Optimization of Submerged Culture Conditions for Mycelial Growth and Exopolysaccharides Production by Agaricus blazei

  • The influences of inoculum size, pH, and medium composition on mycelial growth and exopolysaccharides (EPS) production were investigated in shake flasks and in a bioreactor. The optimum inoculum size for both mycelial growth and EPS production was identified to be 10% (v/v) in shake flask cultures. The optimal initial pH for mycelial growth and EPS production in shake flask cultures were found to be 5.0 and 7.0, respectively. However, the optimal pH was 5.0 for both mycelial growth and EPS production in bioreactor cultures where the pH was regulated. The optimal mass ratio of the two major carbon sources, glucose to dextrin, was 1:4. The optimal mass ratio of the two major nitrogen sources, yeast extract to soy tone peptone, was 2:1. When 500 mg $1^{-1}$ of $MnSO_4-5H_2O$ was added to the bioreactor culture, both mycelial growth and EPS production were enhanced by approximately 10%. Under the optimized conditions, a mycelial biomass of 9.85 g $1^{-1}$ and an EPS concentration of 4.92 g $1^{-1}$ were obtained in 4 days.

Medium Optimization and Application of Affinity Column Chromatography for Trypsin Production from Recombinant Streptomyces griseus

  • The production of Streptomyces griseus trypsin (SGT) by S. griseus IFO13350 transformed with the expression vector pWHM3-TR1R2, containing sprT encoding SGT and the two positive regulatory genes sgtR1 and sgtR2, was investigated in various media. Cultivation in Ferm-0 gave 1.4 times more trypsin activity than in C5/L medium. In addition, replacement of 2% glucose and 1% skim milk in Ferm-0 with 2% dextrin and 1% tryptone (designated Ferm-II) enhanced trypsin activity 4.1-fold. To simplify the purification process, the supernatant from the S. griseus transformant cultured in Ferm-II medium was fractionated with ammonium sulfate (25-55%), then subjected to Hitrap Benzamidine FF affinity column chromatography. The specific activity of SGT purified by one-step chromatography was 69,550 unit/mg protein and the overall purification yield was above 8%, indicating that this method is more effective than those previously reported. Purified SGT was most active at pH 8.0 and $50^{\circ}C$, and it maintained activity between pH 7.0 and 9.0 and at temperatures up to $70^{\circ}C$. These enzymatic properties are very similar to those of authentic eukaryotic trypsin purified from bovine pancreas.

Recombinant Expression and Characterization of Thermoanaerobacter tengcongensis Thermostable $\alpha$-Glucosidase with Regioselectivity for High-Yield Isomaltooligosaccharides Synthesis

  • A novel thermostable $\alpha$-glucosidase (TtGluA) from Thermoanaerobacter tengcongensis MB4 was successfully expressed in E. coli and characterized. The TtgluA gene contained 2,253 bp, which encodes 750 amino acids. The native TtGluA was a trimer with monomer molecular mass of 89 kDa shown by SDS-PAGE. The purified recombinant enzyme showed hydrolytic activity on maltooligosaccharides, p-nitrophenyl-$\alpha$-D-glucopyranide, and dextrin with an exotype cleavage manner. TtGluA showed preference for short-chain maltooligosaccharides and the highest specific activity for maltose of 3.26 units/mg. Maximal activity was observed at $60^{\circ}C$ and pH 5.5. The half-life was 2 h at $60^{\circ}C$. The enzyme showed good tolerance to urea and SDS but was inhibited by Tris. When maltose with the concentration over 50 mM was used as substrate, TtGluA was also capable of catalyzing transglycosylation to produce $\alpha$-1,4-linked maltotriose and $\alpha$-1,6-linked isomaltooligosaccharides. More importantly, TtGluA showed exclusive regiospecificity with high yield to produce $\alpha$-1,6-linked isomaltooligosaccharides when the reaction time extended to more than 10 h.

