한국 미숙아 산모와 제대혈의 IL-10 유전자형 빈도와 신생아 호흡 곤란증 발생과의 연관성 (Association between Maternal and Cord Blood Interleukin-10 (-819T/C and -592A/C) Gene Polymorphisms and Respiratory Distress Syndrome in Preterm Korean Infants)

  • 목 적: 본 연구의 목적은 한국 미숙아에서의 IL-10 유전자형의 빈도와 신생아 호흡 곤란증의 발생 빈도와의 연관성을 조사하고자 하였다. 방 법: 이대목동병원에서 2003년도 11월부터 2008년도 7월에 태어난 214명의 미숙아를 대상으로 하였다. 제대혈과 모체혈에서 IL-10 유전자(IL-10 -1082A/G, -819T/C, -592A/C) 의 다형성을 조사하였고, 임상적인 자료는 의무기록조사를 통해서 분석하였다. 결 과: 미숙아를 분만한 한국 엄마들의 유전자형 빈도는 기존의 보고된 바와 달랐다. IL-10-1082GG homozygote는 발견되지 않았고, 다변량 회귀분석에서 호흡곤란증은 IL-10-592AC/CC 유전형에서 AA 유전자형보다 적게 발현되었다(P=0.033). 호흡곤란증의 위험도가 모체가 IL-10-819TC/CC 유전자형인 경우 TT 유전자형보다 적게 발견되었다(P-0.030). 하지만 제대혈에서는 이러한 차이가 없었다. 각각의 유전자형에서 분석해보면 IL-10 A-1082G/T-819C/A-592C 중, ACC haplotype인 경우, 호흡곤란증에서 보호되는 효과가 있었다(P=0.07). 결 론: 모체의 IL-10-592A/C 과 IL-10-819T/C 유전자 다형성이 미숙아에서 호흡곤란증의 발생에 관여한다고 생각된다.

Association of Genetic Polymorphism of IL-2 Receptor Subunit and Tuberculosis Case

  • Tuberculosis (TB) is infectious disease caused by Mycobacterium tuberculosis (MTB) infection. It is known that not only the property of microorganism but also the genetic susceptibility of infected patients is controlled. Interleukin 2 (IL-2) is a cytokine belonging to type 1 T helper (Th1) activity. In addition, IL-2, when infected with MTB, binds IL-2 receptor and promotes T cell replication and is involved in granuloma formation. The aim of this study was to investigate the genetic polymorphisms of the IL-2 receptor gene in tuberculosis patients and normal individuals. We analyzed 22 SNPs in three genes using the genotype data of 443 tuberculosis cases and 3,228 healthy controls from the Korea Association Resource for their correlation with tuberculosis case. IL2RA, IL2RB, and IL2RG genes were genotyped of 16, 4, and 2 SNPs, respectively. Among three genes, only IL2RA gene polymorphisms showed statistically significant association with tuberculosis case. 6 SNPs with high significance were identified in the IL2RA gene. In addition, the linkage disequilibrium (LD) structure of IL2RA gene was confirmed. SNP imputation of IL2RA gene was performed, it was confirmed that more SNPs were significant between case and control. If we look at the results of IL2RA gene analysis above, we can see that genetic polymorphism in the gene expressing $IL-2R{\alpha}$ will regulate the expression level of $IL-2R{\alpha}$, and the change in the immune system involved in $IL-2R{\alpha}$. In this study, genetic polymorphism that may affect host immunity suggests that susceptibility to tuberculosis may be controlled.

한국인 전반적 급진성 치주염 환자에서 IL-10 promoter 유전자 다변성에 관한 연구 (IL-10 gene promoter polymorphisms in Korean gener-alized aggressive periodontitis patients)

