Streptozotocin 저용량 반복투여로 유도한 당뇨 흰쥐에서 구기자 추출물의 효능연구 (Effect of the Lycii Fructus on multiple low-dose streptozotocin-induced diabetic rats)

  • Objectives : This study was performed to investigate the anti-diabetic effect of the fruit extract of Lycium chinense Mill(Lycii Fructus, LF) on multiple low-dose streptozotocin-induced diabetic rats.Methods : Male Sprague-Dawley rats were divided into three groups; normal, STZ-control, Lycii Fructus extractorally administrated 300 ㎎/㎏ group (STZ-LF). Diabetes was induced in rats by consecutive injection of streptozotocin (STZ) at doses of 30 ㎎/㎏ for 5 days. After 4 weeks, all rats were sacrificed, Fasting blood glucose, total cholesterol (CHO), triglyceride (TG) and HDL-Cholesterol were measured in sera of rats. Histopathological changes of pancreas, kidney, liver and lung tissues were observed by microscope after H&E, Periodic acid-Schiff (PAS) and Masson's trichrome staining. The changes of body weight, blood glucose, and food and water intake were also measured.Results : There were no differences in body, food intake and water intake in LF-administrated groups compared with STZ control group. However, LF extract significantly decreased the levels of serum glucose, CHO, TG and HDL-Cholesterol in diabetic rats. In histopathological analysis of kidney, liver and lung, LF-administrated groups showed the inhibition of morphological damage.Conclusions : These results suggest that LF have a biological action on multi low-dose STZ-induced diabetes in rats via decreasing the serum glucose, TG and TG levels and may protect the morphological changes of kidney, liver and lung.

Effect of Compositae Plants on Protein Levels in Streptozotocin-induced Diabetic Rats

  • The investigation assessed the influence of Compositae plants consumption on the protein profile in streptozotocin (STZ)-induced diabetic rats. Diabetes was induced in Sprague-Dawley rats by injection of STZ (45 mg/kg body weight) into tail vein. The rats were randomly assigned to five groups: normal and STZ-control fed an AIN-93 diet, and groups whose diets were supplemented with 10% Compositae powder containing Artemisia iwayomogi (A. iwayomogi), Atractylodes lancea (A. lancea) or Taraxacum mongolicum (T. mongolicum). To observe the effects of Compositae plants in the animal model, the levels of protein in liver, kidney, lung, pancreas, and brain were determined after 4 weeks. The level of protein in kidney increased significantly in rats receiving the A. iwayomogi- and T. mongolicum-supplemented diet compared to the STZ-control group. The level of protein in lung was increased significantly in the A. iwayomogi-supplemented group. Blood glucose level correlated well with brain protein level but did not correlate with other protein levels. Also, blood glucose correlated inversely with kidney, lung and brain protein levels. It is suggested that supplementation with A. iwayomogi in diabetic rats leads elevates protein in kidney and lung.

인삼백호탕(人蔘白虎湯)이 Alloxan으로 유발(誘發)된 mouse의 당뇨병성(糖尿病性) 신증(腎症)에 미치는 영향(影響) (The Effects of Insambackhotang on Mouse with Diabetic Nephropathy Induced by Alloxan)

  • To investigate recovery effects of Insambackhotang, which have been used clinically in diabetes therapny, on kidney failure of a diabetes-induced mouse by Alloxan administration, body and kidney weight changes of mice, BUN, creatinine, glucose and MDA level in serum, MDA level in kidney tissue. 1. A hyperglycemia(250-400mg/dl) mouse induced by Alloxan(75mg/kg) showed significant decline of kidney function: increase of BUN and creatinine in serum, excretion of glucose, protein, ketone in urine were observed at 4 days after the treatment. 2. Increase of the mouse body and kidney weight and a ratio of the kidney/body weight of Insambackhotang treated group as compared to the control group was significantly inhibited. 3. The BUN, creatinine level in serum of Insambackhotang treated group as compared to the control group were significantly inhibited. 4. The MDA level in serum and kidney tissue of Insambackhotang treated group as compared to the control group were significantly inhibited.

AIIoxan 유도 당뇨병상태에서 과산화지질생성에 미치는 Brazilin의 효과 (Antilipidperoxidative Effects of Brazilin in Alloxan-induced Diabetic Mice)

  • Brazilin was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in liver, kidney and serum were measured by the TBA method. ICR mice receiving alloxan at a dose of 43mg/kg via the tail vein after a 24 hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in liver, kidney and serum of alloxan-induced diabetic mice were dosedependently decreased by the treatment of brazilin at a dose of 10mg/kg, 50mg/kg, 100 mg/kg for 5 days.

