  • 송아지 흉선 nucleohistone의 DNase 1에 對한 susceptibility와 相互作用의 cooperativity에 미치는 spermine의 效果를 nucleostone의 構造變移와 關聯시켜 調査하였다. 이들 data로 부터 nucleohistone은 遊離 DNA와는 對照的으로 spermine에 의하여 monomoclecular condensation을 일으키지 않고 intermolecular aggregation을 이루며 nucleohistone의 DNase 1과의 相互作用 cooperativity가 spermine에 의하여 增加됨을 推理할 수 있다. Nucleohistine의 histone 部分의 cooperativity에 關한 機能的 役割을 究明하기 위하여 histone 部分을 단실化시킨 DNS-nucleohistone을 製造하여 DNase 1 과의 相互作用을 調査하여 보았는바 negative cooperativity로 나타났다. 이로 부터 nucleohistone의 histone部分은 nucleohistone의 cooperativity에 影響을 미치리라고 推理할 수 있다.

Fluorescence Micropatterning Based on the Polymeric Photobase Generator Containing Oxime-Urethane Groups by Dansylation

  • A polymeric photobase generator containing oxime-urethane groups is applied to a fluorescence micropatterning material. Polymer bearing oxime-urethane groups was prepared by copolymerization of methyl methacrylate with methacryloyloxyethyl benzophenoneoxime urethane (MBU). The reaction of amino groups in the irradiated copolymer film with dansyl chloride (Dns-Cl) was monitored by using UV absorption, IR absorption and fluorescence spectroscopy. The fluorescence spectrum of the Dns-Cl-treated irradiated copolymer film shows a strong fluorescence with a fluorescence maximum wavelength at 510 nm. A blue fluorescent micropattern with a line width of $2{\sim}3{\mu}m$ was obtained. Treatment of the irradiated copolymer film with Dns-Cl and rhodamine B mixture led to the formation of green, red, and orange-colored fluorescence micropatterns. Thus, various colored micropatterns on a single polymer film can be obtained by selective excitation of each dye molecules.

액체크로마토그래피를 이용한 수돗물 중 nitrosamine 화합물 분석의 최적화 (Optimization of analytical conditions for the determination of nitrosamines in chlorinated tap water by high performance liquid chromatography)

  • 본 연구는 칼럼 전 유도체화와 HPLC-형광검출을 이용해 염소 소독 음용수 중 7가지 nitrosamine 화합물을 분석하는 방법을 추출 과정(액-액 추출 vs. 고상 추출)과 형광 유도체화 과정(denitrosation 및 dansylation)으로 나누어 평가하여 확립하였다. 최적화된 유도체화 방법으로 두 가지의 추출법을 비교하였을 때, Ambersorb 572를 이용한 고상 추출에 대한 회수율과 재현성(상대표준편차, RSD)이 각각 54.4-88.7%와 1.9-19.4%로 액-액 추출의 51.4-87.7%와 4.2-33.3%에 비해 더 좋게 나타났다. 확립된 방법은 기존의 HPLC를 이용한 분석 방법에 비해 회수율과 재현성 모두 개선되었으며, 방법검출한계(method detection limits, MDLs)는 0.5-4.4 ng/L이었다. 이 방법으로 춘천의 염소로 소독하는 두 곳의 정수장과 10 곳의 수도꼭지에서 채취한 물 시료 중 nitrosamine 화합물을 분석한 결과, N-nitrosodimethylamine (NDMA)이 주요 화합물이었으며, 농도 범위는 26.1-112 ng/L이었다.

Dansyl 유도체화와 역상 고성능액체크로마토그래피에 의한 아미노산의 정량 (Reverse-Phase High Performance Liquid Chromatographic Determination of Amino Acids after Precolumn Derivatization with 1-Dimethyl-aminonaphthalene-5-sulfonyl chloride)

  • This experiment was carried out to determine the separation condition of 1-dimethylaminonaphthalene-5-sulfony(Dansyl) derivatives of amino acids by reverse-phase high performance liquid chromatography with Nova-Pak C18 column. Determined solvent system was solvent A(200mA phosphate buffer pH 6.8 15%, acetonitrile 11%, water 74%) and solvent B(acetonitrile 65%, methanol 28%, water 7%). Linear gradient of solvent B was applied from 12% to 80% for 50min. Complete separation of 20 amino acids including asparagine and glutamine which constitute protein was achieved within 50min. As the detection limit was the range of picomole, the resolution power was excellent. Reproducibility of the retention time was less than mean $\pm$0.05min. According to the above optimum chromatographic conditions, the amino acid composition of some food and human blood was examined. The most affluent amino acid was alanine in human blood, aspartic acid and glutamic acid in soy sauce, alanine and threonine in soy milk and proline in milk and yoghurt.
