Ionic Equilibria in Mixed Solutions of Cuprous and Cupric Chloride (염화 제1구리와 제2구리 혼합용액의 이온평형)

  • The ionic equilibira in mixed solutions of cuprous and cupric chloride were analyzed by considering chemical equilibria, mass and charge balance equations. The activity coefficients of solutes were calculated by using Bromley equation. Required thermodynamic constants and interaction parameters were evaluated from the data reported in the literature. The effect of NaCl and CuCl concentrations on the pH and potential of the mixed solutions was explained in terms of the variation in the concentration of solutes and in the activity of hydrogen ion. The calculated pH values of the mixed solutions agreed well with the measured values. However, the calculated values for the potential of the mixed solutions were lower than the measured values, indicating the necessity of considering the complex formation between cuprous and chloride ion, such as $Cu^2Cl{_4}^{2-}$ and $Cu_3Cl{_6}^{3-}$.

  • Aflatoxin $B_1$ ($AFB_1$) was reacted with ascorbic acid (AA) alone, with AA plue cysteine and with AA plus cupric ion for 5 days (at $37^{\circ}C$ and pH 5), and the mutagenicity of the reaction products was tested with Salmonella typhimurium TA 100. About 10% of AFBl induced mutagenesis was reduced when $AFB_1$ reacted with AA. This decreasing effect was more severe when $AFB_1$ reacted with AA plus cysteine. The mutagenicity of $AFB_1$ when reacted with AA plus cupric ion was almost completely inhibited, however, eupric ion itself was shown to enhance the mutagenicity of $AFB_1$. Therefore, $AFB_1$ may be degraded in the presence of AA under the given reaction condition and the reaction products was observed to have nonmutagenic effects on the bacterial mutagenecity trials.

  • The chitosan solution was prepared by dissolving chitosan into 2 wt % aqueous acetic acid solution and then chitosan beads were made by sol-gel method. The average molecular weight and the degree of deacetylation of the chitosan used here were determined to be $8.2{\times}10^5$ and 85%, respectively. chitosan beads were highly porous which was confirmed by SEM photography and BET. Adsorption equilibrium of $Cu^{2+}$ on porous chitosan beads could be represented by Sips equation. The diffusion of cupric ions in the chitosan beads could be explained by pore and surface diffusion mechanisms. Adsorption dynamics of $Cu^{2+}$ in fixed-bed could be simulated by linear driving force approximation (LDFA). It was proven that porous chitosan beads manufactured in this work are good adsorbents for the removal of $Cu^{2+}$.

  • The frameworks of $(Cu(NH_3)_3OH^+)_x(NH_4^+)_{12-x}-A{\cdot} zH_2O$ which were prepared by the ion-exchange of zeolite A with ammoniac cupric nitrate solution are more stable than those of $Cu_xNa_{12-2x} -A$ obtained by the ion exchange with aqueous cupric nitrate solution are more stable than those of $Cu_xNa_{12-2x} -A$ obtained by the ion exchange with aqueous cupric nitrate solution. An energetic calculation was made on the relatively stable $(CuOH^+)_2(NH_4^+)_{10}-A{\cdot} 2H_2O$ prepared by the partial evacuation of $(Cu(NH_3)_3OH^+)_2(NH_4^+)_{10}-A{\cdot} zH_2O$. The mean stabilization energies of water, OH-, and $NH_4^+$ ions are -30.23 kcal/mol, -60.24 kcal/mol, and -16.65 kcal/mol, respectively. The results of calculation were discussed in terms of framework stability. The $(Cu(NH_3)_3OH^+)_2(NH_4^+)_{10}-A{\cdot} zH_2O$ zeolite shows two step deammoniation reactions. The first deammoniation around 210 $^{\circ}$C (third DSC peak) was attributed to the decomposition of $[Cu(NH_3)_3OH^+]$ ion and the second one around 380 $^{\circ}$C (fourth DSC peak) was ascribed to the decomposition of $NH_4^+$ ion. The activation energies of the first and second deammoniation reactions were 99.75 kJ/mol and 176.57 kJ/mol, respectively.

