Measurement of Hemoglobin Adducts in Female Mice Inhaled with 1,3-butadiene by Using GC/MS

  • 1,3-butadiene(DB) is a well-established rodent carcinogen, and a probable carcinogen to humans. This study was investigated the formation of hemoglobin adduct in ICR female mice inhaled with BD for 3 weeks (5 hr/day, 5 days/week). Body weights of mice were significantly low from onward day 4 or 9 of exposure comparing the control. Hemoglobin adducts formed by DB exposure were (N-2-hydroxy-3-butenyl) valine (HB Val adduct) and (N-2,3,4-trihydroxy-butyl)valine(THB Val adduct). The levels of HB Val adducts were 1.8, 3.7 and 6.2 pmol/mg globin for the 500 ppm BD inhalation group, and 5.7, 7.4 and 16.0 pmol/mg globin for the 1000 ppm BD inhalation group, when observed on the $1^{st},\;2^{nd},\;and\;3^{rd}$ week after inhalation exposure, respectively. The levels of THBVal adducts were 32.0, 42.0 and 55.0 pmol/mg globin for the 500 ppm DB inhalation group, and 67.8, 72.7 and 83.5 pmol/mg globin for the 1000 ppm BD inhalation group, when observed on the $1^{st},\;2^{nd},\;and\;3^{rd}$ week after inhalation exposure, respectively. Ratios of THBVal and HBVal adducts were higher at earlier exposure period and lower concentration. Ratios on the $1^{st},\;2^{nd},\;and\;3^{rd}$ week were 17.8, 11.4 and 8.87 for the 500 ppm BD inhalation group, and 11.9, 9.8 and 5.2 for the 1000 ppm BD inhalation group, respectively. In conclusion, THB Val and HB Val adducts were the important hemoglobin adducts in ICR female mice inhaled with BD, and the latter was more appropriate biomarker than the other for biological monitoring of BD exposure.

차가버섯 추출물이 당뇨생쥐 $(C57BL/KsJ^-m^{+/+}Lepr^{db})$의 혈당수준에 미치는 영향 (Effects of Inonotus Obliqua Extract on Blood Glucose Levels in Genetically Diabetic Mice)

  • This study investigated the therapeutic effects of Inonotus obliqua extract on blood glucose, insulin, and other biochemical parameters in genetically diabetic mice $(C57BL/KsJ^-m^{+/+}Lepr^{db})$. The mice were divided into four groups-control, Chaga 1 (dose of 0.09 mg/kg of body weight), Chaga 5 (5 times of Chaga 1), and Chaga 10 (10 times of Chaga 1) - according to supplemented dose. Inonotus obliqua extract was orally administered to the animals for 6 weeks. The body and organ (liver and kidney) weights were not different among groups. Fasting blood glucose level was significantly lower in the Chaga 5 group compared with the control (p < 0.05). Hemoglobin A1c content was significantly lower in the Chaga 5 group compared with either the control and Chaga 1 group (p < 0.05). There was no significant difference in serum insulin level among groups. The glucose-6-phosphatase activity in liver was significantly the lowest in Chaga 10 group and was significantly lower in Chaga 5 group as compared with those of control and Chaga 1 groups. Therefore, the results of this study demonstrate that Inonotus obliqua extract alleviates many of the symptoms of diabetes in genetically obese mice and may offer a possibility as a therapeutic supplement for the normalization of blood glucose levels in human with hyperglycemia and have beneficial effects in patients with non-insulin-dependent diabetes mellitus.

