Viral Hemorrhagic Septicemia Virus NV Gene Decreases Glycolytic Enzyme Gene Transcription (바이러스성 출혈성 패혈증 바이러스 NV 단백질에 의한 glucokinase 전사 활성의 억제)

  • The viral hemorrhagic septicemia virus (VHSV), which belongs to the Novirhabdovirus genus of the Rhabdoviridae family, is a viral pathogen that causes severe losses in the olive flounder farming industry. Among six encoding VHSV proteins, the non-virion (NV) protein has been shown to have an impact on virulence. In our previous studies, transcriptomics microarray analysis by using VHSV-infected olive flounder showed that VHSV infection significantly down-regulated the mRNA expression of glycolytic enzymes. In addition, VHSV NV protein variants decreased the intracellular ATP level. Based on these results, we have tried to examine the effect of VHSV NV protein on glycolytic enzyme glucokinase expression, which phosphorylates glucose to glucose 6-phosphate. Our results indicated that the NV protein significantly decreased the mRNA expression of glucokinase in olive flounder HINAE cells. Furthermore, the NV protein played a negative role in the promoter activation of glucokinase. Furthermore, glucose uptake was effectively inhibited by VHSV infection and NV protein expression in olive flounder HINAE cells. These results suggest that the VHSV NV protein negatively regulates glycolytic enzyme expression by a transcription level and eventually leads to gradual morbidity of olive flounder through cellular energy deprivation. The present results may be useful for the prevention and diagnosis of VHSV infection in olive flounder.

  • To find out the residual effect of phosphate fertilization on botanical composition, mineral contents and mineral nutrient deprivation of pasture plants, this experiment was arranged as a randomized complete block design with six treatments(0-0, 50-50, 100-65, 150-65, 200-65 and 250-65kg $P_2O_5$/ha), those were composed of three P,O, fertilization level(0, 50 and 65kg P,OJha) after phosphate fertilization trial with six treatments(0, 50, 100, 150, 200 and 250kg $P_2O_5$/ha) from 1989 to 1992, and conducted at hilly land in Kwangju, Kyonggi Province 60m 1993 to 1994. The results obtained are summarized as follows: The botanical composition of pasture plants without phosphate fertilization was very poor. With increasing available phosphate of soil, the percentage of grasses and legumes were remarkably increased and weeds was decreased. With 150-65kg $P_2O_5$/ha application for two years, the average DM yield was composed of 80.6% grasses, 15.4% legume and 4.0% weeds, the percentage was similar to that of 200-65kg $P_2O_5$/ha. As available phosphate of soil increase, P, K and Mg content of pasture plants were increased, but Ca/P ratio was lowed and the other mineral contents tended not to be regular. The mineral nutrient contents deprived by DM yield was increased as available phosphate of soil was increased, but those were not different between 150-65 and 250-65kg $P_2O_5$/ha. However, mineral nutrient of $P_2O_5$,$K_2O$ and MgO were higher in 200-65 and 250-65kg $P_2O_5$/ha. Most of applied mineral elements were accumulated at a subsurface(0-2.5cm) of grasslands, and mineral tended to be decrease than that of soil chemical analysis before the experiment of residual effect of phosphate fertilization.

  • Purpose: Heat shock proteins (HSPs) are a group of proteins that are activated when cells are exposed to a variety of environmental stresses, such as infection, inflammation, exposure to toxins, starvation, hypoxia, brain injury, or water deprivation. The activation of HSPs by environmental stress plays a key role in signal transduction, including cytoprotection, molecular chaperone, anti-apoptotic effect, and anti-aging effects. However, the precise mechanism for the action of small HSPs, such as HSP27 and mitogen-activated protein kinases (MAPKs: extracellular-regulated protein kinase 1/2 (ERK1/2), p38MAPK, stress-activated protein kinase/c-Jun N-terminal kinase (SAPK/JNK), is not completely understood, particularly in application of cell stimulators including platelet-derived growth factor (PDGF), angiotensin II (AngII), tumor necrosis factor $\alpha$ (TNF$\alpha$), and $H_2O_2$. This study examined the relationship between stimulators-induced enzymatic activity of HSP27 and MAPKs from rat smooth and skeletal muscles. Methods: 2-dimensional electrophoresis (2DE) and matrix assisted laser desorption ionizationtime-of-flight/time-of-flight (MALDI-TOF/TOF) analysis were used to identify HSP27 from the intact vascular smooth and skeletal muscles. Three isoforms of HSP27 were detected on silver-stained gels of the whole protein extracts from the rat aortic smooth and skeletal muscle strips. Results: The expression of PDGF, AngII, TNF$\alpha$, and $H_2O_2$-induced activation of HSP27, p38MAPK, ERK1/2, and SAPK/JNK was higher in the smooth muscle cells than the control. SB203580 (30${\mu}$M), a p38MAPK inhibitor, increased the level of HSP27 phosphorylation induced by stimulators in smooth muscle cells. Furthermore, the age-related and starvation-induced activation of HSP27 was higher in skeletal muscle cells (L6 myoblast cell lines) and muscle strips than the control. Conclusion: These results suggest, in part, that the activity of HSP27 and MAPKs affect stressors, such as PDGF, AngII, TNF$\alpha$, $H_2O_2$, and starvation in rat smooth and skeletal muscles. However, more systemic research will be needed into physical therapy, including thermotherapy, electrotherapy, radiotherapy and others.

