Effect of Next-Generation Exome Sequencing Depth for Discovery of Diagnostic Variants

  • Sequencing depth, which is directly related to the cost and time required for the generation, processing, and maintenance of next-generation sequencing data, is an important factor in the practical utilization of such data in clinical fields. Unfortunately, identifying an exome sequencing depth adequate for clinical use is a challenge that has not been addressed extensively. Here, we investigate the effect of exome sequencing depth on the discovery of sequence variants for clinical use. Toward this, we sequenced ten germ-line blood samples from breast cancer patients on the Illumina platform GAII(x) at a high depth of ${\sim}200{\times}$. We observed that most function-related diverse variants in the human exonic regions could be detected at a sequencing depth of $120{\times}$. Furthermore, investigation using a diagnostic gene set showed that the number of clinical variants identified using exome sequencing reached a plateau at an average sequencing depth of about $120{\times}$. Moreover, the phenomena were consistent across the breast cancer samples.

  • The focus of this experimental study is to verify the effect of openings in reinforced concrete deep beam. Main variables are opening depth, width and vertical locations but the shear span-to-overall depth ratio was fixed by 0.5. The experimental results about 8 deep beams which was tested under two equal symmetrically placed point loads are reported.

  • The effect of coating method and binder content on the tensile adhesion strength, water absorption and cl- penetration depth of polymer-modified pastes using redispersible polymer powders and ceramic powder are examined. As a result, the tensile adhesion strength of the polymer-modified pastes tend to increase with increasing binder content and: number of coating. The water absorption and cl- penetration depth of the polymer-modified pastes tend to decrease with increasing binder content and number of coating.

  • Effect of reentrant type bowl geometry on combustion characteristics was investigated in a D.1.diesel engine. The main factor was the aspect ratio (Bowl Diameter/Bowl Depth) of bowl of combustion chamber, and the measured data include the cylinder pressure, engine performance and emissions of the engine using the 4 kinds of the combustion chamber. Experimental results indicate that the effect of dc/H and nozzle protrusion are relatively small and there exists an optimum dc/H according to the combustion conditions. It is also found that the smoke emission is quite sensitive the overall combustion time where the 90 percentage of the combustion heat is released. The smoke mission increases by shortening the 90% combustion time while it decreases by delaying the 90% combustion time.

Improved 3D Resolution Analysis of N-Ocular Imaging Systems with the Defocusing Effect of an Imaging Lens

  • In this paper, we propose an improved framework to analyze an N-ocular imaging system under fixed constrained resources such as the number of image sensors, the pixel size of image sensors, the distance between adjacent image sensors, the focal length of image sensors, and field of view of image sensors. This proposed framework takes into consideration, for the first time, the defocusing effect of the imaging lenses according to the object distance. Based on the proposed framework, the N-ocular imaging system such as integral imaging is analyzed in terms of depth resolution using two-point-source resolution analysis. By taking into consideration the defocusing effect of the imaging lenses using ray projection model, it is shown that an improved depth resolution can be obtained near the central depth plane as the number of cameras increases. To validate the proposed framework, Monte Carlo simulations are carried out and the results are analyzed.

Increase in Color Depth of Polyester Fabric by Resin Treatment (수지처리에 의한 PET직물의 심색화)

  • To improve the deep coloring effect of PET fabrics, the alkali treated and black dyed PET fabrics were treated with 2 kinds of low refractive compounds such as acrylic resin and silicone resin. The color depth effect of treated PET fabrics was evaluated as lightness(L) change by UV-visible spectrophotometer. As the weight loss of PET fiber treated with alkali increased, the color depth of PET fabrics increased. Lightness(L) of PET fabrics treated with deep coloring agent was lower than that of untreated PET fabrics. The optimum concentration of treated PET with deep coloring agent was 4% o.w.s. The deep coloring effect of PET fabrics treated with silicone resin was higher than one treated with acrylic resin. PET fabrics treated with silicone resin only might be more appropriate process than PET fabrics treated with acrylic and silicone resin for giving deep coloring effect for polyester fabrics.

