국내 시중 유통 자연산 및 양식산 활어의 중금속 함량 (Heavy Metal Contents in Wild and Cultured Fishes from the Korean Coasts)

  • We determined the contents of heavy metals in the muscle of wild and cultured fishes, collected from fish markets located in the eastern (Pohang), western (Gunsan), and southern (Tongyeong) coasts of Korea, from 2004 to 2005. As the results of monitoring the heavy metal contents in spring season, the wild fishes contained the range of Cd (0.01-0.08 mg/kg), Cr (ND-0.28 mg/kg), Cu (0.06-1.53 mg/kg), Hg (0.02-0.16 mg/kg), Mn (0.04-1.15 mg/kg), Ni (ND-0.09 mg/kg), Pb (0.03-0.41 mg/kg), and Zn (1.84-6.61 mg/kg). While for the cultured fishes, Cd (0.01-0.05 mg/kg), Cr (ND-0.17 mg/kg), Cu (0.05-0.61 mg/kg), Hg (0.02-0.13 mg/kg), Mn (0.03-0.17 mg/kg), Ni (ND-0.08 mg/kg), Pb (0.03-0.33 mg/kg), and Zn (2.06-6.20 mg/kg) were contained. In summer season, the contents of heavy metal in the muscle of the wild fishes were as follows: Cd (ND-0.11 mg/kg), Cr (0.01-0.37 mg/kg), Cu (0.21-1.31 mg/kg), Hg (0.01-0.11 mg/kg), Mn (ND-1.47 mg/kg), Ni (ND-0.26 mg/kg), Pb (0.06-0.48 mg/kg), and Zn (2.94-14.38 mg/kg). In comparison, the contents of heavy metal in the muscle of cultured fishes were Cd (ND-0.05 mg/kg), Cr (0.13-0.33 mg/kg), Cu (0.19-0.56 mg/kg), Hg (0.05-0.26 mg/kg), Mn (ND-0.14 mg/kg), Ni (ND-0.58 mg/kg), Pb (0.07-0.45 mg/kg), and Zn (2.43-7.53 mg/kg). Also the fall and the winter season, the heavy metal contents in the fishes showed almost similar with other season, however, Pb was lower and Zn was higher than both spring and summer season. The wild fishes contained the heavy metals a little more than the cultured fishes. We could not observe clear seasonal variation in the heavy metal contents of the fishes. The levels of Hg and Pb in all samples tested did not exceed the maximum permissible levels in the fishes set by the Korean Food & Drug Agency for safe human consumption.

  • 운동 사육구와 비운동 사육구에서의 참돔육에 대한 ATP관련 화합물의 변화를 살펴보면 운동 사육구에서는 ATP함량이 약간의 감소를 보이지만 ADP함량은 ATP의 소모만큼 약간의 증가를 보이고 있었다. 반면, 비운동 사육구에서 운동 사육구와 대초적으로 기간이 길어짐에 따라 ATP함량이 큰 변화를 보이고 있지 않았다. AEC수치는 운동 사육구에서 0.88$\pm$0.04으로 나타났고 비운동 사육구에서는 0.89$\pm$0.02으로 나타났으며 외관상으로 참돔의 건강상태는 양호하였다. 참돔의 근원섬유에 있어서 $Mg^{2+}$(+Ca$^{2+}$)-Arpase 활성의 변화는 운동 사육구의 Arpase 활성이 증가하여 운동 20일에는 0.42$\mu$mo1 Pi/min$.$mg의 함량을 나타내었으나, 비운동 사육구에서는 큰 차이를 보이지 않고 있다. $Mg^{2+}$(+Ca$^{2+}$)-ATPase활성은 운동시키 전에는 각각 0.10 $\mu$mol Pi/min$.$mg, 0.14 $\mu$mol Pi/min$.$mg으로 나타나 비운동 사육구가 운동 사육구에 비하여 높았으며 사육기간 동안 활성이 서서히 감소하는 경향을 나타내었다. $Ca^{2+}$-ATPase 활성 또한 사육기간 동안 큰 변화를 보이지 않았다. 운동 횟수에 따른 차이에서는 1회/일 운동시켰을 때에는 ATP관련 화합물의 함량은 2회/일 운동시킨 것과 비슷한 수준이었으며 운동휫수에 따른 ARC수치의 큰 변화는 나타나지 않았다.

조피볼낙(Sebastes schlegeli)에 유행(流行)한 Lymphocystis병에 대(對)하여 (Studies on Lymphocystis Diseases in Sebastes schlegeli)

  • There prevailed lymphocystis disease with 1 to 2cm in size in the skin and pins of cultured Sebastes schlegeli in fish farms in Tongyoung-gun, Sanyang-myeon, Kon-ri from summer, 1987 to fall, 1988. Though there were some difference between each fish farm, this disease prevailed widely from immature fish to mature fish, less than 10cm to 30cm in body length with about 8 through 80% of infection rate. Sebastes schlegeli with the lymphocystis disease showed combined nipple-shaped mass by doubly, triply propagated lymphocystis. But the author considered that lymphocystis disease was not a direct cause of death of Sebastes schlegeli, the fish showed marked inflammation by seconary infection of pathogenic organisms.

