Study on the Alkaloids from Thalictrum fauriei

  • To explore the biological activities of Thalictrum alkaloids, a study was initiated to investigate the alkaloidal constituents from T. fauriei, a species indigenous to Taiwan. Extensive fractionation of the ethanolic extract provided three known aporphines, (+)-oconovine (Ia), (+)-isocorydine (Ib), and (+)-corydine (Ic) from nonphenolic tertiary base fraction. From quaternary base fraction two new protoberberinium salts, thalifaurine (IIa), dehydrodiscretine (IIb), in addition to magnoflorine (III), were isolated. The structure of both new compounds were confirmed by total syntheses. Fractionation of phenolic bases yielded a dihydromorphinandienone, (-)-ocobotrine (IV), as well as a novel aporphine-pavine dimer, tentatively named EP-10. The structure of EP-10 was established by means of 2D NMR studies. Preliminary study indicated a weak activity of lyzing Hela cells, in vitro, for EP-10.

셀루로오즈에서 C.I. Reactive Blue 19의 반응과 확산에 Dimer의 기여 (Contribution of Dimer to Reaction and Diffusion of C.I. Reactive Blue 19 in Cellulose)

  • C.I. Reactive Blue 19에 대한 hydroxylethylsulfonyl type의 수용액에서의 용해성과 안정성을 조사한 결과 이온강력 0.15, pH 5.8과 9.2에서 초기의 용해도를 4시간 동안 유지했으며, 이온강력을 증가시키면 용해도의 단정성이 감소했다. 그러나 용액의 교반하면 안정성이 증가하여 이온강력 0.30에서 초기용해도가 하루 동안 유지되었다. 셀로판 필름을 원주형태의 롤로 만들어 확산과 흡착 거동을 조사한 결과 hydroxylethylsulfonyl type의 용액 농도가 증가하면 표면농도, $C_0$가 증가했으나 확산계수, D는 일정한 값을 유지했다. Bis(arylsulfonylethyl)ether type의 셀룰로우스와의 반응성은 vinylsulfonyl type에 대해 겉보기 반응속도가 1/6 정도였다.

  • 균일 수용액과 polyacrylic acid(PAA) 존재하에서 3,3'-diethyl oxacarbocyanine(DOC)의 spectra변화를 흡수 및 형광분광학적인 방법으로 연구하였다. 균일 수용액에서 DOC의 농도변화에 따른 흡수 spectra변화는 dimer의 형성에 기인하는 것으로 설명하였다. PAA의 존재하에서 P/D변화에 의한 형광세기의 변화는 흡수 분광학적 방법과 유사한 결과를 얻었다. PAA와 DOC의 상호작용의 반응속도론적인 연구를 흡수 및 형광 정지흐름법으로 수행하였다. PAA-DOC계에서 관측된 이완효과는 두 개의 이완과정이 존재한다고 가정함으로써 정량적으로 설명할 수 있었다.

Physiological Influence of Combined Mental Activity with Experimental Subway Noise

  • Numerous psychological stressors playa role in development of the cardiovascular disease. The aim of this study was to determine whether a combined mental activity with experimental subway noise affects hematological physiology. Fifty-four healthy volunteers were divided group I which underwent subway noise (n=24) and group II which underwent a combined mental activity with subway noise (n=30). Venous blood samples were collected for measuring CBC, prothrombine time (PT), activated partial thromboplastine time (APTT), erythrocyte sedimentation rate (ESR), D-dimer and high sensitive C-reactive protein (H-CRP) levels before, 50 min of stress task (S-50m) and 60 min of recovery (R-60m). Changed ratios of granulocyte, lymphocyte, monocyte and platelet counts at S-50m and R-60m were higher in group II compared to group I. RBC count and hematocrit level in group I increased whereas those in group II decreased at S-50m. PT, APTT and ESR in the both groups were shortened at R-60m and the decreased ratios were high in group II compared to group I. H-CRP and D-dimer in the both groups were elevated at S-50m and R-60m while the increased ratios in group II were greater than those in group I. These observations imply that a combined mental activity with experimental subway noise may be a stressor which affects hematological physiology.

Structural and Physiological Characteristics of Rhamnogalacturonan II from Fruit Wines