  • Morus alba extract(MAE) was tested for its ability to inhibit alloxan induced lipidperoxidation. Lipid peroxide contents in serum, liver, kidney and heart were measured by the TBA method. ICR mice receiving alloxan at a dose of 6mg/kg intraperitoneally after a 24hrs starvation showed significantly increased lipid peroxide contents as compared to untreated control. Lipid peroxide contents in serum, liver, kidney of alloxan-diabetic mice were decreased by the treatment of MAE at the dose of 50mg/kg, 100mg/kg for 7 days.

스트렙토조토신-당뇨쥐에서 지질과산화 및 항산화계의 경시적 변화 (Changes in in vivo Lipid Peroxidation and Antioxidant Defense System in Streptozotocin-Induced Diabetic Rats: a Time Course Study)

  • This study was carried out to examine a part of the mechanism for the etiology of diabetic complications. Thirty normal and forty streptozotocin(STZ)-induced diabetic rats were used as the animal models. The animals were sacrificed at the time points of 3 days, 1,2,4 and 6 weeks after STZ-injection and a time course changes in the concentrations of thiobarbituric acid-reactive substances(TBARS) in blood, urine, and tissues, along with the levels of conjugated dienes in tissues were measured as indices of in vivo lipid peroxidation. The activities of antioxidant enzymes, catalase, superoxide dismutase, glutathione peroxidase and the levels of blood retinol and alpha-tocopherol were also measured. The diabetic rats maintained a slightly higher plasma TBARS level throughout the experiment. The urinary TBARS level was significantly higher in diabetic group and gradually increased with time. Concentrations of TBARS in liver, heart, and kidney tissues from diabetic animals were higher than those from the normal group. An increase of conjugated dienes was also observed in the all tissues examined. The kidney tissue of diabetic animals revealed more significant lipid peroxidation state than any other organ tissues. The activities of hepatic antioxidant enzymes such as catalase, superoxide dismutase, glutathione peroxidase were higher in diabetic animals compared to the control ones and increased with the duration of diabetes mellitus. The plasma levels of vitamin A and E were loser in diabetic animals than in normal controls throughout the experimental period. The level of vitamin E in diabetic animals was significantly decreased with the duration of the disease. The results of this study suggest that an effective regimen to suppress the adverse changes in lipid peroxidation and antioxidant defense system is required from the early stage of the disease to prevent the development of diabetic complications. 당뇨쥐 신장조직의 산화적 손상과 사구체 여과율에 미치는 비타민 E의 영향 (Effects of Vitamin E on the Oxidative Damage and Glomerular Filteration Rates of Kidney in Streptozotocin-Inducd Diabetic Rats)

  • The purpose of this study was to investigate the effects of vitamin E on the oxidative damage and glomerular filteration rates(GFR) of kidney in streptozotocin(STZ) induced diabetic rats. Sprague-Dawley male rats weihing 100$\pm$10gm were randomly assigned to one normal and three STZ-induced diabetic groups which were subdivided into vitamin E free diet(DM-0E group)40mg vitamin E per kg diet(DM-40E group) and 400mg vitamin E per kg diet (DM-400E group), Vitamin E level of normal group was 40mg per kg diet. diabetes was experimentally induced by intravenous injection of 55mg/kg of body weight of STZ in citrate buffer(pH 4.3) after 4 weeks feeding of experimental diets. Animals were sacrificed at the 6th day of diabetic states. Activities of xanthine oxidase(XOD) in DM-0E DM-40 and DM-400E groups were significantly increased by 133%, 110%, and 74% respectively compared to normal group. The contents of microsomal superoxide radical(O2) in kidney were 106% and 119% higher of DM-0E and DM-40E groups than normal group respectively but that of DM-400E group was similar to normal group. The level of urnary microalbumin in DM-0E group was increased as 5 times much as normal group at the 6th day. Those of DM-40E and DM-400E groups were decreased to 16% and 36% respectively compared to DM-0E group. The content of urinary $\beta$2-microglobulin in DM-0E DM-40E and DM-400E group were increased by 268%, 181% and 163% respectively compared to normal group. GFR in DM-0E and DM-40E were significantly increased but was nor significantly different from DM-400E groups compared to normal group. In conclusion the supplementation of dietary vitamin E reduced peroxidative damage of renal glomerular and renal dysfunction in diabetic rats.

