Streptozotocin-유도 당뇨모델을 이용한 배양인삼 분말의 혈당 강하 및 지질대사의 개선효과 (Effects of Tissue Cultured Ginseng on Blood Glucose and Lipids in Streptozotocin-Induced Diabetic Rats)

  • 본 연구에서는 streptozotocin-유도 당뇨쥐를 이용하여 배양인삼의 첨가식이가 혈당강하 및 지질대사 등에 미치는 영향을 조사하기 위해 대조군과 당뇨대조군, 5% 및 10% 배야인삼첨가군 등 모두 4군으로 나누어 해당식이로 2주간 사육하였다. 배양인삼은 당뇨쥐의 체중증가와 혈당저하에 효과를 나타냈으며, 수분 및 사료 섭취량도 줄여 당뇨병 증세의 개선효과를 보였다. 또한 당뇨쥐의 지질대사를 개선하여 혈장중성지방, 총 콜레스테롤 및 동맥경화지수를 저하시키고 총 콜레스테롤에 대한 HDL-콜레스테롤의 비율을 증가시켰다. AST 및 ALT의 활성은 5% 첨가군에서만 당뇨대조군에 비해 낮은 값을 보였으며, 10% 첨가군에서는 오히려 높은 값을 보였다. Creatinine의 함량은 모든 첨가군에서 당뇨대조군에 비해 현저히 낮은 값을 보였다.

  • This study investigates the effects of a raw diet (RD) on blood glucose and lipid metabolism in non-diabetic (normal) and streptozotocin (STZ)-induced diabetic rats. Male Sprague-Dawley vats were assigned to four groups (normal control, normal RD, diabetic control, and diabetic RD), for the four-week experimental period. The control groups were fed the AIN diet and the RD groups were fed a diet consisting only of raw materials. Weight gain was statistically lower in the RD group than the control. fasting plasma glucose was significantly lower in the diabetic RD group compared to the diabetic control group. The levels of triglycerides (TG), and of total cholesterol (TC) and LDL-cholesterol in the plasma, were lower in the RD groups than the control groups, but not significantly. There was a statistically significant decrease in the levels of TG and TC in the livers of the diabetic RD group, compared to the diabetic control group. The fecal levels of total lipids, TG, and TC were significantly higher in the RD groups, compared to the non-RD groups. It can be postulated that this raw diet may possess substantial hypoglycemic/hypolipidemic properties in diabetic rats.

  • Diabetic cataract is a major complication of diabetes mellitus. Excess accumulation of sorbitol plays an important role in the pathogenesis of diabetic complications such as cataract formation. In this study, we investigated the inhibitory effect of the extract of the aerial parts of Aster koraiensis (AK) on diabetic cataractogenesis. To examine this further, we evaluated sorbitol accumulation during cataract development using streptozotocin-induced diabetic rat, an animal model of type 1 diabetes. Diabetic rats were treated orally with AK (100 mg/kg and 200 mg/kg body weight) once a day orally for 9 weeks. In vehicle-treated diabetic rats, lens opacity was increased, and lens fiber swelling and membrane rupture were observed. In addition, sorbitol accumulation in diabetic lens was markedly enhanced. However, AK treatment delayed the progression of diabetic cataract through the inhibition of sorbitol accumulation, and prevented lens fiber degeneration in a dose-dependent manner. These observations suggest that AK treatment can delay the progression of lens opacification in the diabetic rats during the early diabetic cataractogenesis.

당뇨쥐의 간 Microsome에서 Phopholipase A_{2} 활성과 지질과산화 (Phospholipase $A_{2}$ Activity and Lipid Peroxidation in Liver Microsome of Streptozotocin Induced Diabetic Rats)

  • The purpose of this study was to investigate phospholipase $A_{2}$ activity and lipid peroxidation I streptozotocin induced diabetic rats. Sprague-Dawley male rats weighting-Dawley male rats weighting 300$\pm$10gm were randomly assigned to normal and STZ-induced diabetic group. Diabetes was induced by intravenous injection of 55mg/kg of STZ in sodium citrate buffer(pH 4.3). Animals were sacrificed at the 6th day of diabetic states. Body weight gains were lower in DM group. Phosphatidylcholine hydrolysis in liver was not significantly different between two groups, whereas phosphatidylethanolamine hydrolysis in liver was increased by 69% in DM group comparing with that of normal group. Liver microsomal phospholipase $A_{2}$ activity and level of TBARS was increased by 91%, 109% in DM group compared with that of normal group, respectively. The present results indicate that phospholipase $A_{2}$ activity is specific to PE hydrolysis, leading to lipid peroxidation process in STZ induced diabetic rats.

