Modulation of Cell Proliferation and Hypertrophy by Gangliosides in Cultured Human Glomerular Mesangial Cells

  • Glomerular mesangial cells (GMCs) in diverse renal diseases undergo cell proliferation and/or hypertrophy, and gangliosides have been reported to play an important role in modulating cell structure and function. This study compared the effects of transforming growth $factor-\beta\; (TGF­\beta1)$ and the effects of the application of exogenous gangliosides on GMCs and investigated whether the application of exogenous gangliosides regulated cellular proliferation and hypertrophy. Human GMCs were cultured with exogenous gangliosides and $TGF-\beta1$ in a media containing $10\%$ fetal bovine serum and in a media without the fetal bovine serum. Exogenous gangliosides biphasically changed the proliferation of human GMCs (0.1-1.0 mg/mL). A low concentration (0.1 mg/mL) of gangliosides mainly increased the number of human GMCs, whereas cellular proliferation was significantly reduced by raising the concentration of exogenous gangliosides. $TGF-\beta1$ greatly reduced the number of human GMCs in a concentration­dependent manner (1-10 ng/mL). Serum deprivation accelerated the gangliosides- and $TGF­\beta1-induced$ inhibition of mesangial cell proliferation to a greater extent. Gangliosides (1.0 mg/ mL) and $TGF-\beta1$ (10 ng/mL) both caused a significant increase in the incorporation of $[^3H]leucine$ per cell in the serum-deprived condition, whereas it was completely reversed in serum­supplemented condition. Similar results to the $[^3H]leucine$ incorporation were also observed in the changes in cell size measured by flow cytometric analysis. These results show that exogenous gangliosides modulate cell proliferation and hypertrophy in cultured human GMCs, and these cellular responses were regulated differently based on whether the media contained serum or not. Results from the present study raise new possibilities about the potential involvement of gangliosides in the development of mesangial cell proliferation and hypertrophy.

Preliminary Report of Use-Effectiveness of Ovulation Method in Korea (자연피임법으로서의 배란법의 피임효과)

  • In recent years, explosive increase in population has been damaging mankind in terms of deprivation of natural resources and more of economical demands. Therefore, we have thought about problems as to counter-balance the increasing population, and reached a resolution of artificial methods of controlling birth. In the past, though now used in some of contraceptive methods, extravaginal ejaculation, condom were commonly used. But recently, pills, IUD and several kinds of operative procedures are quite popular. Though the recent methods are known to be effective compared with the traditional methods, a certain unwanted side effects as well as limited value of usages now must be discussed. On this aspect, we are trying to research for a ideal methods such as symptom free, more of natural way of family planning and try to zero the failure rate. And also it has been suggested that only if those scientific methods of controlling birth can be base on religious concepts of moral being, it will be enlightened. At the Happy Family Planning Clinic of St. Mary's Hospital, we apply Billing's ovulation method to out-patients who want contraception and usually advise them to use it on a self-care basis. For a retrospective study of the presurvey data analysis and use-effectiveness of the ovulation method from April 1, 1975 to Nov., 30, 1975, we have dealt with a total of 1,383 women (urban areas 465, rural areas 918). The results of preliminary survey were as follows; 1) Among 465 women in Seoul areas, the failure rate was 10.3, which signified unplanned pregnancies of 32 women. 27 of the 32 women were pregnant due to the users own failure and the remaing 5 due to the failure of the method. Therefore, the failure of the method accounted for 1.6. 2) Among 918 women in rural areas, the failure rate was 15.2 signifing unplanned pregnancies in 93 women. The cause of the failure in 81 of the 93 women was attributable to the user's own mistake and that in the remaing 12 to the default of the method. Therefore, the failure attributable to the method accounted for 2.0.

