Silicon transporter genes of Fragilariopsis cylindrus (Bacillariophyceae) are differentially expressed during the progression of cell cycle synchronized by Si or light

  • Fragilariopsis cylindrus is one of the most successful psychrophiles in the Southern Ocean. To investigate the molecular mechanism of biomineralization in this species, we attempted to synchronize F. cylindrus growth, since new cell wall formation is tightly coupled to the cell division process. Nutrient limitation analysis showed that F. cylindrus cultures rapidly stopped growing when deprived of silicate or light, while growth continued to a certain extent in the absence of nitrate. Flow cytometry analysis indicated that deprivation of either silicate or light could effectively arrest the cell cycle of this diatom species at the G1 phase, suggesting that synchrony can be established using either factor. Fluorescence labeling of new cell walls was faintly detectable as early as approximately 6 h after silicon repletion or light irradiation, and labeling was markedly intensified by 18 h. It is revealed that the synthesis of girdle bands begins before valve synthesis in this species, with active valve synthesis occurring during the G2 / M phase. Expression profiling revealed that selective member(s) of the F. cylindrus SIT genes (FcSIT) respond to silicate and light, with a different set of genes being responsive to each factor. The Si / light double depletion experiments demonstrated that expression of one FcSIT gene is possibly correlated to transition to G2 / M phase of the cell cycle, when the valve is actively formed.

Sea Level Change during the Middle Holocene at Bibong-ri, Changnyeong-gun, Gyeongsangnam-do, South Korea (창녕 비봉리 지역의 Holocene 중기 해수면변동)

  • The remains and relics such as wood vessel, shell middens and acorn hollows related to marine environments were excavated at Bibong-ri, Changnyeong-gun, Gyeongsangnam-do, South Korea, approximately 70km apart from the Nakdong River mouth. The sea-level changes were reconstructed based on characteristics of shell middens and acorn hollows, sedimentary facies, AMS dating, and diatom analysis. The shell middens and acorn hollows were constructed during the early Neolithic Age and provide information on the paleo-sea level, because of influences of marine processes. The sedimentary facies are classified into a bedrock, base gravel and Holocene sediment (marine, terrestrial and back marsh sediments), upward. The sea level fluctuated during the middle Holocene is in harmony with those in Sejuk-ri, Ulsan and Pyeongtaek. In particular, the sea level at Bibong-ri of study area was higher than the mean high tidal level in Gimhae by approximately 1m during 5,000yr BP and maintained the stable condition during 4,000yr BP.

  • Incorporating renewable fuel into practice, especially from algae, is a promising approach in reducing fossil fuel dependency. Algae are an exceptional feedstock since they produce abundant biomass and oils in short timeframes. Algae also produce high-valued lipid products suitable for human nutrition and supplement. Achieving goals of producing algae fuels and high-valued lipids at competitive prices involves further improvement of technology, especially better control over cultivation. Manipulating microalgae cultivation conditions to prevent contamination is essential in addition to promoting optimal growth and lipid yields. Contamination of algal cultures is a major impediment to algae cultivation that can however be mitigated by choosing extremophile microalgae. This work describes the isolation of alkali-tolerant / alkaliphilic microalgae native to South Florida with ideal characteristics for cultivation. For that purpose, water samples from Lake Okeechobee were inoculated into Zarrouk's medium (pH 9-12) and incubated for 35 days. Selection resulted in isolation of three strains that were screened for biomass and lipid accumulation. Two alkali-tolerant algae Chloroidium sp. 154-1 and Chlorella sp. 154-2 were poor lipid accumulators. One of the isolates, the diatom Fistulifera sp. 154-3, was identified as a lipid accumulating, alkaliphilic organism capable of producing 0.233 g L-1 d-1 dry biomass and a lipid content of 20-30% dry weight. Lipid analysis indicated the most abundant fatty acid within Fistulifera sp. was palmitoleic acid (52%), or omega-7, followed by palmitic acid (17%), and then eicosapentanoic acid (15%). 18S rRNA phylogenetic analysis formed a well-supported clade with Fistulifera species.

Variation Analysis of Phytoplankton Communities in Northern Gamak Bay, Korea (북부 가막만의 식물플랑크톤 군집 변동 해석)

  • Using physiological data which had been already published, we investigated variation of phytoplankton communities due to changes of marine environmental factor at 2 stations of northern Gamak Bay from November 2007 to September 2008. Dominant species (occupied species above 10%) were dinoflagellates 1 species and diatom 11 species. Diatom Skeletonema costatum was observed as dominant species during the periods except in summer (i.e. July and August), especially, which occupied above 90% in December. On the other hand, dominant species during summer were diatom Eucampia zodiacus and Chaetoceros spp., Considering the results of other physiological studies, S. costatum might be non-dominant species during summer by following reasons 1) growth rate of E. zodiacus might be taster than that of S. costatum under the temperature during summer although both S. costatum and E. zodiacus are eurythermal and euryhaline species. 2) Species as E. zodiacus characterized by low affinity with light might have a chance to be dominant with increasing optical transparency due to low suspended solids in July. 3) In aspect of nutrition, species of growth strategist as S. costatum could be dominant in relatively low concentration of phosphate and species of storage strategist as E. zodiacus could be dominant in relatively high concentration of phosphate during summer in this study area. In order to understand the phytoplankton dynamics in detail, the physiological informations about strains isolated from this study area are necessary because physiological conditions are different depending on isolated area.

