Functional Properties of Yogurt Fermented by Bacteriocin-producing Pediococcus acidilactici (박테리오신 생성 Pediococcus acidilactici 를 적용한 요거트 특성 및 항균성 연구)

  • Physical and sensory characteristics of commercial yogurts are important aspects for consumer acceptability. In addition, beneficial functions of commercial yogurts are also emphasized for the probiotic dairy products. The aim of this study was to investigate the functional properties of yogurts with the combination of bacteriocin-producing Pediococcus acidilactici. Yogurts fermented with commercial starter culture (control yogurt) and control yogurt together with P. acidilactici HW01 (yogurt+HW01), P. acidilactici JM01 (yogurt+JM01), or P. acidilactici K10 (yogurt+K10) were prepared. During 28 days after fermentation, the viability of lactic acid bacteria, pH, and brix, in the yogurt samples were assessed with standard methods. Moreover, to investigate the antilisterial activity of the yogurt samples, Listeria monocytogenes was simultaneously inoculated when the yogurts were prepared with lactic acid bacteria, and the viability of L. monocytogenes was determined. Although yogurt+K10 did not completely remove L. monocytogenes, control yogurt, yogurt+HW01, and yogurt+JM01 eradicated L. monocytogenes at day 2 after fermentation. However, yogurt+K10 also removed L. monocytogenes at day 3 after fermentation. Taken together, these findings suggest that the combination of yogurt with P. acidilactici does not affect its quality and they may consequently aid in the development of new probiotic yogurt.

  • Gaesung was the Mecca of Korean ginseng. Factors that Gaesung has been a leading brand of Korean ginseng were multiplicative. Those were natural conditions and huge commercial capital, red ginseng factory, creative business systems and etc. We can quote BongSang Son, SungHak Kong and JeongHo Kim as a famous Gaesung' ginseng merchants. They, as leaders modern ginseng industry had supplied the method of cultivation, prevention of phyto-diseases, excellent ginseng seed, and prepayment system of farming capital. The Gaesung merchants also adopted modern marketing techniques : commercial advertisement, made-order sales, changing package of the ginseng products. The book 'The Dictionary of Korean Companies and Stores' which was published in 1927 introduces BongSang Son as a great businessman in Gaesung. He was not only merchants but also educator and social worker. He practiced the spirit of entrepreneurship. BongSang Son's role of pioneer contributed to the development of Korean ginseng and Gaesung. Due to such efforts of Gaesung merchants, Korean ginseng industry were took a great step forward. This article considered the development of Korean ginseng industry through the life and achievement of the ginseng magnate BongSang Son who was a representative one of Gaesung merchants.

  • Benzo(a)pyrene(B(a)P), an extensively studied polycyclic aromatic hydrocarbon(PAH), is a common contaminant produced through the burning of fossil fuels, particularly coal, and from the exhaust products of internal combustion engines. It produces a wide range of toxicities, including carcinogenicity in experimental animals. B(a)P has been shown to suppress systemic immunity in experimental animals, which may contribute to the growth of the chemical-induced tumors. Using colorimetric MTT assay natural killer(NK) cell-mediated growth inhibition of tomor cell was measured in normal and B(a)P-exposed C57BL/6 mice. Non-adherent splenocytes of normal or B(a)P-exposed mice were cultured with Yac-1 cells at four different effector/target(E/T) cell ratios ranging from 200/1, 100/1, 50/1, and 25/1 in an assay volume of 0.1 ml. After the optical density of culture wells containing MTT solution was measured at a wavelength of 540 nm, the percentage of dead cells relative to the control target cell number was calculated. The NK activity of B(a)P-exposed mice was markedly lower than that of non-exposed mice group at all E/T ratios. These results indicated that suppression of NK cell activity may play a role in allowing for the growth of tumors.

A Case Study of Japanese Local Self-Governments Universal Design Policy (일본 자치체의 유니버설디자인 정책 사례연구)

  • Recently, in each local self-governments Japan, the importation of universal design policy is actively being expanded as a plan to maximize living satisfaction of.citizens within the region. This research investigates the background on how the local self-governments ended up importing the universal design administrative policy, investigates the policies which each self governing bodies have been promoting and presents necessary stipulations following those policies. When we sum up the necessity of promoting universal design by local self-governments, although the we carry various problems in the promoting process in the settlement of provincial era and promoting substantiality of it, we can point out the low birth rate phenomenon, progression of aged people's society, diversification of values as a result of social and economic maturity, etc. as social and economic background. For people's values, as they escape from the economic they attach great importance to economic supremacy, they respect culture, environment, etc. with higher level than economic aspect and the improvement of life's quality is becoming important. Meanwhile, the universal design which is known to many users as a usable design concept as reach to a point where it is getting attention as a basic concept of 21st century design. Today, each of local self-governments in Japan are making various efforts for unperturbed importation and application of universal design to local communities through homepage, universal design guideline or report, various workshops, etc. Especially, there are universal design policies of public areas, facilities, manufacture of other products and information for the contents of the policy. The enforcement of universal design policy puts importance in 1) the process of planning, alteration, policy scheme and decision making 2) the process of policy's enforcement 3) the probation process of all processes and I could find out that equal participation of roles by local citizens, citizen's organizations, companies with the administration hasn't been adopted for each of these processes.

