과산화수소에 노출된 인간 각질형성세포에서 길이가 다른 시스테인 함유 펩타이드의 항산화 효과 (Antioxidant Effects of Cysteine-containing Peptides of Different Lengths in Human HaCaT Keratinocytes Exposed to Hydrogen Peroxide)

  • H2O2는 세포에서 산화 스트레스를 유발하여 세포 성장, 증식, 노화 및 사멸에 영향을 미치는 일종의 활성산소종(ROS)이다. 본 연구의 목적은 H2O2의 세포 독성을 완화시키는 활성 펩타이드를 찾는 것이다. 잠재적인 활성 펩타이드의 서열을 예측하기 위해서 위치 스캐닝 합성 테트라펩타이드 조합 라이브러리를 탐색하였다. H2O2로 유도된 인간 각질형성세포(HaCaT 세포)의 사멸에 대한 펩타이드 풀들의 완화 효과를 비교한 결과, 다양한 활성 펩타이드의 시퀀스가 예측되었다. 특히 시스테인(C) 잔기를 함유한 펩타이드가 활성이 있을 것으로 예측되었다. 후속 실험에서 C-NH2, CC-NH2, CCC-NH2, CCCC-NH2 등의 길이가 다른 시스테인 함유 펩타이드들의 세포 독성과 활성을 조사하였다. C-NH2 및 CC-NH2는 1.0 mM 이하에서 유의한 세포 독성이 없었지만 CCC-NH2 및 CCCC-NH2는 상대적으로 강한 세포 독성을 보였다. C-NH2와 CC-NH2는 H2O2로 유도된 세포독성을 완화시켰다. CC-NH2는 C-NH2, C, N-아세틸 시스테인(NAC) 및 글루타티온(GSH)보다 세포 보호 효과가 높았다. 유세포 분석법으로 세포 내 ROS를 측정하였을 때, H2O2는 ROS의 생성을 증가시켰다.H2O2 노출조건에서 CC-NH2는 C-NH2보다는 더 효과적으로 ROS 생성을 억제하였고, C, NAC, GSH 만큼 효과적이었다. 본 연구의 결과는 다양한 시스테인 함유 펩타이드 중 특히 CC-NH2가 H2O2로 유도된 세포 독성과 ROS 생성을 안전하고 효과적으로 완화시키는 항산화 특성이 있음을 시사한다.

Characterization of Extremely Hydrophobic Immunostimulatory Lipoidal Peptides by Matrix Assisted Laser Desorption Ionization Mass Spectrometry

  • Synthetic lipoidal peptides based on viral protein sequences have been prepared. These peptides contain an N-palmitoyl group at the N-terminal residue, which is a modified cysteine, containing a S-[2,3-bis(acyloxy)-(2-R,S)-propyl] moiety. When this residue (Pam3Cys) is at the N-terminus of a synthetic peptide, it acts as potent immunoadjuvant to enhance both IgM and IgG antibody responses to the attached peptide. Conventional analytical procedures (e.g., Edman degradation and amino acid analysis) are either not applicable due to the N-terminal modification, or do not provide confirmation of the intact structure. Chromatographic analysis is also hindered by the tendency of these lipoidal Pam3Cys peptides to form large aggregates, and in some cases to be permanently adsorbed on reversed phase columns. We have applied several mass spectrometric techniques, including fast atom bombardment (FAB), electrospray ionization (ESI) and matrix assisted laser desorption ionization (MALDI) to characterize the intact structures of a number of different Pam3Cys synthetic peptides. The MALDI-MS has been found to be the most sensitive for the analysis of the structure of Pam3Cys peptides.

  • The trend towards ageing populations has been observed over many years in Europe and the US but has accelerated significantly in developed countries in Asia including Japan and South Korea. In the latter country the elderly population (65+) has increased 5-fold between 1960 and 2000 and this group will comprise 40% of the population by 2050. This creates a new socio-economic group with specific demands and considerable spending power. As ageing occurs a range of changes occur in the body that can be moderated by adjustments in nutrition. A significant body of evidence points to changes in the balance of glutathione synthesis and utilisation as people age. Glutathione is tile most important natural anti-oxidant of the body and the amounts present can become limited by available cysteine in the diet. A cysteine-enriched peptide product, Cysteine Peption$^{TM}$ has been developed by DMV International for dietary supplement and food applications. A qualitative consumer trial has indicated benefits including improved sleep and more energy. Animal and clinical trial will be described that provide indications on bioavailability and possible mechanisms of action of Cysteine Peption$^{TM}$ with particular focus on the ageing population.

The Oxidative Modification of COL6A1 in Membrane Proteins of Ovarian Cancer Patients

  • Ovarian cancer is the most lethal gynecological malignancy, and specific biomarkers are important needed to improve diagnosis, prognosis, and to forecast and monitor treatment efficiency. There are a lot of pathological factors, including reactive oxygen species (ROS), involved in the process of cancer initiation and progression. The oxidative modification of proteins by ROS is implicated in the etiology or progression of disorders and diseases. In this study, a labeling experiment with the thiol-modifying reagent biotinylated iodoacetamide (BIAM) revealed that a variety of proteins were differentially oxidized between normal and tumor tissues of ovarian cancer patients. To identify cysteine oxidation-sensitive proteins in ovarian cancer patients, we performed comparative analysis by nano-UPLC-$MS^E$ shotgun proteomics. We found oxidation-sensitive 22 proteins from 41 peptides containing cysteine oxidation. Using Ingenuity program, these proteins identified were established with canonical network related to cytoskeletal network, cellular organization and maintenance, and metabolism. Among oxidation-sensitive proteins, the modification pattern of Collagen alpha-1(VI) chain (COL6A1) was firstly confirmed between normal and tumor tissues of patients by 2-DE western blotting. This result suggested that COL6A1 might have cysteine oxidative modification in tumor tissue of ovarian cancer patients.

