Cultural Characteristics for the Enhanced Mycelial Growth of Ramaria botrytis

  • The culture conditions for the enhanced mycelial growth of Ramaria botrytis was investigated. The optimal temperature and pH for the mycelial growth were $24^{\circ}C$ and 5.0, respectively. It was shown that starch was best of several carbon sources in Czapek-Dox medium as a minimal medium for the enhanced mycelial growth. Organic nitrogen sources were better than inorganic ones for mycelial growth. The appropriate vitamin and mineral salt were biotin and FeCl3, respectively. When this strain was cultured with $FeCl_3$ for 30 days, 19.23 g/l of dry mycelium of R. botrytis was obtained.

Interspecific Protoplast Fusion between Fusarium poae and Fusarium sporotrichioides (Fusarium poae와 Fusarium sporotrichioides간의 원형질체 융합)

  • In order to develop the protoplast fusion method of the strains of Fusarium, the interspecific protoplast fusion was attempted between Fusarium poae and F. sporotrichioides. Various auxotrophic mutants were isolated by the treatment of N-Methyl-N'-Nitro-N-Nitrosoguanidine. The optimal conditions for the formation and regeneration of protoplasts were examined and the characteristics of a fusant were studied. As a results, protoplasts were readily obtained from 18 hours cultured mycelia by the treatment of driselase for 3 hours and 0.6 M KCl as a best osmotic stabilizer at pH 6.0 for the formation of protoplast. Sucrose was the most suitable for the regeneration. Polyetylene glycol (M.W. 8,000) in $CaCl_{2}$-glycine solution was used to induce the protoplast fusion. The interspecific fusion frequency between protoplasts among the auxotrophic mutants of the two strains ranged from $2.7*10^{-2}$ to $5.7*10^{-3}$ . DNA content and cellulase activity were rather increased in the interspecific fusant. The lag phase of growth curve was slightly elongated in the fusant.

First Report of Postharvest Gray Mold Rot on Carrot Caused by Botrytis cinerea in Korea

  • In February 2014, gray mold rotting symptoms were observed in carrots in cold storage at Gangneung, Gangwon province, Korea. The typical symptom of gray mold rot showed abundant blackish gray mycelia and conidia was observed on the infected root. The pathogen was isolated from infected root and cultured on PDA for further fungal morphological observation and confirming its pathogenicity according to Koch's postulates. Results of morphological data, pathogenicity test and rDNA internal transcribed spacer (ITS 1 and 4) sequence showed that the postharvest gray mold rot of carrot was caused by Botyrtis cinerea. This is the first report of postharvest gray mold rot on carrot in Korea.

Antitumor Activities of the Proteoglycans from the Mycelium of Ganoderma lucidum IY009 (영지버섯 균사체 (Ganoderma lucidum IY009)로부터 추출한 단백다당체의 항암촬성)

  • In this study, the antitumor activities were investigated using $\beta$-lmmunan, a proteoglycan obtained from cultured mycelia of the IY009 strain of Ganoderma lucidum belonging to basidiomycetes. The result showed the significant effect of cytotoxicity test against murine sarcoma 180 and murine lymphocytic leukemia L1210 using immunized macrophage cultures by $\beta$-lmmunan. When intraperitoneally injected at 40 mg/kg/day daily for 10 days, $\beta$-lmmunan inhibited the growth of sarcoma 180 solid tumor in ICR mice by 88.8% (p<0.05). It was also observed that $\beta$-lmmunan increased life span by 85.2% (p<0.01) after treatment of 100 mg/kg/day in BDF1 mice bearing lymphocytic leukemia L1210. And combination therapy with cisplatin (dosage: 4 mg/kg) increased life span by 140.4% (p<0.05) after treatment of 100 mg/kg/day daily in BDF1 mice bearing lymphocytic leukemia L1210.

Antimutagenic Potential of Phellinus igniarius

  • Mutagenic activities of extracts from the filtrate of the cultured broth (PI-I), mycelia (pI-II), and the fruiting bodies (PI-III) of Phellinus igniarius were examined by Ames/Salmonella tests. No mutagenic activity was found in Salmonella typhimurium strains TA98 and TA100, either with or without S9 activation. In contrast, PI-I, PI-II, and PI-III showed inhibitory effects on the mutagenic activities by the directly-acting mutagens, 4-nitro-ο-phenylenediamine(NPD) and sodium azide ($NaN_3$), and also by the indirectly-acting mutagens, 2-aminofluorene (2-AF) and benzo[a]pyrene (B[a]P). These results suggest that P. igniarius possesses some antimutagenic activity and may contain some chemopreventive agents.

