Biocompatibility of Tissue-Engineered Heart Valve Leaflets Based on Acellular Xenografts (세포를 제거한 이종 심장 판막 이식편을 사용한 조직공학 심장 판막첨의 생체 적합성에 대한 연구)

  • Current artificial heart valves have several disadvantages, such as thromboembolism, limited durability, infection, and inability to grow. The solution to these problems would be to develop a tissue-engineered heart valves containing autologous cells. The aim of this study was to optimize the protocol to obtain a porcine acellular matrix and seed goat autologous endothelial cells on it, and to evaluate the biological responses of xenograft and xeno-autograft heart valves in goats. Material and Method: Fresh porcine pulmonic valves were treated with one method among 3 representative decellularization protocols (Triton-X, freeze-thawing, and NaCl-SDS). Goat venous endothelial cells were isolated and seeded onto the acellularized xenograft leaflets. Microscopic examinations were done to select the most effective method of decellularizing xenogeneic cells and seeding autologous endothelial cells. Two pulmonic valve leaflets of. 6 goats were replaced by acellularized porcine leaflets with or without seeding autologous endothelial cells while on cardiopulmonary bypass. Goats were sacrificed electively at 6 hours, 1 day, 1 week, 1 month, 3 months, and 6. months after operation. Morphologic examinations were done to see the biological responses of replaced valve leaflets. Result: The microscopic examinations showed that porcine cells were almost completely removed in the leaflets treated with NaCl-SDS. The seeded endothelial cells were more evenly preserved in NaCl-SDS treatment. All 6 goats survived the operation without complications. The xeno- autografts and xenografts showed the appearance, the remodeling process, and the cellular functions of myofibroblasts, 1 day, 1 month, and 3 months after operation, respectively. They were compatible with the native pulmonary leaflet (control group) except for the increased cellularity at 6 months. The xenografts revealed the new endothelial cell lining at that time. Conclusion: Treatment with NaCl-SDS was most effective in obtaining decellularized xenografts and facilitate seeding autologous endothelial cells. The xenografts and xeno-autografts were repopulated with myofibroblasts and endothelial cells in situ serially. Both of grafts served as a matrix for a tissue engineered heart valve and developed into autologous tissue for 6 months.

Effect of Transplanting Methods on Growth and Yield of Paprika in Coir Culture (코이어 배지 수경재배에서 정식방법이 파프리카 생육과 수량에 미치는 영향)

  • This study was carried out to investigate the effect of transplanting methods on the growth and yield of paprika (Capsicumannuum L. 'Veyron' and 'Coletti') in coir culture during two seasons. The summer type sowed in late winter and harvested from summer and the winter type sowed in summer and harvested from early winter. Control plants grown on the 10cm rockwool block were transplanted on coir slab when the 8 leaves of seedlings were emerged, while plants of the young seedling transplanting(YST) grown on the 7cm rockwool block was put on the slab at time of 2-3 leaves developed. Plants of the temporary transplanting(TT) on the 10cm rockwool block were moved on the slab after 2-3 weeks underpinning cultivation, while plants of the blockless transplanting(BT) were directly transplanted in the slab when the seedlings have 2-3 leaves emerged. The plant height of the control and BT treatment were longer while that of TT showed the shortest among treatments. The bigger leaf size was observed in the YST and BT treatment. Leaf number of the BT treatment was increased, while that of TT was the lowest. There were no differences in fruit size, locules and thickness among treatments. The lower fruit weight was observed in the TT of the winter culture and fruits in the control and YST of the summer culture showed higher fruit weight. The percentage of marketable fruit appeared to be slightly higher in the winter culture than in the summer culture. There were no differences in marketable fruit rate among the treatments of the winter culture but, among the summer culture, the highest marketable fruit rate was observed in the BT with 93%. The yield of the YST and BT was higher and that of the TT was the lowest.

