고불포화 지방식이가 흰쥐의 Plasma High Density Lipoprotein Cholesterol 량과 혈청 및 조직 내의 지방성분에 미치는 영향 (Effects of Dietary Polyunsaturated Fat on HDL - Cholesterol, Total Cholesterol and Triglyceride in Plasma and Tissues of Adult Rats)

  • The study was designed to observe the effects of dietary intake of three different levels of polyunsaturated to saturated fatty acid (P/S) ratio on plasma HDL - cholesterol and triglyceride in plasma and tissues of adult rats. Rats were assigned into the three diet groups, composed of protein 24%, carbohydrate 46%, and fat 30% of total caloric intake. However, the P/S ratio of fat was adjusted by using beef tallow, corn oil and perilla oil to give 0.2, 1.1 and 6.0. All groups were fed ad libitum for 4 week. Plasma cholesterol level was decreased but net significantly with increasing P/S ratio of dietary fat, and showed significant negative correlation coefficient. Plasma TG level was also decreased with increasing PUFA level of dietary fat. However, the effect of P/S ratio (6.0) was not greater than that of P/S ratio (1.1) on plasma TG concentration. This implied that high level of PUFA was not required to change plasma TG level as much in the case of HDL-cholesterol. Plasma HDL-cholesterol was significantly increased only in the group of P/S ratio 6.0 which suggested that high level of dietary PUFA was required to increase HDL level. There was a decreasing trend in the levels of total cholesterol and TG per g liver with increasing P/S ratio of dietary fat. There was no effect in the levels of TG per g skeletal muscle by P/S ratio but the level of cholesterol per unit skeletal muscle was rather increased with increased P/S ratio.

  • The study was to compare the effect of dietary fatty acids on fatty acid profile in tissue and the status of tocopherol and lipid peroxidation, and superoxide dismutase and glutathione peroxidase activities at two fat levels. Male Sprague Dawley rats weighing average 350g(17 weeks) were fed either low fat(LF, 4.3% w/w, 10% kcal) or high fat(HF, 20.8%, w/w, 40% kcal)diet for 6 weeks. The fats used were beef tallow as a source of saturated fatty acid, corn oil for n-6 linoleic acid, perilla oil for n-3 $\alpha$-linolenic acid and fish oil for n-3 eiocosapentatenoic acid(EPA) and n-3 docosahexaenoic acid(DHA). Palsma tocopherol was significantly reduced by fish oil compared to beef tallow at body fat level. However, there was no significant effect on the levels of plasma MDA, RBC MDA and tocopherol, and RBC hempolysis by the type and amount of dietary fat. The peroxidizibility index of fatty acid profile in plasma and liver was increased and liver MDA level was significantly increased by fish oil when dietary fat level was increased. The activities of SOD and GSHPx tended to be increased by perilla oil and fish oil at both fat oil significantly reduced the incorpration of c20:4 and increased the incorporation of c20:5 into liver compared to corn oil. The incorporation of n-3 fatty acids into tissue by perilla oil rich in $\alpha$-linolenic acid was significantly higher tan corn oil and its effect was improved with higher amount of perilla oil in diet by high fat diet. Overall, the lipid peroxidation of tissue could be prevented by tocopherol supplementation when dietary fat level was low in diet. However, at high fat diet, tocopherol supplementation might not be enough to prevent the lipid peroxidation in tissue since the potential for lipid peroxidation was tended to be increased with higher incorporation of higher unsaturated n-3 fatty acids into tissue. Therefore, it could not be recommended to consume large amount of fish oil even with excess amount of tocopherol supplemented to the high fat diet.