  • Many antitumor and immune modulating components have been isolated from fungal extracts. In this study, the authors isolated the proteoglycans from cultured mycelia of Lentin us lepideus, including especially the acidic polysaccharide fraction, named lepidan. It was obtained by extraction with hot water followed by purification using DEAE cellulose anion exchange. To elucidate antitumor effects against different type of tumor, the proteoglycans were tested on sarcoma 180, C3H MCA clone 16 and P388 leukemia in vivo. Lepidan showed 58.3% of tumor inhibition against solid form of sarcoma 180 and 58.6% against MCA clone 16. But lepidan did not affect life span of mice against P388 leukemia. Also when Lepidan was applicated to MTT assay, it did not show any direct cytotoxicity against various tumor cells in vitro.

Development of Non-protoplast transformation System in Aspergillus oryzae

  • Aspergillus oryzae is a filamentous fungus classified in the group Aspergillaceae Ascomycetes. It is an important microorganism for industrial production of enzymes and fermented food productions. It secrets large quantities of proteins or enzymes into the culture medium which makes this organism appealing for the production of heterologous proteins. Recently Electric field-mediated transformation method, electroporation, has been applied to fungal transformation. In this study, fungal transformation was carried out by bypassing the protoplast isolation step, decreasing the culturing time and non-protoplast transformation for the increment of transformation efficiency. Transformants were obtained with electroporation in optimal condition 2,500 voltage, 1,540 ohm and 0.50 capacitance. More than 1,000 transform ants were obtained with 6-10 hrs cultured mycelia without enzyme treatment, called non-protoplast transformation.

  • In the previous report, we described the marked antitumor and immunomodulatory activities of PVp, a protein-polysaccharide fraction of a Korean wild mushroom Psathyrella velutina. In this study, a protein-polysaccharide fraction, PVMP, was prepared from the shake-cultured mycelia of the same mushroom and its immunoactivities as well as chemical compositions were investigated. At 200 $\mu\textrm{g}$/ml, PVMP weakly stimulated the BALB/c mouse splenic lymphocytes to form lymphoblasts and upregulated the expression of CD25 molecules, but failed to stimulate peritoneal macrophages. In chemical analysis these two protein-polysaccharide fractions were found to be quite different in that the carbohydrate contents of PVMP and PVP, respectively, was 85.3% and 41.2%. These results reveals that PVMP, unlike PVP, is a moderate immunostimulator on the immune system.

  • This research was focused on investigation of the general condition for protoplast formation of Trichoderma speues. for protoplast formation, the mycelia cultured in YM medium were collected from each growth phase and were treated with the Iytic enzymes. This procedure was carried out by all strains. The most optimal conditions of NOVOZYM 234 and DRISELASE were determined by T. saturnisporum IAM 12535 and T. longibruchiatum IBM 13107, respectively. The effect of osmotic stabilizers appeared ${KCI}>(NH_4)_2{SO_4}>NaCl>mannitol>{MgSO}_4$ and the optimal concentration of each osmotic stabilizer wns determined by 0.6-0.9 M. The optimal condition of DRISELASE for protoplast formation ; optimal pH 5.0, optimal concentration, 2%, optimal reaction time, 4 hours, and optimal temperature, $30^{\circ}C$. The optimal condition of NOVOZYM 234 for protoplast formation ; optimal pH 5.5, optimal concentration 1%, optimal reaction time 3 hours, and optimal temperature $30^{\circ}C$. The optimal culture period of mycelia for protoplast formation was between the initial and the middle exponential phase. Generally, DUSELASE was more effective than NOVOZYM 234 on protoplast formation except for T. longibruchiatum IAM 13107 and T. viride IAM 5141.

  • To find antitumor constituents in Korean basidiomycetes, the mycelia of Lentinus edodes DMC7, which had shown a good mycelial growth in shakeflasks, were cultured at $27^{\circ}C$ on an orbital shaking incubator at 180 rev/min for 12 days. The medium was composed of glucose (50g/l), yeast extract (9g/l), peptone (9g/l), and seven inorganic salts. A water soluble macromolecular fraction, LF-3, was obtained from the culure filtrate by fractionation with ethanol and dialysis using a Visking tube. When LF-3 was administered i.p. at 50mg/kg/day once daily for 10 consecutive days to female ICR mice which were implanted s.c. with sarcoma 180 $(10^6\;cells/mouse)$, it exerted a highly significant antitumor activity, with the tumor inhibition ratio of 53. 1%.
