The experimental study was made to propose the treatment method of wastewater in the high-density fish culture system. The BOD to COD ratios of effluents were almost same to 0.65 in the eel-farm, but were various in the farm rearing together with tilapia etc. A BOD rate curve of the eel-farm effluent could be described mathematically by the equation, $BODu=14.1(1-10^{-0.222t})+30.9(1-10^{-0.035(t-8)})$. Nitrification in Biological Fluidized Bed(BFB) system to treat the fish-farm wastewater could be reduce ammonium level up to $65{\sim}79\%$ when ammonium loading rates were between 0.014 and 0.075g $NH_4/g$ BVS-day. Nitrification efficiency was decreased by organic matters in the wastewater when ammonium loading was low(0.014 g $NH_4/g$ BVS-day). T-N removal ratios were decreased to increase loading in denitrification process, because of low C/N ratio. Based on much higher biological mass concentrations, BFB system takes many advantages of a practical viewpoint, such as stability of treatment efficiency and reduction of necessary site area for the facility, as compared with conventional treatment systems.

Purpose: The measurement of radiosensitivity of individuals is useful in radiation therapy. Unfortunately, the measurement of radiation survival using a clonogenic assay, which is the established standard, can be difficult and time consuming. The aim of this study is to compare radiosensitivity results obtained from the MTT and clonogenic assays, and to evaluate whether the MTT assay can be used on clinical specimens. Materials and Methods: HCT-8, LoVo, CT-26, and WiDr were the colon cancer cell lines used for this study. The clonogenic assay was performed to obtain the cell survival curves and surviving fractions at a dose of 2 Gy ($SF_2$) as the standard technique for radiosensitivity. Also, the MTT assay was performed for each of the cell lines (in vitro). To simulate clinical specimens, the cell lines were inoculated into nude mice, removed when the tumors reached 1 cm in diameter, and chopped. Next, the tumors were subjected to the same process involved with the MTT assay in vitro. The inhibition rates (IR) of 10 Gy or 20 Gy of irradiation for in vitro and ex vivo were calculated based on the optical density of the MTT assay, respectively. Results: According to $SF_2$ and the cell survival curve, the HCT-8 and WiDr cell lines were more resistant to radiation than LoVo and CT-26 (p<0.05). The IR was measured by in vitro. The MTT assay IR was 17.3%, 21%, 30% and 56.5% for the WiDr, HCT-8, LoVo and CT-26 cell lines, respectively. In addition, the IR measured ex vivo by the MTT assay was 23.5%, 26%, 38% and 53% in the HCT-8, WiDr, LoVo and CT-26 tumors, respectively. Conclusion: The radiosensitivity measured by the MTT assay was correlated with the measures obtained from the clonogenic assay. This result highlights the possibility that the MTT assay could be used in clinical specimens for individual radiosensitivity assays.

An absorbed dose calculation method using a digital phantom is implemented in normal organs. This method cannot be employed for calculating the absorbed dose of tumor. In this study, we measure the S-value for calculating the absorbed dose of each organ and tumor. We inject a radioisotope into a torso phantom and perform Monte Carlo simulation based on the CT data. The torso phantom has lung, liver, spinal, cylinder, and tumor simulated using a spherical phantom. The radioactivity of the actual absorbed dose is measured using the injected dose of the radioisotope, which is Cu-64 73.85 MBq, and detected using a glass dosimeter in the torso phantom. To perform the Monte Carlo simulation, the information on each organ and tumor acquired using the PET/CT and CT data provides anatomical information. The anatomical information is offered above mean value and manually segmented for each organ and tumor. The residence time of the radioisotope in each organ and tumor is calculated using the time activity curve of Cu-64 radioactivity. The S-values of each organ and tumor are calculated based on the Monte Carlo simulation data using the spatial coordinate, voxel size, and density information. The absorbed dose is evaluated using that obtained through the Monte Carlo simulation and the S-value and the residence time in each organ and tumor. The absorbed dose in liver, tumor1, and tumor2 is 4.52E-02, 4.61E-02, and 5.98E-02 mGy/MBq, respectively. The difference in the absorbed dose measured using the glass dosimeter and that obtained through the Monte Carlo simulation data is within 12.3%. The result of this study is that the absorbed dose obtained using an image can evaluate each difference region and size of a region of interest.

