Production and Characterization of Nitrate Reductase Deficient Mutants in Petunia parviflora

  • Nitrate reductase deficient (NR) mutant lines were selected indirectly by their resistance to 100mM chlorate in cell cultures of P. parviflora. A total of 585 chlorate resistant lines were confirmed by a second passage on a high concentration of chlorate. Frequency of spontaneous mutation was $9.7{\times}10^{-7}$ in 3 month old suspension-cultured cells, and in non-selective media containing amino acids as sole nitrogen source. The frequency of mutation could be increased up to 11-fold by culture for 12 months. Out of 40 randomly selected calli, 22 were fully deficient in NR. The rest of the clones contained a decreased level of NR activity. Further characterization was carried out in 13 mutant lines which were fully deficient in NR and in 5 mutant lines containing residual (0-7.0%) NR activity, as compared to wild-type cells cultured on the same medium. The $NR^-$ mutants were tentatively classified as defective in the NR apoenzyme (nia-type; 11 mutant lines including the 5 with residual NR activity) or in the molybdenum cofactor (cnx-type; 7 mutant lines) by the XDH activity. The cnx-type could be further classified into two groups. In one group (5 mutant lines) of these, the NR activity could be partially restored by nonphysiologically high (1.0mM) molybdate in the culture medium. Both types of $NR^-$ mutants were unable to grow on minimal medium containing nitrate as sole nitrogen source, but grew well on amino acids. They also proved to be extremely sensitive to the standard medium ($MSP_1$) containing nitrate and ammonium. Shoot regeneration was obtained only in the $NR^-$ mutants, which contained residual NR activity, but they so far have failed to grow into plants.

태생기 및 신생기의 Phosphatidylcholine 보충기 기억력 향상에 미치는 영향 -전뇌기저부의 Choline성 신경세포 활성에 관한 연구- (Evidence of Memory Improvement by Phosphatidylcholine Supplement at Fetus and Neonate -Studies of Basal Forebrain Cholinerge Neuronal Activities-)

  • To investigate the effect of dietary phosphatidylcholine(PPC) supplement on memory improvement, biochemical study on the brain, and morphometric studies on the cholinergic neurons in the rat basal forebrain were undertaken. The pregnancy rats were divided into the normal control, the choline deficient and the PPC supplemental groups according to quantity of the PPC in diet. According to choline deficiency and PPC supplement after birth, the neonate rate of the normal control group were subdivided into the control diet(N-N) and the PPC supplied (N-S) groups, the choline deficient group were subdivided into the continually deficient (D-D), the control diet(D-N) and the PPC supplied groups(D-S), and the PPC supplemental group were subdivided into the control diet (S-N)and the continually supplied (S-S)group. The PPC supplemented diet was added 2% egg PPC in AIN 76 formula diet. PPC concentrations and cholinesterase(CE) activities were measured in the serum, the liver and the brain, respectively. Immunohistochemical stains for choline acetyltransferase(ChAT) was employed for the morphological and morphometric studies. The maze test was undertaken to evaluate memory improvement. PPC concentration and CE activities in the serum, liver and the brain were high in the PPC supplemental groups and low in the choline deficient groups. ChAT immunoreactivity neurons at the medial septal diagonal bond complex and the basal forebrain nucleus of Meynert were reduced in the choline deficient groups. Average failure rate for the maze test was the lowest in the S-S group and the highest in the D-D group. Insufficient choline suppley during the neuronal development would result in cholinergic neuronal damage, which could be prevented by adequate PPC supplement. It is consequently suggested that PPC supplement may be effective on memory improvement by maintaining the cholinergic neuronal activity in the basal forebrain of the rats.

A Study on the Analysis and Identification of Seafarers' Skill-Rule-Knowledge Inherent in Maritime Accidents

  • The purpose of this study is to classify the deficient abilities of seafarers into SRK (Skill, Rule, and Knowledge) and analyze and identify the SRK by the type of accident and ship. Experimental data used the SRK cumulative frequency for 1,606 marine accident records and two-way ANOVA and t-test were used for the analysis tools. The results of two-way ANOVA showed that it is possible to identify the deficient abilities by using the cumulative frequency of SRK in both accident and ship types. As a result of the t-test, the adoption of the null hypothesis (H=0) that the mean of two pairs is equal and the rejection of the null hypothesis (H=1) were 29.2 % and 70.8 %, respectively. For the ship type, H=0 is 33.3 % and H=1 is 66.7 %. Through this study, it was found that about 70 % of the deficient abilities of seafarers inherent in maritime accidents can be identified using the proposed method.