  • To characterize the polysaccharides which exist as soluble forms in fruit wines, crude polysaccharides were isolated from red, white, raspberry, wild grape, and pear wine, respectively. Among them, the crude polysaccharide (RW-0) in red wine showed the highest yield and considerable amounts of thiobarbituric acid (TBA)-positive materials. The pectic polysaccharide RW-2 was purified to homogeneity from RW-0 by subsequent size-exclusion chromatography using Sephadex G-75 and its structure was characterized. RW-2 consisted of 14 different monosaccharides which included rarely observed sugars in general polysaccharides, such as 2-O-methyl-fucose, 2-O-methyl-xylose, apiose (Api), 3-C-carboxy-5-deoxy-L-xylose (aceric acid, AceA), 3-deoxy-D-manno-2-octulosonic acid (Kdo), and 3-deoxy-D-lyxo-2-heptulosaric acid (Dha). Methylation analysis indicated that RW-2 comprised at least 20 different glycosyl linkages such as 3,4-linked fucose, 2,3,4-linked rhamnose, 3'-linked apiose, and 2,3,3'-linked apiose, being characteristic in rhamnogalacturonan II (RG-II). High performance size-exclusion chromatography indicated that RW-2 mainly comprised RG-II of higher molecular weight (12,000), and that the changes of molecular weight to apparent 7,000 under less than pH 2.0 were observed. These analyses indicated that the higher molecular weight polysaccharide in RW-2 was mainly present as a RG-II dimer.

Atypical presentation of DeBakey type I aortic dissection mimicking pulmonary embolism in a pregnant patient: a case report

  • Aortic dissection in pregnant patients results in an inpatient mortality rate of 8.6%. Owing to the pronounced mortality rate and speed at which aortic dissections progress, efficient early detection methods are crucial. Here, we highlight the importance of early chest computed tomography (CT) for differentiating aortic dissection from pulmonary embolism in pregnant patients with dyspnea. We present the unique case of a 38-year-old pregnant woman with elevated D-dimer and N-terminal pro-brain natriuretic peptide (NT-proBNP) levels, initially suspected of having a pulmonary embolism. Initial transthoracic echocardiography did not indicate aortic dissection. Surprisingly, after an emergency cesarean section, a chest CT scan revealed a DeBakey type I aortic dissection, indicating a diagnostic error. Our findings emphasize the need for early chest CT in pregnant patients with dyspnea and elevated D-dimer and NT-proBNP levels. This case report highlights the critical importance of considering both aortic dissection and pulmonary embolism in the differential diagnosis of such cases, which will inform future clinical practice.

Effect of ABA on Disassembly of Chloroplast during Senescence in Detached Leaves of Zea mays

  • The effect of ABA on the chloroplast disassembly of Zea mays was investigated by measuring the changes in the relative distribution of chlorophyll(Chl) between the Chl-protein complexes in ABA treated and untreated sensecting leaves. The reaction center(RC)-light harvesting complex(LHC) regions were rapidly disassembled in the late stage of dark-induced senescence. Plus, during dark-induced senescence, the disassembly of a reaction center of P700 apoproteins containing mainly Chl a was faster than that of a reaction center of LHCI apoproteins containing both Chl a and Chl b. The increase in the relative distribution of Chl-protein complexes in the RC-Core2 in the late stage of senescence was due to the accumulation of core complexes such as CP47/43 and reaction centers including D1/D2 apoproteins disassembled from the RC-Corel containing the dimer of D1/D2 apoproteins. The LHCII region was more stable than the other Chl-protein complexes throughout leaf senscence. Accordingly, it is suggested that the preferential breakdown of Chl a gives rise to the disassembly of Chl a-binding proteins, particularly reaction centers and core complexes during dark-induced senescence, plus the primary target of the photosynthetic apparatus in sensecing leaves would seem to be Chl a along with the proteins associated with Chl a. The application of ABA promoted the disassembly of the P700 apoproteins in the PSI reaction center and the dimer of D1/D2 apoproteins, and the conversion of the trimeric LHCII apoprotein to the monometirc LHCII apoprotein during the middle stage of leaf senescence, thereby suggesting that ABA accelerates the disassembly of both Chl a-binding and Chl a+b-binding proteins, particularly Chl a-binding proteins during the middle stage of leaf senescence.

결핵성 및 악성 흉막염에서 TNF-$\alpha$, TGF-$\beta$ 및 섬유소용해계의 역할 (TNF-$\alpha$ TGF-$\beta$ and Fibrinolytic Parameters in Tuberculous and Malignant Pleural Effusions)