  • Genetic polymorphisms associated with aggressive periodontitis have previously been reported. Interleukin-10 is an immunoregulatory cytokine that plays a role in the pathogenesis of periodontitis. Individual capacity for IL-10 production appears to be under genetic influence, The aim of present investigation was to explore possible genetic association of IL-10 gene promoter polymorphisms with generalized aggressive periodontitis. The study population consisted of 37 generalized aggressive periodontitis patients from the Department of Periodontology, Chonnam National University Hospital and 27 control subjects, all the subjects were non-smokers, Genomic DNA was obtained from buccal swab. The IL-10promoter -597, -824, -1082 positions were genotyped by amplifying the polymorphic regions using polymerase chain reaction (PCR) , followed by restriction enzyme digestion and gel electrophoresis. IL-10-597 C (allele 1) to A (allele 2) and IL-10-824 C (allele 1) to T (allele 2) and IL-10-1082 G (allele 1) to A (allele 2) polymorphisms were examined. The results were as follows. 1. In patients, the distribution of genotypes C/C, C/A and NA at Il-10-597 was determined to be 13.5%, 37.8% and 48.7%, respectively and the distribution of genotypes at IL-10-824 was the same as that of IL-10-597. The distribution of genotypes G/G, G/A and NA at IL-10-1082 was found to be 2.7%, 16.2% and 81. 4%, respectively. No statistical difference in genotype distribution was found between the patient and control groups. 2. Allele 2 carriage rate at the three position of the IL-10 promoter region was higher in the control group than the patient group. 3. Allele 2 frequencies at IL-10-597 and -824 positions were higher in female group than male group and its difference was statistically significant(p<0.05). No significant difference in genotype distribution between the control and patient groups. Allele frequency between control and patient groups was not significantly different although allele 2 frequency at the three positions in the IL-10 promoter region appeared to be higher in control group. In conclusion, no clear association between IL-10 gene promoter polymorphisms and generalized aggressive periodontitis in Korean was observed.

TSLP 유전자의 다형성은 한국인 류마티스관절염 발생에 영향을 미치치 않는다 (Thymic Stromal Lymphopoietin (TSLP) Gene Polymorphisms are not Associated with Rheumatoid Arthritis in a Korean Population)

  • TSLP 유전자는 IL-7와 유사한 새로운 조혈성 사이토카인이다. 인간의 TSLP는 상피세포, 기질세포 및 비만세포에서 만들어진다. TSLP는 류마티스관절염 환자의 윤활성 활액에서 높은 발현을 나타낸다. 이전 연구에서 우리들은 사람의 TSLP유전자에서 4개의 유전자다형성 및 한 개의 변이를 발굴하였다. 이 연구에서는, 우리들이 발굴한 TSLP유전자의 유전자다형성의 유전자형 및 대립형질의 비율을 건강한 정상인과 류마티스관절염 환자에서 비교분석하였으며, 류마티스관절염 환자에 있어서 유전자형에 따른 RF 및 anti-CCP의 정도를 비교 분석하였다. 또한, 양쪽 그룹에서 이들 유전자다형성에 의한 일배체형 비율을 비교 분석하였다. 그 결과, 류마티스관절염 환자군과 건강한 정상인 군 사이에 있어서 유전자형, 대립형질 비율뿐만 아니라 일배체형 비율에 큰 차이를 보이지 않았다. 이 결과는 TSLP유전자의 유전자다형성은 류마티스관절염 감수성에 영향을 미치지 않음을 암시한다.

Single Nucleotide Polymorphisms of Cytokine Genes are Associated with Fibrosis of the Intrahepatic Bile Duct Wall in Human Clonorchiasis

  • This study examined the association of cytokine gene polymorph isms with intrahepatic bile duct wall fibrosis in human clonorchiasis. A total of 240 residents in Heilongjiang, China underwent ultrasonography, blood sampling, and stool examination. Single nucleotide polymorphism (SNP) sites for $IFN-{\gamma}$ (+874 T/A), IL-10 (-1,082 G/A, -819 C/T, -592 C/A), $TNF-{\alpha}$ (-308 G/A), and $TGF-{\beta}1$ (codon 10 T/C, codon 25 G/C) genes were observed with the TaqMan allelic discrimination assay. No significant correlation was observed between individual cytokine gene polymorphisms and intrahepatic duct dilatation (IHDD). Among individuals with clonorchiasis of moderate intensity, the incidence of IHDD was high in those with $IFN-{\gamma}$ intermediate-producing genotype, +874AT (80.0%, P=0.177), and in those with $TNF-{\alpha}$ low-producing genotype, -308GG (63.0%, P=0.148). According to the combination of $IFN-{\gamma}$ and $TNF-{\alpha}$ genotypes, the risks for IHDD could be stratified into high (intermediate-producing $IFN-{\gamma}$ and low producing $TNF-{\alpha}$), moderate, and low (low-producing $IFN-{\gamma}$ and high producing $TNF-{\alpha}$) risk groups. The incidence of IHDD was significantly different among these groups (P=0.022): 88.9% (odds ratio, OR=24.0) in high, 56.5% (OR=3.9) in moderate, and 25.0% (OR=1) in low risk groups. SNP of $IFN-{\gamma}$ and $TNF-{\alpha}$ genes may contribute to the modulation of fibrosis in the intrahepatic bile duct wall in clonorchiasis patients.