Myxococcus xanthus socD500 mutation causes Sporulation and Induction of two C-signal Specific Genes (Myxococcus xanthus socD500에 의한 포자 형성 및 CsgA신호에 특이적 유전자의 발현에 관한 연구)

  • Myxococcus xanthus is a Gram negative, rod-shaped, soil bacterium that displays a social behaviors, and multicellular development upon nutrient deprivation. The csgA gene encoding a cell surface protein is essential for developmental behaviors including rippling, aggregation, fruiting body formation and sporulation. csgA mutants show normal vegetative growth, but lack all these developmental phenotypes. Expression of the CsgA (C-signal) specific genes are eliminated or dramatically reduced in csgA mutants. In order to identify components of C-signal transduction pathway, second site mutations were introduced into csgA mutants and were identified which can fully or partially restore development of csgA mutants (Rhie, H. G. et. al. 1989. J. Bacteriol. 171, 3268-3276). One of such csgA suppressor mutations, socD500 restores only sporulation to csgA mutants at 15$^{\circ}C$. The socD500 mutaion however eliminates the three basic developmental requirements, starvation, high cell density and a solid surface. Only sporulation, not accompanied with fruiting body formation is induced simply by shifting the temperature of vegetatively growing cells from $32^{\circ}C$ to $15^{\circ}C$. Spores induced by socD500 mutation is not as thick as that of wild-type fruiting body. In socD500 genetic background, two of ten C-signal dependent genes, $\Omega$DK4506 and $\Omega$DK4406 are more highly expressed in growing cells at $15^{\circ}C$. These results indicate that the socD500 mutation may be partly involved in the regulation of expression of two C-signal dependent genes and genes for sporulation in this transduction pathway.

A Study on Records Management of Public Enterprises Supervised by National Archives of Korea (국가기록원 직접관리기관 공기업의 기록관리에 관한 연구)

  • This study examined Records Management(RM) practice of public enterprises supervised by National Archives of Korea(NAK). These enterprises are included among public institutions mandated to perform RM according to Public Records Management Act of 2006. The study performed telephone interviews of 8 records managers employed in public enterprises and investigated current status of RM practices, perception of the practices and records managers within the enterprises and treatment to them, as well as opinions of the NAK's assessment of RM in public institutions, While non-electronic records were managed in an appropriate manner, electronic records needed to be transferred, disposed of, and preserved properly. The majority of interviewees expressed positive opinions on the contribution of RM to effectiveness and transparency of the enterprises. However, RM was perceived as irksome work conducted by those in a low position. More interviewees were permanent contract workers or contract personnel than regular employees. They felt a sense of deprivation due to the discrepancy of annual incomes among public enterprises and the differences in treatment and attitude toward between contract workers and regular workers. Interviewees suggested that NAK's assessment of RM was useful since it provided a basis for RM in their institutions. Yet, they pointed out that its measurements were lack of consistency and relevance to public enterprises because those focused on RM in governmental organizations. They also noted that RM practices that the assessment do not cover might be ignored.

Neuroprotective and Anti-Oxidative Effect of Puerariae Radix on Hippocampal Neurons and BV-2 Microglia Cells (갈근(葛根)의 뇌해마(腦海馬) 신경세포 손상보호와 항산화(抗酸化) 효능에 대한 연구)