Hepatoprotective and Anti-diabetic Effects of Pelvetia siliquosa, a Marine Algae, in Rats

  • The effects of various fractions from the whole plant of Pelvetia siliquosa Tseng et Chang (Fucaceae) on the $CCl_4$-induced hepatotoxicity as well as on streptozotocin (STZ)-induced diabetes in rats were investigated. The ether fraction exhibited a potent rat lens aldose reductase (RLAR) inhibition in vitro and showed a significant inhibition of not only serum glucose concentrations but also sorbitol accumulations in the lens, red blood cells and sciatic nerves in the STZ-induced diabetic rats. When administered orally in Sprague-Dawley rats, $H_{2}O$ fraction was found to cause a significant inhibition of the rise in the serum transaminase activities in $CCl_4$-intoxicated rats. These results suggested that this plant might possess constituents with hepatoprotective, anti-diabetic effects and those effects on diabetic complications.

  • To evaluate the hypoglycemic effect of nano powder propolis, streptozotocin (STZ) induced diabetic rats were divided into 2 groups : diabetic control group and nano powder propolis (0.9ml) group. Then the rats were fed with nano-powder-propolis for 4 weeks. After 4 weeks, oral glucose tolerance test (oral GTT) was performed and blood sugar levels after 16 hours fasting, body weights, and blood lipid levels were measured. Finally, pancreas were collected and examined by histopathology and immunohistochemistry. In conclusion, the nano-powder-propolis was effective in the treatment of diabetes due to the reduction of blood sugar level and the regeneration of damaged ${\beta}$-cells observed in streptozotocin-induced diabetic rats.

Characterization of $ET_B$ Receptor-mediated Relaxation in Precontracted Mesenteric Artery from Streptozotocin-induced Diabetic Rats

  • Diabetes mellitus is associated with vascular complications, including an impairment of vascular function and alterations in the reactivity of blood vessels to vasoactive substances in various vasculature. In the present study, the authors have observed endothelin-B ($ET_B$) receptor agonist-induced relaxation in precontracted mesenteric arterial segments from streptozotocin (STZ)-induced diabetic rats, which was not shown from control rats or in other arterial segments from diabetic rats. Accordingly, the goal of this study was to investigate in what way STZ-induced diabetes altered reactivity of the mesenteric arterial bed and to examine the causal relaxation, if any, between this $ET_B$ receptor-mediated relaxation and endothelial paracrine function, especially nitric oxide (NO) production. The relaxation induced by $ET_B$ agonists was not observed in mesenteric arteries without endothelium. The relaxation to $ET_B$ agonists was completely abolished by pretreatment with BQ788, but not by BQ610. $N_{\omega}-nitro-L-arginine$ methyl ester and soluble guanylate cyclase inhibitors, methylene blue or LY83583 significantly attenuated the relaxant responses to $ET_B$ agonists, respectively. When the expression of eNOS and iNOS was evaluated on agarose gel stained with ethidium bromide, the expression of eNOS mRNA in diabetic rats was significantly decreased, but the expression of iNOS was increased compared with control rats. Furthermore, the iNOS-like immunostaining was densely detected in the endothelium and slightly in the arterial smooth muscle of diabetic rats, but not in control rats. These observations suggest that $ET_B$ receptor may not play a role in maintaining mesenteric vascular tone in normal situation. However, the alterations in $ET_B$ receptor sensitivity were found in diabetic rats and lead to the $ET_B$ agonist-induced vasorelaxation, which is closely related to NO production. In the state of increased vascular resistance of diabetic mesenteric vascular bed, enhanced NO production by activation of iNOS could lead to compensatory vasorelaxation to modulate adequate perfusion pressure to splanchnic area.

