Effects of Jinmutang (JMT) on Hypothyroidism in Rats (진무탕(眞武湯)이 백서의 갑상선기능저하증에 미치는 영향)

  • Objectives : This study was done to investigate the effects of JMT on hypothyroidism in rats. Materials and Methods : After pretreatment with 50mg/kg thiouracil for 30 days, the experimental group was treated with 0.56g/kg JMT orally and the control group was treated with 0.56g/kg of normal saline instead for 20 days. The various indicators related to hypothyroidism were measured, such as T3-uptake content, T3 content, T4 content, TSH content, hematological values, enzyme activity, total cholesterol content, Na+ content change in the serum and body weight of rats induced by thiouracil. Results : The JMT-treated group showed significant increase in comparison with the control group in serum T3-uptake and T4 content. The JMT-treated group also showed significant decrease in comparison with the control group in serum TSH content. However, the JMT-treated group did not show significant increase in comparison with the control group in serum T3 content. The JMT-treated group showed significant increase in comparison with the control group in serum RBC. The JMT-treated group showed significant decrease in comparison with the control group in serum CPK, ALP, LDH content and serum total cholesterol content. The JMT-treated group did not show significant increase in comparison with the control group in serum Na+ content. The JMT-treated group showed significant decrease in comparison with the control group in body weight. Conclusions : These results suggest that JMT is useful in treatment of hypothyroidism.

Combined training improves body composition, balance, and muscle function in sarcopenia elderly

  • Purpose: Sarcopenia is defined as a decrease in muscle mass, strength, and function with age that affects overall body function. We aimed to investigate the effect of combined training on body composition, balance, and muscle function in sarcopenia elderly. Research design, data, and methodology: Twenty-eight sarcopenia elderly (age 74.9±4.5 years) were randomly assigned to an exercise, EG (n=14), or a control, CG (n=14), group. The EG performed an intervention consisting of combined exercise training (60-75 min) for a total of 12 weeks, three times a week. The CG maintained their usual daily lifestyle during the intervention period. We measured body weight, body mass index (BMI), % body fat, free fat mass, balance ability, peak torque in shoulder, knee, and lumbar joints normalized for bodyweight in one second. Results: The EG showed improved body composition (i.e., BMI, fat-free body mass, fat mass; all p < 0.031, η2 > 0.179), balance (i.e., right and left of static and dynamic balance and fast 10 m walk; all p < 0.049, η2 > 0.152), and muscular function (i.e., 90°/sec and 180°/sec peak power per kg bodyweight, 90°/sec average power per kg bodyweight, 180°/sec total work, and 180°/sec endurance ratio; all p < 0.045, η2 > 0.158). Conclusions: Combined exercise training improves muscle mass and strength, body composition, balance, and muscle function in sarcopenia elderly.

Cissus quadrangularis Extracts Decreases Body Fat Through Regulation of Fatty acid Synthesis in High-fat Diet-induced Obese Mice

  • The current study investigated the anti-obesity effect of Cissus quadrangularsis extracts (CQR-300) and its molecular action mechanism on obese mice induced high-fat diet (HFD). To induce the obesity, mice were fed a HFD for 6 weeks and then fed HFD only or HFD with CQR-300 at 50 and 200 mg/kg. Then, body weight gain and white adipose tissue weights were measured. We investigated the reduction in body fat and the regulation of fatty acid synthesis was measured by dual energy X-ray absorptiometry and real-time PCR with Western blot, respectively. In vitro study, CQR-300 inhibited pancreatic lipase activity. The CQR-300 treatment was significantly decreased the body weight gain and adipocytes size as well as white adipose tissues weights in HFD-induced obese mice. Furthermore, CQR-300 reduced the body fat and fat mass with regulating of adipose tissue hormones as leptin. Treatment with 50 mg/kg CQR-300 showed effectively lower expression levels of adipogenesis/lipogenesis related genes and proteins such as CCAAT/enhancer binding protein ${\alpha}$ ($C/EBP{\alpha}$), peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), Sterol regulatory element binding protein-1c (SREBP-1c), and fatty acid synthase (FAS) in white adipose tissue (WAT) as compared with the HFD fed only mice. These results suggest that the CQR-300 has an anti-obesity effect via inhibition of lipase activity, decrease the body fat mass by regulating the adipogenesis and lipogenesis related genes and proteins in epididymal adipose tissue with evaluate body fat reduce in the HFD-induced obese mice.