  • 서론 : 결핵성 흉막염은 치료후에도 약 50% 정도에서 흉막비후가 발생한다. 일부 연구에서 흉막액내 TNF-$\alpha$의 증가 및 섬유소융해의 장애등이 흉막비후의 원인으로 제시되었지만, 아직 흉막비후의 발생기전 또는 예측인자에 대하여는 잘 알려져 있지 않다. TGF-$\beta$는 섬유화를 촉진시키며 결핵성 흉막액에서 증가되어 있음이 보고되었다. 한편 결핵성 흉막염 및 악성 흉막염은 림프구 우세성 삼출액 소견을 나타내는 질환으로 감별진단이 중요하다. 본 연구자는 결핵성 흉막염과 악성 흉막염 환자를 대상으로 TNF-$\alpha$, TGF-$\beta$ 그리고 섬유소 융해계 인자들을 측정하여 양 군간에 비교하였고, 결핵성 흉막염에서 치료 후 흉막비후 발생의 예측인자를 찾고자하였다. 방법 : 1997년 2월부터 1999년 8월까지 서울중앙병원에서 결핵성 흉막염과 악성 흉막염으로 진단받고, 흉막액 검체가 보관되었던 각각 35명과 14명을 대상으로 하였다. 결핵성 흉막염은 모두 4제병용 1차 항결핵 치료를 시행하였다. 각 군에서 임상상을 비교하였고, 흉막액에서 일반 흉막액 검사를 시행하고, TNF-$\alpha$, TGF-$\beta$1, tPA, PAI-1, plasminogen, $\alpha$2-antiplasmin, 그리고 D-dimer를 측정하였다. 결핵성 흉막염 환자 중에서 치료를 종료한 30명을 대상으로 흉막비후가 2mm 미만인 군과 2mm 이상인 군으로 나누어 양 군간에 흉막액 검사소견 및 임상상을 비교하였다. 결과 : 결핵성 흉막염 환자에서 TNF-$\alpha$ tPA, PAI-1, plasminogen, $\alpha$2-antiplasmin, 그리고 D-dimer 모두 말초혈액보다 흉막액에서 증가되어 있었고, 악성 흉막액 환자에서는 plasminogen, $\alpha$2-antiplasmin, 그리고 D-dimer가 말초혈액보다 흉막액에서 증가되어 있었다. TNF-$\alpha$, TGF-$\beta$, PAI-1, plasminogen, 그리고 $\alpha$2-antiplasmin 는 악성 흉막액 보다 결핵성 흉막액에서 유의하게 증가되어 있었다. 항결핵치료 종료 후 2mm 이상의 흉막비후가 발생한 군과 발생하지 않은 군 사이에 TNF-$\alpha$, TGF-$\beta$ 그리고 섬유소융해계의 측정치들간의 차이는 발견할 수 없었다. 결론 : 결핵성 흉막염 환자의 흉막액 TNF-$\alpha$ TGF-$\beta$ 및 섬유소융해계가 흉막비후에 관여할 것으로 추측되었으나, 흉막비후 발생의 예측인자는 찾지 못하였다. 또한 양 질환의 감별에도 기존의 검사들에 비하여 추가적 유용성은 없었다.

Tobacco mitochondrial small heat shock protein NtHSP24.6 adopts a dimeric configuration and has a broad range of substrates

  • There is a broad range of different small heat shock proteins (sHSPs) that have diverse structural and functional characteristics. To better understand the functional role of mitochondrial sHSP, NtHSP24.6 was expressed in Escherichia coli with a hexahistidine tag and purified. The protein was analyzed by non-denaturing PAGE, chemical cross-linking and size exclusion chromatography and the $H_6NtHSP24.6$ protein was found to form a dimer in solution. The in vitro functional analysis of $H_6NtHSP24.6$ using firefly luciferase and citrate synthase demonstrated that this protein displays typical molecular chaperone activity. When cell lysates of E. coli were heated after the addition of $H_6NtHSP24.6$, a broad range of proteins from 10 to 160 kD in size remained in the soluble state. These results suggest that NtHSP24.6 forms a dimer and can function as a molecular chaperone to protect a diverse range of proteins from thermal aggregation.

Silver Oxide를 이용한 1,2-벤조티아진 유도체의 비대칭 중합체 합성 및 결정 구조 (Synthesis and Crystal Structure of Asymmetric Dimer of 1,2-Benzothiazine Derivatives Using Silver Oxide)

  • 새로운 비대칭 중합체인 7,7'-substituted (or H)-4-oxo-2,2'-dialkyl-1,2',2,2'-dibenzothiazine-3,3'-dicarboxylic acid methyl ester-1,1,1',1'-tetraoxide 3,4'-yl ethers 2a-d를 silver oxide($Ag_2O$)를 이용한 산화적인 중합반응에 의해 7-substituted (or H)-4-hydroxy-2-alkyl-1,2-benzothiazine-3-carboxylic acid methyl ester 1,1-dioxide 1a-d로부터 합성하였다. 4-Oxo-e,e'-dialkyl-1,1',2,2'-dibenzothiazine-3,3'-dicarboxylic acid methyl ester-1,1,1',1'-tetraoxide 3,4'-yl ether 2c의 구조를 X-ray 결정 구조 분석에 의해 확인하였다. Y의 혼입량, 소결 온도 및 냉각 속도를 실험 인자로 설정하여 BSYT의 상온비저항을 위한 모델식을 반응표면분석법으로 구하였다. 그 결과 Y의 혼입량 변화가 상온비저항에 가장 크게 영향을 미치며, Y의 함량이 약 0.24 mol% 일 때 상온비저항은 최소값을 나타내었다. 실제 실험으로부터 구한 상온비저항 값과 모델식으로부터 계산한 상온비저항 값을 비교한 결과 잘 일치함을 확인하였다.
