  • The cytochrome c oxidase subunit I (COI) gene region of mitochondrial DNA (mtDNA) was examined in four abalone species to estimate its utility as a genetic marker using restriction fragment length polymorphism (RFLP) analysis. The utility was evaluated in terms of genetic divergence and relationships among Haliotis discus hannai, H. rufescens, H. rubra, and H. midae in both hemispheres of the world. There was clear genetic divergence in the mtDNA COI region between all pairs of the four species. Moreover, relationships among the abalone species were reflected in their geographical distributions and morphological characteristics. Therefore, RFLP analysis of the mtDNA COI region is a suitable genetic marker for the estimation of genetic divergence and relationships among abalone species. However, it is not effective for the evaluation of genetic differences within abalone species.

  • Apicidin, a histone-deacetylase inhibitor, has been successfully used to inhibit the growth of cancer cells. In this study, the apoptotic potential and mechanistic insights of apicidin were investigated in human myeloid leukemia U937 cells. Treatment of U937 cells with apicidin resulted in a decrease of cell viability with apoptotic characteristics, including chromatin condensation and ladder-pattern fragmentation of genomic DNA. Apicidin converted the procaspase-3 protease to catalytically active effector protease, resulting in subsequent cleavage of poly (ADP-ribose) polymerase (PARP) and inhibitor of caspase-activated deoxyribonuclease (ICAD). In addition, apicidin induced the activation of caspase-9 protease and the cytosolic release of mitochondrial cytochrome c with mitochon-drial membrane potential transition. Moreover, apicidin transiently increased the expression of Fas and Fas ligand proteins. Taken together, the results suggest that apicidin induces apoptosis of U937 cells through activation of intrinsic caspase cascades and Fas/FasL system with mitochondrial dysfunction.

  • Platorchestia monodi (Mateus et al., 1986), the beach hopper, commonly lives in sea shores and was previously reported only from the Atlantic regions, such as Mid-Atlantic islands, Brazilian coast, and Israel. We present the first record of this species in the Pacific region. The present species can be clearly distinguished from related species by the shapes of gnathopods 1-2 and development of sexual dimorphism in antenna 2 and pereopod 7. Descriptions of the diagnostic characteristics of the species are provided on the text. In addition, this paper also provided the partial sequence of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene of the species for its molecular characteristics.

  • Fifty-five specimens of Octopus longispadiceus, belonging to the family Octopodidae, were collected for the first time from the East Sea of Korea and identified by DNA barcoding. This species is characterized by its long right third arm and ligula, the presence of enlarged suckers in the mature male, small white spots on the mantle, head, and arms, and no cirrus above the eye. A molecular analysis of the partial cytochrome c oxidase subunit I gene showed that these specimens are all the same species and have the smallest genetic distance with O. longispadiceus (Kimura- two-parameter distance = 0.002-0.003). A new Korean name, "Bal-mun-eo" is proposed for this species.

  • A sea cucumber was collected from Gonghyeonjin in the East Sea of Korea at a depth of 50 m on 22 June 2011 and was identified as Psolus phantapus (Strussenfelt, 1765). This species belongs to the family Psolidae of the order Dendrochirotida based on morphological characteristics and mitochondrial cytochrome c oxidase subunit I sequence analysis. Psolus phantapus, which widely distributes in the Arctic and North Atlantic Oceans, is newly recorded in the Korean fauna. Two Psolus species including the previously reported P. squamatus are recorded in the East Sea of Korea.

  • We previously reported that a synthetic naphthoquinone analog. 2.3-dichloro-5.8-dihydroxy-1, 4-naphthoquinone (NA). effectively induces apoptosis in human leukemic HL-60 cells. However. the cellular mechanism by which NA induces cell death remain unclear. In this study. we show that NA induces activation of capases. release of cytochrome c and upregulation of proapoptotic Bax protein. Futhermore. NA suppressed phosphorylation of Akt and Bad. suggesting that Akt regulates NA-induced apoptosis. Expresson of a dominant negative Akt enhancde NA-induced apoptosis. suggesting that naphthoquinone analog induces apoptosis through activating proapoptotic pathway and by the inactivation of antiapoptotic pathway.

  • Two marine isopods, Elaphognathia sugashimaensis (Nunomura, 1981) and Metacirolana shijikiensis Nunomura, 2008, have been reported for the first time in South Korea. Specimens of E. sugashimaensis and M. shijikiensis were collected using light traps from Yeongdeok-gun and Gageo-do Island in South Korea, respectively. The genera Elaphognathia Monod, 1926 and Metacirolana Kussakin, 1979 are new to South Korea. In this paper, we provide descriptions of diagnostic characteristics and illustrations of their morphologies. Additionally, the partial sequences of mitochondrial cytochrome c oxidase subunit 1 (CO1) of two species are provided as molecular characteristics.

  • Two feather mites, Zachvatkinia larica Mironov, 1989 and Ardeacarus ardeae (Canestrini, 1878) are reported for the first time in Korea. Specimens of Z. larica and A. ardeae were collected from the blacktailed gull Larus crassirostris and little egret Egretta garzetta, respectively. The genera Zachvatkinia Dubinin, 1949 and Ardeacarus Dubinin, 1951 are new to this country. Here, we provided morphological descriptions and illustrations of these two species. Additionally, we provide the partial sequences of the mitochondrial cytochrome c oxidase subunit I (COI) as DNA barcode markers.

  • The branchiobdellidan species Holtodrilus truncatus (Liang, 1963) is reported for the first time in Korea. The genus Holtodrilus Gelder and Brinkhurst, 1990 is monotypic within the family Xironodrilidae, and the genus and family of this species are also new to the Korean fauna. The branchiobdellidan specimens were collected from the exoskeletons of the freshwater atyid shrimp, Neocaridina sp., sampled from the 'Andeok Valley', Jeju-do Province, South Korea. Holtodrilus truncatus is characterized by its terete body form, 7/7 dental formula (with a large medial tooth and three pairs of smaller lateral teeth), and no trunk appendages. In addition, we determined a partial sequence of the mitochondrial cytochrome c oxidase subunit 1 (CO1) gene as a DNA barcode marker for H. truncatus. The present study is the first record of the branchiobdellidan occurrence on a non-crayfish host (shrimp) in Korea. To date, seven species (four genera in three families) of branchiobdellidans have been reported in the Korean fauna.

  • 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD) is known to induce cytochrome p450 1A1 and to activate c-Src kinase and p21 Ras. This study examined the molecular interactions of adaptor proteins including Shc, Grb2, and Sos in rat primary hepatocytes and their relationship to the induction of CYP1A1 by TCDD. TCDD induced CYP1A1 level and EROD activity in a dose-dependent mode. Sos/Grb2 association isincreased by TCDDㅑㅜ a dose dependent mode. Tyrosine phosphorylated Shc, mainly p152, onloads to Grb2/Sos complex upon TCDD stimulation. The electrophoretic mobility shift of Sos is showed by TCDD. These results indicate that TCDD modulated the molecular interaction features of adaptor compoes proteins including Shc, Grb2, and Cnl in early signaling pathway of TCDD-mediated CYP 1A1 induction of rat primary hepatocyte.
