보건소 방문 제2형 당뇨병환자의 식행동 영향 요인 분석 (Analysis of Dietary Behavior of Type 2 Diabetic Patients Visiting Public Health Center)

  • The purpose of this study was to investigate the dietary behavior of people with type 2 diabetes mellitus and to improve their quality of life through medical nutrition therapy. The subjects were 38 persons with type 2 diabetes mellitus visiting a public health center to participate in a dietary education program from Jun, 2003 to Nov. 2003 in Daegu, Korea. The interviews were tape-recorded and analyzed attitude, knowledge, and awareness of patients by focus group interview. Most of the patients were mainly dependent on drug therapy and had little experience of diet education. Barriers to dietary practice adherences were limitations in food selection, lack of will and feel of burden. Barriers to follow guidelines were lack of self-control, confliction with food habits of their family, accessibility, economical problems, fear for the change after dietary practice, food difficulties in meal distribution and difficulties for eating out. After 4 weeks of intensive nutrition education, fasting blood sugar levels were decreased and postprandial and waist circumference were significantly decreased in all patients and 26.9% of patients were under decreased oral hypoglycemic agent dosage due to improved blood sugar level. dietary knowledge of subjects were greatly improved in such items as dietary intake, saturated fat, HbA1C, ideal body weight, and waist circumference.

  • This study was aimed to investigate the relations between self-selected intake of 3 macronutrie-nts and body weight gain and body fat accumulation in male rats given three isocaloric diets differing carbohydrate protein and fat contents concurrently. Also the effect of dietary restriction was observed. Forty two male rats of Sprague-Dawley strain weighing 68.7$\pm$6.1g were randomly divided into 2 groups and were allowed to have foods from 3 different cups for 8-hours a day or ad libitum, After 12 weeksthey were decapitated and their brains were quickly removed and frozen until they were assayed for serotonin and its metabolite 5-HIAA The carcass was dried at 105$\pm$2$^{\circ}C$ and measured the contents of body water and body fat. The animals chose a moderately high and constant carbohydrate level and showed the increase of percent protein intake with age and great individual variations. Protein in the diet seemed to trigger appetite and increase food intake which resulted in higher weight gains and in more fat deposition in the body. The concentration of brain serotonin did not show any correlations with the intake of nutrients. the accumulation of body fat and the gain of body weight.

Effects of dietary onion (Allium cepa) extract supplementation on performance, apparent total tract retention of nutrients, blood profile and meat quality of broiler chicks

  • Objective: This study was conducted to investigate the effect of dietary onion (Allium cepa) extract (OE) supplementation on growth performance, apparent total tract retention (ATTR), blood profile, carcass characteristics and meat quality in broilers. Methods: Four hundred male broiler chicks (Ross 308, 3-d old) were randomly allocated to four dietary treatments for 28 d feeding trial. Each treatment has five replications with 20 birds each. Four dietary treatments were designated according to the OE supplementation levels (0 as control, 5, 7.5, and 10 g of OE per kg of basal diet respectively). On d 28, a total of 20 birds from each treatment were subjected for ATTR, serum biochemical assay, carcass characteristic and organ weight measurement. Results: Overall weight gain of OE 7.5 g/kg group was higher (p = 0.04) than control group. The ATTRs of dietary energy (p<0.01) and ether extract (p = 0.04) linearly increased with increasing levels of dietary OE. However, no difference in ATTR of dry matter and crude protein was evident. Furthermore, serum IgG concentration increased linearly (p<0.01) and quadratically (p = 0.03) with increasing OE supplementation. No differences in carcass dressing weight and amount of abdominal fat by treatments were observed. Also, the weight of organ including immune organ was not different among the treatments. The TBARS values of 10 d stored breast meat decreased linearly (p<0.01) and quadratically (p<0.01) with increasing dietary OE levels. The meat color was also affected, with lower (p<0.01) redness score in meats from OE supplemented groups. This study showed that dietary OE improved broiler weight gain presumably by increasing feed intake and ATTR of both energy and ether extract. The dietary OE increased serum IgG level and meat anti-oxidation capacity. Conclusion: This study implies that the recommended level of dietary OE supplementation could be beneficial for improving broiler performance and meat quality.