Agronomic Characteristics of A Mutant for Genic Male Sterility-Chalky Endosperm and Its Utilization on $F_1$ Hybrid Breeding System in Rice (벼 유전자적 웅성불임-심백 돌연변이체의 특성과 그를 이용한 일대잡종 육성체계)

  • A mutant for chalky endosperm and genetic male sterility (GMS) was newly developed in rice. The two characters were found to be controlled by single recessive gene which has pleiotropic effect, indicating that chalky seeds should be GMS seeds in segregating populations. Chalky seeds showed the same shape and size as normal seeds. However, starch composition of central part of endosperm was looser and shape of starch granules was rounder compared with normal endosperm, resulting in significantly lower grain weight, absolute density and grain hardness in chalky grains than in normal ones. Amylose content and gel consistency of chalky grains were much lower and harder, respectively. Male sterile plants showed much shorter plant height, poorer panicle exsertion and lesser panicle number compared with normal plants. Microspore abortion stage in pollen developmental process was observed as before meiosis. Male sterility of the mutant was stable regadless of temperature and day length. A system breeding hybrid rice using this mutant was discussed, comp ring with other systems utilizing cytoplasmic-genic male sterility(CGMS) and environment sensitive GMS(EGMS). Separation of GMS seeds in mixed seed bulks by specific gravity (1.14∼1.16g / cm3) was successful about 85∼90%. But some mixed normal plants were seemed to be easily removed by the apparent difference in growth characters at seedling stage. The highest natural outcrossing rate of this GMS line was as 17.3 % in a plot treated with 2-row pollinator, I-row GMS, and GA3 + cutting of flag leaf + pollen-scattering by rope.

Comparison of Micronutrients and Flavor Compounds in Breast Meat of Native Chicken Strains and Baeksemi for Samgyetang (삼계용 토종닭과 백세미 가슴살의 미량영양소 및 풍미물질 비교)

  • The micronutrients and flavor compounds of three new native chicken strains (A, C, and D) being developed in a breeding program (Golden Seed Project) were compared with a commercial native chicken strain (H) and the Baeksemi (W, white semi broiler). After 100 male chicks in each strains were reared for 5 weeks, the breast muscles from randomly selected 40 birds were analyzed at 8 replications. Native chicken strain A had greater amounts of α-tocopherol and α-tocotrienol compared with strain W. Native chicken strains showed higher contents of vitamin B12 than Stain W. Stain H strain had the highest values of cholesterol content and strain D did the lowest. There was no different content of most minerals between native chickens (A, C, D, and H) and W, but Cu were more contained in native chicken strains compared with W. In terms of nucleotide-related flavor compounds, Stain A had the lowest content of hypoxanthine and strain D had highest inosine monophosphate. Native chicken strains had higher contents of umami-related free amino acids (glutamate and aspartate) than W. Among native chickens, strain A had more amounts in the contents of taurine, tasty flavor compounds, and certain vitamins, despite of the relatively low growth productivity. This result will provide information to select a strain with characteristic meat quality in a chicken breeding program.

A Study on the Forecasting Model on Market Share of a Retail Facility -Focusing on Extension of Interaction Model- (유통시설의 시장점유율 예측 모델에 관한 연구 -상호작용 모델의 확장을 중심으로)

  • In this chapter, we summarize the results on the optimal location selection and present limitation and direction of research. In order to reach the objective, this study selected and tested the interaction model which obtains the value of co-ordinates on location selection through the optimization technique. This study used the original variables in the model, but the results indicated that there is difference in reality. In order to overcome this difference, this study peformed market survey and found the new variables (first data such as price, quality and assortment of goods, and the second data such as aggregate area, and area of shop, and the number of cars in the parking lot). Then this study determined an optimal variable by empirical analysis which compares an actual value of market share in 1988 with the market share yielded in the model. However, this study found the market share in each variables does not reflect a reality due to an assumption of λ-value in the model. In order to improve this, this study performed a sensitivity analysis which adds the λ value from 1.0 to 2.9 marginally. The analyzed result indicated the highest significance with the market share ratio in 1998 at λ of 1.0. Applying the weighted value to a variable from each of the first data and second data yielded the results that more variables from the first data coincided with the realistic rank on sales. Although this study have some limits and improvements, if a marketer uses this extended model, more significant results will be produced.

