-control clinical trail. Current fatty acids intake status was determined by 3-day food record method. Blood sample were collected from 30 subjects in each group and blood fatty acid composition and immune parameters were analysed. AD patients consumed less PUFA and their n-6/n-3 PUFA ratio was higher than that of HC. Both the ratios of PUFA and MUFA were positively correlated with SCORAD in AD patients (p < 0.05). In the AD patients, there were abnormalities in the fatty acid composition of the RBC and WBC, SFA being significantly high and most n-3 PUFA being significantly low. Moreover, both the ratios of EPA and DHA in WBC were negatively correlated with dietary n-6/n-3 PUFA ratio in AD patients (p < 0.05). Serum total IgE and IL-4 levels of AD patients increased significantly compared with the levels of HC (p < 0.01). Ratios of monocyte and eosinophil in WBC of AD patients increased significantly compared with the levels of HC including total WBC count (p < 0.01), and ratios of Iymphocyte and basophil in WBC of AD patients decreased significantly compared with the levels of HC (p < 0.05). Moreover, the ratios of eosinophil in WBC were positively correlated with dietary P/M ratio (p < 0.05), and the ratios of monocyte in WBC were positively correlated with n-6/n-3 PUFA ratio (p < 0.05) in AD patients. This results indicated that AD patients had significantly high intake of dietary n-6/n-3 PUFA compared with HC. Imbalance of intake of dietary fatty acids affected fatty acid compositions in the RBC and WBC, and these lead to immune imbalance and grow worse of AD.
${\varepsilon}$-Polylysine (EPL) is used as a natural preservative in food. However, few studies have been conducted to assess the beneficial functions of dietary EPL. The purpose of this study was to elucidate the mechanism underlying the inhibition of neutral and acidic sterol absorption and hepatic enzyme activity-related fatty acid biosynthesis following EPL intake. EPL digest prepared using an in vitro digestion model had lower lipase activity and micellar lipid solubility and higher bile acid binding capacity than casein digest. Male Wistar rats were fed an AIN-93G diet containing 1% (wt/wt) EPL or L-lysine. After 4 weeks of feeding these diets, the marked decrease in serum and liver triacylglycerol contents by the EPL diet was partly attributed to increased fecal fatty acid excretion. The activities of hepatic acetyl-coenzyme A carboxylase and glucose-6-phosphate dehydrogenase, which are key enzymes of fatty acid biosynthesis, were enhanced in rats fed EPL diet. The increased fatty acid biosynthesis activity due to dietary EPL may be prevented by the enhancement of fecal fatty acid excretion. The hypocholesterolemic effect of EPL was mediated by increased fecal neutral and acidic sterol excretions due to the EPL digest suppressing micellar lipid solubility and high bile acid binding capacity. These results show that dietary EPL has beneficial effects that could help prevent lifestyle-related diseases such as hyperlipidemia and atherosclerosis.
To compare the effect of three kinds of n-3 fatty acids-eicosapentaenoic acid(EPA), docosahexaenoic acid(DHA) and perilla oil (PO)-on serum and liver lipid levels and fatty acid composition of liver phospholipid(PL) at low fat level(5%, w/w), 4-weeks old Sprague-Dawley rats were fed with one of five different oil diets for 4 weeks. Beef tallow(BT) and corn oil(CO) was used as control for sturated or n-6 fatty acid respectively. Se겨m concentrations of cholesterol(TC) and phospholipid(PL) were lower in PO DHA and EPA groups than in BT and CO groups. HDL-cholesterol levels were higher in CO and PO groups than in EPA, DHA and BT groups. Liver PL concentrations were higher in DHA and EPA groups than in CO, PO and BT groups, but liver TC and heal PL and TC concentrations did not show any significant difference among groups. Hepatic fatty acid composition of phosphatidylcholine (PC) and phosphatidylethanolamine (PE), two major phospholipids in liver, reflected their dietary fatty acid composition. In PC and PE, total percentage own-6 series was higher in CO group than in any other groups, and that own-3 series was higher in DHA and PO groups than in EPA, CO and BT groups. Moreover, the ratio of 20 : 4/18 : 2 was lower in PO and DHA groups than in EPA, CO and BT groups. On the contrary, the percentage of C22 : 6 was lower in EPA, CO and BT groups than in PO and DHA groups. These results revealed that n-3 series(EPA, DHA and PO) were more effective in lowering um lipids than n-6 fatty acids or saturated fatty acid. Based on the results of fatty acid composition of hepatic phospholipid, we suggest that the dietary effect of PO and DHA on antiatherogenic characteristics seems to be similar extent. In addition, the effect of EPA might not be significantly different from that of BT or CO in the view of eicosanoids production from the precursor fatty acid. These difference of hepatic fatty acid composition might come from other characteristics of dietary oil as well as the type of unsaturation, not from the carbon chain length or the degree of unsaturation of n-3 fatty acid.