Anti-Oxidative and Neuroprotective Effects of Rhei Rhizoma on BV-2 Microglia Cells and Hippocampal Neurons (대황(大黃)의 항산화와 신경세포손상 보호효능에 대한 연구)

  • This study demonstrated anti-oxidative and neuroprotective effects of Rhei Rhizoma. Anti-oxidative effects were studied on BV-2 microglia cells damaged by $H_2O_2$ and nitric oxide. Neuroprotective effects were studied by using oxygen/glucose deprivation of the organotypic hippocampal slice cultures. The results obtained are as follows; The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of neuronal cell death area and cell death area percentages in CA1 region of ischemic damaged hippocampus cultures during whole 48 hours of the experiment. The group treated with 50 mg/ml of Puerariae Radix demonstrated decreases of neuronal cell death area and cell death area percentages in CA1 region, but these were not significant statistically. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of neuronal cell death area and cell death area percentages in dentate gyrus of ischemic damaged hippocampus cultures during whole 48 hours of the experiment. The group treated with 50 mg/ml of Puerariae Radix demonstrated decreases of neuronal cell death area and cell death area percentages in dentate gyrus, but these were not significant statistically. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of TUNEL-positive cells in both CA1 region and dentate gyrus of ischemic damaged hippocampus cultures. The group treated with 50 mg/ml of Puerariae Radix demonstrated significant decrease of TUNEL-positive cells in CA1 region, but not in dentate gyrus of ischemic damaged hippocampus. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant decreases of LDH concentrations in culture media of ischemic damaged hippocampus cultures. The group treated with 50 mg/ml of Puerariae Radix demonstrated decrease of LDH concentrations in culture media, but it was not significant statistically. The groups treated with 0.5 and 5 mg/ml of Puerariae Radix revealed significant increases of cell viabilities of BV-2 microglia cells damaged by $H_2O_2$. The group treated with 50 mg/ml of Puerariae Radix demonstrated increase of cell viability of BV-2 microglia cells, but it was not significant statistically. The group treated with 0.5 mg/ml of Puerariae Radix revealed significant increase of cell viability of BV-2 microglia cells damaged by nitric oxide. The groups treated with 5 and 50 mg/ml of Puerariae Radix demonstrated increases of cell viabilities of BV-2 microglia cells, but these were not significant statistically. These results suggested that Puerariae Radix revealed neuroprotective effects through the control effect of apoptosis and oxidative damages.

Overexpression of CD44 Standard Isoform Upregulates HIF-1α Signaling in Hypoxic Breast Cancer Cells

  • Cluster of differentiation 44 (CD44), a cell surface receptor for hyaluronic acid (HA), is involved in aggressive cancer phenotypes. Herein, we investigated the role of the CD44 standard isoform (CD44s) in hypoxia-inducible $factor-1{\alpha}$ ($HIF-1{\alpha}$) regulation using MCF7 overexpressing CD44s (pCD44s-MCF7). When pCD44s-MCF7 was incubated under hypoxia, levels of $HIF-1{\alpha}$, vascular endothelial growth factor, and the $HIF-1{\alpha}$ response element-derived luciferase activity were significantly increased compared to those in the control MCF7. Incubation of pCD44s-MCF7 cells with HA further increased $HIF-1{\alpha}$ accumulation, and the silencing of CD44s attenuated $HIF-1{\alpha}$ elevation, which verifies the role of CD44s in $HIF-1{\alpha}$ regulation. In addition, the levels of phosphorylated extracellular signal-regulated kinase (ERK) was higher in hypoxic pCD44s-MCF7 cells, and $HIF-1{\alpha}$ accumulation was diminished by the pharmacological inhibitors of ERK. CD44s-mediated $HIF-1{\alpha}$ augmentation resulted in two functional outcomes. First, pCD44s-MCF7 cells showed facilitated cell motility under hypoxia via the upregulation of proteins associated with epithelial-mesenchymal transition, such as SNAIL1 and ZEB1. Second, pCD44s-MCF7 cells exhibited higher levels of glycolytic proteins, such as glucose transporter-1, and produced higher levels of lactate under hypoxa. As a consequence of the enhanced glycolytic adaptation to hypoxia, pCD44s-MCF7 cells exhibited a higher rate of cell survival under hypoxia than that of the control MCF7, and glucose deprivation abolished these differential responses of the two cell lines. Taken together, these results suggest that CD44s activates hypoxia-inducible $HIF-1{\alpha}$ signaling via ERK pathway, and the $CD44s-ERK-HIF-1{\alpha}$ pathway is involved in facilitated cancer cell viability and motility under hypoxic conditions.

