We examined the effect of the turbid water on the periphytic diatom community in an artificial stream system. The artificial stream was constructed with transparent acryl and composed of four channels. Each channel ($20\;cm{\times}200\;cm{\times}40\;cm$) was supplied continuously with eutrophic lake water. In order to the freely colonize and grow diatoms, artificial substrate was installed with commercial slide glass soaked in 1% agar. Prior to introducing turbid water, the artificial stream was operated with lake water for 6 days to permit the propagation of diatom community on the substrates. The turbid water prepared with sediment sieved with ${\varphi}$$64\;{\mu}m$ at $2\;g\;L^{-1}$ (final concentration, 300 NTU) was provided daily for 50 minute duration. The experiment was conducted for 7 days with manipulated experimental condition of light ($50{\sim}80\;{\mu}mol\;m^{-2}s^{-1}$, light:dark=24:0), temperature ($10{\pm}1^{\circ}C$), and flow rate ($0.31\;cm\;s^{-1}$). Sampling and analysis were conducted daily for water quality and diatom. Turbidity of the water varied 162.2~173.2 NTU during the experiment. After introduction of turbid water, DO, pH and TN were decreased, while SS and TP increased significantly. A total of 14 genera and 47 species of diatoms was observed on the artificial substrates during the experimental period. Of these, Navicula appeared to be a most dominant genus with 10 species, followed by Cymbella (6 species), Fragilaria (6 species) and Gomphonema (5 species). Achnanthes minutissima was the most dominant species (>70% of total frequency) in both control and treatment experiments. Increase in diatom abundance lasted for three days since turbid water introduction, after that they gradually decreased by the termination of the experiment. These results suggest that frequent supply of highly-concentrated turbid water significantly decreases the periphytic diatom community, and retard the recovery of the stable food-web within the stream.

The community dynamics of benthic diatoms in the hypersaline environment are investigated to advance our understanding how salinity impacts marine life. Diatoms were sampled in the two salterns encompassing salt Ponds, ditches, and seawater reservoirs (n = 11), along the salinity gradient (max = 324 psu), and nearby tidal flats (n = 2). The floral assemblages and distributions across sites and stations showed great variations, with a total of 169 identified taxa. First, not surprisingly, higher diversity of benthic diatoms was found at natural tidal flats than salterns. The saltern diatoms generally showed salinity dependent distributions with distinct spatial changes in species composition and dominant taxa. Biota-environment and principal component analysis confirmed that salinity, mud content, and total nitrogen were key factors influencing the overall benthic community structure. Some dominant species, e.g., Nitzschia scalpelliformis and Achnanthes sp. 1, showed salinity tolerance / preference. The number of diatom species at salinity of >100 psu reduced over half and no diatoms were found at maximum salinity of 324 psu. The highest salinity for the observed live diatoms was 205 psu, however, a simple regression indicated a theoretical salinity threshold of ~300 psu on the survival. Finally, the indicator species were identified along the salinity gradient in salterns as well as natural tidal flats. Overall, high species numbers, varying taxa, and euryhaline distributions of saltern diatoms collectively reflected a dynamic saltern ecosystem. The present study would provide backgrounds for biodiversity monitoring of ecologically important microalgal producers in some unique hypersaline environment, and elsewhere.
Diatoms were studied from the trench sediments around Bangudae petroglyphs in order to better understand the depositional environment before and after the construction of Sayeon dam in Ulsan. There were no diatoms produced from the sediments before the dam construction while the diatoms were produced from the sediments (depth of trench 228 cm) after the construction of the dam. Seventy-five species of diatoms of 27 genera were identified in the trench sediments. The number of diatom valves per gram of dry sediment ranged from $0.2-5.8{\times}10^5g^{-1}$. Four diatom assemblage zones were identified according to the frequency of critical taxa as follows: assemblage zone I, from 228 to 150 cm; assemblage zone II, from 150 to 122 cm; assemblage zone III, from 122 to 62 cm; and assemblage zone IV, from 62 to 0 cm. In addition, based on the environmental indicator species, an analysis was carried out to measure eutrophication, acidity and $Cl^-$ value. Results of the eutrophication and $Cl^-$ values were as follows. Based on the lower 74 cm horizons, the degree of eutrophication middle-high to $Cl^-$ values were lower, upper horizons appeared to eutrophication in the low, and $Cl^-$ values were high. Acidification from low horizons of 122 cm showed a neutral-alkaline degree whereas it exhibited acid in the upper part. In particular, regarding nutrients (TP and TN), the index taxa showed a higher TP value at 175 cm while higher TN value at 62 cm.