Effect of Undegradable Dietary Protein Level and Plane of Nutrition on Lactation Performance in Crossbred Cattle

  • An experiment was conducted in order to assess the effect of level of RDP:UDP ratio and level of feeding concentrate on milk yield, milk composition and nutrient utilization in lactating crossbred cattle. Twenty four medium producing (-10 kg/d, 45 to 135 days postpartum) lactating crossbred cows were divided into four groups of six animals each in a 2${\times}$2 factorial completely randomized design. The cows in group 1 were fed concentrate mixture I containing 59:41 RDP:UDP ratio (low UDP) at normal plane (LUDP+NP), in group 2 were fed low UDP ration at 115% of NRC (1989) requirements (LUDP+HP), whereas cows in group 3 were fed concentrate mixture II containing 52:48 RDP:UDP ratio (high UDP) at normal plane (HUDP+NP) and in group 4 were fed high UDP ration at 115% of NRC (1989) requirements (HUDP+HP). Green jowar was fed ad libitum as the sole roughage to all the animals. The experimental feeding trial lasted for 105 days. The total dry matter intake (DMI), DMI/100 kg body weight, DMI/kg $W^{0.75}$, digestibilities of DM, OM, CP, CF, EE and NFE and intakes of TDN and DCP did not differ significantly among the different groups and also due to both UDP level and plane of nutrition and also due to their interaction. The total dry matter intake varied from 145 g in group 1 (LUDP +NP) to 152.57 g/kg $W^{0.75}$ in group 2 (LUDP+HP) diet. However, increase in milk yield with increased UDP level and also with increased plane of nutrition was observed consistently throughout the experimental period. The average milk yield was 7.66, 8.15, 8.64 and 9.35 kg in groups 1, 2, 3 and 4, respectively and there was no significant difference in milk yield among different groups of cows. The overall daily average milk yields in cows fed with low and high UDP diets were 7.91 and 8.99 kg, respectively and at normal and higher plane of feeding the milk yields were 8.15 and 8.75 kg/day, respectively. Thus, there was 13.65% increase in milk yield due to high UDP level and 7.36% due to higher plane of feeding. The daily 4% FCM yields were 9.20 kg for low UDP diet and 10.28 kg for high UDP diet, whereas it was 9.11 kg at normal plane of feeding and 10.37 kg at higher plane of feeding. Fat yields for the corresponding treatment groups were 0.37, 0.43, 0.41 and 0.48 kg, respectively. The 4% FCM yield and also fat yield did not differ significantly among different dietary treatments and also due to UDP level and plane of nutrition, however, 4% FCM yield was increased by 11.74% with high UDP level and 13.83% with higher plane of feeding. The values for total solids, fat, lactose, solids-not-fat and gross energy contents in milk differed significantly (p<0.05) among the different groups and were significantly (p<0.05) higher in milk of cows fed LUDP+HP diet followed by HUDP+HP diet. Total solids (14.65 and 13.83%), lactose (5.44 and 4.92%), solids-not-fat (9.44 and 8.83%) and gross energy (887 and 838 kcal/kg) of milk decreased significantly (p<0.05) with increased UDP level while total solids (13.84 and 14.64), fat (4.84 and 5.36%) and gross energy (832 and 894 kcal/kg) increased significantly (p<0.05) with increase in plane of feeding. Gross and net energetic efficiencies and also gross and net efficiencies of nitrogen utilization for milk production were not significantly different among different groups and also were not affected significantly due to either UDP levels or plane of feeding. Results of the present study suggest that, increasing UDP level from 41% to 48% of CP in concentrate mixture and also increasing plane of feeding from normal (100%) to 115% of NRC requirements maintain a consistently higher milk production.

EFFECTS OF CIMATEROL ON THE GROWTH PERFORMANCE, CARCASS CHARACTERISTICS AND TISSUE METABOLISM IN BROILER CHICKS FED DIFFERENT DIETARY ENERGY

  • This study was a sequential experiment consisting if feeding trial and in vitro culture studies. Feeding was conducted by $2{\times}2{\times}2$ factorial design with two cimaterol levels (0, 0.25 mg/kg), two energy levels (3,200, 2,900 ME kcal/kg) and two sexes. In starting period (0-21 days) broilers were fed diets containing two energy level without dietary supplementation of cimaterol. During finishing period (21-42 days) cimaterol groups were fed cimaterol supplemented diets. In vitro cultures were carried out to study the cellular metabolism of protein and fat in liver and adipose tissues prepared from chicks used in feeding trials. Body weight gain was significantly improved by the administration of cimaterol to experimental diets by 2.4% (p < 0.05). Feed intake was reduced by cimaterol administration at the high energy level, but this trend was reversed at low energy level. Feed efficiency was improved by cimaterol administration and at high energy level the difference (5.7%) was significant(p < 0.05). The administration of cimaterol had no effects on percentage of abdominal fat content, giblet and neck. There was little difference in carcass yield between control and cimaterol treated group. The administration of cimaterol had no effects on nutrient metabolizability or carcass composition. The results of in vitro studies with liver tissues showed that cimaterol increased the lipolytic activities (p < 0.05) and decreased lipogenic activities (p < 0.05). In in vitro studies with acinar cell of liver tissues. cimaterol increased the amount of retained protein and decreased secreted protein at high energy level. but the trend was opposite at low energy level.