Bromobenzene으로 유도된 간 손상 마우스에 대한 Microcluster수의 효과 (Detoxification Effect of Microcluster-Water on Bromobenzene-Induced Liver Damaged Mice)

  • McW의 간 해독효과를 조사하였다. 실험군은 총 4개군 즉, 증류수급여군으로 대조군(DC)과 증류수를 3주간 섭취케 한 후 희생직전에 BB를 처리한 군(DB), McW 급여군으로는 McW 대조군(MC)과 McW를 3주간 섭취케 한 후 희생직전에 BB를 처리한 군(MB)의 4개 군으로 구분하였다. DC군과 MC군의 혈청 ALT 및 AST 활성은 뚜렷한 차이를 보이지 않았으나 MB군에서는 DB군에 비하여 유의하게 낮았다. 간 AH 활성은 모든 실험군에서 유의한 변동이 없었으나 GST 활성은 MC군이 DC군에 비하여, MB군이 DB군에 비하여 각각 높았다. 간 조직의 LPO 함량은 DC 및 MC군이 유사하였으며, DB 및 MB군 모두 DC군보다 유의하게 증가하였으나 그 증가율은 MB군이 DB군에 비하여 낮았다. McW의 전자공여능은 DW에 비하여 유의하게 높았다. 이상의 결과 McW는 간 해독효소의 일종인 GST의 활성을 증가시킴과 동시에 McW의 지속적인 섭취에 의한 항산화 작용에 의해 해독작용을 나타내는 것으로 생각되나 추후 계속적인 연구검토가 행해져야할 것이다.

Poly-N-acetyl-glucosamine이 당뇨병 쥐에서 창상치료에 미치는 영향 (Effects of Poly-N-acetyl Glucosamine(pGlcNAc) Patch on Wound Healing in db/db Mouse)

  • Purpose: Poly-N-acetyl glucosamine(PGlcNAc) nanofiber-based materials, produced by a marine microalga, have been characterized as effective hemostatic and angiogenic agents. The similarity between PGlcNAc patch and the natural extracellular matrix allows it to support new healthy tissue growth in an injured area and to encourage fluid absorption. In this study, we hypothesized that a poly-N-acetyl glucosamine fiber patch(PGlcNAc patch) may enhance wound healing in the db/db mouse. Methods: PGlcNAc patches were applied on one square centimeter, full-thickness, skin wounds in the db/db mouse model. Wounds(n=15 per group) were dressed with a PGlcNAc nanofiber patch for 1 hour(1 h), 24 hours(24 h) or left untreated(NT). After the application time, patches were removed and wounds were allowed to heal spontaneously. The rate of wound closure was evaluated by digital analysis of unclosed wound area in course of time. At day 10, wounds(n=7 per group) were harvested and quantified with immunohistochemical markers of proliferation(Ki-67) and vascularization (platelet endothelial cell adhesion molecule, PECAM-1). Results: Wounds dressed with PGlcNAc patches for 1 hour closed faster than control wounds, reaching 90% closure in 16.6 days, nine days faster than untreated wounds. Granulation tissue showed higher levels of proliferation and vascularization following 1 h treatment than the 24 h and NT groups. In addition to its hemostatic properties, the PGlcNAc material also appears to accelerate wound closure in healing-impaired genetically diabetic mice. Conclusion: This material, with its combination of hemostatic and wound healing properties, has the potential to be effective agent for the treatment of complicated wounds.

An extract of the root of Lithospermun erythrorhison accelerates wound healing

  • Metabolic disease such as diabetes, which is caused by stress or imbalanced diet, has been increasing. A diabetic tend to suffer from a delay or difficulty of wound healing. The extract of SHIKON (SK), that is the root of Lithospermun erythrorhison, has been reported to have an effect on healing for normal wound, but has never studies for intractable wound so far. Therefore we examined the effect of SK extract on wound healing with healing impaired mouse model. Full-thickness round wounds were created on the backs of db/db mice and applied SK, and we observed neovascularization and collagen synthesis, distribution of apoptotic cells, and vascular endothelial growth factor (VEGF)- positive cells in granulation tissue. After two weeks, a number of capillary vessel and collagen synthesis were increased in SK-treated wounds. Infiltration of VEGF-positive neutrophils was also seen in the wound, besides apoptotic fibroblasts and endothelial cells were appeared in the granulation tissue. After three weeks, the wound closed completely with SK-treated but not in control. These results suggest that SK enhanced neovascularization by VEGF and this kind of apoptosis process makes the scar smooth. In this study, it is obvious that SK also accelerates healing of intractable wound.