Chemical Composition of Seed from Inbred Lines and Hybrids of Maize Recently Developed in Korea (국내 육성 주요 옥수수 자식계통 및 교잡종의 종실 성분 특성)

  • This study was carried out to evaluate the quality properties, fatty acid composition and amino acid composition of major Korean inbred lines and hybrids of maize for corn breeding. Protein content of $F_2$ seeds was 9.0%, which was slightly lower than those of inbred lines, 11.4%, and $F_1$ seeds, 10.9%. in hybrid maizes. Lipid content of $F_2$ seeds of hybrid maizes was 4.2%, slightly higher than those of inbred lines, 3.3%, and $F_1$ seeds, 3.6%. However, there was no significant difference in ash content. The amount of linoleic acid in inbred lines, $F_1$ and $F_2$ seeds of hybrid maizes was highest among all fatty acids and followed by oleic acid, palmitic acid, stearic acid and linolenic acid. The composition of saturated fatty acid of $F_2$ seeds was 17.6%, similar to $F_1$ seeds but it was lower than that of inbred lines. The composition of unsaturated fatty acid of $F_2$ seeds was 82.4%, similar to $F_1$ seeds but was higher than inbred lines. The content of leucine of $F_2$ seeds was lower than inbred lines and $F_1$ seeds. The content of valine, serine, threonine, cysteine, aspartic acid, lysine, arginine, histidine of $F_2$ seeds was higher than inbred lines and $F_1$ seeds. MMA (monoamino monocarboxylic acid) in inbred lines, $F_1$ and $F_2$ seeds of hybrid maizes was highest among all amino acids and followed by DMA (diamino monocarboxylic acid) and AAA (aromatic amino acid).

Current status and prospects of molecular marker development for systematic breeding program in citrus (감귤 분자육종을 위한 분자표지 개발 현황 및 전망)

  • Citrus is an economically important fruit crop widely growing worldwide. However, citrus production largely depends on natural hybrid selection and bud sport mutation. Unique botanical features including long juvenility, polyembryony, and QTL that controls major agronomic traits can hinder the development of superior variety by conventional breeding. Diverse factors including drastic changes of citrus production environment due to global warming and changes in market trends require systematic molecular breeding program for early selection of elite candidates with target traits, sustainable production of high quality fruits, cultivar diversification, and cost-effective breeding. Since the construction of the first genetic linkage map using isozymes, citrus scientists have constructed linkage maps using various DNA-based markers and developed molecular markers related to biotic and abiotic stresses, polyembryony, fruit coloration, seedlessness, male sterility, acidless, morphology, fruit quality, seed number, yield, early fruit setting traits, and QTL mapping on genetic maps. Genes closely related to CTV resistance and flesh color have been cloned. SSR markers for identifying zygotic and nucellar individuals will contribute to cost-effective breeding. The two high quality citrus reference genomes recently released are being efficiently used for genomics-based molecular breeding such as construction of reference linkage/physical maps and comparative genome mapping. In the near future, the development of DNA molecular markers tightly linked to various agronomic traits and the cloning of useful and/or variant genes will be accelerated through comparative genome analysis using citrus core collection and genome-wide approaches such as genotyping-by-sequencing and genome wide association study.

Growth Characteristics, Seed and Forage Productivity of New Naked Oat (Avena nuda L.) Cultivar "Jungmo2005" (귀리 품종 '중모2005'의 생육특성과 종실 및 조사료 생산성)

  • A new naked oat cultivar, 'Jungmo2005'(Avena nuda L.), was developed for food and forage use by National Institute of Crop Science, RDA in 2010. It was derived from a cross between $F_1$['Early80'/'Gwiri33'] and $F_1$['Early80'/'Gwiri23']. 'Early80', a covered oats, has early heading and high yielding, while 'Gwiri23' of covered type and 'Gwiri33' of naked type has early heading with large-size grain. 'Jungmo2005' has the characteristics of narrow and long leaves of pale green color, middle diameter culm of yellow color and medium grain of whitish yellow color. Cultivar 'Jungmo2005' had 2 days earlier heading date (May 9) than the check cultivar 'Seonyang' (May 11) in field condition. The 'Jungmo2005' showed better winter hardiness than that of the check cultivar, and similar to the check cultivar in respect to lodging resistance. The 'Jungmo2005' had 105 cm of culm length, 21.5 cm of spike length, 658 spikes per $m^2$, 82 grains per spike, 23.3 g of 1,000-grain weight, and 622 g of test weight. The grain yield of 'Jungmo2005' was averaged $3.38MT\;ha^{-1}$, which was 6% higher than that of the check. Average forage fresh and dry matter yield of 'Jungmo2005' harvested at milk-ripe stage were 44.8 and $12.6tone\;ha^{-1}$, respectively, compared with 47.5 and $12.5tone\;ha^{-1}$ of the check. The protein content of the 'Jungmo2005' was similar to the check (7.6% and 7.5%, respectively), while ADF (28.6%) and NDF (51.5%) were lower than the check (31.4% and 57.0%, respectively). TDN content and RFV were higher than those of the check (66.3%, 120.3 and 64.1, 105.2, respectively). 'Jungmo2005' is recommended for fall sowing cropping only in the south area where daily minimum mean temperatures are averaged higher than $-4^{\circ}C$ in January, and it should not be cultivated in mountain areas, where frost damages is likely to occur. The areas would do better only to sow in spring season.