This study was designed to investigate the changes in energy substrates, glucose and non-esterified fatty acid(NEFA), and fatty acid compositions in serum, following physiolgical stress in rats fed diets containing various fatty acids. Forty two Sprague-Dawley strain male rats, weighing 108$\pm$2.1g, were fed 3 different experimental diets for 4 weeks. The diets were composed of 105 fat(w/w) of either corn oil(CO;18:2 n6:57%), plant perilla oil(PO;18:3 n3:59%), or tuna fish oil(FO;20:5 n3:17%%, 22:6 n3:19%). After 4 weeks of feeding, each group wa subdiveided into (a) control, (b) 2 min swim in ice-cold water. Animals wer decapitated 20min after commencing the swim; trunk blood, brain, liver and epididymal fat pad were obtained. The levels of serum corticosterone, glucose, NEFA, triglyceride, fatty acid compositions, brain serotonin and 5-hydroxyindoleacetic acid were determined. Basal levels of corticosterone na NEFA of serum were significantly lower in fish oil fed animals than those of any other oil fed animals. Compared to either perilla oil-fed or corn oil-fed rats, cold swim stress in fish oil fed rats produced significantly smaller NEFA and larger corticosterone responses. However, there was no significant difference in basal levels of serum glucose. Stress increased serum glucose levels slightly, and the amount of increment was larger in fish oil rats than those of any other oil fed rats than those of any other oil fed rats, although all the values were normal level. Dietary fats and stress did not affect serotonin metabolism. In additions, the composition of fatty acids in serum was significantly affected by the dietary compostion of fatty acids and stress. Stress induced decreases in monounsaturated fatty acid and non-polyunsaturated fatty acid concentration in either perilla oil fed or fish group, but did not in corn oil fed group. Stress resulted in changes in fatty acid metabolism similar to that associated with essential fatty acid(EFA) dificiency, when feeding animals n-3 fatty acids in diet. In conclusion, feeding fish oil was more effective to decrease NEFA in serum than feeding perilla oil or corn oil and improved lipid metabolism, when the rats were maintained in normal or exposed to stressful environment. However, the fact that feeding diet containing n-3 fatty acids decreased EFA status under stress suggests that the requirement of n-6 PUFA should be increased in these groups.
Serum and liver lipid levels and fatty acid composition of liver phospolipids (PL) were investigated in 36 rats which consumed either one of five different dietary fats or a high carbohydrate diet for 4 weeks. As the sources of five dietary fats, concentrated cicosapentaenoic acid(EPA), fish oil (FO), perilla oil(PO), corn oil(CO) and beef tallow (BT) were provided to the rats. As a control group, cron starch (CS) replaced dietary fat. The FO group showed lower serum total cholesterol (TC), high density lipiprotein cholesterol(HDL-C) and serum PL levels than those of the CO group(p<0.05). There were no significant differences in serum TC and serum HDL-C levels between the polyunsatured fatty acid(PUFA) groups and the EPA, FO and PO groups. The CS group showed the highest level serum TC. Compared with the CS group, both the EPA and CO groups showed significantly lower atherogenic indices(AI). However, there were no significant differences in AI among different dietary fat groups. No significant differences in liver triglyceride (TG) , TC and PL levels were detected among the six experimental groups. Phosphatidylcholine(PC) and phosphatidylethanolamine(PE) composed 30-40% and 15-20% of total liver PL, respectively. The fatty acid composition of liver PC and PE reflected dietary fatty acid composition . Compared to the different dietary fat based diets used in our study, the high carbohydrate diet had the most adverse effects on serum lipid profiles. However, we can not conclude from this result that long chain n-3 PUFA diets such as the EPA and FO based diets have more beneficial effects on serum lipid profiles than n-6 PUFA diet such as the CO based diet or shorter chain n-3 PUFA diets like the PO based diet.