Effect of different sources and inclusion levels of dietary fat on productive performance and egg quality in laying hens raised under hot environmental conditions

  • Objective: This experiment aimed to investigate the effect of different sources and inclusion levels of dietary fat on productive performance and egg quality in laying hens raised under hot environmental conditions. Methods: A total of 480 Hy-Line Brown laying hens at 31 wk of age were randomly allotted to 1 of 5 experimental diets. The control diet contained 2,800 kcal/kg nitrogen-corrected apparent metabolizable energy with no fat addition. Four additional diets were prepared by adding 2.0% or 4.0% of animal fat (AF) or soybean oil (SO). Energy and nutrient concentrations were consistent among all diets. Diets were fed to hens for 4 weeks. Average daily room temperature and humidity were $26.7^{\circ}C{\pm}1.52^{\circ}C$ and $77.4%{\pm}4.50%$. The heat stress index was approximately 76, indicating that hens were raised under heat stress conditions. Results: Final body weight (BW) was greater (p<0.05) for hens fed diets containing 2.0% or 4.0% AF than for those fed the control diet or diets containing 2.0% or 4.0% SO. The BW gain and feed intake were greater (p<0.05) for hens fed diets containing additional AF or SO than those fed the control diet. Eggshell thickness was the greatest (p<0.05) for hens fed the control diet, but the least (p<0.05) for hens fed diets containing 4.0% SO. Egg yolk color was the greatest (p<0.05) for hens fed the control diet, but the least (p<0.05) for hens fed diets containing 4.0% SO. Conclusion: Inclusion of supplemental fat (AF and SO) in diets exhibits preventative effects on BW loss for hens raised under hot environmental conditions when energy and nutrient concentrations in diets were maintained. The effects were greater for AF than for SO. However, inclusion of supplemental fat in diets decreases eggshell thickness and egg yolk yellowness, possibly due to a reduction in Ca absorption and intake of egg yolk colorants.

Effect of dietary supplementation with Spirulina on the expressions of AANAT, ADRB3, BTG2 and FASN genes in the subcutaneous adipose and Longissimus dorsi muscle tissues of purebred and crossbred Australian sheep

  • Background: The demand for healthy, lean and consistent meat products containing low saturated fatty acid content and high quality polyunsaturated fatty acids (PUFA), especially long-chain (${\geq}C_{20}$) omega-3 PUFA, has increased in recent times. Fat deposition is altered by both the genetic background and dietary supplements, and this study aimed to assess the effect of dietary Spirulina supplementation levels on the mRNA expression patterns of genes controlling lipid metabolism in the subcutaneous adipose tissue (SAT) and Longissimus dorsi (ld) muscle of Australian crossbred sheep. Methods: Twenty-four weaned lambs belonging to four breeds under the same management conditions were maintained on ryegrass pasture and fed three levels of Spirulina supplement (control, low and high). In terms of nutrient composition, Spirulina is a nutrient-rich supplement that contains all essential amino acids, vitamins and minerals. It also is a rich source of carotenoids and fatty acids, especially gamma-linolenic acid (GLA) that infer health benefits. After slaughter, subcutaneous adipose tissue (SAT) and ld samples were subjected to mRNA extraction and reverse transcription using quantitative polymerase chain reaction (RT-qPCR) to assess the mRNA expression levels of the Aralkylamine N-acetyltransferase (AANAT), Adrenergic beta-3 receptor (ADRB3), B-cell translocation gene 2 (BTG2) and Fatty acid synthase (FASN) genes, which are associated with lipid metabolism. Results: Both low and high Spirulina supplementation levels strongly up-regulated the transcription of all the selected genes in both SAT and ld tissues (mostly in the subcutaneous adipose), but sheep breed and sex did not influence the gene expression patterns in these tissues. Conclusions: The evidence indicates that high Spirulina supplementation level resulted in a decrease in intramuscular fat content in Australian purebred and crossbred sheep due to the enhanced production of melatonin in sheep muscle tissues and strong up-regulation of mRNA expression of BTG2 in SAT which negatively affected fat deposition. In contrast, low Spirulina supplementation level strongly up-regulated the ADRB3 and FASN genes responsible for fat production. These findings are consistent with the observed phenotypic data suggesting that low Spirulina supplementation level can increase lamb production, with higher long-chain PUFA content.