Spatio-temporal microalgal and environmental variations of the Hajeonri tidal flat, Gochang, Korea (한국 고창군 하전 갯벌의 미세조류 및 환경의 시공간적 변화)

  • We studied the spatio-temporal microalgal (phytoplankton and benthic diatoms) and environmental changes of the Hajeonri intertidal zone from February to November 2014. Seawater and phytoplankton analysis were conducted all through the months. The species, composition and abundance of phytoplankton and environmental factors were measured. As a result, diatom was dominant among a total of 113 species identified. On a seasonal basis (Feb. May, Aug., Nov.), we carried out studies on benthic diatoms on the surface of the sediments and their habitats at nine stations on the transect line at Hajeonri at low tide. The grain of the surface sediments was mainly composed of sandy silt and sandy mud. Organic pollution level was low. Benthic microalgal biomass (chlorophyll ${\alpha}$) was high in the spring and summer. A total of 163 benthic diatom species were identified. Navicula sp.1 and Paralia sulcata were dominant over the study period. Five diatom species were observed both in water column and on surface sediment at the same time.

Assessment of Biological Water Quality Using Epilithic Diatoms in the Upper Region of Nakdong River (낙동강 상류 수계에서 부착돌말류를 이용한 생물학적 수질 평가)

  • This study assessed biological water quality using epilithic diatoms in the Yeong river, Naeseong stream and Wi stream in the upper region of the Nakdong river from May to October 2016. Epilithic diatoms were not mobile, so they could reflect long-term water quality. The sampled epilithic diatoms were identified a total 158 taxa which were composed to 2 orders, 3 suborders, 8 families, 34 genera, 143 species and 15 varieties. Dominant species were Achnanthes convergens and Achnanthes minutissima at Yeong river, Nitzschia inconspicua at Naeseong stream, and Achnanthes minutissima, Cocconeis placentula var. lineata and Navicula minima at Wi stream. As a result of the CCA, Electrical conductivity, total nitrogen and total phosphorus were important factors determining the diatom species composition in the upper region of the Nakdong river. The correlation between diatom indices (DAIpo & TDI) measured to be high in the correlation coefficient (0.87) from the result of correlation analysis. In the result of the assessment of biological water quality using DAIpo and TDI, Yeong river was rated as class A at most sites. Naeseong stream was rated as class C to D at all sites except for N1 which was rated as Class A. Wi stream was rated as class B to C for DAIpo of W1, and TDI was rated as class D. The assessment of biological water quality at this site showed inferior TDI result compared to that of DAIpo. DAIpo and TDI of W2 were rated as class A to D, and the water quality has changed a lot. W3 and W4 were mostly rated as class B and C respectively.

  • Spatial and temporal variations of environmental factors and phytoplankton community were investigated in the three stations of Andong Reservoir in 1997 and 2003. The changes of physico-chemical water quality and phytoplanktonic biomass were higher in 2003 than that of 1997, due to rainfall difference. The concentration of total nitrogen (TN) and total phosphorus (TP) in the reservoir decreased, but total nitrogen fell relatively more between them. TN/TP ratio decreased from 109 to 90 showing no change at the downstream but a big decrease at the midand upstream. Predominant phylum of phytoplankton in Andong Reservoir were six genus that composed to Anabaena, Aphanizomenon and Microcystis of Cyanophyceae, Cosmarium and Scenedesmus of Chlorophyceae and Synedra of Bacillariophyceae, respectively. Among the observed phytoplankton, diatom Synedra occured as the maximum amount of 3,400 cells mL$^{-1}$ even at the above 30°C. Green algae Scenedesmus observed along with Microcystis. It seemed to be compete with Microcystis during the high water temperature period. Although trophic state of Andong Reservoir was decreased, the standing crops of phytoplankton were increased. Moreover bluegreen algae, Aphanizomenon and Microcystis in the region of upstream to midstream and diatom, Synedra in the region of midstream to downstream were increased until to reach the algal bloom, respectively. It seemed necessary to attention the changes of blue-green algae Aphanizomenon, that has an ability of nitrogen fixation.