  • This study demonstrated neuroprotective and anti-oxidative effects of Puerariae Radix for cerebral ischemia. Neuroprotective effects were studied by using oxygen/glucous deprivation of the organotypic hippocampal slice cultures to complement limitations of in vivo and in vitro models for cerebral ischemia study. Anti-oxidative effects were studied on BV-2 microglia cells damaged by $H_2O_2$ and nitric oxide. The results obtained are as follows; The groups treated with 0.5 and $5{\mu}g/m{\ell}$ of Puerariae Radix revealed significant decreases of neuronal cell death area and cell death area percentages in CA1 region of ischemic damaged hippocampus cultures during whole 48 hours of the experiment. The groups treated with 0.5 and $5{\mu}g/m{\ell}$ of Puerariae Radix revealed significant decreases of neuronal cell death area and cell death area percentages in DG region of ischemic damaged hippocampus cultures during whole 48 hours of the experiment. The groups treated with 0.5 and $5{\mu}g/m{\ell}$ of Puerariae Radix revealed significant decreases of TUNEL-positive cells in both CA1 region and DG region of ischemic damaged hippocampus cultures. The group treated with $50\;{\mu}g/m{\ell}$ of Puerariae Radix demonstrated significant decrease of TUNEL-positive cells in CA1 region. The groups treated with 0.5 and $5{\mu}g/m{\ell}$ of Puerariae Radix revealed significant decreases of LDH concentrations in culture media of ischemic damaged hippocampus cultures. The groups treated with 0.5 and $5{\mu}g/m{\ell}$ of Puerariae Radix revealed significant increases of cell viabilities of BV-2 microglia cells damaged by $H_2O_2$. The group treated with $5{\mu}g/m{\ell}$ of Puerariae Radix revealed significant increase of cell viability of BV-2 microglia cells damaged by nitric oxide. These results suggested that Puerariae Radix of cerebral ischemic revealed neuroprotective effects through the control effect of apoptosis and oxidative damages.

EGCG induces Apoptosis under Hypoxic State in B16F10 Melanoma Cancer Cells (저산소증 상태에서 B16F10 피부암 세포에 EGCG를 처리하였을 때의 apoptosis 효과)

  • EGCG, catechins in green tea, is a kind of phytochemical. Through the regulation of signal pathways, EGCG has been known to show anti-oxidant and anti-tumor effects in cells. In this study, we investigated the apoptotic effects of EGCG through AMP-activated protein kinase (AMPK) signal pathways, including hypoxia inducible factor-1 alpha (HIF-$1{\alpha}$). The experiments were performed in B16F10 melanoma cells in a hypoxic state. AMPK is activated by ATP consumption such as nutrient deficiency, exercise, heat shock, etc. The activated AMPK that plays an important role as an energy sensor inhibits proliferation of cancer cells, as well as inducing apoptosis. HIF-$1{\alpha}$, the primary transcriptional regulator of the response to oxygen deprivation, plays a critical role in modulating tumor growth and angiogenesis in a hypoxic state. The apoptotic effects of EGCG were studied in B16F10 cells in a hypoxic state. The results show that EGCG inhibits the transcriptional activity of HIF-$1{\alpha}$ and induces apoptosis. These observations suggest that EGCG may exert inhibitory effects of angiogenesis and control tumor cell growth in hypoxic melanoma cells.

The Study of the Correction and Perceived Social Support by the Juvenile Delinquents in the Juvenile Justice Process (소년사법처리과정에 있는 청소년이 지각한 사회적 지지와 교정성에 관한 연구)

  • The purpose of this study is to investigate correction-level associated with the social support perceived by juvenile delinquents. To accomplish these purposes, this study used sample of 546 juvenile delinquents who are in the process of the beginning stage in the police and prosecution. Social support and correction were assessed with an instrument designed for this study. Juvenile delinquents completed instruments assessing the following variables: (1) perceived social support scale, specially social support types(esteem support, emotional support, informational support) and social support providers (parents, relatives, friends, teachers, community) (2) correction scale, specially social stigma, social deprivation, self reflection, legal consciousness, self concept. The resulting scale of the instrument had good internal reliability(Cronbach's alpha=. ) and was scored so that high scores indicated a willingness to access. To examine this purpose, One way ANOVA, multiple regression analysis were conducted. And the results are as follows: The relationship ratings of the correction factors were positively associated with their rating of the type of social support. Informational support was positively associated with self reflection and esteem support was positively associated with self concept, legal consciousness, self reflection and emotional support were positively associated with esteem. The type of support according to support providers was significant, explaining 25% of the variance in self concept scores. The significant predictor was esteem support from friends and emotional support from a community. The type of support according to support providers was significant, explaining 20% of the variance in legal consciousness scores. The significant predictor was informational support from teachers and esteem support from a community. The type of support according to support providers was significant, explaining 21% of the variance in self reflection scores. The significant predictor was esteem support and emotional one from relatives.