Synthesis of 18F-labeled 2-cyanobenzothiazole derivative for efficient radiolabeling of N-terminal cysteine-bearing biomolecules

  • This article provides an efficient 18F-labeling protocol based on a rapid condensation reaction between 2-cyanobenzothiazole (CBT) and N-terminal cysteine-containing biomolecules. The 18F-labeled CBT (18F-1) was prepared by radiofluorination of the tosylated precursor 4 with 18-crown-6/K+/[18F]F- complex. Using the purified 18F-1, 18F-labeled peptide (18F-7) and protein (18F-8) could be synthesized efficiently under mild conditions. This strategy would provide a convenient approach for rapid and site-specific 18F-labeling of various peptides and proteins for in vivo imaging and biomedical applications.

  • 노인 인구 증가는 유럽과 미국에 걸쳐 몇 년간 관찰되어 왔지만, 일본과 한국과 같은 개발도상국에서도 증가 추세가 현저히 나타나고 있다. 개발도상국에서는 2000년, 65세 이상 노인의 인구비율이 1960년에 비해 약 5배 정도 증가했으며, 2050년에는 전체 인구의 40% 정도를 차지할 것으로 예상되고 있다. 이런 급격한 인구 변화는 이들이 특정 권리와 구매력을 지니는 새로운 사회경제적 집단으로 성장하게 하는 계기가 되었다. 노화가 일어나면, 식이요법으로 어느 정도 조절할 수 있는 인체의 변화가 다양하게 일어난다. 그 중 대표적인 것은 글루타치온 합성과 이용의 균형에 변화가 생기는 것이다. 글루타치온은 인체에서 가장 중요한 항산화 물질이고, 식이 내 시스테인 아미노산에 의해 체내 함량이 결정될 수 있다. 시스테인이 풍부한 펩티드 제품이 기능성 식품 및 식품원료로 개발되었다. 소비자 조사 결과, 이 제품은 숙면과 활력을 제공하는 등 장점이 있음이 밝혀졌다. 동물 임상 실험 결과를 통해 특별히 노인 인구를 대상으로 하여 시스테인 펩타이드의 생리 활성과 대사 과정을 소개하고자 한다.

Molecular Cloning and Characterization of a Flower-specific Thionin in Chinese Cabbage

  • Thionins are a family of low molecular weight cysteine-rich antimicrobial peptides. We isolated a cDNA encoding thionin gene from a flower bud cDNA library of Chinese cabbage (CFT). The gene contains 611 by nucleotides with 60 bp, and 150 by untranslated regions at its N- and C-terminal, respectively. The deduced amino acid sequence encoded 133 amino acids containing precursor polypeptide. The protein reveals that the precursor has a tripartite structure: a putative signal sequence at the N-terminus, followed by a mature thionin peptide, and a C-terminal acidic domain, which facilitates transport of the mature thionin through membrane. Genomic Southern blot analysis suggests that the CFT gene may be present as a single or two copy gene in the Chinese cabbage genome. Northern blot analysis shows that the gene is specifically expressed in flowers, but not in leaves, stems, or roots. When we analyzed the antifungal activity of the recombinant CFT protein, which was expressed in E. coli using the truncated cDNA region corresponding to the mature protein part, it was not active on fungal growth inhibition.

Antioxidant and Antihypertensive Activities of Grains Grown in South Korea in Relation to Phenolic Compound and Amino Acid Contents

  • Hypertension is characterized by excessive renin-angiotensin system activity, leading to blood vessel constriction. Several synthetic compounds have been developed to inhibit renin and angiotensin-converting enzyme (ACE). These drugs often have adverse side effects, driving the exploration of plant protein-derived peptides as alternative or supplementary treatments. This study assessed the phenolic compound and amino acid content and the antioxidant and antihypertensive activity of 5 South Korean staple crops. Sorghum had the highest phenolic compound content and exhibited the highest antioxidant activity. Millet grains, particularly finger millet (38.86%), showed higher antihypertensive activity than red beans (14.42%) and sorghum (17.16%). Finger millet was found to contain a large proportion of branched-chain, aromatic, and sulfur-containing amino acids, which are associated with ACE inhibition. In particular, cysteine content was positively correlated with ACE inhibition in the crops tested (r=0.696, p<0.01). This study confirmed that the amino acid composition was more correlated with the antihypertensive activity of grains than the phenolic compound content. Finger millet mainly contained amino acids, which have higher ACE inhibitory activity, resulting in the strongest antihypertensive activity. These findings underscore the antihypertensive potential of select crops as plant-based food ingredients, offering insight into their biological functions.