Reproductive Toxicity Study of SM-101(sulbactam.metampicillin): Fertility Study in Rats (복합항생제 SM-101(설박탐.메탐피실린)의 생식독성연구: 랫트 수태능력시험)

  • A new composite antibiotic, SM-101(sulbactam.metampicillin), was at dose levels of 0, 250, 500 and 1000 mg/kg/day administered intravenously to Sprague-Dawley male rats from predating to mating period and to females from premating to early gestation period. Effects of test agent on general findings and reproductive performance of parent animals and embryonic development were examined. In male parents, two deaths occurred at 1000 mg/kg. The increase in kidney weight of the 1000 mg/kg group were also observed. The decrease in body weight and food consumption were found at 500 and 1000 mg/kg. The decrease in spleen weight were seen at 250, 500 and 1000 mg/kg. In female parents, three deaths were found at 1000 mg/kg. Mating performance and fertility of parent animals were not adversely affected by all doses tested. F1 fetuses showed no changes related to treatment of SM-101. The results show that the no effect dose level(NOEL) for general toxicity of parent animals is under 250 mg/kg/day and NOELS for reproductive capability and fatal development are over 1000 mg/kg/day.

On the Growth, Feeding Rates and the Efficiency of Food Conversion for Cuttlefishes and Squids (오징어, 꼴뚜기류의 성장 및 섭이율과 이료의 전환효율)

  • Studies on the rate of growth, the rate of feeding and the efficiency of food conversion on the stage of new-born fries to the near adult size for three species of cuttlefishes , Sepia esculenta, Sepia subaculeata, Sepiella maindroni and two species of squids, Sepioteuthis lessomiana, Euprymna berryi were carried out in the process of artificial raising, and then argued about a feasibility of the propagation of cuttlefishes and squids. 1. The relation between the daily age (D) and the body weight(W) of Sepia exculent is expressed in a logarithmic equation, log W=3.0649 log D-4.2768. The daily rates of growth through 121 days of the raising period were 1.46 per cent for the man시 length and 1.67 percent for the body weight. The raipidest growth of Sepia esculenta is observed at the stage of 1 to 4 cm in the mantle length . At that time the daily rates of growth reach 3.3 to 5.5 percent for the mantle length and 10.4 to 12.0 percent for the body weight, respectively. The growth of Sepia esculenta varies a great deal to the bait. When fed on a dead bait the rates of growth decrease 17 per cent for the mantle length and 26 percent for the body weight compared with those fed on a live bait. 2. The relation between the daily age and the body weight of Sepia subaculeata is expressed in a logarithmic equation, log W=3.7447 log D-4.9003. The daily rates of growth through 110 days of the raising period were 1.63 percent for the mantle length and 1.83 percent for the body weight. The rapidest growth of Selia subaculeata is observed at the stage of 1.5 to 9.0 cm in the mantle length. At that time the daily rates of growth reach 3.1 to 7.4 percent for the mantle length and 6.8 to 16.7 percent for the body weight , respectively. 3. The relation between tehdaily age and the body weight of Sepiella maindroni is expressed in a ogarithmic equation , log W=2.9332 log D-3.8224 . The daily rates of growth through 133 days of the rearing period were 1.39 percent for the mantle length and 1.51 percent for the body weight . The rapidest growth of Sepiella maindroni is observed at the stage of 0.4 to 5.8 cm in the mantle length. At that time the daily rates for growth reach 4.6 to 7.3 percent for the mantle length and 8.5 to 15.4 percent for the body weight , respectively. 4. The daily rates of growth onthe stage of 0.5 to 6.0 comin the mantle length of Sepioteuthis lessoniana were 4.1 to 5.9 percent for the mantle length and 7.1 to 10.7 percent for the body weight . 5. During the rearing period of 31 days immediately after the hatching , the daily rateof feeding of Sepia esculenta marked 11.0 to 39.4 percent (28.2 percent in an average), and the efficiency of food conversion of this species reached 9.0 to 71.0percent (38.7percent in an average). Even at the more growing stage of 4.5to 6.2 cm in the mantle length, the daily rate of feeding of three species of cuttlefishes wee maintained 17.7 percent for Sepia esculenta, 30.8 percent for Sepia subaculeata and 34.7percent for Sepiella maindroni on an average. 6. The efficiency of food coversion of cuttlefishes and squids are larger than those of other fishes, and all the species are rapid in their growth. Four to five months are thought to be enough for their growing into a fair commercial size.