The Metabolic Effects of FGF21: From Physiology to Pharmacology (생리, 약학적 관점에서 fibroblast growth factor 21 (FGF21)의 대사 효과 고찰)

  • Fibroblast growth factor 21 (FGF21) is an atypical member of the FGF protein family which is highly synthesized in the liver, pancreas, and adipose tissue. Depending on the expression tissue, FGF21 uses endo- or paracrine features to regulate several metabolic pathways including glucose metabolism and energy homeostasis. Different physiologically stressful conditions such as starvation, a ketogenic diet, extreme cold, and mitochondrial dysfunction are known to induce FGF21 synthesis in various tissues to exert either adaptive or defensive mechanisms. More specifically, peroxisome proliferator-activated receptor gamma and peroxisome proliferator-activated receptor alpha control FGF21 expression in adipose tissue and liver, respectively. In addition, the pharmacologic administration of FGF21 has been reported to decrease the body weight and improve the insulin sensitivity and lipoprotein profiles of obese mice and type 2 diabetes patients meaning that FGF21 has attracted huge interest as a therapeutic agent for type 2 diabetes, obesity, and non-alcoholic fatty liver disease. However, understanding FGF21 remains complicated due to the paradoxical condition of its tissue-dependent expression. For example, nutrient deprivation largely increases hepatic FGF21 levels whereas adipose tissue-derived FGF21 is increased under feeding condition. This review discusses the issues of interest that have arisen from existing publications, including the tissue-specific function of FGF21 and its action mechanism. We also summarize the current stage of a clinical trial using several FGF21 analogs.

A Case of Primary Pulmonary Histiocytosis-X Associated with Central Diabetes Insipidus (중추성 요붕증을 동반한 원발성 폐 조직구종 X 1예)

  • Pulmonary histiocytosis X is a granulomatous disorder of the lung of unknown cause. Patients with this disease often complain of cough, dyspnea on exertion and, occasionally, chest pain from pneumothorax or bone involvement. However, DI is uncommon in these patients. We report a case of primary pulmonary histiocytosis X with central diabetes insipidus. A 23-year-old man presented with dyspnea suffered from dry cough, exertional dyspnea, polydipsia and polyuria for 4 months. He was a heavy smoker. He was found to have reticulonodular interstitial opacities on chest X-ray film. High-resolution computed tomography revealed thin-walled cysts of various sizes in both lungs. Open lung biopsy was done. On light microscopic examination revealed proliferation and infiltration of Langerhans cells. Immunohistochemically, Langerhans cells showed strong cytoplasmic staining with S-100 protein and electronmicroscopic examination showed Birbeck granules in Langerhans cells. Water deprivation test showed central-type diabetes insipidus and brain MRI showed no abnormal lesion on suprasellar region. Smoking cessation was recommended. He was treated with oral desmopressin.

A study on the Institutional Characteristics of Artisanal Fisheries (입어관행과 관행어업의 제도적 성격에 관한 고찰)

  • This study is concerned with the identification of the concepts and characteristics of the traditional access of small scale fishery and traditional small scale fishery based on the classification of fisheries under Fisheries Law, As we called, these fisheries are the small scale of artisanal fisheries. These traditional small scale of artisanal fisheries have been developed throughout the most coastal fisheries but have never been included the institutional fishery system and policy since industrial fisheries have been exploited. By this reason there was no studies to understand and identify these fisheries as fisheries under category of institutional fishery system. Recently by the Fisheries Law, my Person who desires to operate these fisheries, shall report to the provincial governor and then these fisheries were incorporated into the category of fisheries to be reported. But this case of the administrative practices should be not consistent with the classification of fisheries under Fisheries Law. The traditional access of small scale fishery is a derivative concept of fishery categories under the institutional fakery system Because this fishery under institutional fishery system can not be establish the right of fishing by fishery right system technically but the right of fishing is supported by the fishing access system only. Therefore, It is a mistake for the provincial fishery governor to adopt such a policy that the fishing right of the traditional access of small scale fishery which is different from the fishing based right is restricted by the factors of fisheries to be reported. On the other hand the traditional small scale fishery is coincident with the traditional access of small scale fishery and the common fishery under the style of utilization of fishing gear and fishing gear and methods. But this fishery has never been included the fishery categories supported by the government subsidy policies and considerations and has been remained with indifference during the last few decades. Anyhow the fishing right of these fisheries have been assured under the institutional fishery system, any person who desires to operate this fishery should have a fishing right against the public or private uses of fishing ground such as land reclamations and other coastal zone development. A deprivation of the fishing right of this fishery by the conflicts of these public and private uses of fishing ground is not regarded as appropriate according to the social welfare policy. Also it is the administrative expediency to support the fishing right of this fishery under the fisheries lobe reported.