Evaluation Potential Antidiabetic Effects of Ferula latisecta in Streptozotocin-Induced Diabetic Rats

  • Objectives: The aim of the present work was to evaluate the possible beneficial effects of F. latisecta on blood glucose, lipids, and diabetes-related changes in the liver and kidney of streptozotocin-induced diabetic rats. Methods: Male Wistar rats were randomly allocated into four groups (n = 6): normal control rats, diabetic control rats, diabetic rats treated for 4 weeks with F. latisecta root (400 mg/kg/day), and diabetic rats treated with F. latisecta aerial parts (400 mg/kg/day). Results: Induction of diabetes significantly (p < 0.05) increased the levels of fasting blood glucose (FBG), triglyceride, total cholesterol, low-density lipoprotein (LDL), blood urea nitrogen (BUN), aspartate aminotransferase (AST), and alanine aminotransferase (ALT). Diabetes also increased (p < 0.05) oxidative stress in the kidney and liver (decrease of thiol and increase of superoxide dismutase). The root and aerial parts of F. latisecta significantly reduced the level of LDL (p < 0.05) and restored the content of thiol (p < 0.05) and superoxide dismutase (p < 0.01) in the kidney and liver. F. latisecta had no significant effect on the levels of FBG, BUN, AST, and ALT. The root of F. latisecta also reduced the serum level of total cholesterol (p < 0.05) and prevented the progression of hyperglycemia. Conclusion: These findings suggest that F. latisecta may improve diabetic dyslipidemia by reducing serum LDL. Further studies are needed to confirm our findings.

메밀급여가 Streptozotocin유발 당뇨쥐의 장기무게 및 당질과 지질대사에 미치는 영향 (Effects of Buckwheat on Organ Weight, Glucose and Lipid Metabolism in Streptozotocin-Induced Diabetic Rats)

  • The present study was undertaken to evaluate the effects of 50% buckwheat diet on the body weight, organ weight, urine albumin, urine glucose, plasma glucose and plasma lipid in normal rats and diabetic rats treated with streptozotocin(STZ). The food intake, body weight, the level of urine glucose in diabetic buckwheat groups were not significantly different with diabetic control group. The level of urine albumin was lower in raw and steam buckwheat group than in the diabetic control group. Compared to the normal control group, liver and kidney weights were heavier in the diabetic groups. Pancreas weight was heavier diabetic buckwheat groups than in normal and diabetic control groups. Fasting plasma glucose level of diabetic buckwheat groups significantly decreased by 18-37% compared with the diabetic control group. Plasma triglyceride level of diabetic buckwheat groups significantly decreased by 34-50% compared with the diabetic control group. Plasma total cholesterol level of diabetic buckwheat groups decreased by 15-27% compared with the diabetic control group. The level of HDL-cholesterol was not affected by buckwheat diet. These results indicate that buckwheat is an effective therapeutic regimen for the control of metabolic derangements in diabetics.

한약재 함유 대두발효 추출물이 streptozotocin으로 유발된 당뇨 흰쥐의 혈당 및 조직형태학적 변화에 미치는 영향 (Effect of Fermented Soy Bean Extract Containing Herbal Medicines (Godjang) on Blood Glucose Levels and Histomorphology in Streptozotocin-induced Diabetic Rat)

  • Objective : This study was designed to investigate anti-diabetic effects of fermented soy bean extract with herbal medicines (Godjang) in diabetic rat models induced by streptozotocin (STZ) injection. Method : Changes in body weight, drinking water, and food intake were observed for 4 weeks before and after induction of diabetes mellitus in rats. The anti-diabetic capacity of Godjang was analyzed by fasting blood glucose (FBG) every week. Also, after 4 weeks of administration, the oral glucose tolerance test (OGTT) was performed, and then blood levels of insulin were checked. And serum levels of total cholesterol and triglycerides were determined. Histomorphological changes of liver, kidney and pancreatic tissues were also observed in STZ-induced diabetic rats and Godjang administered rats. Result : In Godjang administered group, body weight and water intake were more lower than that of STZ-induced diabetic rats. FBG was decreased in the Godjang administered group than STZ-induced diabetic group. According to OGTT, blood glucose levels at 30 minutes and 60 minutes significantly decreased in Godjang administered group than in STZ-induced diabetic control group. Administration of Godjang extract for 4W significantly decreased levels of serum glucose, total cholesterol (TC), triglycerides (TG) in diabetic rats. In histomorphological analysis of kidney, liver, Godjang administrated groups showed the inhibition of pathological damage. Conclusion : These results suggest that Godjang extract has an anti-diabetic action through decrease in serum glucose, TC, TG levels and recovery of the morphological changes in kidney and liver in STZ-induced diabetic rats.
