Anxiolytic effect of chronic ginseng treatment using elevated T-maze in mice

  • The roots of the plant Korean ginseng have been extensively used in the traditional Chinese herbal medicine. The effects of chronic administration of Korean ginseng extract (KGE) were investigated on two different anxiety models: the elevated T-maze (for inhibitory avoidance and escape measurements) and the open field test (OFT). Diazepam (1 mg/kg), KGE (10, 30 and 100 mg/kg) were administered orally for 15 days. On the 14th day, mice were previously exposed for 30 min to one of the open arms of the T-maze, 24 h before the test. On 15th day, mice had two exposures to the enclosed and open arm of the elevated T-maze followed by exposure to the open field apparatus. The number of line crossings in the apparatus was used to assess locomotor changes. Cumulative Concentration Response Curve of 5-HT was plotted using rat fundus which were pre-treated in a similar way. Treatment with Diazepam (1 mg/kg) and KGE (10, 30 and 100 mg/kg) significantly (P < 0.05) impaired inhibitory avoidance performance but did not impair escape latency. In OFT, diazepam facilitated locomotion as compared to vehicle and other treatment groups. KGE at any of the selected doses did not impair locomotion. Concentration response curve of 5-HT was shifted towards the right with suppression of maxima in rats treated with KGE. The results suggest that KGE exerts anxiolytic like behaviour in a specific subset of defensive behaviour, particularly those related to generalized anxiety disorder.

  • Recently, jawbone osteonecrosis has been largely reported as a potential adverse effect of bisphosphonate (BP)administration. Currently available published incidence data for BRONJ are based on retrospective studies and estimates of cumulative incidence range from 0.8 to 12%. The mandible is more commonly affected than the maxilla (2:1 ratio), and 60-70% of cases are preceded by a dental surgical procedure. The signs and symptoms that may occur before the appearance of clinical evident osteonecrosis include changes in the health of periodontal tissues, non-healing mucosal ulcers, loose teeth and unexplained soft-tissue infection. Tooth extraction as a precipitating event is a common observation. The significant benefits that bisphosphonates offer to patients clearly surpass the risk of potential side effects; however, any patient for whom prolonged bisphosphonate therapy is indicated, should be provided with preventive dental care in order to minimize the risk of developing this severe condition.

Characteristics of Non-point Pollution Discharge on Stormwater Runoff from Lake Doam Watershed (도암호 유역의 강우시 비점오염물질 유출 특성)

  • Lake Doam watershed was surveyed to evaluate non-point source discharge characteristics and discharge load including several water quality parameters in Song Stream from July 2009 to July 2011. Concentrations of water pollutants were high during the rainfall period, especially, SS, TP and COD showed increasing tendencies toward cumulative water discharge but TN did not show much difference. SS, TP and COD had an initial flush effect of over 50 mm rainfall event but there was no clear tendency for rainfalls below that level. Event mean concentration (EMC) regarding the rainy and dry period showed large differences. Especially rainy season EMC (SS, TP, COD) demonstrated an increasingly high tendency. EMCs of COD, SS, TN and TP measured for twelve rain events were as high as 26.1, 866.0, 4.68 and 0.605 mg $L^{-1}$, respectively. COD, SS, TN and TP loadings from the highland agricultural region of the Song Stream watershed were 34,263, 1,250,254, 2,673 and 933 kg $yr^{-1}\;km^{-2}$, respectively, which were relatively higher than the results of other stream systems. Therefore, it is strongly recommended that long-term monitoring and non-point pollution reduction programs for the highland agricultural area to continue. Furthermore, this non-point source pollution loading research acquired from the highland agricultural area could be the base for reassessment.