  • Background: Celecoxib, a COX-2 specific inhibitor, has recently been used for the treatment of rheumatoid arthritis. However, the molecular and cellular mechanisms of celecoxib against RA inflammation remain to be defined. To elucidate the action mechanism of celecoxib on inflammatory cells, we investigated the effect of celecoxib on the production of two important mediators of inflammation, nitric oxide and PGE2 Methods: RAW 264.7 cells stimulated with LPS were preincubated with various concentrations of celecoxib (from $10^{-8}$ to $10^{-5}$ M) and $10{\mu}M$ hydrocortisone, respectively. The production of NO and PGE2, the end products of iNOS and COX-2 genes, were estimated in culture supernatants by Greiss method and EIA, respectively. The expression of iNOS gene, COX-2 gene, $NF-{\kappa}B$, and $I-{\kappa}B$ were determined by RT-PCR and western blot analysis. Results: Celecoxib and hydrocortisone inhibited the production of NO and PGE2 in dose dependent manner, when RAW 264.7 cells were stimulated with LPS. The expression of iNOS was also down-regulated by celecoxib and hydrocortisone. Interestingly, COX-2 gene differentially expressed according to the dose of celecoxib, a decrease with lower dose ($10^{-8}$ M) but an increase with higher dose ($10^{-5}$ M). $NF-{\kappa}B$ binding activity was decreased by lower dose of celecoxib, whereas was not affected by higher dose of it. The expression of $I-{\kappa}B$ was suppressed by higher dose of celecoxib. Conclusion: The celecoxib strongly suppressed the production of NO and PGE2 in LPS-stimulated RAW264.7 cells. The decrease of NO seems to be linked to the inhibition of iNOS by celecoxib. The lower and higher dose of celecoxib differentially regulated the COX-2 expression and $NF-{\kappa}B$ activity.

  • Mountain destination plays a vital role in satisfying the demand of natural tourism products, provides exclusive leisure, adventure and unique cultural experience. The Himalayan region includes some of the most biologically diverse habitats on Earth as well as homes to a bewildering variety of culture. The purpose of this study is to figure out the mountain tourism as a tool of sustainable development and explore the tourism destination development strategies with case study of Himalayan area. In this study, we recognized the importance of tourism for the isolated developing communities, for which, tourism provide the way of developing its economic and society. It also generates new opportunities of employment and change the soclo-economic environment of region. The hilly terrain of the state lacks reliable transportation infrastructure, therefore tourism is the only industries to support the economy and to uplift the standard of living for the people of Himalayan region. The region has large potential in the natural and cultural based tourism. The tourism development of the state may have cultural and ecological impact in future. Therefore, the planning and successfully promoting long-term policy and developing more sustainable tourism is important.

Effects of Tomatoes and Lycopene on Prostate Cancer Prevention and Treatment (토마토와 라이코펜이 전립선암의 예방과 치료에 미치는 영향)

  • Prostate cancer is a leading cause of cancer death in American men and evidences point to significant life style/diet components as risk factors for its development or prevention. Two large cohort studies have identified the consumption of tomatoes or high Plasma levels of Iycopene as associated with reduced risk. A number of other substances such as quercetin, phytoene, phytofluene, cyclolycopene, salicylates and tomatine in tomato besides lycopene could have anticancer activity and may be acting synergistically with lycopene. Lycopene at almost physiologically feasible concentrations, reduces cell viability by cell cycle arrest and apoptosis and modulates the cyclin pathways as well as increasing intercellular communication. However, it is not clear whether lycopene or its oxidation products are more bioactive. Tomato product supplementation results in plasma accumulation of phytoene, Phytofluene, the lycopene oxidation product, and cyclolycopene at significant concentrations and lycopene supplementation, either as a tomato product or as beadlets, results in maximal mean plasma lycopene concentrations of ∼ 1 $\mu$M which is at the lower limit of its activity in cell culture. Rats and mice are poor accumulators of lycopene and other carotenoids making them poor models for the study of cancer prevention and control. Of the 19 animal studies for various cancer sites, lycopene showed a positive effect in 10 studies but negative in 2 prostate cancer studies. In vivo prevention of leukocyte DNA damage in humans has been mostly studied using tomato product supplementation but lycopene supplementation appeared to reduce oxidative DNA damage as well as tomato product supplementation. Lycopene appears to be bioactive in intefering with carcinogenesis but the actions of phytoene, phytofluene or cyclolycopene cannot be ruled out since these compounds were present in most of the lycopene material used for these studies. Although lycopene remains as a promising agent, especially for cancer control, exploring interactions with other tomato phytochemicals and with current prostate cancer therapies should be encouraged.

