Characterization of a Late Gene, ORF60 from Bombyx mori Nucleopolyhedrovirus

  • Open reading frame 60 of Bombyx mori nucleopolyhedrovirus (Bm60) is located between 56,673 and 57,479 bp in the BmNPV genome which encodes 268 amino acid residues with predicted molecular weight of 31.0 kDa. Bm60 and its homologues have been identified in 11 completely sequenced lepidopteran NPVs. The transcript of Bm60 was detected by RT-PCR at 18-72 h post-infection (p.i.), while the corresponding protein could be detected at 24-72 h p.i. in BmNPV-infected BmN cells by Western blot analysis using a polyclonal antibody against Bm60. The expression of Bm60 was inhibited in the presence of Ara-c, an inhibitor of viral DNA synthesis. These results together indicated that Bm60 was a late gene. The size of Bm60 product was found to be a 31 kDa in BmNPV-infected BmN cells, consistent with predicted molecular weight. Immuno-fluoresence analysis showed that the Bm60 product was first detected in the cytoplasm at 24 h p.i and also located in nucleus during later infection. In conclusion, the available data suggest that Bm60 is a functional ORF of BmNPV and encodes a 31kDa protein expressed in the later stage of infection cycle.

  • 뽕나무버섯 추출물에서 fibrinolytic enzyme을 분리 정제하였다. 이 효소의 분자량은 약 19800Da 이고 pH 7.0에서 최고의 활성을 보여주는 neutral protease로 $55^{\circ}C$에서 최대 활성을 나타냈다. EDTA에 의해 활성이 저해되는 것으로 보아 metalloprotease로 추정되며 $Hg^{+2}$의 영향을 받아 활성이 상실되었다. N-terminal amino acid 분석 결과 15번째까지의 아미노산 잔기 순서는 X-X-T-N-G-X-T-X-S-R-Q-T-T-L-V 이었으며 지금까지 알려진 것과는 다른 새로운 효소이다.

84-kDa의 폐렴구균 열충격단백질 ClpL의 Cloning 및 면역특성에 관한 연구 (Cloning and Immunological Characterization of the 84-kDa Heat Shock Protein, ClpL, in Streptococcus pneumoniae)

  • Heat shock proteins serve as chaperone by preventing the aggregation of denatured proteins and promote survival of pathogens in harsh environments. In this study, heat shock gene encoding a 84-kDa (p84) protein, which is one of the three major heat shock proteins in S. pneumoniae, was cloned and characterized. PCR with a forward primer derived from N-terminal amino acid sequence of the p84 and a reverse primer derived from the conserved second ATP-binding region of Clp family was used for amplification of the gene encoding the p84 and subsequently the PCR product was used for sequence determination. Sequence analysis of the p84 gene demonstrated that it is a member of ClpL. The deduced amino acid sequence of pneumococcal ClpL shows homology with other members of the Clp family, and particularly, even in variable leader region, with bovine Clp-like protein and L. lactis ClpL. S. pneumoniae clpL is the smallest clop member (701 amono acids) containing the two conserved ATP-binding regions, and hydrophilic N-terminal variable region of pneu-mococcal Clp ATPase is much shorter than any known Clp ATPases. Histidine tagged ClpL was overexpressed and purified from E. coli. Immunoblot analysis employing antisera raised against pneumococcus p84 demonstrated no cross-reactivity with Clp analog in Eschericha coli, Staphylococcus aureus and human HeLa cells. Preimmunization of mice with ClpL extended mice life partially but did not protect them from death.

  • 이 논문에서 우리는 분산 실시간 소프트웨어 설계방법인 CODARTS/DA(Concurrent Design Approach for Real-Time Systems/Distributed Application)를 위한 타스크 행위 명세방법을 제안한다. 각 타스크는 CODARTS/DA 모듈들, 즉, 다수 클라이언트/서버 모듈(Multiple Client/Sever Module), 단일 클라이언트/서버 모듈(Single Client/Server Module), 그룹 통신모듈(Group Comm. Module), 우선 순위 모듈등과 통신을 수행하는데, 분산환경의 동기, 비동기 문제들을 제안한 방법으로 설계 명세할 수 있다. 또한 제안된 명세방법은 C언어와 비슷한 문법구조를 제공하므로 쉽게 문서들이 구조적 프로그램으로 변환될 수있다.