청폐사간탕(淸肺瀉肝湯)이 Streptozotocin으로 유발(誘發)된 흰쥐의 실험적(實驗的) 당뇨(糖尿)에 미치는 영향(影響) (Effects of Chungpyesagantang on the Diabetic Rats induced by Streptozotocin)

  • In order to study the effects of Chungpyesagantang(淸肺瀉肝湯) on the diabetic rats induced by streptozotocin, during 15 days rats were administered Chungpyesagantang extract after streptozotocin injection (50mg/kg) (sample group). On 2nd, 9th, and 15th day, I investigated the levels of body weight, serum glucose, serum total cholestrol, serum triglyceride. The last day, I killed rats and investigated hepatic lipid peroxidase. The results were obtained as follows: 1. Body weight of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 15th day, increased effectively. (p〈0.05 respectively) 2. Glucose levels in serum of the diabetic rats induced by streptozotoc in and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 3. Total cholesterol levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively ) 4. Triglyceride levels in serum of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.05 respectively) 5. On hepatic lipid peroxidethe of the diabetic rats induced by streptozotocin and Chungpyesagantang, as compared with the control group on the 9th and 15th day, decreased effectively.(p〈0.01 respectively)

Effect of Ichnocarpus frutescens extract on antihyperglycemic, antihyperlipidemic and antioxidant status in streptozotocin-induced diabetic rats

  • The present study was carried out to investigate the antihyperglycemic, antihyperlipidemic and antioxidant effect of chloroform and methanol extract of whole plant of Ichnocarpus frutescens (CEIF and MEIF) in streptozotocin (STZ)-induced diabetic rats. Administration of CEIF and MEIF orally at the dose of 200 mg/kg and 400 mg/kg body weight resulted in significant (P<0.01) reduction in blood glucose levels. The body weights were significantly (P<0.001) reduced in STZ-induced diabetic rats when compared to normal rats while the extracts significantly (P<0.01) prevented the decrease in body weight in the CEIF and MEIF treated rats. The study was further undertaken to evaluate the antioxidant and antihyperlipidemic potential of CEIF and MEIF in STZ-induced diabetic rats. The increased levels of lipid peroxidation in the liver tissues of diabetic rats were significantly reverted back to normal levels and a significant increase in activity of antioxidant enzymes such as superoxide dismutase, catalase and the level of reduced glutathione in the liver of diabetic rats after the treatment with CEIF and MEIF was noticed. These results clearly indicate that CEIF and MEIF exhibit significant antihyperglycemic, antihyperlipidemic and in vivo antioxidant activity in STZ-induced diabetic rats and the results were found to be in a dose dependent manner.

Effects of Functional Extracts Made from Fermented Plants on Serum Glucose and Lipids Level in Streptozotocin-Induced Diabetic Rats

  • This study was performed to explore the effects of the fermented functional extracts (FE) on blood glucose and lipid levels in diabetes. FE were created by mixing 9 kinds of plants with sea water and then allowing the mixture to ferment for 1 year. FE were supplemented in the feed of streptozotocin (STZ)-induced diabetic rats at 1%, 3% and 5%. The 1% feeding group showed the lowest weight loss of the three experimental groups. The blood glucose and glycosylated hemoglobin level were significantly decreased in the FE fed rats compared to the diabetic control (DMC) group. The lipid levels in serum were decreased in 1% and 3% FE fed rats in comparison to the DMC group, and there was no significant difference in triglyceride levels due to the FE concentration. The HDL-C level was significantly increased in rats with FE supplemented diets, compared to the DMC group. The levels of lipid peroxides in liver tissue were significantly decreased in FE fed diabetic rats, and the hepatic glycogen content was increased in rats receiving supplements. As a result of these studies, we believe 1% FE may be the optimum level for controlling blood glucose and alleviating hyperlipidemia in STZ-induced diabetic rats.