양식넙치(Paralichthys olivaceus)의 복수증에 관한 연구 - Edwardsiella tarde의 일부 특성과 병원성 및 대책 - (Studies on the Edwardsiellosis of cultured flounder, Paralichthys oliuaceus;- Characteristics, pathgenicity and control of Edwardsiella tarda -)

  • Characteristics, pathogenicity and control of the causative organisms isolated front diseased cultured flounder, Paralichthys olivaceus were studied. The causative organisms were identified as E tarda by biochemical and biophysical characteristics. Also, it strains were named as E tarda KBF-1 and E tarda KMF-1, and optimal pH of E tarda KBF-1 and E tarda KMF-1 were 8.0, and optimal concentration of NaCl. E tarda KBF-1 was 0% and E tarda KMF-1 was 1%. In the pathogenicity test, 0~10 of the flounders of artificially infected group(E tarda KBF-1) with $1.0{\times}10^7$ cfu/fish were died within 60 hrs, but 0~9 flounders infected group with 41.0{\times}10^6$ cfu/fish were died within 60 hrs. Also, 0~10 flounders infected group(E tarda KMF-1) with $1.0{\times}10^7$ cfu/fish were died within 36 hrs, while 0~7 flounders infected with $1.0{\times}10^6$ cfu/fish were died within 60 hrs. Drug sensitivity of E tarda KBF-1 strain was resistant to AM, CF and N, and intermediate to E, K and S, and sensitivity to C, G, SxT and FF. But E tarda KMF-1 strain was resistant to CF, E and V, and intermediate to AM, C, N and SxT, and sensitivity to GM and FF.

Anaesthetic Tolerance of Juvenile Black Rockfish Sebastess schlegeli, Produced for Wild Stock Enhancement

  • The strength of juvenile black rockfish, Sebastes schlegeli, raised in different hatcheries for wild stock enhancement was evaluated in terms of resistance to an anesthetizing agent, tricaine methane-sulfonate (MS-222), and exposure to drying. The working dosage of MS-222 varied significantly with fish size and hatchery population. Smaller fish were less resistant to the chemical than larger ones. MS-222 effects also differed with fish growth history. The fish cultured in embanked populations showed stronger resistance, earlier recovery, and lower mortality, compared to those cultured in land-based tanks or collected from wild stocks. Similar results were seen in juveniles challenged to dry exposure. These results suggest that an embanked population of black rockfish is more resistant to anesthetic stress, expressed as anesthesia recovery and mortality, and that this population is healthier than others.

양식 동자개(Pelteobagrus fulvidraco)의 Edwardsiella ictaluri 감염 (Edwardsiella ictaluri Infection in Cultured Yellow Catfish Pelteobagrus fulvidraco Fingerlings in Korea)

  • We observed yellow catfish Pelteobagrus fulvidraco fingerlings cultured in land ponds in Korea swimming in a corkscrew spiral pattern while hanging head-up and tail-down at the water surface, before eventually dying. Externally, these fish displayed “hole in the head” disease, pale gills, and hemorrhages in the base of the pectoral and caudal fins; internally they had liver hemorrhages and kidney discoloration. The bacterium Edwardsiella ictaluri (YCK-01 and YCL-01) was identified in the kidneys and livers of diseased fish via phenotypic characteristics and PCR analysis using the ictaluri-specific primers IVS (an intervening sequence) and IRS (the inter-ribosomal spacer). Infectivity challenges by intraperitoneal and immersion routes showed that a representative bacterial strain (YCK) exhibited strong virulence to yellow catfish, with an LD50 of 3.2×104 CFU/fish and 2.5×106 CFU/mL, respectively. This is the first report of E. ictaluri infection in yellow catfish from Korea.

우리나라 양식 자주복, Takifugu rubripes (Temminck & Schlegel, 1850)에서 발생한 Vibrio atlanticus의 첫 감염 사례 보고 (The first case report on Vibrio atlanticus infection in cultured tiger puffer, Takifugu rubripes)

  • 본 연구는 육상 순환 양식장, 약 $20^{\circ}C$의 수온에서 양식 중이던 자주복에서 체표에 백탁과 궤양을 보이는 개체가 다수 발생하여 그 원인을 밝히고자 하였다. TCBS 선택배지에서 순수 배양된 균체를 확인하고 계대 배양하여 16S rRNA와 rpoA 유전자의 염기서열 분석을 실시하였으며, 그 결과는 Vibrio atlanticus와 99.77%의 상동성을 가지는 것으로 나타났다. 생화학적 특징을 이미 보고된 같은 균종과 비교한 결과, Voges-Proskauer test와 Gelatin 가수분해능에 대한 결과에서 차이를 나타내는 것으로 확인하였다.

