가미옥액탕이 즉시형 과민반응에 미치는 영향 (Effect of Gamiokyaek-tang on Immediate Type Hypersensitivity)

  • Object : We investigated the effect of Gamiokyaek-tang (GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl (DNP) IgE-mediated passive cutaneous anaphylaxis and acetic acid-induced vascular permeability in rodents. Also perfonned MTT assay and b-hexosaminidase activity is measured in RBL-2H3. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5 mg/ml didn't have an effect on cell viability and cytotoxicity. b-hexosaminidase release in RBL-2H3 was significantly reduced by 2 and 5 mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity .

  • Object : We investigated the effect of Gamiokyaek-tang(GOYT) on immediate type hypersensitivity. Methods : We investigated anti-dinitrophenyl(DNP) IgE-mediated passive cutaneous anaphylaxis(PCA) and acetic acid-induced vascular permeability in rodents. Also we measured MTT assay, ${\beta}-hexosminidase$ activity and IL-4 from RBL-2H3 and nitric oxide from Raw264.7. Results : GOYT inhibited passive cutaneous anaphylaxis and acetic acid-induced vascular permeability by oral administration. All the concentrations of GOYT from 0.1 to 5mg/ml didn't have an effect on cell viability and cytotoxicity. In RBL-2H3, ${\beta}-hexosminidase$ release and IL-4 production were significantly reduced by 1, 2 and 5mg/ml of GOYT. In Raw264.7, nitric oxide level was decreased by 5mg/ml of GOYT. Conclusion : These results indicate that GOYT have inhibition effects on immediate type hypersensitivity.

행인 제제의 독성 및 항암효과의 자연살해세포 활성에 미치는 효과 (Study on toxicity, anti-cancer and NK cell activity of Lateril oil)

  • In this experiment, we examined the the effect of Laetrile Oil to the immune system and tried to disclose the anti-cancer mechanism of this. The result is listed as below. 1. After per-os, sub-cutaneous and into the peritoneal injection of Laetrile Oil to the SPF mice, the LD50 is above 5000mg/kg at even group. 2. Mean survival days of mice treated with laetril oil, after S-180 cells transplantation into the peritoneal cavity decreases 1.5 days(-6.8%) compared with the Mean survival days of the control group(22days.) 3. Effects of laetril oil on natural killer cell activity at Effector/Target Cell Ratio with 100:1, 50:1, 10:1 into methotrexate-pretreated mice is like this.: Compared with $29.22\pm12.7\%$ Cytotoxicity of the control group, sample group's Cytotoxicity had $38.83{\pm}12.5%$ of meaningful acrease. At 100:1 Effecr/Target Cell Ratio. At 50:1 Effector/Target Cell Ratio, control group has $20.02{\pm}9.6%$ Cytotoxiciy and sample group had $31.53{\pm}13.4%$ Cytotoxicity. At 25:1 Effector/Target Cell Ratio control group has $13.60{\pm}6.6%$ Cytotoxicity and sample group had $20.81{\pm}9.8%$ Cytotoxicity According to the above results, the Laetrile Oil represents nontotoxic to a SPF mice. non-effective to transplantable Sarcoma 180 tumors, and activaion in NK cell activity.

자울약침액(紫菀藥鍼液)의 폐유(肺兪) 처치(處置)가 Type I Hypersensitivity에 미치는 영향 (Effect of Radix Asteris Herbal Acupuncture at $BL_{13}$ on the Type I Hypersensitivity)

  • Objectives : We studied the effects of Radix Asteris herbal acupuncture solution (RAHAS) on the type I hypersensitivity. Methods : In vivo, we measured compound 48/80 induced active systemic anaphylactic shock, anti-DNP IgE induced passive cutaneous anaphylaxis (PCA) and acetic acid induced microvascular permeability using ICR mice. In vitro, we showed effects on cytotoxicity and ${\beta}$-hexosaminidase release from RBL-2H3 cells. Results : In vivo, RAHAS pretreatments at $BL_{13}$ and optional points inhibited active systemic anaphylactic shock induced by compound 48/80 and microvascular permeability increased by acetic acid. PCA was only inhibited by RAHAS pretreatments at $BL_{13}$. In vitro, RAHAS treatments inhibited ${\beta}$-hexosaminidase release. Conclusion : These results suggest that RAHAS may be beneficial in the prevention of type I hypersensitive inflammatory response.

황금약침(黃芩藥鍼)이 Type 1 Hypersensitivity에 미치는 영향 (Effect of scutellariae radix pharmacopuncture on the type 1 hypersensitivity)

  • Objectives : We studied the effects of Scutellariae Radix pharmacopuncture solution (SRHAS) on the type 1 hypersensitivity. Methods : In vivo, we measured compound 48/80-induced active systemic anaphylactic shock using ICR mice and anti-DNP IgE-induced passive cutaneous anaphylaxis (PCA) using Sprague Dawley rats. In vitro, we showed effects on cytotoxicity and ${\beta}-hexosaminidase$ release from RBL-2H3 cells. Results : In vivo, SRHAS pretreatments (100% or 50%) at BL13 inhibited active systemic anaphylactic shock induced by compound 48/80. PCA was only inhibited by pretreatments of SRHAS at optional points. In vitro, $0.1{\sim}2%$ SRHAS treatments did not affect cell viability while ${\beta}$-hexosaminidase release was significantly inhibited. Conclusions : These results suggest that SRHAS may be beneficial in the inhibition of type I hypersensitive inflammatory response.