  • The factors to influence the travelling distance of sprayed particles for the medium range nozzle may by the groove depth of swirl plate, the cap slope, the diameter of cap hole, and pressure. 1. This study was conducted to examine Interaction effects among four factors to the travelling distance. The results of this study are summarized as followa; a) Interaction effects among four factors the groove depth(G), cap slope(C), diameter of cap hole(D), and pressure (P), were significant to influence the travelling distance except for $G{\times}P,\;C{\times}D{\times}P\;and\;G{\times}C{\times}D{\times}P$. b) Interaction effects with the pressure were very smaller than interaction effects among the other factors. c) Effect of change of the groove depth of swirl plate on the travelling distance of sprayed particles was generally a linear, the increasing rate of the change was about 0.345, which was very significant. d) Effect of change of cap slope on the travelling distance was generally a linear or a dull quadratic, the increasing rate was very small. e) Main effect of change of cap slope in the medium range nozzle was very smaller than that of the close range nozzle on the travelling distance, which was estimated by the changing of turning radius of flowing course in nozzle. f) Interaction effect between two factors in the medium range nozzle was more significant than that of the close range nozzle on the travelling distance.

A Study on the Influence of Induction Coil Movement Speed and Frequency on Induction Hardening of SCM440 Steel (SCM440 강의 유도 경화에 미치는 유도코일 이동속도 및 주파수의 영향에 관한 연구)

  • In this study, microstructure, hardening layer hardness, and case depth were evaluated after induction hardening(IH) of base metal specimen(BM) treated with annealing and quenching-tempering specimen(QT) treated with quenching and tempering. The microstructure after IH was significantly influenced by the microstructure before IH and the induction coil heating movement speed, but the effect of the induction frequency was very small. The hardness of the hardened layer at an induction coil heating movement speed of 15 mm/s or less was more influenced by the microstructure before IH than the induction coil travel speed and induction frequency. The induction coil travel speed has the significantly effect on the case depth, the induction frequency has effect and the microstructure before IH has a small effect.

Effect of Surface Charging on the SIMS Depth Profile of Bismuth Titanate Thin Film (SIMS 분석조건이 Bismuth Titanate 박막의 깊이방향 조성 해석에 미치는 영향)

  • The effect of SIMS analysis conditions such as mesh grid, offset voltage and ion species on the in-depth profile for bismuth titanate thin film was examined in terms of charging effect and detection limit. The results shows that the use of offset voltage -40 V reduces the charging effect and the detection limit. The employment of mesh grid in sample preparation leads to the reduction of the charging effect in small amount, but deteriorate the detection limit. Utilization of primary $O^-$ ion for SIMS analysis of bismuth titanate thin film showed almost the same effect as using offset voltage -40 V. However, it takes approximately triple acquisition time than using $O_2{^+}$ ion due to the poor beam current of the source in the experiment.

The Effects of Intention Inferences on Scarcity Effect: Moderating Effect of Scarcity Type, Scarcity Depth (소비자의 기업의도 추론이 희소성 효과에 미치는 영향: 수량한정 유형과 폭의 조절효과)