High Vitamin E Supplement is Needed to Have an Anticarcinogenic Effect of Fish Oil (어유와 비타민 E 보강 수준이 쥐간의 전암성 병변에 미치는 영향)

  • The influences of fish oil and different levels of vitamin I supplement on hepatocellular chemical carcinogenesis have been studied. Male Sprague-Dawley rats received diethylnitrosamine (DEN)(200mg/kg body weight) and were subjected to two-thirds partial hepatectomy to induce murine chemical hepatocarcinogenic procedure. Placental glutathione S-transferase(GST-P) positive foci area, antioxidant enzymes(Cu/Zn-superoxide dismutase(SOD), catalase, glutathione reductase (GR), total- glutathione peroxidase (TGPx), glutathione S -transferase (GST)), glucose 6-phosphatase (G6Pase) activities, and lipid peroxidation of microsomes(thiobarbituric acid reactive substances (TBARS)) were measured. Experimental animals were fed 15% corn or fish oil with 0, 40, 1,000, 10,000IU vitamin E /kg diet for 8 weeks. Vitamin E supplements decreased the area of GST-P positive foci in both groups. The higher the vitamin E levels, the smaller the area of GST-P positive foci were noticed. Compared to 0 IU vitamin E, 40 IU in corn oil and 1,000 IU in fish oil groups were effective in decreasing G57-P positive foci area. Fish oil groups tended to have smaller area of GST-P positive foci. fish oil groups showed lower body weight, lower activities of Cu/Zn-SOD and TGPx, higher TBARS contents, higher activities of GST, catalase, G6Pase, GR and higher liver/body ratio than corn oil groups. As the level of vitamin I increased, GST-P positive foci count, catalase activities, and TBARS tended to decrease. G6Pase activities tended to increase in both groups. At higher vitamin E levels, GST activities tended to decrease in fish oil groups. These results suggest that vitamin I has suppressive offects on hepatocellular chemical carcinogenesis probably through antioxidant eH:cts decreasing TBARS contents, $H_2O$$_2$, and organic peroxides. fish oil tended to have greated suppressive offects than corn oil on hepatocellular carcinogenesis. Effects of Gamgung-tang Gamibang on 3,5,3-triiodothyronine-induced Hyperthyroidism in rats (감궁탕 가미방이 갑상샘기능장애에 미치는 효과)

  • In this study, the effects of Gamgung-tang gamibang on the hyperthyroidism induced by the intraperitoneal injection of 3,5,3-triiodothyronine was examined by the measurement of physical changes, body weight, the volume of food intake and rectal temperature, and heart weight, heart beat, blood pressure with contrast to propranolol, one of beta-blocking agents. the obtained results were as follows. The Gamgung-tang gamibang extract showed to inhibit the decrease of body weight and rectal temperature, and decrease the food intake, so the inhibitory effects of Gamgung-tang gamibang extract on the experimental hyperthyroidism were exhibited. The Gamgung-tang gamibang extract showed the inhibitory effects on the circulatory functions changed and enhanced by the experimental induced hyperthyroidism, the action of Gamgung-tang gamibang extract was less effective than the propranolol of D-CONT group. The Gamgung-tang gamibang extract showed significant effects to inhibit the concentration of serum thyroid houmone, more effective than the propranolol, beta-blocking agents. The Gamgung-tang gamibang extract showed the effective inhibitory reaction on the biochemical changes in serum, cholesterol, ketone bodies, free fatty acid, glucose in hyperthyroid rats induced by 3,5,3-triiodothyronine.