Dietary Supplementation of Betaine (Betafin) and Response to High Temperature Stress in Male Broiler Chickens

  • The effects of supplemental betaine ($Betafin^{(R)}$) in the drinking water (50 g/kg) (WB) or feed (100 g/kg) (FB) were investigated on male broiler chickens ($Cobb{\times}Cobb$) exposed to 4 h episodes of heat stress at $34{\pm}1^{\circ}C$ on day (d) 35 and $36{\pm}1^{\circ}C$ from d 36 to 41. Prior to (d 1 to 34) and following heat exposure (d 35 to 41), betaine supplementation had no significant effect on body weight, total feed intake and cumulative feed conversion ratios of broilers. The total water intake of WB chicks was lower compared to controls. Prior to heat exposure, there was no difference in percentage of mortality among the three dietary groups. Following the heat challenge period, although higher percentage of control chicks succumbed to the heat challenge as compared to those of WB, it was not significantly different. The WB and FB chicks were less hyperthermic than controls in response to the heat challenge. Irrespective of treatment groups, the heat treatment resulted in a marked elevation in heterophil/lymphocyte ratios (HLR). The WB birds, however, had smaller increase in HLR than those of controls during heat exposure. Antibody production against Newcastle disease vaccine on day 35 was not affected by betaine supplementation. On d 42, WB birds had higher antibody production than those of FB. It is concluded that the WB treatment, as measured by HLR, antibody production and mortality rate, has advantages over the FB group under heat stress conditions.

Transarterial Chemoembolization Monotherapy in Combination with Radiofrequency Ablation or Percutaneous Ethanol Injection for Hepatocellular Carcinoma

  • Purpose: To evaluate whether combined transarterial chemoembolization (TACE) with radiofrequency ablation (RFA) or percutaneous ethanol injection (PEI) for hepatocellular carcinoma (HCC) have superior efficacy to transarterial chemoembolization (TACE) alone a retrospective review was conducted. Methods: During January 2009 to March 2013, 108 patients with hepatocellular carcinoma underwent TACE or combined therapies (TACE+RFA or TACE+PEI). The long-term survival rates were evaluated in those patients by various statistical analyses. Results: The cumulative survival rates in the combined TACE+RFA/PEI group were significantly superior to those in the TACE alone group. When the comparison among the groups was restricted to patients with two or three tumors fulfilling the Milan criteria, significantly greater prolongation of survival was observed in the combined TACE+ RFA/PEI group than in the RFA/PEI alone group. Conclusions: In terms of the effect on the survival period, combined TACE+ RFA/PEI therapy was more effective than TACE monotherapy, and also more effective than PEI or RFA monotherapy in cases with multiple tumors.

Managing Red Oak (Quercus rubra L.) Reduces Sensitivity to Climatic Stress

  • This study was conducted in a long-term experimental forest in the central hardwoods region of southwestern Michigan to retrospectively examine the role of past forest management practices and climate on red oak (Quercus rubra L.) productivity. Initially, in 1971, plots within the experimental forest were treated separately with a clearcut and shelterwood regeneration harvest in an attempt to increase red oak regeneration. From 1987-1989, a new study was initiated within a portion of the clearcut and shelterwood plots to evaluate the effectiveness of additional oak crop tree release using mechanical and chemical applications. Cumulative diameter and mortality rates of 719 red oaks were monitored across the four silvicultural treatments: Clearcut-A (clearcut without additional release treatment), Clearcut-B (clearcut with additional release treatment), Shelterwood-A (shelterwood without additional release), and Shelterwood-B (shelterwood with additional release) plus an untreated control. Increment cores were obtained from red oak trees and neighboring competitor species. Tree-ring analyses (dendrochronology) were applied to examine the effect of these silvicultural treatments and climatic factors (temperature and precipitation) on red oak productivity. The results indicated that crop tree release following a clearcut or shelterwood harvest reduced mortality rates and thus increased survival of red oak. Red oak in control plots or plots only receiving the initial regeneration harvesting treatment and no additional competition release were negatively affected by climatic stress, which included summer moisture stress. In contrast, red oak in plots that received the competition release treatment from shade tolerant tree species not only had higher tree level productivity (i.e., tree basal area) and lower mortality rates, but were also relatively more resilient to climatic stress by showing limited or no associations between climate and growth.