Dissolution Profiles of Solid Dispersions Containing Poorly Water-Soluble Drugs and Solubilizing Compositions (가용화 조성물과 난용성 약물군을 함유하는 고체분산체의 용출양상)

  • Polymer based physical mixtures or solid dispersions containing solubilizing compositions[OA, tween80 and SLS] were prepared using a spray-dryer. Lovastatin(LOS), simvastatin(SIMS), aceclofenac(AFC) and cisapride(CSP) were selected as poorly water-soluble drugs. Dextrin, poly(vinylalcohol) (PVA), poly(vinylpyrrolidone)(PVP) and polyethylene glycol(PEG) were chosen as solubilizing carriers for solid dispersions. The solid dispersions containing solubilizing compositions without drug were prepared without using organic solvents or tedious changes of formulation compositions. This system could be used to quickly screen the dissolution profiles of poorly water-soluble drugs by simply mixing with drugs thereafter. In case of solid dispersion containing drug, organic solvent systems could be used to solubilize model drugs. The dissolution rates of the drugs were higher when mixed with drug and solid dispersions containing solubilizing compositions. However, solid dispersions of LOS, AFC, and CSP simultaneously containing drug and solubilizing compositions in organic solvent systems were more useful than physical mixtures of drug and solid dispersions without drug except SIMS. Based on solubilizing capability of polymer based physical mixtures in gelatin hard capsules, optimal solid dispersion system of poorly water-soluble drugs could be formulated. However, it should be noted that dissolution rate of poorly water-soluble drugs were highly dependent on drug properties, solubilizing compositions and polymeric carriers.

  • Cosmetic industries have recently developed sun-block products, which are composed of W/O or O/W emulsion system. It was very difficult for waterproofing product to show the stability in W/O emulsion with $TiO_{2}$. To enhance the stability of W/O emulsion, it needs to be combined with the water and oil soluble components as the gelling agents. The emulsifiers used in W/O were 3.0% of cetyl dimethicone copolyol, 2.0% of sorbitan sesquioleate as the basic emulsifiers, and 0.6% of quaternium-18 bentonite and 1.5% of dextrin palmitate as stabilizer were used. The content of titanium dioxide was optimized up to 8.0%. Titanium dioxide was used as the UV scattering powder coated with $Al_{2}O_{3}$(UV-sperse T40/TN). The sunscreen cream prepared with W/O emulsion system by using QB and DP showed higher stability than that of W/O emulsion system by using each QB and DP. W/O emulsion from Formula 3 for passing one year was very durable more than F1 and F2. Within W/O emulsion by observing F1, F2 and F3 for one year, F3 was more excellent than F2 and F3 when they were observed at RT, $4^{\circ}C$, $40^{\circ}C$, because F3 used the mixed QB and DP in W/O emulsion. The zeta potential for F1, F2, and F3 after one year were 21, 30 and 43, respectively. From these result F3 was found best stable emulsion. The in-vitro SPF value for F3 was 35 for the initial product at room temperature and also, the in-vitro SPF values of F3 was 32 for after one year. Finally, the mean in-vivo SPF value of 10 volunteers for F3 was 27.3 by the Korea cosmetic association made the rules of SPF.

Conservation of Excavated Lacquer-wares for using artificially water-soaked Lacquer-wares (인공수침 칠기를 이용한 고대칠기 보존연구)

  • Among the treatment results of test samples of the antique lacquer-ware, the treatment with PEG#3,350 40% solution displayed excellent effect with low shrinkage ratio; in weight gain the treatment with Sucrose 19%+Glycerin 1%(t-butanol 5% in water) solution showed consistent increase. However during the impregnation process of Sucrose, the weight of the testing samples decreased by dehydration because the inner part of the test samples and the treatment solution showed concentration gradient. Therefore, we concluded longer impregnation period should be necessary to prevent dehydration. Since both higher and lower molecular weight treatment chemicals could penetrate into the wood of the lacquer-ware, air drying and conditioning after impregnation treatment with high concentration chemicals would be possible, as well as vacuum freeze-drying.