Acupuncture Effects of Fatty Mice Induced by High Fat Diet (침 자극이 고지방식이로 유발된 비만 백서에 미치는 영향)

  • Objectives The aim of this study was to investigate the effects of acupuncture on weight loss, food intake, lipid metabolism, adipogenesis. Methods Four week-old C57BL/6J mice acclimatized to the laboratory environment for 1 week were allocated into four groups of 6~8 mice each: normal diet group, high fat diet group, high fat diet group and treated with acupuncture at HT7, high fat diet group and treated with acupuncture at ST36 for 90days. High fat diet group was used to control group. Results 1) Body weight, food intake of the ST36, HT7 groups decreased compared with those of control group. ST36 group was more effective with body weight decrease, and HT7 group was more effective with food intake decrease. 2) NEFA, TG, TC, ALT, AST of the ST36, HT7 groups decreased compared with those of control group. ST36 group was more effective. 3) The expression of SREP-1c, SREBP-2 of the ST36 group decreased compared with those of control group. These decreased rates were statistically significant. SREBP-2 of the HT7 group decreased significantly compared to the control group. 4) LXR, FAS, SCD of the ST36 group decreased significantly compared with control group. 5) p-ACC, HMGCR of the ST36 group decreased significantly compared with those of control group. HMGCR of the HT7 group decreased significantly compared with control group. Conclusions These results suggest that ST36, HT7 acupuncture may be used prevent or treat the obesity induced by high fat diet.

Nephroprotective Effects of Manjeonmoktong-san on Cisplatin-induced Rat Acute Renal Failure (만전목통산(萬全木通散)이 Cisplatin으로 유발(誘發)된 흰쥐의 급성신부전(急性腎不全)에 미치는 영향(影響))

  • Purpose : The object of this study was to observe the nephroprotective effects of Manjeonmoktong-san(MJMTS), which has traditionally been used as Korean medicine for treating various renal diseases, on cisplatin-induced rat acute renal failure. Methods : At first, three different dosages of MJMTS extract were orally administered once a day for 28 days. On the 23rd day after MJMTS extract treatment, cisplatin was treated. Then, 5 days after cisplatin treatment, all the rats (6 groups of 8 rats each) were checked in the present study. The changes to the body weight, kidney weight, serum BUN and creatinine levels were observed with changes on the kidney MDA and GSH contents. The results were compared with captopril 100mg/kg in which the effects on cisplatin-induced acute renal failures were already confirmed. Results : It showed dramatical decrease on the body weight, increase of serum BUN and creatinine levels were detected in cisplatin control as compared with intact control. In addition marked increase of kidney MDA contests and decrease of kidney GSH contents were also detected in cisplatin control as compared with intact control. it means that cisplatin induced ARF are induced by oxiidative stress and related lipid peroxidation in the present study. However, these ARFs and inhibition of antioxidant effects induced by cisplatin were dose-dependently reduced by treatment of all three different dosages of MJMTS extracts. Conclusion : This study suggests that MJMTS extracts have favorable effects on the cisplatin-induced rat ARF.

Antihyperlipidemic effects of alcoholic extract of Pongamia pinnata Linn. leaves on high fat diet fed rats

  • The objective of the study was to investigate the antihyperlipidemic activity of alcoholic extract of Pongamia pinnata Linn. (PPAE) leaves in rats fed with high fat diet (HFD). PPAE was administered orally in the divided doses of 250 and 500 mg/kg/day for 30 days in HFD fed rats. Body weights were observed and the analysis of serum lipid profile was carried out on day 30. Marked decrease in the body weight, total cholesterol (TC), triglycerides (TG), low density lipoprotein (LDL) and very low density lipoprotein (VLDL) whereas significant increase in the levels of high density lipoprotein (HDL) were observed after treatment with PPAE. However, PPAE in a dose of 250 mg/kg did not show significant (P < 0.05) increase in HDL levels. The PPAE also lowered TC: HDL-c and LDL: HDL-c ratios significantly suggesting it's antihyperlipidemic and cardioprotective potential. The present work reveals that PPAE at the dose of 500 mg/kg/day exhibited significant (P < 0.01) antihyperlipidemic effects.