Morphological Adaptation of Zostera marina L. to Ocean Currents in Korea (한국산 거머리말(Zostera marina L.)의 해류에 대한 형태적 적응)

  • The main purpose of this research is to prepare and provide basic materials for the propagational strategy of eelgrass by investigating on the morphological adaptation of Korean Zostera marina to ocean currents. An eelgrass plant mainly consists of rhizome, leaf sheath, leaves and roots. The rhizome is the horizontal stem of the plant that serves as the backbone from which the leaves and roots emerge. The leaf sheath is the bundle at the base of the leaves that holds the leaves together, protecting the meristem, the primary growth point of the shoot. Leaves originate from a meristem which is protected by a sheath at the actively growing end of the rhizome. As the shoot grows, the rhizome elongates, moving across or within the sediment, forming roots as it progresses. The aggregated leaves from the leaf sheath are found to have two cell layers on one side and multiple layers of airy tissues called aerenchyma on the other. The aerenchyma tissues are developed in multi-layered cell structures surrounding the veins which are formed in the leaf sheath. Generative shoots are made of rhizomes, which are circular or ovoidal, stem, and spathe and spadix. The transverse section of rhizome and the stem and central floral axis is found to be circular, ovoid and in the shape of convex respectively, and the vascular bundle, which is a part of transport system, has one large tube in the center and two small tubes on both sides. The layers of collenchyma cells numbered from 12 to 15 in the stem, and from 7 to 12 in the rhizome. The seed coat is composed of sclereids, small bundles of sclerenchyma tissues, which prevent the influx of sea water from the outside and help endure the environmental stress. In conclusion, alternative multi-layer structure in circular, convex type aggregated leaf base are interpreted to morphological adaption as doing tolerable elastic structure through movement of seawater. The generative shoots develop long slim stem and branches in circular or ovoidal shapes to minimize the adverse impacts of sea current, which can be interpreted as the plant's morphological adaptation to its environment.

Taxonomic Studies of Genus Juniperus (향나무속(屬)의 분류학적(分類學的) 연구(硏究))

  • In order to solve the taxonomic problems of the genus Juniperus growing in South Korea, an identification key of the genus and species was developed bayed un flower structure, cane and seed shape, branching habit, tree form, leaf characteristics etc. of the 7 native species and the a exotic cultivars. The typical pattern of karyotype found by chromosome analysis of the species was used for the identification among morphologically similar species. The length of chromosome were ranged $9{\sim}15{\mu}m$ in all studied specie. J. chinensis, var. procumbens, and var. kaizuka sere tetraploid, 4n=44, var. globosa, var. procumbens, var. horizontalis, J. virginiada, J. rigida, J. rigida var. longicarpa, and J. coreana were diploid, 2n=22. The species in the Sabina section showed large variation in the length of chromosome and kinetochore position. The species in the Oxycedrus section showed the cytological characteristics that the 11th chromosome t-type(acrocentric), and the m-type abundant chromosome set was relatively uniform as compared to those of the Sabina section. The species in the Sabina section, which are planted in the large city area, show great morphological variation because many different ecotypes were mixed and often crossed among them. In summary, this study was able to make clear identification and to find out similarity among Juniperus, species by the morphological and cytological analysis.
