Characterization plan for National Competency Standards and cultural contents curriculum (직무능력표준과 문화콘텐츠계열 교육과정 특성화 방안)

  • 이 연구의 목적은 최근 학문분야별로 체계적인 직무능력 매뉴얼이 개발되고 있는 상황에서 전문대학원 교육이 현장에 필요한 맞춤형 인재양성이라는 교육목표의 수월성을 위해 역량 있는 교육과정 방안을 직무능력에 근거하여 살펴보고자 한다. 연구 목적을 달성하기 위하여 먼저 국내외 전문대학원 문화콘텐츠(융합콘텐츠 포함) 관련 학과들의 교육과정을 분석하고, 이를 토대로 전문대학원 교육의 목표, 내용, 방법에 대한 종합적인 시사점을 도출한다. 다음으로, 전문대학원의 고유한 전문성과 주체성을 가진 현장밀착형 교육과정 사례를 통해 교육과정의 특성화 방안을 제시하고자 한다. 국가의 직무능력표준이 정한 문화콘텐츠 기획, 제작, 유통 및 서비스별로 요구되는 능력수준, 정의, 능력단위 요소를 체계적으로 제시함으로써 교육과정의 개념틀(framework of curriculum)을 마련할 수 있을 것이다.

Biocontrol of Fusarium Crown and Root Rot and Promotion of Growth of Tomato by Paenibacillus Strains Isolated from Soil

  • In this study, bacterial strains were isolated from soils from 30 locations of Samcheok, Gangwon province. Of the isolated strains, seven showed potential plant growth promoting and antagonistic activities. Based on cultural and morphological characterization, and 16S rRNA gene sequencing, these strains were identified as Paenibacillus species. All seven strains produced ammonia, cellulase, hydrocyanic acid, indole-3-acetic acid, protease, phosphatase, and siderophores. They also inhibited the mycelial growth of Fusarium oxysporum f. sp. radicis-lycopersici in vitro. The seven Paenibacillus strains enhanced a range of growth parameters in tomato plants under greenhouse conditions, in comparison with non-inoculated control plants. Notably, treatment of tomato plants with one identified strain, P. polymyxa SC09-21, resulted in 80.0% suppression of fusarium crown and root rot under greenhouse conditions. The plant growth promoting and antifungal activity of P. polymyxa SC09-21 identified in this study highlight its potential suitability as a bioinoculant.

Characterization of the purple nonsulfur bacterium, rhodopseudomonas palustris strain P-1, degrading ferulate

  • Photosynthetic bacteria which can utilize ferulate as a sole carbon source for their metabolic activities were isolated from soils by liquid enrichment culture technique. The strain P-1 was selected by the highest capability of degrading ferulate in aerobic and anaerobic conditions. The strain P-1 was rod-shaped with its motility, strained gram negatively and could not utilize sulfur compounds. This strain has the bacteriochlorophyll a group I carotenoid and membrane structures like lamellae. As the results of physiological, morphological and cultural charactderistics, the isolate was identified as Rhodopseudomonas plaustris, one of the purple nonsulfer bacteria. The strain P-1 utilized 2mM/day in aerobic condition and 0.86 mM/day in anaerobic condition.

  • Colony morphology, growth responses on some simple carbon sources and taxonomic position were established for three stock cultures of National Collection of Industrial Bacteria, Scotland. It was confirmed that NCIB 8077 belonged to the Cellulomonas species and that NCIB 8633 and NCIB 8634 belonged to the Pseudomonas species. Taxonomy of other cellulolytic bacteria published on various journals was also discussed.

  • The endophytic fungal populations of different tissues of Taxus baccata grown at high altitudes in West Bengal, India were explored. These isolated fungal populations represented different genera, which were screened for taxol production using immunoassay technique. The culture AAT-TS-$4_1$ that produced taxol was identified as Gliocladium sp. based on its cultural, morphological characteristics, internal transcribed spacer, and 18S rRNA sequence analysis. Kinetics of taxol production as a function of culture growth were investigated.

Characterization of a Brown Rot Fungus Isolated from Dwarf Flowering Almond in Korea

  • The fruits showing brown rot symptom on dwarf flowering almond were found in Gongju, Chungchungnam-Do in Korea in July 2005. Small water-soaked lesions on the fruits were initiated, and gradually developed to soft rot covered with gray conidia. Then the diseased fruits were shrunk and became grayish-black mummies. A fungus was isolated from the diseased fruit and its morphological, cultural and molecular genetic characteristics were investigated. Typical blastospores of Monilinia spp. were observed under a light microscope both from tissues of the diseased fruits and from PDA-grown cultures. The fungus grew well at $25^{\circ}C$ and on PDA. The ITS ribosomal DNA region (650 bp) of the fungus was amplified by PCR and analyzed. Comparative data on ITS sequence homology among Monilinia spp., ITS sequence-based phylogram and morphological characteristics showed that the fungus is Monilinia fructicola. This is the first report on Monilinia fructicola causing brown rot on fruits of dwarf flowering almond in Korea.

  • This study was carried out to obtain the basic information for improving the flavor quality of yakju. Three kinds of yakju were prepared with different yeast strains to investigate the effects of those strains on flavor components. A total of 23 strains were isolated from fruits such as apple, pear, persimmon and citron and two strains were excellent in producing ethanol and flavors. They were identified as Saccharomyces cerevisiae S 2 and Saccharomyces cerevisiae S 6 from morphological cultural test and physiological quality. Yakju A, B and C were prepared with S 2, S 6 and Saccharomyces cerevisiae IFO 1950, respectively. Flavor components of yakju were analyzed by gas chromatography and mass spectr ometry. A total of 57 peaks were detected and 13 compound were identified. They were 4 alcohol, 2 esters, 7 acids and miscellaneous compounds.

  • Several microorganisms which degrade phenol and trichloroethylene(TCE) were isolated from the activated sludge of a wastewater treatment plant. Among them, one isolate EL-04J showed the highest degradability and was identified as a Pseudomonas species according to morphological, cultural and biochemical properties. The phenol-induced cells of Pseudomonas EL-04J, which were preincubated in the mineral salts medium containing phenol as a sole carbon source, degraded 90% of 25$\mu$M TCE within 20h. This strain could also utilize some of methylated phenol derivatives (o-cresol, m-cresol and p-cresol) as the sole source of carbon and energy. Cresol-induced cells of Pseudomonas EL-04J also cometabolized TCE.

  • Many photosynthetic bacteria capable of hydrogen production were isolated from samples of mud flats of paddy field collected in Kim Hae and Dae Jeo. A strain 230 was selected for the high capability of hydrogen evolution. As the results of examination in physiological, morghological and cultural characteristics, the strain 230 was identified as Rhodopseudomonas sphaeroides.