녹각교 함유 인공피부를 이용한 피부독성도의 검사 (Assessment of Skin Toxicity Using Skin Equivalents Containing Cervi cornus Colla)

  • 피부 자극과 부식 검사에 사용되는 동물실험을 대체하기 위한 방법으로 인공피부가 개발되어 왔다. 최근에 본 연구진은 녹각교를 함유하는 새로운 인공피부를 구축하였다. 현재 연구에서는 녹각교를 포함하는 인공피부를 사용하여 물질의 독성도 검사를 수행하였다. 그리하여 sodium dodecylsulfate (SDS) 또는 sodium carbonate를 인공피부에 도포하였고 표피의 손상 정도를 H&E와 면역조직화학 염색을 통하여 평가하였다. 인공피부의 표피는 SDS와 sodium carbonate에 의해 농도 의존적으로 영향을 받았다. 더 나아가 이들 물질에 의하여 p63의 발현이 감소하였다. 그러므로 녹각교를 함유하는 인공피부는 동물실험을 대체할 수 있는 모델로써 사용될 수 있고 in vitro에서 자극과 부식 검사 시험법의 발전에 도움을 줄 것으로 예상된다.

Lipolytic Enzymes Involved in the Virulence of Human Pathogenic Fungi

  • Pathogenic microbes secrete various enzymes with lipolytic activities to facilitate their survival within the host. Lipolytic enzymes include extracellular lipases and phospholipases, and several lines of evidence have suggested that these enzymes contribute to the virulence of pathogenic fungi. Candida albicans and Cryptococcus neoformans are the most commonly isolated human fungal pathogens, and several biochemical and molecular approaches have identified their extracellular lipolytic enzymes. The role of lipases and phospholipases in the virulence of C. albicans has been extensively studied, and these enzymes have been shown to contribute to C. albicans morphological transition, colonization, cytotoxicity, and penetration to the host. While not much is known about the lipases in C. neoformans, the roles of phospholipases in the dissemination of fungal cells in the host and in signaling pathways have been described. Lipolytic enzymes may also influence the survival of the lipophilic cutaneous pathogenic yeast Malassezia species within the host, and an unusually high number of lipase-coding genes may complement the lipid dependency of this fungus. This review briefly describes the current understanding of the lipolytic enzymes in major human fungal pathogens, namely C. albicans, C. neoformans, and Malassezia spp.

생인공피부를 이용한 독성 반응 시험 (Use of Cultured Bioartificial Skins as in vitro Models for Cutaneous Toxicity Testing)

  • Cytotoxicity assays using artificial skins have been proposed as in vitro alternatives to minimize animal ocular and dermal irritation testing. Accordingly, the responses of artificial skins to the well-characterized chemical irritants toluene, glutaraldehyde, and sodium lauryl sulfate (SLS), and the nonirritant polyethylene glycol were studied. The evaluation of the irritating and non-irritating test chemicals was also compared with the responses observed in human dermal fibroblasts and human epidermal keratinocytes grown in a monolayer culture. The responses monitored included an MTT mitochondrial functionality assay. In order to better understand the local mechanisms involved in skin damage and repair, the production of several mitogenic proinflammatory mediators, interleukin-l$\alpha$, 12-HETE, and 15-HETE, was also investigated. Dose-dependent increases in the levels of かIn and the HETEs were observed in the underlying medium of the skin systems exposed to the two skin irritants, glutaraldehyde and SLS. The results of the present study show that both human artificial skins can be used as efficient in vitro testing models for the evaluation of skin toxicity and for screening contact skin irritancy.

牛蒡子추출물 및 분획층이 항알레르기에 미치는 實驗的 硏究 (The effects of Arctii fructus extract on the allergenic inflammation reactions.)

  • The effects of Arctii fructus extract on allegenic inflammation were investigated using in vivo and in vitro test models. Firstly, the cytotoxicity of Arctii fructus extract was validated using MTT assay. As a result, Arctii fructus extract showed no cytotoxic potential, while SDS, a positive control, revealed strong cytotoxic effect. In LLNA assay, Arctii fructus extract showed no skin allergenicity. Next, the anti-allergic actions of Arctii fructus extract were evaluated using rodent experimental models. The oral, intraperitoneal and intradermal administration of Arctii fructus extract significantly inhibited the compound 48/80-induced vascular permeability documented by Evans blue extravasation. In addition, Arctii fructus extract showed potent inhibitory effect on passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE when orally administered. In an in vitro study, Arctii fructus extract revealed to possess inhibitory potential on the compound 48/80-induced histamine release from rat peritoneal mast cells. Moreover, Arctii fructus extract inhibited the IL-4 and TNF-${\alpha}$ mRNA induction by PMA and A23187 in human leukemia mast cells, HMC-1. Finally, it revealed that Arctii fructus extract significantly suppressed histamin-provoked antigenic inflammation reactions in human prick test. Taken together, these results suggest that anti-allergic action of Arctii fructus extract may be due to the inhibition of histamine release and cytokine gene expression in the mast cells.

Glabridin Liposome Ameliorating UVB-Induced Erythema and Lethery Skin by Suppressing Inflammatory Cytokine Production

  • Glabridin, a compound of the flavonoid, has shown outstanding skin-whitening and anti-aging properties, but its water insolubility limits its wide application. Therefore, glabridin liposome (GL) has been developed to improve its poor bioavailability, while there are few studies to evaluate its amelioration of UVB- induced photoaging. This study is performed to investigate the amelioration of GL against UVB- induced cutaneous photoaging. The prepared GL has a spheroidal morphology with an average diameter of 200 nm. The GL shows lower cytotoxicity than glabridin, but it has a more effective role in inhibition of melanin. Moreover, the application of GL can effectively relieve UV radiation induced erythema and leathery skin, associated with the down-regulated expression of inflammatory cytokines (TNF-α, IL-6 and IL-10). Taken together, these results demonstrate that GL has potentials as topical therapeutic agents against UVB radiation induced skin damage through inhibiting inflammation.
