TBP (TATA-binding protein)-related factor 2 (TRF2) regulates transcription during a nuber of cellular processes. We previously demonstrated that it is localized in the cytoplasm and is translocated to the nucleus by DNA-damaging agents. However, the cytoplasmic localization of TRF2 is controversial. In this study, we reconfirmed its cytoplasmic localization in various ways and examined its nuclear migration. Stresses such as heat shock, redox agents, heavy metals, and osmotic shock did not affect localization whereas genotoxins such as methyl methanesulfonate (MMS), cisplatin, etoposide, and hydroxyurea caused it to migrate to the nucleus. Adriamycin, mitomycin C and ${\gamma}$-rays had no obvious effect. We determined optimal conditions for the nuclear migration. The proportions of cells with nuclei enriched for TRF2 were 25-60% and 5-10% for stressed cells and control cells, respectively. Nuclear translocation was observed after 1 h, 4 h and 12 h for cisplatin, etoposide and MMS and hydroxyurea, respectively. The association of TRF2 with the chromatin and promoter region of the proliferating cell nuclear antigen (PCNA) gene, a putative target of TRF2, was increased by MMS treatment. Thus TRF2 may be involved in genotoxin-induced transcriptional regulation.
Using BHK cells with stable expression of cardiac $Na^+/Ca^{2+}$ exchanger(BHK-NCX1), reverse mode(i.e. $Ca^{2+}$ influx mode) of NCX1 current was recorded by whole-cell patch clamp. Repeated stimulation of reverse NCX1 produced a cytosolic $Ca^{2+}$-dependent long-term inactivation of the exchanger activity. The degrees of inactivation correlated with NCX1 densities of the cells and were attenuated by reduced $Ca^{2+}$ influx via the reverse exchanger. The inactivation of NCX1 was attenuated by(i) inhibition of $Ca^{2+}$ influx with reduced extracellular $Ca^{2+}$, (ii) treatment with NCX1 blocker($Na^{2+}$), and (iii) increase of cytoplasmic $Ca^{2+}$ buffer(EGTA). In BHK-NCX1 cells transiently expressing TRPV1 channels, $Ca^{2+}$ influx elicited by capsaicin produced a marked inactivation of NCX1. We suggest that cytoplasmic $Ca^{2+}$ has a dual effect on NCX1 activities, and that allosteric $Ca^{2+}$ activation of NCX1 can be opposed by the $Ca^{2+}$-dependent long-term inactivation in intact cells.
The author has studied the quantitative distribution of mast cells in the skin and proventriculus of the developing chicks and changes of their cytoplasmic granules after administration of a single dose of hyaluronidase 400$\mu$/kg into the yolk sac of 10 day-old chick embryos and obtained the following conclusions. 1. The administration of hyaluronidase considerably retarded the appearance of mast cells in the skin and proventriculus of the developing chicks. 2. In 6 to 7 days after treatment of hyaluronidase the number of mast cells showed only one half of those in the control and then began to show gradual resumption and returned to an almost normal status in 7 day chicks. 3. By treatment of hyaluronidase any degranulation of the mast cells could scarcely be observed but marked decrease of their stainability by resolution of the metachromatic granules. According to this experiment it is fairly clear that hyaluronic acid may play a role in the formation of cytoplasmic granules of mast cells.
This study was conducted to investigate the effects of various supplements added into maturation medium of immature porcine oocytes on quantity of cytoplasmic lipid droplets(LD), subsequent fertilization and development to the blastocyst stage in vitro. The basic maturation medium was TCM 199 + 1 ㎍/㎖ FSH, 0.57 mM cystein, 10 ng/㎖ EGF and was supplemented various supplements(10% FBS, 10% pFF, 0.4% BSA, 1.0% BSA, 0.4% PVP, 1.0% PVP). (omitted)

Ionizing radiation is enough energy to interact with matter to remove orbital electrons, neutrons, and protons in the atom. Ionizing radiation like this leads to oxidizing metabolism that alter molecular structure through direct and indirect interactions of radiation with the deoxyribonucleic acid in the nucleus and cytoplasmic organelles or via products of cytoplasm radiolysis. These ionization can result in tissue damage and disruption of cellular function at the molecular level. Consequently, ionizing radiation-induced modifications of ion channels and transporters have been reported. When the harmful effects exceed those of homeostatic biochemical processes, induced biological changes persist and may be propagated to progeny cells. Also, Reactive oxygen species formed on the effect of ionizing radiation can get across into neighboring cells through the cell junctions that are responsible for intercellular chemical communication, and may there bring about changes characteristic to radiation damage. Depending on radiation dose, dose-rate and quality, these protective mechanisms may or may not be sufficient to cope with the stress. This paper briefly reviewed reports on ionization radiation effects on cellular level that support the concept of radiation biology. A better understanding of the biological effects of ionizing radiation will lead to better use of and better protection from radiation.