The effects of age and dietary fatty acid composition on prostagladin production was investigated in Sprague-Dawley strain male rats. Animals weighing 88.6$\pm$2.2g were fed 10% dietary fat(W/W, 20% of total energy). The P/S ratios of dietary lipid were three levels(0.5, 1, 2) and there were three different levels of n-6/n-3 fatty acid ratio(2, 4, 8) in each P/S ratio. The experimental period were 1 month and 12 months, respectively. The results of this study were as follows. As the age of rats increased, the plasma thromboxane B2 production increased, but aorta 6-keto prostaglandin F1$\alpha$ decreased. When a higher amount of n-3 fatty acid was fed in each P/S ratio, the relative percentage of linolenic acid and EPA in platelet increased.
This paper examines the process and evidence used to create the Dietary Reference Intake (DRI) of alpha-linolenic acid (ALA) and eicosapentaenoic acid (EPA) + docosahexaenoic acid (DHA) for Koreans. ALA (18:3n3) is an essential fatty acid, and EPA and DHA are known to have beneficial effects on cardiovascular disease risk and reduction of triglyceride levels. Various international organizations have suggested dietary recommendations for n-3 polyunsaturated fatty acids (PUFAs), including ALA, EPA, and DHA. A DRI for Koreans was established for the first time in 2020, specifically for the adequate intake (AI) of ALA and EPA + DHA. This recommendation was based on the average intake of ALA and EPA + DHA from the Korea National Health and Nutrition Examination Survey 2013-2017. For Korean infants, the AI of ALA and DHA was based on the fatty acid composition of maternal milk. Estimated average requirement and a tolerable upper intake level have not been set for n-3 PUFA due to insufficient evidence. In addition, the intake level of n-3 PUFA for prevention of chronic disease has also not been determined. Future studies and randomized controlled trials are required to establish the UL and to define the level for disease prevention.
Serum lipid levels and fatty acid composition of serum phospholipids(PL) were investigated in 24 healthy male subjects who consumed either corn oil(CO) rich In linoleic acid(LA), perilla oil(PO) rich in $\alpha$-linolenic acid($\alpha$-LNA), or canola oil acid(OA) as a major fat source fir 5 weeks. The PO and the CNO groups showed significant(P <0.05) increases in serum high density lipoprotein-cholesterol(HDL-C) levels and in ratios of HDL- C/total cholesterol(TC) compared with initial values measured at the beginning of the study. Significantly(p <0.05) increased concentrations of serum triglycerides(TG) ware observed after 5 weeks of the CO based diet compared with both its intial value and the concentration observed after 5 weeks with the PO-based diet. fatty acid composition of serum PL reflected changes in dietary fatty acid composition and metabolism. Compared with the initial levels, significandy increased contents of eicosapentaenoic acid(EPA) and docosahexaenoic acid(DHA) were observed in serum PL of the PO group and significantly increased contents of $\alpha$-LNA and EPA were observed in the CNO group. Arachidonic acid(AA) content of serum PL did not change in the CO group during the study period, although, the increase in LA was significant(P <0.05). Compared with the CO-based diet, both the PO and the CNO-based diets seem to have beneficial effects on atherosclerosis by influencing the serum lipid profile and fatty acid composition of serum PL.
This study was aimed at evaluating the lipid status of 122 pregnant women, who were attending peripheral community clinics in low-income areas and were vulnerable in terms of public health, by anthropometric measurements, estimating dietary intakes and analyzing serum lipids content and fatty acids composition. Regarding the skinfold thickness, it has increased more significantly in suprailliac than in triceps, biceps, subscapular, which means body fat has been accumulated in central parts more than in distal parts of the pregnants. It is found that intakes of energy, protein, lipid and carbohydrate are desirable while calcium, zinc and copper are low and sodium is excessive. Regarding dietary fat intake, the energy $\%$ of fat was within the same range as RDA(20%) for adults, but energy $\%$ intake from each fatty acid was usually low, especially energy % of monoun-saturated fatty acid intake was below RDA for adult women. And intake ratio of $\omega$6$\omega$3 fatty acids was desirable. Accordingly, it is thought that dietary fat intake was balanced in quality. Serum lipids concentrations were increased with the length of pregnancy and were higher as compared with non-pregnant women. Polyunsaturated fatty acids, especially $\omega$6 fatty acids, have gradually decreased with the length of pregnancy. Serum lipids content and fatty acids composition were not greatly influenced by dietary fat but serum free fatty acid level was negatively related to energy, carbohydrate, fatty acids and cholesterol intake. On the basis of the above results, it can be predicted that lipid metabolism for mother and fetus is changed by pregnant status and multilateral research on maternal nutrition in terms of areas and income levels will have to be made.