Effects of dietary energy sources on early postmortem muscle metabolism of finishing pigs

  • Objective: This study investigated the effects of different dietary energy sources on early postmortem muscle metabolism of finishing pigs. Methods: Seventy-two barrow ($Duroc{\times}Landrace{\times}Yorkshire$, DLY) pigs ($65.0{\pm}2.0kg$) were allotted to three iso-energetic and iso-nitrogenous diets: A (44.1% starch, 5.9% crude fat, and 12.6% neutral detergent fibre [NDF]), B (37.6% starch, 9.5% crude fat, and 15.4% NDF) or C (30.9% starch, 14.3% crude fat, and 17.8% NDF). After the duration of 28-day feeding experiment, 24 pigs (eight per treatment) were slaughtered and the M. longissimus lumborum (LL) samples at 45 min postmortem were collected. Results: Compared with diet A, diet C resulted in greater adenosine triphosphate and decreased phosphocreatine (PCr) concentrations, greater activity of creatine kinase and reduced percentage bound activities of hexokinase (HK), and pyruvate kinase (PK) in LL muscles (p<0.05). Moreover, diet C decreased the phosphor-AKT level and increased the hydroxy-hypoxia-inducible $factor-1{\alpha}$ ($HIF-1{\alpha}$) level, as well as decreased the bound protein expressions of HK II, PKM2, and lactate dehydrogenase A (p<0.05). Conclusion: Diet C with the lowest level of starch and the highest levels of fat and NDF could enhance the PCr utilization and attenuate glycolysis early postmortem in LL muscle of finishing pigs.

Effect of Methionine Source and Dietary Crude Protein Level on Growth Performance, Carcass Traits and Nutrient Retention in Chinese Color-feathered Chicks

  • A total of 1,200 LinNan Chinese color-feathered chicks were used to study the effects of methionine source [DL-2-hydroxy-4-methylthio-butanoic acid (HMTBa) or DL-methionine (DLM)] and dietary crude protein (CP) level on growth performance, carcass traits, and whole-body nitrogen and fat retention. The trial was designed as a $2{\times}2$ factorial arrangement, including two CP levels (adequate and low) and two methionine sources (HMTBa and DL-methionine). Diets were formulated for three phases, starter (0-21 d), grower (21-42 d), and finisher (42-63 d). Chicks fed HMTBa had higher daily gain and improved feed efficiency than DLM during the grower phase (p<0.05). A significant two-way interaction was observed for growth performance during the finisher phase and overall (0-63 d). Growth performance was greater for chicks fed HMTBa than DLM on adequate-CP diets (p<0.05), but this was not observed at low-CP level (p>0.05). Chicks fed low-CP diets grew slower, used feed less efficiently during the grower, finisher phase and overall. On d 42, regardless of dietary CP levels, birds fed HMTBa had higher carcass weights, breast and thigh weights than DLM-fed birds (p<0.04). Birds fed low-CP diet had lighter carcass weights and less breast muscle, thigh muscle, and dressing percentage at the end of starter, grower and finisher phases (p<0.05). Whole body composition analyses found that birds fed HMTBa tended to contain more protein and less fat compared to those chicks fed DLM at the end of the starter phase (p<0.10). Low-CP diets increased CP concentration in the whole body at the end of the finisher phase (p = 0.05). HMTBa supplementation increased whole-body N retention rate during the finisher phase and overall (p<0.01), and low-CP diets reduced N intake and whole-body fat retention during the finisher phase and overall (p<0.05). In summary, HMTBa was better than DLM on an equimolar basis for growth performance, carcass traits, and N retention in Chinese color-feathered chicks. Low-CP diets lowered growth performance as well as carcass traits in color-feathered birds, probably due to imbalanced AA profiles.