The purpose of the present study was to examine the effect of naltrexone, an opioid antagonist, on secretion of catecholamines (CA) evoked by cholinergic nicotinic stimulation and membrane-depolarization from the isolated perfused rat adrenal gland and to establish the mechanism of its action. Naltrexone $(3{\times}10^{-6}M)$ perfused into an adrenal vein for 60 min produced time-dependent inhibition in CA secretory responses evoked by ACh $(5.32{\times}10^{-3}M)$ , high $K^+$$(5.6{\times}10^{-2}M)$ , DMPP ($10^{-4}$ M) and McN-A-343 $(10^{-4}M)$ . Naltrexone itself did also fail to affect basal CA output. In adrenal glands loaded with naltrexone $(3{\times}10^{-6}M)$ , the CA secretory responses evoked by Bay-K-8644, an activator of L-type $Ca^{2+}$ channels and cyclopiazonic acid, an inhibitor of cytoplasmic $Ca^{2+}-ATPase$, were also inhibited. However, in the presence of met-enkephalin $(5{\times}10^{-6}M)$ , a well-known opioid agonist, the CA secretory responses evoked by ACh, high $K^+$, DMPP, McN-A-343, Bay-K-8644 and cyclopiazonic acid were also significantly inhibited. Collectively, these experimental results demonstrate that naltrexone inhibits greatly CA secretion evoked by stimulation of cholinergic (both nicotinic and muscarinic) receptors as well as that by membrane depolarization. It seems that this inhibitory effect of naltrexone does not involve opioid receptors, but might be mediated by blocking both the calcium influx into the rat adrenal medullary chromaffin cells and the uptake of $Ca^{2+}$ into the cytoplasmic calcium store, which are at least partly relevant to the direct interaction with the nicotinic receptor itself.
The present study represents the investigation of in vitro immunopotentiating activities of cellular fractions of major lactic acid bacteria found in kimchi (KLAB) and bifidobacteria. The macrophage cells, RAW264.7, were stimulated with heat-killed whole-cell, cell-wall, and cytoplasmic fractions of four strains of KLAB (Leuconostoc mesenteroides, Leuconostoc citreum, Lactobacillus plantarum, and Lactobacillus sake) and two strains of bifidobacteria (Bifidobacterium longum and Bifidobacterium lactis) each, and then the production of nitric oxide (NO) and cytokines including tumor necrosis $factor-\alpha\;(TNF-\alpha)$ and interleukin-6 (IL-6) was measured by Griess and ELISA assays, respectively. Heat-killed wholecell and cell-wall fractions-but not the cytoplasmic fraction-from all strains of KLAB significantly increased the production of NO in RAW264.7 cells, and all fractions from bifidobacteria exerted similar effects. In the production of $TNF-\alpha$, heat-killed whole-cell and cell-wall fractions of L. plantarum showed the strongest effect, followed by L. sake and B. lactis, whereas other KLAB fractions did not exert any effect. In the production of IL-6, only whole-cell and cell-wall fractions of L. plantarum were effective. These results, taken together, indicate that L. plantarum might playa critical role in the immunopotentiating activities of kimchi.
To breed new superior strains, collected strains were characterized and then several white strains were selected as parents. Monokaryons from the parents were isolated and studied. All tested white strains showed same mating genotype. Growth rate of monokaryons were lower than collected dikaryons. New dikaryotic strains were derived from inbreeding method, which means mating between monokaryotic isolates from different white strains having same mating genotype. Some of them showed higher yields of fruitbody than their parents. Specially Fv 4-1 strain showed the best productivity. Furthermore some mating combination showed cytoplasmic effect, when they mated reciprocally.
Objective : In this study, the medicine herbs above were directly treated to cultured normal lung cells and lung carcinoma cells, and the effects were investigated to develope new cancer treatments with increased anti-cancer efficiency as well as decreased side-effects and to suggest more useful clinical therapies. Materials and Methods : In the experiment, WI-26 VA lung normal cell line and A-427 lung carcinoma cell line were cultured. To observe the morphological change of the treated cells, were subjected to Giemsa staining and observed under Reflected Fluorescence microscope. To examine whether cell death occurred, cells observed under Reflected Fluorescence microscope, To investigate the degree of cell death in the nucleus, cells were screened by Laser cytometry ACAS 570. Results : Samgibopye-tang(Shenqibufei-tang) stimulated not only the growth of the normal cells but also that of the carcinoma cells, Wikyeung-tang(Weijing-tang) induced morphological change such as cytoplasmic constriction in the normal cells and the carcinoma cells, but it did not show any strong inhibitory effect on the cell growth. Samso-eum(Shensu-yin) caused severe cell damage in both cell lines, Eunkyo San(Yinqiao-san) significantly damaged the nuclei and caused weak cytoplasmic constriction in both cell lines, Normal cells treated with Gilgyeung-tang(Jingeng-tang) did not show any significant morphological change while some Gilgyeung-tang(Jingeng-tang) treated carcinoma cells were observed to have a normal cell-like shape, interestingly, conclusions : As the results above, Samgibopye-tang(Shenqibufei-tang), Wikyeung-tang(Weijing-tang), and Gilgyeung-tang(Jingeng-tang) helped the growth of both cell lines, and especially Samgibopye-tang(Shenqibufei-tang) showed the best effect, However, Samso-eum(Shensuyin) and Eunkyo-san(Yinqiao-san) caused lethal damage in the normal cells and also showed strong toxicity in the carcinoma cells.