  • This study was designed to observe the effects of both control and atherogenic diets on the cholesterol and triglyceride (TG) in serum and liver of adult rats fed diets supplying two levels of dietary fat and two different sources of dietary protein in early life. For the first experimental period, the rats were assigned into the four diet groups: High fat, casein (HC); High fat, gluten (HG); Low fat, casein (LC): Low fat, gluten (LG). Each group was subdivided into control and atherogenic groups for the second experimental period. Cholesterol and TG were determined in serum and liver after 7 hr fasting. The body weight gain was greater in the rats of the casein groups than those of the gluten groups tut not influenced by the level of the dietary fat. The difference in body weight from the quality of dietary protein in the first period was not disappeared even after the second period. After the first period, higher serum cholesterol was observed in the rats fed either casein or high fat diets. With the second experimental diet, rats fed atherogenic diet showed higher serum cholesterol concentration but lower serum TG levels compared to those fed control diet, regardless the diets fed in the first period. Serum cholesterol level of the rats of both groups which had been fed high fat diets in early life was increased compared to those of the low fat diet groups. This effect was more pronounced with the atherogenic diet groups than control groups. However, no differences were found in serum cholesterol levels resulted from the different types of dietary protein fed in the first period. Serum TG concentration was not influenced by the quality of protein and level of fat in the diet but seemed to be mere affected by the amount of carbohyrates in the diet. Liver cholesterol per unit weight was greater in the gluten diet groups than in the casein groups but total cholesterol was higher in casein fed rats. There were no differences in liver TG among the groups.

중년기 성인의 식습관과 영양 및 건강상태에 관한 연구 (A study on the relationship between dietary habits and health status of the middle-aged adults)

  • This study was performed to investigate the dietary habits, nutrient intake, and health status and to explore the relationship between dietary habits and health status in the middle-aged men and women. The subjects consisted of 220 men and 220 women aged 40-59 years old. Dietary habits and nutrient intake were assessed by self-administered questionnaire and 24 hours dietary recall method by personal interview. Percent of body fat, serum cholesterol, and serum triglyceride level were measured. The middle aged adults who had breakfast and meals irregularly were more than 60% of the study subjects. The middle aged men had meals habits significantly more irregularly than the middle aged women. The middle aged men had high fat meat intake more frequently than the middle aged women. The middle aged men had significantly high energy and protein intake more than the middle aged women but the middle aged women had significantly high carbohydrate intake more than the middle aged men and tended to consume milk higher than the middle aged men. The middle aged adults who had high body fat tended to overeat food, to have high fat meat and processed food intake more frequently than those who had low body fat. The middle aged adults who had high serum cholesterol and triglyceride level tended to overeat food and to have processed food intake more frequently than those who had low serum cholesterol and triglyceride level. The middle aged adults who had regular meals habits. tended to have low serum triglyceride level than those who had irregular meals habits.

Effect of dietary soluble fiber on neurohormonal profiles in serum and brain of rats

  • This study was conducted to investigate the effect of dietary soluble fiber administration and/or high fat diet on serum and brain neurohormonal profiles, adipose tissue mass and body weight gain in Sprague-Dawley rats. Four groups of rats were respectively fed 10% fat diet (C), 10% fat plus pectin diet (P), 20% fat diet (HFC) and 20% fat plus pectin diet (HFP) for 4 weeks. In HFP group, the food and energy intake, body weight gain, FER including fecal excretion were the smallest (p<0.05). Serum HDL-cholesterol, triglyceride and glucose level were also the lowest in HFP group (p<0.05). The weight of brain, epididymal fat pad and adrenal gland except liver didn't show any significant differences among groups. Interestingly serum norepinephrine concentration of HFP group tended to be higher, but dopamine concentration tended to be lower than those of HFC group. However serum catecholamine concentration didn't show any significant differences among all groups. Norepinephrine and epinephrine contents of right portion of midbrain of P and HFP groups were remarkably lower than those of the C group. These results suggested that soluble fiber pectin consumption might affect neurohormonal profiles in serum and brain according to dietary fat level.

지방과 콜레스테롤 섭취 제한이 고콜레스테롤혈증에 미치는 효과 (The Effects of Lowering Dietary Fat and Cholesteol on Hypercholessterolemic Men)