  • The scarcity is pervasive aspect of human life and is a fundamental precondition of economic behavior of consumers. Also, the effect of scarcity message is a power social influence principle used by marketers to increase the subjective desirability of products. Because valuable objects are often scare, consumers tend to infer the scarce objects are valuable. Marketers often do base promotional appeals on the principle of scarcity to increase the subjective desirability their products among consumers. Specially, advertisers and retailers often promote their products using restrictions. These restriction act to constraint consumers' ability th take advantage of the promotion and can assume several forms. For example, some promotions are advertised as limited time offers, while others limit the quantity that can be bought at the deal price by employing the statements such as 'limit one per consumer,' 'limit 5 per customer,' 'limited products for special commemoration celebration,' Some retailers use statements extensively. A recent weekly flyer by a prominent retailer limited purchase quantities on 50% of the specials advertised on front page. When consumers saw these phrase, they often infer value from the product that has limited availability or is promoted as being scarce. But, the past researchers explored a direct relationship between the purchase quantity and time limit on deal purchase intention. They also don't explored that all restriction message are not created equal. Namely, we thought that different restrictions signal deal value in different ways or different mechanism. Consumers appear to perceive that time limits are used to attract consumers to the brand, while quantity limits are necessary to reduce stockpiling. This suggests other possible differences across restrictions. For example, quantity limits could imply product quality (i.e., this product at this price is so good that purchases must be limited). In contrast, purchase preconditions force the consumer to spend a certain amount to qualify for the deal, which suggests that inferences about the absolute quality of the promoted item would decline from purchase limits (highest quality) to time limits to purchase preconditions (lowest quality). This might be expected to be particularly true for unfamiliar brands. However, a critical but elusive issue in scarcity message research is the impacts of a inferred motives on the promoted scarcity message. The past researchers not explored possibility of inferred motives on the scarcity message context. Despite various type to the quantity limits message, they didn't separated scarcity message among the quantity limits. Therefore, we apply a stricter definition of scarcity message(i.e. quantity limits) and consider scarcity message type(general scarcity message vs. special scarcity message), scarcity depth(high vs. low). The purpose of this study is to examine the effect of the scarcity message on the consumer's purchase intension. Specifically, we investigate the effect of general versus special scarcity messages on the consumer's purchase intention using the level of the scarcity depth as moderators. In other words, we postulates that the scarcity message type and scarcity depth play an essential moderating role in the relationship between the inferred motives and purchase intention. In other worlds, different from the past studies, we examine the interplay between the perceived motives and scarcity type, and between the perceived motives and scarcity depth. Both of these constructs have been examined in isolation, but a key question is whether they interact to produce an effect in reaction to the scarcity message type or scarcity depth increase. The perceived motive Inference behind the scarcity message will have important impact on consumers' reactions to the degree of scarcity depth increase. In relation ti this general question, we investigate the following specific issues. First, does consumers' inferred motives weaken the positive relationship between the scarcity depth decrease and the consumers' purchase intention, and if so, how much does it attenuate this relationship? Second, we examine the interplay between the scarcity message type and the consumers' purchase intention in the context of the scarcity depth decrease. Third, we study whether scarcity message type and scarcity depth directly affect the consumers' purchase intention. For the answer of these questions, this research is composed of 2(intention inference: existence vs. nonexistence)${\times}2$(scarcity type: special vs. general)${\times}2$(scarcity depth: high vs. low) between subject designs. The results are summarized as follows. First, intention inference(inferred motive) is not significant on scarcity effect in case of special scarcity message. However, nonexistence of intention inference is more effective than existence of intention inference on purchase intention in case of general scarcity. Second, intention inference(inferred motive) is not significant on scarcity effect in case of low scarcity. However, nonexistence of intention inference is more effective than existence of intention inference on purchase intention in case of high scarcity. The results of this study will help managers to understand the relative importance among the type of the scarcity message and to make decisions in using their scarcity message. Finally, this article have several contribution. First, we have shown that restrictions server to activates a mental resource that is used to render a judgment regarding a promoted product. In the absence of other information, this resource appears to read to an inference of value. In the presence of other value related cue, however, either database(i.e., scarcity depth: high vs. low) or conceptual base(i.e.,, scarcity type special vs. general), the resource is used in conjunction with the other cues as a basis for judgment, leading to different effects across levels of these other value-related cues. Second, our results suggest that a restriction can affect consumer behavior through four possible routes: 1) the affective route, through making consumers feel irritated, 2) the cognitive making route, through making consumers infer motivation or attribution about promoted scarcity message, and 3) the economic route, through making the consumer lose an opportunity to stockpile at a low scarcity depth, or forcing him her to making additional purchases, lastly 4) informative route, through changing what consumer believe about the transaction. Third, as a note already, this results suggest that we should consider consumers' inferences of motives or attributions for the scarcity dept level and cognitive resources available in order to have a complete understanding the effects of quantity restriction message.