  • 천연 및 인공 넙치 종묘의 성장 단계에 따른 체지질 조성 변화를 비교 검토한 결과는 다음과 같다. 1) 어체의 지질 및 비타민 E 함량은 천연 종묘 보다 인공 종묘에서 높은 값을 나타내었다. 2) 어체 성장에 따른 지질, 지방산(18: 2n-6, 22: 6n-3) 및 비타민 E 함량 변화는 부화 후 전장 5-10mm 전후의 배합 사료 공급기에서 급격한 증감 현상을 보였다. 3) 어체 지질, 지방산 조성 및 비타민 E 함량은 먹이 조성에 직접적인 영향을 받았다. 4) 천연 종묘는 인공 종묘에 비해 자연 상태에서 지질 함량이 낮고 고농도의 DHA를 함유하고 있는 천연 먹이를 섭취한 것으로 추정되었다.

양식 볼락류에서 비특이적 방어인자의 활성 (Activities of non-specific defense factors in cultured oblong rockfish(Sebastes oblongus) and rockfish(S. schlegeli))

  • To understand the activity of non-specific defence factors in cultured Sebastes, the antibacterial effect of the serum, skin mucus and homogenate of various organs from cultured oblong rockfish (Sebastes oblongus) and rockfish(Sebastes schlegeli) against pathogenic bacteria, Aeromonas hydrophila, Edwardsiella tarda, Vibrio anguillarum, and Streptococcus sp. was compared with that of flounder(Paralichthys olivaceus) and seabass(Leteolabrax japonicus). And the activities of proteolytic enzyme, chitinolytic enzyme and haemolycin as non-specific defence factor were investigated on the oblong rockfish and rockfish. Samples from oblong rockfish showed the highest antibacterial activity by lysoplate assay on agar plate mixed with pathogens, followed in descending order by rockfish, seabass, and flounder. Turbidimetric assay was carried to evaluate the lysozyme activity of fish samples against lyophilized cells of Micrococcus lysodeiktikus. The serum, kidney, liver, stomach, intestine and eyeball of oblong rockfish and the mucus and gill of rockfish appeared to have the highest lysozyme activity among the fish strains investigated. All samples except skin mucus, liver, and eyeball of oblong rockfish and rockfish showed proteolytic enzyme activity. Chitinolytic enzyme activity was showed in random sampling and haemolytic activity was remarkable in oblong rockfish. Therefore, Sebastes strain was proved to have effective defense mechanisms based on the antibacterial activities, and lysozyme, proteolytic enzyme, chitinolytic enzyme, and haemolycin were considered to act as the non-specific defence factor of Sebastes.

이리도 바이러스 감염에 의한 양식 돌돔, Oplegnathus fasciatus의 대량폐사 (Mass Mortalities of Cultured Striped Beakperch, Oplegnathus fasciatus by Iridoviral Infection)

  • 1998년 8~10월에 남해안 일원 해상 가두리 양식장에서 사육중인 돌돔, Oplegnathus fasciatus이 60%이상 폐사하였다. 병어의 체색은 검어지거나 퇴색되며 아가미는 빈혈증상이 심하고 비장은 특이적으로 비대되어 있었고, 비장, 신장, 심장 및 간장 조직에서 퓰겐양성과 호염기성의 비대세포가 관찰되었다. 병어의 비장조직 마쇄액을 GF세포에 접종배양한 결과, 세포가 구형화 되면서 커지는 세포변성효과가 나타나 감염된 GF 세포로부터 바이러스가 분리되었다. 분리된 바이러스 배양액($10^4TCID_{50}$/0.1 ml)을 건강한 돌돔 치어에 복강주사하면 자연감염어와 유사한 증상을 나타내면서 폐사가 일어났고, 또한 병어의 비장조직을 전자현미경으로 관찰한 결과, 조직내 비대세포의 세포질에서 직경이 120~130 nm이며 외막이 있는 정 20면체의 바이러스 입자가 관찰되었다. 그리고 genebank로부터 얻은 참돔 이리도바이러스(RSIV)의 ATPase gene의 DNA염기서열을 주형으로 제작된 primer를 사용하여 PCR을 수행한 결과, 500 bp의 PCR 생성물이 병어나 인위감염된 GF세포에서 얻어졌는데, 이들 PCR 생성물은 RSIV의 ATPase cDNA gene과 95%의 상동성을 나타내었다. 따라서 양식돌돔에 대량 폐사를 일으킨 원인 바이러스는 참돔 이리도바이러스(RSIV)와 유사한 이리도바이러스로 밝혀졌다.