Amphora Ehrenberg ex Kutzing sensu lato is a common and widespread benthic diatom genus with a taxonomy that has been under continual revision, particularly based on molecular analyses. Although Amphora species have been studied using modern microscopy in recent years, there has not been much progress on molecular characterization of the species, especially in Asia. In this study of Amphora, sampling was carried out from September 2009 to August 2010 in Korean coastal waters. The morphological and molecular characteristics of eight Amphora sensu lato were examined: Amphora marina, A. proteus, Halamphora costata, H. coffeaeformis, H. eunotia, H. holsatica, H. terroris, and Halamphora sp. Based on previous accounts, morphology suggested that A. marina and A. proteus belong to the subgenus Amphora Cleve, which have smooth girdle bands and rather coarse and very distinct areolae on the valve. The other species, H. coffeaeformis, H. costata, H. eunotia, H. holsatica, H. terroris, and Halamphora sp. belong to the subgenus Halamphora Cleve, which was recently elevated to generic status by Levkov 2009, have plicate girdle bands, puncta which do not form straight longitudinal lines, valves which have a narrow ventral portion and apices that are generally rostrate-capitate and recurved. In agreement with analysis based on morphological characteristics, phylogenetic analysis based on small subunit rDNA suggested that the eight Amphora sensu lato species were not a monophyletic group as the morphological classification. Also, the results of molecular work and statistical analysis on all these Amphora sensu lato combined with phylogenic analysis on our geographically representative samples give strong evidence that Halamphora Levkov is independent of Amphora Cleve. Furthermore, in this study, Amphora terroris was transferred Halamphora as Halamphora terroris (Ehrenberg) Wang comb. nov. and Amphora marina was recorded for the first time in Korea.
The genus Chaetoceros provides highly diversified diatoms in marine systems. Morphological descriptions of the genus are well-documented, yet the DNA taxonomy of Chaetoceros has not been satisfactorily established. Here, the molecular divergences of the 18S-28S rDNA of Chaetoceros were assessed. DNA similarities were relatively low in both 18S (93.1 $\pm$ 3.9%) and 28S rDNA (81.0 $\pm$ 4.6%). Phylogenies of the 18S, 28S rDNAs showed that Chaetoceros was divided according to individual species, clustering the same species into single clades. Statistical analysis with corrected genetic (p-) distance scores showed that nucleotide divergence of Chaetoceros 28S rDNA significantly differed from that of 18S rDNA (Student's t-test, p < 0.05). This finding suggests that the 28S rDNA may be treated as a more suitable marker for species-level taxonomic distinctions of Chaetoceros.

Surirella atomus Hustedt 1955, surirelloid diatom, were obtained from Hampyeong Bay, Mu-an, in the west coast of Korea. We describe the first record of S. atomus in Korea and report its ultrastructure and molecular characteristics for the first time. For molecular analysis, rbcL gene and cob gene were sequenced. This species is very small in size, making it difficult to observe its morphological features under a light microscope. Even with a scanning electron microscope, it has no unique structure in the valve. It is morphologically very simple. This rare species was known as a marine benthic species. However, it appears to be widely distributed in marine, freshwater, and brackish environments according to the literature documenting this species. In terms of phylogeny, S. atomus has been tentatively linked to Petrodictyon gemma, but the phylogenetic placement of S. atomus seems to be still uncertain, in a manner that is similar to S. febigeri. However, morphological observations derived from this study suggest that this species belongs to the genus Surirella.
The sedimentary environments and biostratigrapy of the tidal sediments off the Mankyung-Dongjin River estuary were studied based on sedimentary facies and diatom assemblage analysis. Sediment facies from the five vibracores are mainly clay, silt, and sand facies. The clay and silty sediment facies are more dominant than the sandy facies, and contain diatom frustules. The frequency of the diatom frustules are rate to common, but not found in sandy sediment facies. Bigeneric structures such as burrow and non-bigeneric primary sedimentary structures such as laminated sand and mud or silt and mud couplets, flaser bedding, ripple-cross lamination are found in several stratigraphic levels of the sedimentary sequences. A total of 219 species and varieties, belonging to 61 genera has been identified in the present study. Among them, paralia sulcata is the most abundant species about 30 to 50% of the total diatom frustules. Another predominant species are Cyclotella striata. Thalassionema nitzschioides. Actinoptychus undulatus. Delphineis surirella, Raphoneis amphiceros. Most of the diatoms occurred in this study area are marine, marine-brackish water, and brackish water species, and are benthic and tychopelagic or meroplanktonic species. Also, most of the species are coastal to littoral and littoral to inner neritic species. The occurrences of freshwater species, about 1 to 5% is higher than that of the Namyang Bay tidal sediments. The ecological properties of the diatoms occurred in the study area and primary sedimentary structure such as flaser bedding ripple cross bedding indicate that the deposits are formed under coastal or littoral to subeditorial environments such as tidal zone which was subjected to the influenced of stream water and was more strongly influenced by temperate to warm water than cold water. The sedimentary environments have not been changed distinctively during the time of deposition. The vertical distribution pattern of diatoms in the study area is studied by Q-mode cluster Analysis using spss/pc+ (ver.4.0). The results show that the three cores (GE-3, GE-11, GE-12) are divided into two diatom assemblages, respectively. Biostratigraphic correlation using the data of Q-mode Cluster Analysis are attempt in this study.