  • 본 연구는 지방의 종류와 함량이 다른 식이의 섭취가 하지 고정 흰 쥐의 근육 내 화학 조성, PPAR${\delta}$ 및 관련 효소의 발현에 미치는 영향을 근육 종류에 따라 알아 보기 위해 수행되었다. 연구결과를 요약하면 다음과 같다. 1) 10일 간의 하지고정과 식이 중재에 의해 실험동물의 체중, 식이섭취량 및 혈청 지방산에는 유의적인 차이가 없었다. 2) 10일 간의 하지고정에 따라 근육 양은 하지 근육 종류에 따라, 각각 soleus 에서 약 57%, plantaris에서 약 39% 그리고 gastrocnemius에서 약 38%로 달리 감소되었다 (p < 0.01). 특히 soleus에서 가장 많은 감소 현상을 보였다. 이러한 감소 경향은 식이 지방의 종류 및 함량에 영향을 받지 않았다. 3) 10일 간의 하지고정에 따라 근육 내 조성 변화의 경우, gastrocnemius에서 분석한 근육 총 단백질량은 정상 다리(contralateral)와 비교하여 하지고정에 따라 유의적으로 감소 (CO, 41%; FO, 40%; HCO, 37%; HBT, 38%)하였다. Plantaris, gastrocnemius에서 분석한 근육 내 중성지방량의 경우에도, 정상다리와 비교하여 하지고정에 따라 plantaris(CO, 39%; FO, 27%; HCO, 38%; HBT, 35%)와 gastrocnemius(CO, 38%; FO, 34%; HCO, 33%; HBT, 34%) 모두에서 유의적으로 감소하였다. 그러나, 근 단백질량과 중성지방량의 감소 현상은 모두 식이군 간의 차이는 나타나지 않았다. 4) PPAR${\delta}$ 단백질은 하지고정에 의해 plantaris에서는 유의적으로 증가 (p < 0.05)하였고, gastrocnemius, soleus에서는 증가하는 경향을 나타내었다. PPAR${\delta}$ 단백질과는 반대로 PPAR${\delta}$ mRNA는 하지고정에 의해 plantaris에서는 유의적으로 감소 하였고, soleus에서는 FO군만 PPAR${\delta}$ mRNA 발현 양이 유의적으로 감소하였다. PPAR${\alpha}$의 경우 하지고정에 따라 두 근육에서 모두 영향을 받지 않았다. mCPT1 mRNA는 하지고정에 따라 soleus와 plantaris 모두에서 변화가 없었지만, PDK4 mRNA는 하지고정에 따라 plantaris에서 유의적으로 감소하였고, 반대로 soleus에서는 유의적으로 증가하였다. 5) PPAR${\delta}$ 단백질은 세 근육의 FO군에서 모두 크게 증가하는 경향을 보였지만, 식이에 따른 유의적인 차이를 보이지는 않았다. PPAR${\delta}$ mRNA의 경우, plantaris의 CO군과 비교해 FO군에서 mRNA 발현이 유의적으로 낮은 수준이었고, 다른 군에서는 유의적인 차이가 없었다. Soleus의 PPAR${\delta}$ mRNA는 CO군과 비교해 FO, HCO 그리고 HBT군 모두에서 발현 양이 유의적으로 낮은 수준을 보였다. 반면, PPAR${\alpha}$의 mRNA 발현은 두 근육에서 모두 식이에 의한 영향을 받지 않았다. mCPT1 mRNA 발현의 경우, plantaris에서는 CO군과 비교해 FO군에서 발현 양이 유의적으로 낮은 수준을 보였지만, 고지방 식이 군에서는 유의적인 차이가 없었다. 반대로, soleus에서는 CO군과 비교해 FO군에서 발현이 유의적으로 높은 수준을 보였지만, 고지방 식이 군에서는 CO군과 비교해 유의적인 차이는 없었다. PDK4 mRNA의 경우, plantaris에서는 CO군과 비교해 FO군의 mRNA 발현 양이 유의적으로 높은 수준을 보였다. 고지방 식이 군에서는 CO군과 비교해 발현 양의 유의적인 차이가 없었다. 한편, soleus에서는 FO군에서 발현이 낮은 경향을 보였을 뿐, 식이에 따른 mRNA 발현에는 모든 군에서 유의적인 차이가 없었다. 본 연구에서는 10일간의 하지고정에 의해서 모든 근육에서 유의적인 근 위축이 유발되었고, 이에 따라 근육 양의 감소와 함께 근육 총 단백질의 유의적인 감소가 나타났다. 하지만 근육 총 단백질과 중성지방의 유의적인 감소에도 불구하고, 하지고정에 의한 근육 내 중성지방 축적은 일어나지 않았다. 이는 하지고정에 의해 모든 근육에서 증가된 PPAR${\delta}$ 단백질이 하지고정 초기에 근육 내 중성지방의 축적을 억제했을 것으로 사료된다. 결론적으로, 하지고정에 따른 근 위축에 의해 근육 내 지질대사를 조절하는 PPAR${\delta}$의 mRNA 발현은 감소하였지만, ${\omega}$-3 PUFA를 함유한 어유 식이는 PPAR${\delta}$ 단백질 및 관련 효소의 발현 수준과 활성에 근 섬유별특성에 따라 긍정적인 영향을 줌으로써 근육 내 지질축적을 비롯한 근위축의 경감에 도움을 줄 수 있다는 가능성을 제시하였다.