Asymptomatic maternal 3-methylcrotonylglycinuria detected by her unaffected baby's neonatal screening test

  • 3-methylcrotonyl-coenzyme A carboxylase (3MCC) deficiency is an autosomal recessive disorder in which leucine catabolism is hampered, leading to increased urinary excretion of 3-methylcrotonylglycine. In addition, 3-hydroxyisovalerylcarnitine levels increase in the blood, and the elevated levels form the basis of neonatal screening. 3MCC deficiency symptoms are variable, ranging from neonatal onset with severe neurological abnormality to a normal, asymptomatic phenotype. Although 3MCC deficiency was previously considered to be rare, it has been found to be one of the most common metabolic disorders in newborns after the neonatal screening test using tandem mass spectrometry was introduced. Additionally, asymptomatic 3MCC deficient mothers have been identified due to abnormal results of unaffected baby's neonatal screening test. Some of the 3MCC-deficient mothers show symptoms such as fatigue, myopathy, or metabolic crisis with febrile illnesses. In the current study, we identified an asymptomatic 3MCC deficient mother when she showed abnormal results during a neonatal screening test of a healthy infant.

  • Color-normal observers (CNOs) and color-deficient observers (CDOs) have different preferences and emotions for color images. A color-image quality-enhancement algorithm for a CDO is developed to easily adjust images according to each observer's preference or image quality factors. The color-perception differences between CDO and CNO are analyzed and modeled in terms of the YCbCr chroma ratio and hue difference; then the color-shift method is designed to control the degree of color difference.

  • Effects of dammarane glycosides of Panax ginseng on primary cultured chicken embryonic brain cells were studied by microscopic observation and determination of the activity of pyruvate dehydrogenase complex (PDHC). Brain cells were prepared from the brain of 10-day-old chicken embryo and cultured with either a standard medium consisted of 85% Dulbecco's Modified Eagle Medium (DMEM), 10% horse serum and 5% chicken embryonic extracts or a deficient medium consisted of 90% DMEM and 10% horse serum. It was observed that dammarane glycosides of Panax ginseng seemed to show the tendency to stimulate the neurite outgrowth of brain cells which were cultured with a deficient medium under microscopic observation. The activity of PDHC in brain cells cultured with a deficient medium was increased by dammarane glycosides of Panax ginseng.

PbO 과잉 및 결핍이 PZT 세라믹스의 소결 및 전기적 특성에 미치는 영향 (Effect of Excess and Deficiency of PbO on the Sintering and Electrical Properties of PZT Ceramics)

  • Effect of excess and deficiency of PbO on the sintering and electrical properties of tetragonal Pb(Zr0.5Ti0.5)O3 were investigated. Fired density revealed maximum value with 4 wt% of excess PbO, and decreased with increasing amount of both excess and deficient PbO. Grain size increased with excess PbO, and decreased with deficiency of PbO. It appeared that excess PbO forms a liquid phase to enhance grain growth and completely volatilizes after sintering, whereas ZrO2 particles formed in PbO-deficient PZT inhibit grain growth. The change in the values of the equivalent circuit elements of PZT corresponded well with thar of porosity formed by excess and deficiency of PbO; kp, Co and Cm decreased while Qm, Rm and Lm increased conosiderably with porosity. This change was more pronounced in PbO-deficient type probably due to change in Zr/Ti ratio of PZT.

  • Osteoporosis is characterized by bone loss and mobidity with osteoporotic fracture. This study was performed to evaluate the effect of on the bone mass and its related factors in estrogen-deficient animal model. The model rats of osteoporsis showed a significant decrease in bone density, bone ash density, calcium content of femur bone. At the 14th day after ovariectomy-surgery, rats were administered with DBE, extract of Dioscorea batatas, per orally, and continued for 10 weeks. And osteoporosis related parameters were determined to investigate the effect of DBE. Osteoporetic rats showed lower serum estrogen level, higher body weight than normal rats, and showed atrophy of uterine horns. DBE showed inhibitory effect on bone loss in osteoporetic condition, and reduced the increase of ALP activity and osteocalcin level in serum, and reduced the increase of OH-proline level in urine. But, DBE had no effect on cell proliferation and ALP activity in rat calvarial cell culture.

  • The natural history of the anterior cruciate ligament(ACL)-deficient knee remains controversial. although numerous investigation have tried to ascertain the course that the knee would follow once the ACL has lost functional integrity. An extensive review of the literature performed according to symptoms. physical examinations, associated surrounding tissue injuries. returns to activity level and radiological changes in the knee joint. An active individual with a non-functioning ACL was susceptible to injury to the menisci and deteriorate the articular cartilage, followed radiographic changes. An activity levels in general also changed after injury. The most common symptom was pain. But instability varied in these individuals. Conclusively we believe that all these factors will eventually, if not initially, result in a symptomatic knee. which will result in significant limitations to the individual's desired level. So we recommend an aggressive approach in person who desired to return to a relatively active life style in young person as well as in middle aged individuals who have significant symptomatic ACL deficient knee.

Comparison of Nutrient Deficient Stress Diagnoses of Cucumber Plant Using Non-Destructive Physiological Instruments

  • This paper represents methods to diagnose nutrient deficient stresses of cucumber plants using several physiological instruments. The stresses could be detected by measuring and analyzing the difference of chlorophyll content, photochemical efficiency (Fv/Fm), temperature difference (DT) and light absorbance at wavelengths of 480nm, 560nm and 710nm between deficient and control plants. From the all over experiments, the stresses could be first diagnosed in the 3rd to 5th day in general after treatment and the overall diagnosis rate was estimated at more than 50% up to 100%.