  • In this study, we investigated the effects of lowering the fat and cholesterol in the diets of 26 Korean hypercholesteolemic men($\geq$240mg/dl). They consumed 2378kcal/day with 20.9% of the energy deriving from fat, and a cholesterol intake of 282mg(118mg/1000kcal). The experimental diet consisted of 2400kcal, the same as their usual diet, but the fat content was restricted to 15%, and the cholesterol level to below 100mg/1000kcal. The subjects kept to this diet for four weeks and were asked to maintain their usual life activities during the experimental period. The dietary intake and levels of plasma lipid, lipoprotein-cholesterol and apoprotein of the subjects were analyzed before, two weeks into, and after four weeks dietary intervention. After two of the dietary intervention, there were no significant changes of plasma total cholesterol or triglyceride levels but there was some changes of phospholipid level. However, after four weeks, the levels of plasma total cholesterol, triglyceride, and phospholipid had decreased significantly: 18.2%, 32.9%, and 11.9%, respectively. And the LDL-cholesterol and VLDL-cholesterol levels also showed a marked reduction of 18.1%, and 33.0% respectively without change of HDL-cholesterol level. There were no changes in the levels of Apo-A 1, Apo-B, or Lp(a). The changes of the plasma lipid levels were significantly associated with the changes in dietary fat intake but not the cholesterol intake. In conclusion, although the responses to the dietary intervention varied among the individual subjects, the lowering of dietary fat component from 21% to 15% of energy intake seems to be an effective way to reduce plasma cholesterol and triglyceride levels without decreasing HDL-cholesterol level. It was also found that the restriction of dietary cholesterol to below 100mg/1000kcal not seem to be effective for the hypercholesterolemic patient who already consuming below 300mg/day of cholesterol.

  • The adipose tissue hormone leptin has been proposed to be involved in the regulation of flood intake and energy expenditure via thermogenesis by uncoupling protein(UCP) in brown adipose tissue(BAT). The objective of the study was to examine the effects of high fat diet on the serum leptin levels, BAT UCPl expression and the body fat mass in rats after weaning. During experimental period of 12 weeks, 4 male Sprague-Dawley rats were killed for the baseline experiment at 4 weeks of age while the remaining rats were fed the two different diets: the control diet AIN-76A(n = 20), high fat(beef tallow) diet(n = 20) ad libitum, which provided 11.7% or 40% of calories as fat, respectively. At 16 weeks of age, the increase in the food efficiency ratio(FER) was related to fat mass in rats on high fat diet. Serum leptin level was increased by age and dietary high fat. There was no difference in serum insulin level between groups until 10 weeks of age, but rats fed high fat diet for 12 weeks showed hyperinsulinemia. The amount of body fat pads was increased significantly in high fat group compared to normal diet group. Visceral fat mass affected acutely by high fat diet, as a result, it was higher in rats fed high fat diet for 2 weeks than normal diet. At 16 weeks of age, BAT and visceral fat mass were significantly high in high fat group. Also, the serum leptin levels reflected the amount of body fat mass. BAT UCPI mRNA expression increased with age and dietary high fat. This study demonstrates that dietary high fat increased serum leptin levels, BAT UCPI expression and body fat mass. Futhermore, in rats fed high fat diets, the increases in leptin and UCPI expression counteracts only in part the excess adiposity and obesity.

Anti-Obesity and Hypolipidemic Effects of Dietary Levan in High Fat Diet-Induced Obese Rats

  • We found previously that dietary high fat caused obesity, and levan supplementation to the regular diet reduced adiposity and serum lipids. In the present study, we examined the effects of levan [high-molecular-mass $\beta$-(2,6)-linked fructose polymer] supplement on the development of obesity and lipid metabolism in rats fed with high-fat diet. Thus, to determine whether the dietary levan may have the anti-obesity and hypolipidemic effects, 4-wk-old Sprague Dawley male rats were fed with high-fat diet for 6 wk to induce obesity, and subsequently fed with 0, 1, 5, or 10% levan supplemented high-fat diets (w/w) for another 4 wk. For the comparison, a normal control group was fed with AIN-76A diet. Supplementation with levan resulted in a significant reduction of high-fat-induced body weight gain, white fat (i.e., epididymal, visceral, and peritoneal fat) development, adipocyte hypertrophy, and the development of hyperinsulinemia and hyperlipidemia in a dose-dependent manner. Serum triglyceride and free fatty acid levels were greatly reduced by levan supplementation. Serum total cholesterol level was reduced, whereas the HDL cholesterol level was increased by dietary levan. The expression of uncoupling protein (UCP) was increased by dietary high fat, and was further induced by levan supplementation. The mRNA level of UCP1, 2, and 3 in brown adipose tissue (BAT) and UCP3 in skeletal muscle was upregulated in rats fed with dietary levan. In conclusion, upregulated UCP mRNA expression may contribute to suppression of development of obesity through increased energy expenditure. The present results suggest that levan supplementation to the diet is beneficial in suppressing diet-induced obesity and hyperlipidemia.