The present research is focused on the effect of porous graphite what is influenced on the 4H-SiC crystal growth by PVT method. We expect that it produces more C-rich and a change of temperature gradient for polytype stability of 4H-SiC crystal as adding the porous graphite in the growth cell. The SiC seeds and high purity SiC source materials were placed on opposite side in a sealed graphite crucible which was surrounded by graphite insulator. The growth temperature was around $2100{\sim}2300^{\circ}C$ and the growth pressure was 10~30 Torr of an argon pressure with 5~15 % nitrogen. 2 inch $4^{\circ}$ off-axis 4H-SiC with C-face (000-1) was used as a seed material. The porous graphite plate was inserted on SiC powder source to produce a more C-rich for polytype stability of 4H-SiC crystal and uniform radial temperature gradient. While in case of the conventional crucible, various polytypes such as 6H-, 15R-SiC were observed on SiC wafers, only 4H-SiC polytype was observed on SiC wafers prepared in porous graphite inserted crucible. The defect level such as MP and EP density of SiC crystal grown in the conventional crucible was observed to be higher than that of porous graphite inserted crucible. The better crystal quality of SiC grown using porous graphite plate was also confirmed by rocking curve measurement and Raman spectra analysis.

This study examined how much chicken meat was in sausage made with pork. Both real-time polymerase chain reaction (PCR) and internal standard addition were used. Fifty ng of chicken DNA was added to the sausages as an internal standard. The addition of standard DNA increased the amplification efficiency of PCR and confirmed the possibility of quantitative analysis. A QIAamp DNA Micro Kit was used to improve the DNA recovery and amplification efficiency. The density of template DNA and primer were suitable for $3.0{\sim}5.0{\mu}L$ and $0.5{\mu}L$, respectively. Each DNA of pig and chicken was diluted in 10-fold from steps 50 ng to 0.05 ng. The detection limit of both pig and chicken meat was more than 0.05 ng and the correlation coefficient of the standard curve was at least 0.98. The result of the quantitative analysis after heat treatment of 3 samples of pigs and chickens mixed at 70:30 showed a 5.7% difference (64.3:35.7) between the expected value and measured value. The quantitative value was changed by affecting the DNA according to the heat treatment ($70^{\circ}C$, 10 min). An analysis of the pork and chicken content in sausages showed that it was difficult to detect chicken meat and the quantitative value of DNA according to the Ct value was very low. On the other hand, when adding standard material (50 ng of chicken DNA) to the sausages, the Ct value decreased gradually with increasing chicken mixing ratio. Thus, the mixing ratio of chicken in sausages could be estimated.
The effects of light quality and irradiance on the growth of centric diatom, Skeletonema costatum (Jinhae Bay strain) were investigated in the laboratory. At 20$^{\circ}C$ and 30 psu, the irradiance-growth curve showed the maximum growth rate of 1.17 day$^{-1}$ with half-saturation photon flux density (PFD) (K$_s$) of 92.4 $\mu$mol photons $m^{-2}s^{-1}$, $\mu$=1.17 (I-5.28)/(I+81.8), (r=0.98), and a compensation PFD (I$_0$) was 5.28 $\mu$mol photons $m^{-2}s^{-1}$. The 10 equated to a depth of 3$\sim$5 m from March to May, 11 m in June and 4 m from July to September in Jinhae Bay. These responses suggested that irradiance at the depth near the surface layer in Jinhae Bay would provide favorable conditions for S. costatum. To assess the effects of light (i.e. wavelengths) on the growth, nine wave-lengths were used ranging from near ultraviolet to near-infrared supplied by light emitting diode. At an irradiance level of 25 $\mu$mol photons $m^{-2}s^{-1}$, S. costatum grew under wavelengths of 405, 470, 505, 525, 568 and 644 nm, but did not grow at 590 and 623 nm; whereas S. costatum grew at all wavelengths at 100 $\mu$mol photons $m^{-2}s^{-1}$. This implies that S. costatum is likely to grow well in enclosed water bodies where suspended particles absorbs most of the blue wavelengths, and dominated by yellow-orange wavelengths.