사람의 Interleukin-29 유전자의 새로운 변이의 단리 및 그들의 연관 (Novel Variations in Human Interleukin-29 and Their Association)

  • 사이토카인과 그들 수용체의 유전자 다형성은 면역작용에 의한 질병들의 발병원인에 있어서 유전적인 인자로 여겨지는 후보물질들로서, 자가면역질환 및 염증성 그리고 감염질환에 민감하게 연관되어 있다고 알려져 있다. 최근 새롭게 보고된 Interleukin-29유전자는 유전학적 질병들의 복잡한 특성을 해결할 수 있는 중요한 후보유전자이지만 이 유전자에 대한 다형성에 대한 연구는 아직 보고된바 없다. 우리는 이 연구에서 처음으로 프로모터부분을 포함한 Interleukin-29 유전자의 전체 지름 DNA에서 유전자의 다형성을 염기서열 분석 방법을 이용하여 탐색하였다. Interleukin-29 유전자의 다형성들이 한국인의 알레르기성 비염의 감염력과 관련되어 있는지를 알아보기 위하여 알레르기성 비염환자 및 알레르기성 비염이 걸리지 않은 정상인의 다형성을 유전자형과 대립유전자의 빈도를 비교분석 하였다. 우리는 이 연구에서 사람의 Interleukin-29 유전자의 한 개의 신규의 다형성 (1184C>A)을 intron 2에서 그리고 한 개의 신규의 변이부위 (-1842_-1841dupGA)를 프로모터에서 찾아냈다. 우리들의 연구 결과는 이들 유전자 다형성 부위 및 변이부위가 알레르기성 비염과 연관은 없는 것으로 밝혀졌다.

Interleukin-10 Gene Promoter Polymorphisms and Risk of Gastric Cancer in a Chinese Population: Single Nucleotide and Haplotype Analyses

  • Objectives: Interleukin (IL) -10 is a potent cytokine with a dual ability to immunosuppress or immunostimulate. We aimed to explore the association of IL10 promoter polymorphisms with risk of gastric cancer (GC) in a Han population in Southwestern China. Methods: We enrolled 308 pairs of GC and control subjects from four hospitals and a community between October 2010 and August 2011 in a 1:1 matched case-control design. Demographic information was collected using a designed questionnaire. IL10-592 A>C and IL10-1082 A>G polymorphisms were determined by Sequenom MassARRAY analysis. Results: Patients with GC reported statistically higher proportions of family history of cancer (29.9% versus 10.7%, P<0.01) and alcohol drinking (54.6% versus 43.2%, P<0.01) than did controls. Similar results were observed in comparison between non-cardia GC patients and controls (P<0.01 and P=0.03). Variant genotypes of IL10-592 A>C and IL10-1082 A>G were not associated with overall GC risk (adjusted OR, 0.94, 95% CI, 0.66-1.33; adjusted OR, 1.00, 95% CI, 0.62-1.60). Sub-analysis showed that the IL10-592 AC/CC variant genotype was associated with decreased non-cardia GC risk (adjusted OR, 0.58; 95% CI, 0.36-0.95). No association was found between any of the IL10 haplotypes established from two polymorphisms and risk of non-cardia GC. Conclusions: In conclusion, our data do not link the two SNPs of IL10-592 and IL10-1082 with overall GC risk. We demonstrate that IL10-592 polymorphism is associated with protective effect against non-cardia GC. Our findings may offer insight into risk associated with the development of GC in this region.