Effects of Lower Dietary Lysine and Energy Content on Carcass Characteristics and Meat Quality in Growing-finishing Pigs

  • Fifty-four PIC barrows were used to evaluate the effects of lower dietary lysine content and energy level on carcass characteristics and meat quality in slaughter pigs. Pigs were allotted to one of three treatments by body weight with six replicate pens in each treatment. The dietary treatments for body weights of 20-50 kg, 50-80 kg and 80-90 kg were as follows, respectively: control diet (digestible energy 14.22 MJ/kg, lysine/DE 0.67 g/MJ, 0.53 g/MJ and 0.42 g/MJ); a low lysine group (digestible energy 14.22 MJ/kg, lysine/DE 0.49, 0.38 and 0.30 g/MJ); and a low lysine-low energy group or low nutrient group (digestible energy 13.11 MJ/kg, lysine/DE 0.49, 0.38 and 0.30 g/MJ). The daily weight gain, daily feed intake and feed efficiency were calculated in the overall growth period (nearly 12 weeks). Meanwhile, carcass characteristics and meat quality were evaluated at 60 and 90 kg body weight respectively. During the overall growth trial, lowering dietary lysine and nutrient level both decreased weight gain (p<0.05) and feed efficiency (p<0.01). At 60 kg body weight, decreasing dietary lysine and nutrient level noticeably decreased dressing percentage (p<0.01) and back fat depth at last rib of PIC pigs (p<0.01), but enhanced marbling scores (p<0.10), intramuscular fat content (p<0.10) and water loss rate (p<0.01) of the longissimus dorsi muscle. At 90 kg body weight, lean percentage (p<0.01) was evidently reduced by both lowering lysine content and nutrient level in the diet. However, the shoulder back fat depth (p<0.05) and marbling scores of the loin eye muscle (p<0.05) were increased; Lowering dietary nutrient level could improve back fat depth of 10th rib (p<0.01) and last rib (p<0.01), intramuscular fat content (p<0.10), redness (p<0.01) and water loss rate of the loin eye muscle (p<0.05), but decrease loin area (p<0.05). Finally, when comparing the 60 kg and 90 kg slaughter weights, it was found that the shoulder back fat depth (p<0.01, p<0.10), 6th-7th rib (p<0.01, p<0.01), 10th-rib (p<0.01, p<0.01) and last rib back fat depth (p<0.01, p<0.01) of the low lysine and low nutrient group were all obviously increased comparing with the control group. Taken together, the results showed that decreasing dietary lysine content and nutrient level increased intramuscular fat content and water loss rate of longissimus dorsi muscle; On the other hand, both lowering dietary lysine and nutrient level markedly compensated to increase back fat deposition in the later finishing period (body weight from 60 to 90 kg) in contrast to the control group.

  • To investigate effects of ethanol and dietary fat on growth and bichemical indices of liver tissue and blood in rats 40 male rats of Sprague-Dawley wtrain weighing about 160g were divided into 5 groups (low-fat diet group ethanol-administered low-fat diet group high-fat diet group ethanol-administered high-fat diet group and commercial diet group) and fed expe-rimental diets for 8 weeks. Ethanol-administered groups consumed ethanol corresponding to 22 cal% which was considered as moderate drinking. Neither the ethanol intake nor the dietary fat level affected calorie intake. Nonetheless the low-fat diet group with ethanol had the lowest growth rate and 2-fold increase in the concentration of plasma triglyceride. There was no effect of ethanol and dietary fat level on contents of protein lipid and lipid composition of liver tissue. The level of lipid peroxide of liver tissue tended to be increased by ethanol intake but the increase was statistically insignificatnt. The low-fat ethanol group had lowered hepatic mitochondrial respiration rate and deformed structure of mitochondria of hepatocytes.