Effects of graded levels of cupric citrate on growth performance, antioxidant status, serum lipid metabolites and immunity, and tissue residues of trace elements in weaned pigs

  • Objective: The goal of this study was to investigate the effects of cupric citrate (CuCit) on growth performance, antioxidant indices, serum lipid metabolites, serum immune indices, and tissue residues of copper (Cu), zinc, and iron in weaned pigs. Methods: A total of 180 weaned pigs ($Duroc{\times}Landrace{\times}Large$ White) with an average body weight of $8.98{\pm}1.21kg$ were randomly assigned to a corn-soybean meal control ration, or 4 similar rations with 30, 60, 120, or 240 mg/kg Cu as CuCit. All diets contained 10 mg/kg Cu as cupric sulfate from the vitamin-mineral premix. The experiment was divided into two phases: 0 to 14 d (phase 1) and 15 to 28 d (phase 2). Results: Average daily gain (ADG; linearly, p<0.01) and average daily feed intake (ADFI; linearly and quadratically, p<0.05) were affected by an increase in CuCit during phase 2. Overall period, ADG (p<0.05) and ADFI (p<0.01) were linearly increased with increasing dietary levels of CuCit. Serum malondialdehyde concentrations (p<0.05) and glutathione peroxidase activity (p<0.01) linearly decreased and increased respectively with an increase in CuCit. Serum levels of Cu-Zn superoxide dismutase were linearly affected with an increase in CuCit (p<0.01). Hepatic malondialdehyde levels decreased with an increase in CuCit (linearly and quadratically, p<0.01). Serum total cholesterol concentrations were quadratically affected (p<0.05) and decreased in pigs fed Cu as CuCit at 60 and 120 mg/kg and increased in pigs fed 240 mg/kg Cu as CuCit. Serum high-density lipoprotein concentrations were linearly affected with an increase in CuCit (p<0.01). Serum $IL-1{\beta}$ levels were quadratically affected (p<0.05) by dietary treatment. Compared with other treatments, 240 mg/kg Cu from CuCit quadratically increased hepatic (p<0.01) and renal (p<0.05) Cu concentrations, and quadratically decreased hepatic and renal iron concentrations (p<0.05). Conclusion: Cu administered in the form of CuCit at a dosage range of 30 to 60 mg/kg, effectively enhanced the growth performance and antioxidant status of weaned pigs.

Microfluidic System for the Measurement of Cupric Ion Concentration using Bilayer Lipid Membrane on Silver Surface (은 표면의 이중층 지질막에 의한 구리 이온 농도 측정용 마이크로플루이딕 시스템)

  • A microfluidic system has been developed using biomaterial for the measurement of cupric ion concentration. The cell-membrane-mimicking bilayer lipid membrane(BLM)-coated silver electrode was used for the sensing of cupric ion concentration. The silver-supported BLM could increase its stability. A silver-supported bilayer lipid membrane(s-BLM) was easily obtained using its self-assembling characteristics by immersing silver wire into lipid(phosphatidylcholine; PC) solution and then dipping into aqueous KCl solution. These s-BLMs were used to determine the relationship between $Cu^{2+}$ concentration and current crossing s-BLM. Their relationship showed high linearity and reproducibility. The calibration curve was constructed to express the relationship between $Cu^{2+}$ concentration and current in the $Cu^{2+}$ concentration range of 10 and $130{\mu}M$. This calibration curve was used to measure $Cu^{2+}$ concentration in an unknown sample. Microfluidic system with s-BLM was made of PDMS(polydimethyl siloxane) using typical soft photolithography and molding technique. This integrated system has various functions such as activation of the silver surface without cutting silver wire, coating of BLM on silver surface, injection of KCl buffer solution, injection of $Cu^{2+}$ sample and measurement of $Cu^{2+}$ concentration in the sample.