Production and Rheological Properties of the Polysaccharide from Bacillus sp. A29 (Bacillus sp. A29에 의한 다당류의 생산과 물성)

  • A bacterial strain producing highly viscous polysaccharide(A29 POL) was isolated from soil and identified sa Bacillus sp. A29. The cultural conditions of the Bacillus sp. A29 for the polysaccharide prouction were dextrin 12%, soytone 0.2%, SnCl$_{2}$ $\cdot $2H$_{2}$O 0.02%, Na$_{2}$HPO$_{4}$ $\CDOT $12H$_{2}$O 0.36%, L-alanine 0.01%, initial pH6.8, and 30$\circ $C at pH 3 FOR 4 days. Final viscosity of the culture broth was 65, 000 cp and then the amount of produced polysaccharide was 8.3 g/l. A29 POL was composed of glucose and xylose. A29 POL showed high viscosity at low concentration(0.1%) and in the presence of the salts such as NaCl or CaCl$_{2}$. A29 POL showed high viscosity acid condition and at alkali condition and high pseudoplasticity in the presence of a NaCl or CaCl$_{2}$. It was shown that the viscosity at high temperature(80$\circ $C) was decreased but it was recovered at low temperature (20$\circ $C. A29 POL was able to from film and gel in the presence of MgSO$_{4}$ $\CDOT $7H$_{2}$O, Na$_{2}$CO$_{3}$ \CDOT $H$_{2}$O, MnSO$_{4}$ $\CDOT $ 7H$_{2}$O. A29 POL had anionic charge.

  • Effects of some sources on the vegetative growth of Naematoloma sublateritium (Fr.) Karst. were investigated using liquid and solid culture media. Temperature, pH, carbohydrates as carbon sources, amino acids as nitrogen sources, the optimal carbon/nitrogen ratio, mineral element and organic acids were studied for good mycelial growth. We could improve a new semisynthetic medium for mycelial growth in N. sublateritium.

Sensory Characteristics of Granular Tea and the Components of Mulberry Fruit Extracts by Different Extraction Process (오디의 추출 공정에 따른 성분 변화 및 분말 과립차의 관능 특성)

  • In the present work, mulberry fruit extracts by four extraction processes, namely wet pressing extraction (WPE), hot-water extraction (HWE), enzymatic hydrolysis (EH), and lactic-acid bacteria fermentation (LBF) by Lactobacillus plantarum TO-2100, were analyzed for nutrients and functional compounds. The sugar contents of extracts by WPE, HWE, EH, and LBF were 12.0, 10.9, 14.5, and 14.3 brix, respectively, and the extraction yields by EH and LBF were 1.65 and 1.50 times higher than those by WPE. Among the organic acids, tartaric acid and malic acid contents were the highest in the extracts by WPE. Acetic acid was best extracted by LBF, and citric acid was best extracted by EH. Lactic acid was detected only in LBF. The extracts by EH showed the highest contents of all vitamins with an exception that the extracts by LBF showed the highest contents of the folic acid, vitamin B12, and vitamin C. We also noted that vitamin B group was not detected in the extracts by LBF. The extracts by EH showed the highest contents of all the amino acids, whereas LBF showed the lowest. Polyphenol contents of extracts by EH and LBF were 3.05 and 2.51 times more than those by WPE respectively. Anthocyanin contents were 7.66, 7.14 times higher for EH and LBF compare to WPE. We manufactured mulberry fruit granular teas with different compositions and tested them for their sensory characteristics. We found that 15% mulberry fruit extracts by enzymatic hydrolysis and 85% dextrin composition gave the most satisfactory result.

  • This study was excuted to decide the physiological characteristics of Kuehneromyces mutabilis. K. mutabilis was tested to select pertinent substract, temperature and pH range for the growth. Mycelial growth of K. mutabilis was mostly supported on MYPA among other tested synthetic or semi-synthetic media. The temperature range for pertinent mycelial growth was about 25~32$^{\circ}C$ and mostly stimulated at $25^{\circ}C$. And the pertinent pH range of MYPA was 5.0~6.0. The required carbon and nitrogen source for mycelial growth of K. mutabilis was tested. The mycelial growth was mostly stimulated by soluble starch at content. The carbon sources for pertinent mycelial growth was mannose or glucose. And the nitrogen source for pertinent mycelial growth was yeast extract or peptone.