Helicobacter pylori 감염과 Interleukin $1\beta$ 유전자의 다형성에 따른 위암 발생 위험도 (Risk of the Gastric Cancer Associated with the Interleukin $1\beta$ Gene Polymorphism and Helicobacter pylori)

  • Purpose: According to the recent studies, it is shown that the polymorphism of Interleukin $1\beta$ gene is associated with the incidence of gastric cancer caused by the Helicobacter pylori infection. Interleukin $1\beta$ is a cytokine markedly inhibiting gastric acid secretion. Interleukin $1\beta$ production associated with Helicobacter pylori gastric infection may exacerbate mucosal damage including chronic gastritis and atrophic gastritis, may induce eventual neoplasia. Among these Interleukin $1\beta$ gene polymorphisms, polymorphisms at -31 portion and -511 portion may associated with these processes, eventually increase the risk of gastric cancer. We investigated the risk of gastric cancer according to the Helicobacter pylori infection and genetic polymorphism of Interleukin $1\beta$ in gastric cancer patients. Materials and Methods: 176 individuals with gastric cancer and 40 healthy controls were analyzed. Each group was divided into two groups whether they infected with Helicobacter pylori or not. DNA was extracted from the peripheral blood in all groups. The PCR-RFLP method was used for investigating the distribution of genotype of C/C, C/T, T/T at -31 portion and -511 portion. Results: T/T genotype at -511 portion was $19.3\%$ in gastric cancer cases and $10\%$ in controls, which was statistically significant. (P=0.0432) The risk of gastric cancer was increased 4.86 ($1.26\∼18.77$) in group which had T/T genotype. In gastric cancer cases, C/C genotype at 31 portion was $27.6\%$ in group with Helicobacter pylori infection and $12.8\%$ in group without infection, which was statistically significant. (P=0.0047) The risk of gastric cancer was increased 4.82 ($1.81\~12.81$) in group which had C/C genotype. Conclusion: T genotype at -511 portion among the Interleukin $1\beta$ genetic polymorphisms may be the risk factor of gastric cancer. And, with Helicobacter pylori infection, C genotype at -31 portion may be the risk factor of gastric cancer.

사람의 IL-27p28 유전자 다형성은 알레르기성 비염 환자 혈청의 IgE 양과 연관됨 (Human IL-27p28 Gene Polymorphisms are Associated with the Serum Total IgE Levels of Allergic Rhinitis Patients)

  • 인터루킨 27(IL-27)은 인터루킨 12계열의 이성이량체 사이토카인으로 EBI3와 p28의 두 서브유니트로 구성 되어 있다. 인터루킨 27은 Th1 개시의 초기 조절에 관여하는 사이토카인으로 Th2 인자인 GATA-3의 작용을 억제하는 역할을 한다. 이 사이토카인은 $CD4^+$ T 림프구와 자연살해세포에서 아주 높게 발현하는 수용체(WSX-1)에 의하여 매개된다. 우리들은 사람의 IL-27p28유전자에서 네 개의 유전자다형성 부위를 찾아서 이들 유전자다형성이 기관지 천식의 감수성에 영향을 미치는 것을 보고한 바 있다. 이 연구에서는 IL-27p28 유전자의 유전자다형성이 알레르기성 비염의 감수성에 영향을 미치는 지를 알아보기 위하여 이들 다형성에 있어서 알레르기성 비염환자 군과 정상인군 사이의 유전자형 및 대립형질의 빈도를 비교분석 하였다. 비록 알레르기성 비염환자 군에서 IL-27p28 유전자의 유전자다형성의 유전자형 및 대립형질의 빈도의 차는 정상인 군의 그것과 큰 차이가 없었으나, g.2905T>G SNP에서 두 그룹간에 주목할만한 차이(P=0.037)를 발견하였다. 이 결과는 IL-27p28 유전자의 g.2905T>G 유전자 형성이 알레르기성 비염환자에서 IgE의 생산에 영향을 준다는 것을 암시한다.