Synthesis, Fastness and Spectral Properties of Some New Azo Pyrazole and Pyrazolotriazole Derivatives (Pyrazole과 Pyrazolotriazole 유도체의 합성 및 특성 연구)

  • Coupling of 5-amino-1,3-diaryl-pyrazoles 1a-c with diazonium salts of different aryl amines gave a series of novel 1,3-diaryl-5-amino-4-arylazopyrazoles 3a-l. Such compounds could be also obtained by reaction of 5-amino-1,3-diaryl-4-nitroso- 1H-pyrazoles 2a-c with different aryl amines in alkaline medium. Oxidation of azo derivatives 3a-l with cupric acetate, in dimethyl formamide and stream of air, gave 2,4,6-triaryl-2,4-dihydropyrazolo [4,3-d]-1,2,3-triazoles 4a-l. and the fluorescence properties of the cyclic triazoles were studied. Diazotization of 5-amino-1,3-diaryl-1H-pyrazoles 1a-c by sodium nitrite in ortho-phosphoric acid followed by coupling with some aryl amines gave o-aminoazo compounds 5a-f. Cyclisation of compounds 5a-f in pyridine and cupric acetate gave the corresponding triazoles 6a-f. The coupling of compounds 6a-f with different aryl diazonium salts gave compounds 7a-j. The synthesized dyes were applied to polyesters as disperse dyes and the fastness properties were evaluated.

Effect of Copper Source (Cupric Citrate vs Cupric Sulfate) and Level on Growth Performance and Copper Metabolism in Pigs

  • Two experiments were conducted to evaluate the efficacy of cupric citrate (Cu-citrate) relative to cupric sulfate $(CuSO_4)$ as a Cu source for weanling and grow-finish pigs. In addition, the use of liver and bile Cu concentrations as indices of the bioavailability of Cu sources was investigated. Experiment one consisted of a nursery phase (35 d; initial BW=6.4 kg, final BW=21.4 kg) followed by a grow-finish phase (103 d; initial BW=21.5 kg, final BW=111.7 kg). Experiment two only consisted of a nursery phase (35 d; initial BW=6.3 kg, final BW=18.6 kg). Dietary treatments were identical for both experiments and consisted of: control (10 ppm $CuSO_4$); control+66 or 225 ppm $CuSO_4$; control+33, 66, or 100 ppm Cu-citrate. An antibiotic was included in diets for Exp. 1 but not Exp. 2. In both experiments, growth performance variables were similar for pigs receiving Cu-citrate and $CuSO_4$; however, growth performance was not improved by high concentrations of $CuSO_4$. Liver and bile Cu were increased (p<0.05) by 225 ppm $CuSO_4$; however, lower dietary concentrations of Cu from either $CuSO_4$ or Cu-citrate did not affect the Cu concentration of liver or bile relative to that observed in the control pigs. Irrespective of Cu source, there was no linear (p>0.10) increase in plasma Cu with increasing Cu concentrations in the diet for both experiments. However, the plasma Cu concentrations were highest (p<0.10) in pigs receiving diets supplemented with 225 ppm $CuSO_4$. Sixteen randomly chosen pigs per treatment in Exp. 1 were continued through the grow-finish phase. Body weight gain and feed intake were improved (p<0.10) by 66 ppm $CuSO_4$, but other dietary Cu treatments did not alter pig performance compared to the control diet. Plasma Cu concentrations were increased (p<0.10) by 225 ppm $CuSO_4$ in the growing phase and by 225 ppm $CuSO_4$ and 100 ppm Cu-citrate in the finishing phase. These data reveal no consistent effect of $CuSO_4$ on performance; therefore, it is difficult to assess the efficacy of these two Cu sources. In addition, these studies demonstrate that liver and bile Cu are not good indicators of Cu bioavailability in pigs fed adequate to pharmacological concentrations of Cu.