Media Violence and Delinquent Behavior (폭력매체와 청소년 비행)

  • The effects of media violence on the delinquent behavior and violence of children and adolescent are controversial. However, a small but genuine association appears to exist between media violence and aggression or violence. At the present, for a considerable proportion of the population of children and adolescent, delinquent behavior and violence has become a major problem and a way of life. One factor contributing to this problem has been assumed to be the negative influence of mass media including television, drama, videotapes and fiction magazines and so on. Therefore, this paper is intended to discriminate the causal relationship of influence of mass media and juvenile delinquent behavior and violence and to provide nine hypotheses derived from reviewing the literature related to mass media and delinquent behavior. The nine hypotheses are as follows : 1. The presence of modeling mass media crime themes and the extent of interest in and exposure of children and adolescent to media violence themes will be positively correlated to their delinquent(aggressive) behavior. 2. A higher positive correlation will be revealed between interest in and exposure to media violence themes and aggressive behavior among adolescent having parental rejection than among adolescent not experiencing parental rejection. 3. A higher positive relationship will be found between interest in and exposure to mass media crime themes and delinquent behavior among youth having need deprivation than among youth not having need frustration. 4. A higher positive relationship will be presented between the presence of imitating mass media crime themes and interest in and exposure to media violence themes and delinquent or aggressive behavior among youth dysplaying their maladaptive character tendency than among those adolescent dysplaying adaptive character tendency. 5. A higher positively correlating relationship will be shown between interest in and exposure to mass media crime themes and delinquent behavior among youth with a higher scores of depressive trend than among those youth having few or no depression. 6. A higher positive relationship will be found between interest in and exposure to media violence themes and aggressive behavior among adolescent complaining of a high degree of psychosomatic complaints than among those youth having few or no psychosomatic complaints. 7. A higher positive correlation will be appeared between interest in and exposure to mass media crime themes and delinquent behavior among youth displaying aggressive impulsiveness than among those youth having few or no aggressive impulsiveness. 8. A higher positive relationship will be found between interest in and exposure to media violence themes and aggressive behavior among youth having antisocial character or neurotic character with weak ego functioning than among those adolescent not having antisocial character or neurotic character with weak ego functioning. 9. A higher positive correlation will be existed between interest in and exposure to mass media crime themes and delinquent behavior among adolescent displaying the lack of sociality than among those youth not having the lack of sociality. The above nine hypotheses will be tested by statistical methods including Chi-square test, simple correlation, principal component analysis, principal component regression analysis and LISREL path analysis.

Pulmonary Langerhans Cell Histiocytosis in an Adult Male Presenting with Central Diabetes Insipidus and Diabetes Mellitus: A Case Report

  • Pulmonary Langerhans cell histiocytosis is an uncommon diffuse cystic lung disease in adults. In rare cases, it can involve extrapulmonary organs and lead to endocrine abnormalities such as central diabetes insipidus. A 42-year-old man presented with polyphagia and polydipsia, as well as a dry cough and dyspnea on exertion. Magnetic resonance imaging of the hypothalamic-pituitary system failed to show the posterior pituitary, which is a typical finding in patients with central diabetes insipidus. This condition was confirmed by a water deprivation test, and the patient was also found to have type 2 diabetes mellitus. Computed tomographic scanning of the lungs revealed multiple, irregularly shaped cystic lesions and small nodules bilaterally, with sparing of the costophrenic angles. Lung biopsy through video-assisted thoracoscopic surgery revealed pulmonary Langerhans cell histiocytosis. On a follow-up visit, only 1 year after the patient had quit smoking, clinical and radiological improvement was significant. Here, we report an uncommon case of pulmonary Langerhans cell histiocytosis that simultaneously presented with diabetes insipidus and diabetes mellitus.