Pfiesteria piscicida is one of heterotrophic dinoflagellate having toxic metaboliges, and it is difficult to detect and quantify this dinoflagellate via light microscope due to small size and morphological similarity with Pfiesteria-like dinoflagellate (PLD) species. Alternatively, we developed quantitative real-time PCR assay based on EvaGreen and determined field accessibility throughout the investigation of distribution in the entire Korean coastal waters and population dynamics in Shihwa Lake. The P. piscicida-specific primers based on internal transcribed spacer 1 (ITS 1) were designed and the specificity of primers was confirmed by PCR with other genomic DNAs which have genetic similarity with target species. Through real-time PCR assay, a standard curve which had a significant linear correlation between log cell number and $C_T$ value ($r^2{\geq}0.999$) and one informative melting peak ($88^{\circ}C$) were obtained. These results implies that developed real-time PCR can accurately detect and quantify P. piscicida. Throughout the field applications of real-time PCR assay, P. piscicida was distributed in western (Mokpo and Kimje) and easthern (Gangneng) Korean coastal water even though light microscopy failed to identify P. piscicida. In the investigation of population dynamics in Shihwa Lake, the density of P. piscicida was peaked in June, July and August 2007 at St. 1 where salinity (${\leq}15psu$) was lower than the other 2 sites. In this study, we successed to develop EvaGreen bassed real-time PCR for detection and quantification of P. piscicida in fields, so this developed assay will be useful for various ecological studies in the future.
In Seoul, it has been confirmed that the duration of rainfall is shortened and the frequency and intensity of heavy rains are increasing with a changing climate. In addition, due to high population density and urbanization in most areas, floods frequently occur in flood-prone areas for the increase in impermeable areas. Furthermore, the Seoul City is pursuing various projects such as structural and non-structural measures to resolve flood-prone areas. A disaster prevention performance target was set in consideration of the climate change impact of future precipitation, and this study conducted to reduce the overall flood damage in Seoul for the long-term. In this study, 29 GCMs with RCP4.5 and RCP8.5 scenarios were used for spatial and temporal disaggregation, and we also considered for 3 research periods, which is short-term (2006-2040, P1), mid-term (2041-2070, P2), and long-term (2071-2100, P3), respectively. For spatial downscaling, daily data of GCM was processed through Quantile Mapping based on the rainfall of the Seoul station managed by the Korea Meteorological Administration and for temporal downscaling, daily data were downscaled to hourly data through k-nearest neighbor resampling and nonparametric temporal detailing techniques using genetic algorithms. Through temporal downscaling, 100 detailed scenarios were calculated for each GCM scenario, and the IDF curve was calculated based on a total of 2,900 detailed scenarios, and by averaging this, the change in the future extreme rainfall was calculated. As a result, it was confirmed that the probability of rainfall for a duration of 100 years and a duration of 1 hour increased by 8 to 16% in the RCP4.5 scenario, and increased by 7 to 26% in the RCP8.5 scenario. Based on the results of this study, the amount of rainfall designed to prepare for future climate change in Seoul was estimated and if can be used to establish purpose-wise water related disaster prevention policies.

Diverse native tree species are distributed in Korean forests; however, information on their regeneration strategies is limited. This study analyzed seed rain from 2011 to 2013, seedling emergence, seedling survival, lifespan distribution, and the relationship between seedling mortality and herbaceous vegetation for 3 years, from 2012 to 2014, to understand the natural regeneration and early survival characteristics of Fraxinus rhynchophylla Hance in natural broad-leaved forests. Seed rain and emerged seedling density significantly differed among the years (p < .05). An increase in seed rain was followed by an increase in seedling emergence in the following year. Seed rain in 2013 and seedling emergence in 2014 showed a significant correlation (p < .05), confirming the trait of F. rhynchophylla seeds that germinate in the following year after seed production. Seedlings emerged in late spring and early summer. Of the seedlings, 78% emerged in the early summer of 2014. Most seedlings died immediately after emergence. Of the seedlings that emerged in August 2012, 56% died within 2 months, and 38% of those that emerged in July 2014 died within 1 month. Shade and competition against ground vegetation chiefly affected the seedling survival of F. rhynchophylla. Fraxinus rhynchophylla showed mast seeding, with a large number of seed production in a particular year. A surge of seedling emergence followed in the early summer of the next year. However, most seedlings died in the same year of emergence. Fraxinus rhynchophylla has a high seedling mortality, but mast seeding and the resulting high seedling emergence produce a few survived seedlings, enabling the continuation of the F. rhynchophylla population. Efforts and research on the natural regeneration of native tree species are required to effectively manage natural forests in Korea.

We investigated the effects of water temperature, salinity and irradiance on the growth of the harmful algae Chattonella ovata isolated from South Sea, Korea. C. ovata grew under all combinations of water temperatures and salinity, except for all the salinity conditions at the water temperature of $10^{\circ}C$, with the salinity of 7.5 psu and 10 psu at $15^{\circ}C$, and 7.5 psu at $20^{\circ}C$ and $30^{\circ}C$. The maximum specific growth rate was $0.62\;day^{-1}$ at the combination of $30^{\circ}C$ and 30 psu. The results of two-way ANOVA indicated that growth rate depended greatly on the water temperatures while not being affected by interactions with the salinity. This indicates that C. ovata is a stenothermal and euryhaline organism, preferring high water temperatures. C. ovata did not grow at irradiance ${\leq}30\;{\mu}mol$ photons $m^{-2}s^{-1}$. Photoinhibition did not occur at $800\;{\mu}mol$ photons $m^{-2}s^{-1}$, which was the maximum irradiance used in this study. The irradiance-growth curve was described as $\mu$ = 0.74(I-16.0)/(I+43.9) at $30^{\circ}C$ and 30 psu. The half-saturation light intensity ($K_s$) was $75.9\;{\mu}mol$ photons $m^{-2}s^{-1}$ and compensation photon flux density ($I_c$) was $16.0\;{\mu}mol$ photons $m^{-2}s^{-1}$, especially this value was comparatively lower than those of Skeletonema costatum and other flagellates previously reported. Therefore, our results indicate that C. ovata has advantageous physiological characteristics for interspecific competition at the embayment and coastal areas of Korea in summer.