Ondansetron (1 mg/kg), FE (10, 30 &100 mg/kg) and TF (10 & 30 mg/kg) were administered orally for 15 days. On the $14^{th}$ day mice were previously exposed for 30 min to one of the open arms of the T-maze, 24 h before the test. On $15^{th}$ day mice had two exposures to the enclosed and open arm of the ETM followed by exposure to the open field apparatus. The number of line crossings in the apparatus was used to assess locomotor changes.   • In this study, the subsidence behavior caused by groundwater ex-flow in a limestone cavity encountered during tunnel excavation was quantified based on numerical analysis and the effect was analyzed. Based on the groundwater level and surface subsidence surveyed at the site, a numerical analysis technique was applied to analyze the characteristics of the subsidence behavior according to the tunnel passage of the geological vulnerabilities. The results of groundwater seepage-coupled analysis were analyzed to reflect the actual ground subsidence behavior. As a result of the study, it was analyzed that the ground subsidence due to the tunnel excavation in the limestone common section(the geological vulnerable zone) was analyzed that the dramatical decrease in groundwater level was the main cause. As a result of numerical analysis, it was analyzed that the long-term cumulative settlement of the asphalt surface after the groundwater ex-flow was 76~118mm due to the reduction of the volume of the soil layer due to the decrease in the groundwater level, and the settlement amount increased as the depth of the soil layer increased.

  • Objectives: This study aimed to develop the attention-deficit hyperactivity disorder (ADHD)-After School Checklist (ASK) to evaluate the severity of ADHD symptoms and self-management ability in children and adolescents in South Korea. Additionally, we evaluated the reliability and validity of the scale. Methods: We developed the ASK to evaluate the effect of ADHD psychopathologies on self-management and interpersonal impulsivity. We investigated the reliability and validity of the scale with 1349 parents (male 56.9%; 1202 parents of non-ADHD children, 147 parents of children with ADHD) in Seoul and Gyeonggi Province, Korea. Results: According to the construct validity test using principal constant analysis with the varimax rotation method, two factors explained 60.7% of the cumulative variance in ASK scores. Cronbach's alpha for the whole scale was 0.71. There was no statistical difference between mean ASK scores at test and retest. Mean total ASK scores of the ADHD group were significantly higher than those of the non-ADHD group (p<0.001). Conclusion: The ASK can be used as a reliable and valid tool to evaluate not only self-management capability of children and adolescents with ADHD in their academic and everyday life, but also their impulsiveness in interpersonal relationships.

Acute toxicity evaluation of drought-tolerant transgenic rice Agb0103 to Daphnia magna

  • A drought-tolerant transgenic rice (Agb0103) was developed using a pepper methionine sulfoxide reductase (CaMsrB2) under the control of rice Rab21 promoter with a selection marker, the phosphinothricin acetyltransferase (PAT) gene. Commercialization of genetically modified (GM) crops will require the evaluation of risks associated with the release of GM crops. With the potential problems associated to GM crops safety testing, the investigation of their effects on non-target organisms is necessary for environmental risk research. This study was carried out to assess acute toxicity of a GM crop using the water flea (Daphnia magna) for non-target organism risk evaluation. The effect of acute toxicity on Daphnia magna of Agb0103 rice and a non-GM rice, Ilmibyeo, were investigated at different concentrations (0, 625, 1,250, 2,500, 5,000, and 10,000 mg/L). The Agb0103 rice used for the test was confirmed to express the CaMsrB2/PAT gene by the PCR and ELISA. Daphnia magna feeding tests showed no significant differences in cumulative immobility or abnormal response with either Agb0103 rice or non-GM rice. The 48hr-EC50 values showed no difference between Agb0103 rice (2243 mg/L) and non-GM rice (2694 mg/L). These results suggest that there is no significant difference in toxicity to Daphnia magna between Agb0103 rice and its non-GM counterpart.