Fermented Ginseng Attenuates Hepatic Lipid Accumulation and Hyperglycemia through AMPK Activation

  • Fermented ginseng (FG) is an ethanol extract of ginseng radix processed with $\beta$-galactosidase. It was hypothesized that FG may exert anti-hyperlipidemic and anti-diabetic activities through modulating AMP-activated protein kinase (AMPK) in HepG2 human hepatoma cells. In this study, we showed that AMPK phosphorylation was stimulated by FG. These effects were abolished by pretreatment with an AMPK inhibitor, compound C. In addition, FG regulated the expression of genes associated with lipogenesis and lipolysis, thus causing suppression of hepatic triglyceride accumulation. In vivo study using db/db mice, FG reduced fasting plasma glucose, HbAlc, and insulin resistance index, when compared to diabetic control. FG also increased the phospho-AMPK and glucose transporter 4 (GLUT4) expressions in liver and skeletal muscle, respectively. In liver, expressions of lipogenic gene were decreased whereas expressions of lipolytic genes were induced, when compared to diabetic control. Taken together, we may suggest that FG ameliorates hyperglycemia and hyperlipidemia through activation of AMPK and could be developed as a health functional food or therapeutic agent for type 2 diabetic patients.

산약의 항당뇨 특성 연구(2) - NGF 유도 신경병증예방 - (Characteristics of Antidiabetic Effect of Dioscorea rhizoma(2) - Prevention of Diabetic Neuropathy by NGF Induction -)

  • The main cause of diabetic neuropathy, one of the most debilitating complications, is the chronic hyperglycemia, the increase sorbitol or the decrease of nerve growth factor(NGF). NGF, a protein, plays a major role in the development and maintenance of peripheral nervous system. Systemic administration of NGF prevents manifestations of neuropathy in rodent models of diabetic neuropathy. In the previous investigation, we report the hypoglycemia effect of Dioscorea rhizoma extract(DRE) in diabetic mice. The present study shows protective effect of DRE on diabetic neuropathy by induction of NGF protein. We investigated the NGF level in salivary gland and sciatic nerve of normal mouse and the effect of DRE on sciatic nerve conductivity and thermal hyperalgesia test in Type 2 db/db mouse. DRE increased endogenous NGF level in salivary gland and sciatic nerve of mouse. And sensory nerve conductivity velocity(SNCV), motor nerve conductivity velocity(MNCV) and thermal hyperalgesia increased in DRE treatment mice compared with control group. On the basis of our results, we conclude that DRE increase induction of endogenous NGF level and have protective effect on diabetic neuropathy by induction of NGF. Therefore, we propose that long-term use of DRE might help prevention of diabetes-associated complication; diabetic neuropathy.

Effect of Garlic and Aged Black Garlic on Hyperglycemia and Dyslipidemia in Animal Model of Type 2 Diabetes Mellitus

  • Control of hyperglycemia and dyslipidemia is strongly correlated with decreased risk for cardiovascular disease, the most common and fatal diabetic complication. The purpose of this study is to determine the effects of garlic and aged black garlic on glycemic control and blood lipid profile in animal model of type 2 diabetes. Three week-old db/db mice (C57BL/Ks, n=21) were fed AIN-93G semipurified diet or diet containing 5% freeze-dried garlic or aged black garlic for 7 weeks after 1 week of adaptation. Fasting serum glucose, insulin, triglyceride, total cholesterol, and HDL-cholesterol and blood glycated hemoglobin were measured. Body weight and food intake of garlic and aged black garlic group were not significantly different from those of the control group. Fasting serum glucose and blood glycated hemoglobin levels were significantly decreased and insulin level was significantly increased in garlic group compared with control group (p<0.05). Consumption of aged black garlic significantly decreased homeostasis model assessment for insulin resistance (HOMA-IR) and tended to decrease serum glucose. Garlic consumption significantly decreased total cholesterol, while aged black garlic significantly reduced serum total cholesterol and triglyceride and increased HDL-cholesterol levels. These results suggest that garlic exerts hypoglycemic and hypocholesterolemic effect and aged black garlic improved insulin sensitivity and dyslipidemia in db/db mice.