Field Bioassays On Shellfish To Assess Environmental Pollution Levels Of The Masan Bay (마산만(馬山灣)의 환경오염(環境汚染) 평가(評價)를 위한 야외(野外) 생물(生物) 오염(汚染) 시험(試驗) 연구(硏究))

  • A study on field bioassay test using four species of commercially important shellfish was carried out to assess the effect of pollutants and determine the extent of marine environmental pollution of the Masan Bay from 9 to 15 August 1978. Water quality analysis and planktological examination of sea water were made during the experiment and the examination gave the following results. The water temperature was so high at 31.7$^{\circ}C$ in maximum and rather subject to change on weather condition of the land than on the effect of the water mass from outer bay. The range of DO,COD and SS at the stations were 0.3-7.08cc/l, 0.07-3.31ppm and 5.5-117ppm, respectively with the high values of COD and SS at the stations 7 and 1. The concentrations of the dissolved inorganic nitrogen in sea water, NH$\sub$4/-N, NO$\sub$2/-N, NO$\sub$3/-N, and PO$\sub$4/-P were 18.90-99.80, 2.48- 19.60, 13.00-39.00 and 1.04-14.0$\mu\textrm{g}$ at/l, respectively with decrease of their values in the outer part of the Bay. The high values mentioned above were caused by the sewage and industrial activities. The effects of organic waste are increased oxygen demand, nutrient concentration, turbidity and a higher input of pathogens, leading to structural changes in the marine ecosystems and to a considerable hazard to public health. The percentage composition of phytoplankton standing crop between diatom and dinoflagellate was characterized by making a difference between the two groups in respect of location: a decrease of diatom and a increase of dinoflagellate in numerical abundance toward inner part from outer part of the bay. Namely phytoplankton organisms were composed of 80% of diatom and 20% of dinoflagellate in outer bay, on the contrary, only 4% of diatom and 96% of dinoflagellate occupied by 94% of prorocentrum micans known as tolerant species to polluted reaas in the inner bay. On the occurrence and composition of zooplankton, there are two significant communities in the bay:one is characterized by the predominance of Oithona nana and the other by Favella sp They were composed of a range from 84% to 90% of the total organisms and monotonously constituted of themselves only at most inner station 3 even small numbers. From the results mentioned above, Oithona nana, Favella and prorocentrum micans recommed themselves as valuable indicators for judging the extent of the marine pollution.During the period of the biossays Mytilus edulis showed the highest mortality and Tapes japonica the lowest one between the four test species. The highest death rate by stations was found at most inner stations 3'and 4near Masan Free Export Zone with the most sensitive response and the lowest one occured at outer station 13 where no death specimen of oyster and arkshell was found during the whole test period.As for mussel,85 percent death rate appeared after 72 hours and 100percent rate after 120hours at station 4. It was found that the significant high mortality of the test shellfish mentioned above was caused by severe pollution with mainly organic pollutants from domestic sewage and industrial wastes from the results of too much higher concentrations of dissolved inorganic nitrogen especially ammonia-N,COD,SS and lack of dissolved oxygen,and furthermore occurrence and abundance composition of Prorcentrum,Favella and Oithona nana by stations, valuable indicator species of coastal pollution by orgnic and boilogical pollutants.

Community Dynamics of Phytoplankton in Lake Daecheong (대청호의 식물플랑크톤 군집 변화)

  • Temporal variability in lake phytoplankton is controlled by a complex between hydrological and chemical factors, and biological interactions. We explored annual change of phytoplankton in Lake Daecheong, using phytoplankton analysis data from 1997 to 2002 (except 2000). The standing crop of phytoplankton was ranged from 3.5 x 10 to 1.5 x 106 cells mL$^{-1}$ and the highest mean value was at site 1. The class composition ratios of phytoplankton standing crop were divided into three classes. From January to March, diatoms showed a dominance (68.1-77.7%). From April to June, diatoms were mixed with cryptomonad etc. or blue-green algae. From July to October, blue-green algae showed a dominance (54.7-84.0%). In the case of green algae, the class composition ratios were below 10%. But green algae appeared all the year round.

Chemical and Mineralogical Properties on the Ulsan Marine Deposited Clay (울산 퇴적 점토의 화학적, 광물학적 특성)

  • 울산지역에 널리 분포하고 있는 해성 퇴적토에 대하여 깊이별로 물리, 화학 그리고 광물학적 특성을 파악하였으며, 이 지역에서 채취된 시료에 대하여 깊이별로 퓨준압말시험을 실시하였다. 본 논문을 위하여 울산 해성 퇴적토의 생성과정을 추정하고, X-선 회절 분석, X-선 형광분석 및 에너지분산분광분석을 실시하여 깊이에 따른 변화를 비교하였다. 울산 해성 퇴적토의 점토광물에 대한 정량분석결과, 일라이트, 카올리나이트, 녹니석, 스멕타이트 순으로 많게 나타났음, 화학성분 분석결과, SiO$_2$, $Al_2$O$_3$그리고 Fe$_2$O$_3$가 대부분을 차지하는 것으로 나타났다. 주사전자현미경 관찰결과, 유공충과 규조류가 관찰되었다. 표준압밀시험 결과, 울산지역의 해성 퇴적 점토는 깊이에 따른 변형거동이 화학적, 광물학적 분석결과와 마찬가지로 깊이에 따른 변화를 보이지 않아 같은 시디에 퇴적인 것으로 판단된다.
