흰쥐 정자형성과정에 미치는 Di-(2-ethylhexyl)phthalate의 영향 (Effect of Di-(2-ethylhexyl)phthalate(DBHP) on Spermatogenesis in Rat Testes)

  • 플라스틱 제품의 가소제로 널리 사용되며, 최근 내분비 교란물질로 알려져 있는 di-(2-ethylhexyl)phthalate(DEHP)를 흰쥐에 15일 동안 구강 투여(1g/kg/day, 2g/kg/day, 3g/kg/day)한 후, 정자형성과정에 연관된 정소의 기능과 구조에 미치는 영향을 조사하였다. DEHP 처리군에서는 대조군에 비하여 체중 증가율이 감소하였을 뿐만 아니라 정소의 무게도 감소하였다. 또한 세정관의 직경이 고농도군으로 갈수록 작아지는 경향을 보였으며 세정관내의 세포층이 감소하는 현상이 나타났다. 미세구조의 변화를 관찰한 결과, 실험군은 세정관내 세포사이 공간이 증가하였으며 정원세포와 정모세포의 수가 감소하였고, 세포질내 공포가 증가하는 것이 관찰되었다. 또한 세정관내 Sertoli 세포의 전체 세포질 양이 감소하는 경향을 보였고, 정원세포의 경우 핵막 이중층이 분리되는 현상을 보였다. 특히 고농도군의 세정관내에서는 Sertoli 세포 이외의 세포는 거의 관찰되지 않았으며, 세포사이 공간과 공포들이 상당히 증가하였다. Sertoli 세포의 핵막은 심하게 함입된 형태를 나타냈으며 이질염색질이 증가하여 염색질의 덩어리를 이루고 있었고, Sertoli 세포들을 지지하는 기저판은 심하게 굴곡된 형태를 보였다. Leydig 세포의 미세구조에 있어서도 실험군은 대조군과 비교하여 현저한 차이를 보였다. 세포질내의 활면소포체와 핵막 이중층이 심하게 팽대되는 경향을 보였으며 핵내에 이질염색질이 상당히 증가하고 세포질내 리소솜이 증가하는 특징을 보였다.또한 혈청내 테스토스테론 함량에 있어서도 실험군은 현저히 낮은 수치를 나타냈다. 결론적으로, DEHP는 정소의 발달을 농도의존적으로 저해하고, Leydig 세포의 테스토스테론 합성기능을 저해하며, 이어서 세정관내부의 Sertoli 세포의 구조와 기능을 손상시켜 생식세포들의 괴사를 유도하는 과정을 통해 일련의 정자형성과정을 억제하는 것으로 사료된다.

  • 플라스틱 제품의 가소제로 널리 사용되며, 내분비 교란물질로도 알려져 있는 di-(2-ethylhexyl) phthalate (DEHP)를 사춘기 이전 흰쥐에 1주일 동안 구강 투여 (1g/kg, 3g/kg, 5g/kg)한 후 분화중인 정소의 미세구조에 미치는 영향을 조사하였다. DEHP 처리군에서는 대조군에 비하여 세정관 직경이 매우 작았으며, Leydig 세포, Sertoli 세포 및 분화중인 생식세포들의 증식이 억제되었다. 실험군에서 세정관 사이에 존재하여 테스토스테론을 분비하는 Leydig세포는 이질염색질이 증가하고, 세포질내에서 활면소포체의 발달이 미흡하였으며, 리소솜과 지방적이 증가하는 특징을 보였다. 세정관 내에서 Sertoli 세포는 세포질의 크기가 감소하고, 핵막이 불규칙해지거나, 염색질이 응축되어 있는 모습이 나타나며, 세포질내에서 리소솜과 액포들이 증가하였다. 실험군 흰쥐 정소내에서 분화중인 생식세포들의 핵은 이질염색질이 증가하거나, 응축되어 있고, 인의 분리현상이 관찰되기도 하며, 심한 경우 핵내에 공포가 형성되어 있거나, 괴사과정에 있는 생식세포들도 관찰되었다. 이와 같은 정소 미세 구조의 변화는 DEHP 투여량의 농도에 의존성을 보이며, 이 화합물은 Leydig 세포의 테스토스테론 합성 기능을 방해하고, 이어서 Sertoli 세포의 구조와 기능이 손상되어 정자형성과정 중인 생식세포들의 증식과 분화가 억제되는 것으로 사료된다.