Biodegradation of Crude Oil and Petroleum Products by Crude Oil-degrading Microorganism (미생물을 이용한 원유 및 원유제품의 분해 특성)

  • Two kinds of crude oil-degrading microorganisms from soil and one kind from sea were isolated and named strain Al32, strain F722 and strain OM1, respectively. These microorganism were identified Acinetobacter sp., Pseudomonas aeruginosa and Acinetobacter calcoaceticus, respectively. The optimum cultivation temperature of Acinetobacter sp. A132 and P. aeruginosa F722 was $35^{\circ}C$ and optimum growth pH was 8 and 9, respectively. The growth was the highest at 2.0% (w/v) substrate concentration when crude oil was only carbon source. The growth of A. calcoaceticus OM1 isolated from sea was the highest at 3.0% (w/v) of crude oil. In inspection of crude oil degradability, strain Al32 showed 5.49 g/L.day with Eleuthera (OMAN), 2.0% (w/v). P. aeruginosa F722 showed 1.19 g/L g/L.day with L-Zakum (AFRICA). In case of kerosene $nC_9\simnC_{20}$ and diesel $nC_9\simnC_{28}$, A. calcoaceticus OM1 was degraded 95% and 75%, respectively, for 7 days culture, and P. aeruginosa F722 was 80% after 10 days.

  • This study was conducted to investigate the biochemical properties of isolated bacteria, low temperature tolerant methane-producing clostridia which were selected for using them as inoculum to anaerobic fermentation of agricultural and livestock wastes at low temperature. The results were; 1. Low temperature tolerant methane-producing clostridia were isolated from the samples which showed the high methanogenesis rate by enrichment culture at low temperature in cellulose medium. These clostridia, Clostridium botulinum SRC-64, Clostridium scatologens SRC-91 and Clostridium tyrobutyricum SRC-100, were isolated from swampy sediment at latitude $56.9^{\circ}N$, lake sediment IV at latitude $55.0^{\circ}N$, and tidal land soil II at latitude $37.0^{\circ}N$, respectively. The optimum growth temperature for these isolates was $37^{\circ}C$ and the minimum, around $10^{\circ}C$. They all had detectable amount of $F_{420}$, specific coenzyme of methanogens. 2. As anaerobic fermentation products of glucose SRC-64 produced $H_2$, acetic, isovaleric and caproic acid, SRC-91 produced $H_2$, propionic, butyric, valeric, and caproic acid, and SRC-100 produced only acetic and propionic acid. The isolates were produced $CH_4$ ranged from 2.6 to 8.68 n moles/ml for 2 days at $13^{\circ}C$.

  • Loop-mediated isothermal amplification (LAMP) is an emerging detection technology for the amplification of DNA under isothermal conditions. The aim of this study was to develop a rapid and reliable LAMP technique for the detection of Cronobacter sakazakii in milk powder. In order to enhance the sensitivity and specificity, LAMP primers targeting outer membrane protein A (ompA) gene of C. sakazakii were designed using Explorer V4 software. Thirty seven C. sakazakii strains and 13 pathogenic microorganisms were used for comparative detection of C. sakazakii using polymerase chain reaction (PCR), real-time PCR, and LAMP. LAMP developed in this study could specifically detect C. sakazakii strains without cross-reactivity with other foodborne pathogens. LAMP products amplified from ompA gene of C. sakazakii were digested with with HhaI and NruI enzyme. The specificity of LAMP was confirmed by restriction fragment length polymorphism (RFLP) analysis. LAMP could detect C. sakazakii within 1 h without bacterial culture and its detection limit was as low as 1 CFU/mL C. sakazakii in milk. In the comparison of the sensitivity, LAMP showed 10,000- and 100-times higher detection limit than PCR or real-time PCR, respectively. Therefore, this study can conclude that LAMP is a rapid and reliable detection technique for C. sakazakii contaminated in powdered milk.