Creep Crack Growth Properties of Low Pressure Turbine Rotor Steel under Constant Load and Ct

  • The propagation rate(da/dt) prediction parameter and the microstructure properties of creep crack in domestic 3.3NiCrMov steel were investigated at 550$\^{C}$ by using 0.5" CT specimen under constant load(4090N) and constant Ct(300∼4000N/mhr) condition that was maintained during crack growth of 1mm distance. C* usually increased with crack length though load was reduced in order to maintain constant Ct value as crack growth and considerably showed the scatter band, but Ct depended on load line displacement rate and represented a good relation with da/dt. At constant toad and Ct region, crack growth slope was 0.900 and 0.844 each, in the other hand C* slope was 0.480. Fully coalesced area(FCA) ahead of crack tip was increased as Ct value increase to the critical value, and after that value FCA decreased. The average diameter ditribution of cavity in FCA showed the greatest value about 1.5 ㎛ when Ct=2000N/mhr. The increasing of Ct in FCA view point enlarged the size of damage area and the size reached to maximum 800 ㎛ when Ct=2000N/mhr.

  • The fatigue crack propagation behavior of the SA516-Grade 70 steel which is used for pressure vessels was experimentally examined under the condition of at room temperature, $150^{\circ}C$, $250^{\circ}C$ and $370^{\circ}C$ with stress ration of R=0.1 and 0.3. The fatigue crack propagation rate , da/dN, related with the stress intensity factor range, $\vartriangle$N, was influenced by the stress ration within the stable growth of fatigue crack(Region II) with an increase in $\vartriangle$N. The resistance to the fatigue crack growth at high temperature is higher in comparison with that at room temperature, and the resistance attributed to the extent of plasticity-induced by compressive residual stress according to the cyclic loads. Fractographic examinations revels that the differences of the fatigue crack growth characteristics between room and high temperatures are mainly explained by the crack and oxide-induced by high temperature.

Purification and Characterization of Cop, a Protein Involved in the Copy Number Control of Plasmid pE194

  • Cop protein has been overexpressed in Escherichia coli using a T7 RNA polymerase system. Purification to apparent homogeneity was achieved by the sequential chromatography on ion exchange, affinity chromatography, and reverse phase high performance liquid chromatography system. The molecular weight of the purified Cop was estimated as 6.1 kDa by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). But the molecular mass of the native state Cop was shown to be 19 kDa by an analytical high performance size exclusion chromatography, suggesting a trimer-like structure in 50 mM Tris-HCI buffer (pH 7.5) containing 100 mM NaCl. Cop protein Was calculated to contain $39.1% {\alpha}-helix, 16.8% {\beta}-sheet$, 17.4% turn, and 26.8% random structure. The DNA binding property of Cop protein expressed in E. coli Was preserved during the expression and purification process. The isoelectric point of Cop was determined to be 9.0. The results of amino acid composition analysis and N-terminal amino acid sequencing of Cop showed that it has the same amino acid composition and N-terminal amino acid sequence as those deduced from its DNA sequence analysis, except for the partial removal of N-terminal methionine residue by methionyl-aminopeptidase in E. coli.

Immunoblot technique을 이용한 폐흡충의 발육단계별 항원 특이성 분석 (Analysis of antigenic specificities of Puragonimus westeymuni developmental stages using immunoblot technique)

  • 폐흡충증의 진단은 객담검사법이 가장 확실한 진단 방법이기는 하지만 폐흡충이 폐에 기 생한 경우에도 충란 검출이 어려울 때가 있으므로 혈청학적 진단법이 이용되고 있다. 혈청학적 진단법에 사용되는 항원인 기생충 추출물, 즉 조항원은 분류학적으로 유사한 기생충과 서로 공유하고 있는 공통 항원 때문에 교차 반응을 일으키는 경우가 있다. 이 연구는 발육단계 별 폐흡충에서 만든 조항원을 SDS-PAGE(sodium dodecyl sulfate polyacrylamide gel electrophoresis)로 전기영동한 다음 EITB(enzyme-linked immunoelectrotransfer blot)를 이용하여 항원대별 항원성 및 특이성을 관찰하여 시기별로 채취한 고양이 혈청에 대한 특이 반응대를 관찰해 보고자 시행하였다. 실험에 사용한 항원은 실험적으로 고양이에 감염시켜 3, 5, 8및 12주 만에 얻은 폐흡충의 식염수 추출액 (SEPn; n=감염된 시기)이며 3∼20% linear gradient gel에서 SDS-PAGE하였다. Silver stain결과 폐흡충 조항원은 최소한 30개 이상의 band들로 구성되어 있었는데 각 발육단계 에 공통된 항원대로 203, 63, 35, 21, 19, 13 kDa band들이 관찰되었고, 단계별로 차이점도 관찰 되었다. SEPl2에서는 새로운 229 kDa band가 관찰되었다. 주요 항원대에 대하여 EITB를 한 결과 각 항원과 감염 후 5주 이상 된 혈청과 공통적으로 반응한 항원대는 203, 115, 91, 85, 67, 63, 48, 39, 35 및 25 kDa band들이었고, 8주 이상 된 혈청과의 반응에서는 19, 13및 10 kDa 항원대와 공통적으로 일관성 있게 반응하였다. SEP12는 12주 된 혈청과 229 kDa에서 특이하게 반응하였다.