고체상 추출법을 이용한 Phthalate Esters의 분석방법 연구 (A Study on Analytical Method of Phthalate Esters in Water by SPE(Solid-Phase-Extraction))

  • The study was carried out to evaluate the new analytical method of phthalate esters(diethylphthalate, di-n-butylphthalate, butylbenzylphthalate, bis(2-ethylhexyl)phthalate), one of the endocrine disruptors, which were performed by GC/MS-SIM(selected ion monitoring). The phthalate esters were extracted from water samples using solid-phase extraction on $C_{18}$ columns. It investigated that the extraction recovery rate of phthalate esters with different solvents and solvent volume. The optimal solvent was dichloromethane and proper volume of dichloromethane for recovery of phthalate esters was 4 mL. There were good linearities(above $R^2$=0.9975) in the range 0.01~0.50mg/L, and the detection limits were below 0.01~0.03$\mu\textrm{g}$/L. The recovery rates, RSD and MDLs for phthalate esters were 80~114%, 5.0~8.1% and 0.03~0.11$\mu\textrm{g}$/L, respectively. This method shows a good precision of phthalate esters.

PVC포장재에 사용된 가소제에 관한 조사연구 (A Study of Plasticizer in Food and Drug PVC Packaging)

  • This study was Performed to survey and evaluate the contents of Plasticizers such as DEP(diethyl phthalate), DPrP(di-n-Phthalate), DBP(di-n-butyl Phthalate), DPP(di-n-pentyl Phthalate), DCHP(dicyclohexyl phthalate), BBP (butylbenzyl phthalate), DEHP(di-(2-ethylhexyl) phthalate) and DEHA(di-(2-ethylheryl) adipate), which are suspected as endocrine disruptors, in food and drug PVC packaging. Tested samples were 5 food wraps, 35 food containers, 40 food and drug packages(type of tablet and capsule) in Gyeonggi-Do area. The contents of DEHA in wrap were 188.9g/kg, 203.1g/kg, 238.4g/kg, 290.9g/kg and 308.3g/kg, respectively, while the other plasticizers were not detected. DEHP was used in 4 samples of food containers and DEHP contents were 4.7g/kg, 30.7g/kg, 35.8g/kg and 53.4g/kg, respectively. In food and drug packaging materials(type of tablet and capsule), the plasticizers were not detected.

  • Migration levels of plasticizers, di-n-butyl phthalate (DBP), benzyl-butyl phthalate (BBP), di-n-octyl phthalate(DNOP), di-iso-decyl phthalate (DIDP) and di-iso-nonyl phthalate (DINP), di-(2-ethylhexyl) adipate (DEHA), from 46 poly(vinyl chloride) (PVC) wrap films and 54 PVC gaskets into food simulants were determined using gas chromatography/mass spectrometry (GC/MS). The method was validated with limit of detection (LOD) of $0.01{\sim}0.02\;{\mu}g/mL$ for DBP, BBP, DNOP and DEHA, and $2\;{\mu}g/mL$ for DIDP and DINP. The linearity were found to be > 0.99 for all the compounds in concentration range of $0.1{\sim}81.4\;{\mu}g/mL$, and overall recoveries were ranged from 90.4 ~ 99.6%. DBP, BBP, DNOP, DEHA, DIDP and DINP were not detected in food simulants, except 1 wrap sample from which 0.28 and $0.99\;{\mu}g/mL$ of DEHA were detected respectively when tested with 20% ethanol and n-heptane as food simulants. These values were below the regulatory limitation in European Union (EU).

Altered Gene Profiles using KISTCHIP-400 in MCF-7 cells after Exposure to Di(2-ethylhexyl) Phthalate (DEHP) and Dibutyl Phthalate (DBP)

  • There are many synthetic chemicals, such as di(2-ethylhexyl) phthalate (DEHP) and dibutyl phthalate (DBP), used in chemical reaction processes in industry. The establishment of toxicity and detection of synthetic chemicals that may pose a genetic hazard in our enviornment is subjects of great concern at present DEHP, a ubiquitous phthalate plasticizer, induces a wide range of developmental and reproductive toxicities in mammals. DEHP belongs to the large diverse class of peroxisome proliferator compounds, which include herbicides, hypolipidemic drugs. DBP is a plasticizer used to products containing nitrocellulose, polyvinyl acetate, and polyvinyl chloride such as food wraps and blood bags. DBP is also used in cosmetics as a solvent and fixative for perfumes, a suspension agent for solids, an antifoamer, a skin emollient, and hair spray The present study was performed to examine patterns of gene expression in MCF-7 cells following DEHP and DBP exposure. Changes in gene expression were determined by microarray analysis using KISTCHIP-400 including 401 endocrine related genes based on public database and research papers. Of the genes analysis, we determined that genes detected by array showed a 2-fold or greater change in their expression level(increase or decrease). The results of this study demonstrate that a number of genes were differentially expressed in MCF-7 cells but these changes were not significant. Therefore, we keep going this study using microarray analysis and future studies will examine changes of gene expression on time-course and does treatment in variable cell lines.