Quality of Life of Poor Women - Focused on the Discretionary Time (재량시간(discretionary time)을 중심으로 본 빈곤여성의 삶의 질)

  • The main purpose of this paper is to analyze the quality of life of poor women. This study utilized data combined KLIPS with Time Use Survey for an analysis and analyzed time at the intersection of the class and gender. With discretionary time as center, this study investigated poverty status from the quality of life. And in order to make comparison more clearly, object was classified into 4 groups; non-poor male, poor male, non-poor female and poor female. Study results are as follows; First of all, poor women had the shortest discretionary time among all the groups and also had highest poverty rate of living quality. And this study found that deprivation of non-poor female headed householder is serious. Second, analysis of inequality level between and within groups through Theil index indicated that gender influence in poor strata was 3 times higher than that of non-poor strata. This study found that poor women experienced mixed exclusion at the point where gender and class crossed. And this study also has a meaning that an empirical analysis was conducted through above matters on secondary poverty and hidden poverty of poor women which existing researches were unable to discover.

A Review of Improvements for Providing Safe and Secure Environments for Medical Treatment (안전한 진료환경 구축을 위한 정책 개선과제)

  • On December 31, 2018, an incident occurred where a doctor was attacked and killed by a patient carrying a lethal weapon in the outpatients' clinic of the psychiatric department of a tertiary general hospital. The suspect was diagnosed with bipolar affective disorder (manic depressive disorder) and has been hospitalized and cared for in the psychiatric ward of this hospital. This incident illustrates the necessity of more active cures and therapeutic intervention for mental patients with intellectual developmental disorders who require treatment considering the fact that a radical outcome has been caused by such a patient. However, on the other hand, there is also a need for an approach and analysis from the perspective of crime prevention for all medical departments. The reason for this is that even a tertiary general hospital equipped with the largest human resources, medical devices, facilities, and so forth, is susceptible to violence. As for illegal actions perpetrated against health and medical service personnel in medical institutions, such as verbal abuse, assault, injury, etc. there have neither been understanding shown for the current extent of damage in detail, nor discussions of active institutional improvement related to the seriousness of the act. It can be said that violence in the field of medical treatment is a realm requiring serious discussion and appropriate remedial actions. This is because when such incidents take place, if a patient who is supposed to get treatment from the damaged health care provider is in an urgent situation or on the waiting list of serious cases, he or she could suffer serious damage caused by deprivation of treatment opportunity, or secondary damage might be caused to the patient and/or a guardian who can hardly have an opportunity to take action. Accordingly, in this review, we would like to help create the necessary conditions for both health and medical service personnel and patients/guardians, respectively, to provide and receive medical treatment in a more secure environment. Therefore, objective assessment of the institution and issues relating to this aforementioned incident and general cases of violence occurring in medical institutions, and by suggesting legal and institutional improvements and solutions.

A Comparative Study on Visibility Performance Between Pole Type Lightings and Low-Level Lightings (등주식 조명과 낮은 조명의 성능 비교에 관한 연구)

  • Road lighting is highly preferred as the major nighttime road safety countermeasures. When it comes to the pole-type road lighting, as it's installed at a certain height from the road surface, the driver is dazzled by the light source, interrupting the driver's visibility and furthermore, the light leaks to the area outside the road boundary, worsening the energy efficiency as well as generating the light pollution to the surrounding environment including the animals and plants. The study developed the low-level lighting systems, so it is possible to prevent the lights from being strayed, so there is no any side effects in terms of the sleep deprivation. The study compared the performance as the lightings between low-level lightings and conventional pole based lighting systems. As the results, the low-level lighting systems showed the higher performance in terms of average surface luminance, uniformity, glare compared to the conventional lighting systems.