Environmental factors and epilithic diatom communities in the lower streams near the harbor region of South Korean peninsula were examined during no monsoon period in May 2013. The sampling of water and epilithic diatoms was conducted at both streams, 19 regulated streams (RS) that there are one or several dams constructed in the river system, and 19 un-regulated streams (US) that there are no dams within the river. A cluster analysis based on the number of species and abundance of epilithic diatoms through the stations, divided into three groups such as groups I (mainly US), II (mixed with US and RS) and III (mainly RS), respectively. Group I showed that water quality is good and high diversity of diatom, while Group II and III was water quality is relatively poor, but not differed in biomass of diatom from Group I. In addition, Group II that had high conductivity, nitrogen and phosphorus, was the lowest in diatom diversity among them. Dominant species were Nitzschia palea (17%) and Navicula seminuloides (11%) in Group I, Nitzschia inconspicua (19%) and Navicula perminuta (9%) in Group II, and Nitzschia inconspicua (15%) and Nitzschia palea (14%) in Group III, respectively. These taxa were widely distributed in brackish water, and not closely related with specific water quality, like eutrophic water. However, the groups II and III belonged to RS, had not only little biomass, but bad water quality such as high concentrations of nutrient and chlorophyll-a. Therefore, to determine the effect of dam construction on the lower water ecosystem, the planktonic algae, which can occur algal bloom in the estuary, also was considered to be a parallel investigation.
The purpose of this study was to analyze the habitat status and species diversity of benthic diatoms and estimate the applicability of TDI (Trophic Diatom Index) to obtain the basic data for the identification and management of created wetlands in the Nakdong River. We observed a total of 38 families and 173 species of benthic diatom during the survey period, and spring and autumn showed a similar number of species of 156 and 154, respectively. The result of the SOM (Self-Organizing Map) analysis showed that the distribution of benthic diatom was sensitive to environmental factors such as nutrient concentration and rainfall in each wetland. The cluster 1 was characterized by the survey sites of autumn mostly and consisted of points of high TDI, although the nutrients such as total phosphorus and total nitrogen were low, and the species number and abundance of diatoms were low. Conversely, cluster 4 was characterized by the survey sites of spring mostly and consisted of points of low TDI, even though total nitrogen was high. Considering that most of the created wetlands had the reduced inflow and outflow, the increased flow rate in the summer lowers nutrient values in autumn, and the species number and abundance of benthic diatom decreases due to the increase of turbidity, which reduces the light penetrations to the substrates. On the contrary, the TDI value is low in spring because the low water level causes insufficient substrate surface to the benthic diatoms, and it is too early for the establishment and development of saprophilous species. Although various studies have used TDI as an indicator for evaluating the habitat environment and water quality, it is not a good evaluation indicator in this study since the nutrient concentration in the wetlands mostly high as they have a low flow rate and are close to the stagnant area. Nevertheless, additional periodic surveys that comprehensively reflect the fact that the summer rainfall and inflow/outflow regulating function might affect the species diversity and distribution of benthic diatoms are necessary.

Red tide has caused massive fish kills in Korean coastal waters with devastating economic loss in the aquaculture industry since 1995. Remote sensing technique has shown to be effective for the detection of red tide in wide areas, where the absorption property of red tide water plays a central role in understanding the red tide reflectance. This study analyzed the optical absorption property of sea waters dominated by the dinoflagellate specie of Alexandirum affine, off the Tongyeong area in August, 2017. Water samples collected from 20 stations in the ship-based campaign were measured for absorption by pigment, suspended solid, and dissolved organic matter, with the corresponding water quality variables such as chlorophyll concentration and total suspended solid. The analysis showed that Alexandrium-dominated water exhibits strong absorption in the spectral range below 400 nm unlike that of diatom-dominated waters, and greater fluctuations in the range of 400 nm - 500 nm. The packaging effect in pigment absorption was stronger in Alexandrium-dominated waters, and the exponent in the absorption by detritus and gelbstoff is disparate for diatom and Alexandrium. In the model for the detritus and gelbstoff absorption (adg(λ)=adg(λ0)e-s(λ-λ0)), the optimal exponent coefficient(s) for the Alexandrium was close to 0.01 rather than to 0.015, which was commonly use for modelling diatom waters.
