  • We evaluated the antioxidant compounds and activity of the methanolic extracts of foxtail millet (FM), proso millet (PM), and sorghum (SG) using different pulverizing methods (pin mill and ultra fine). The particle size of the FM, PM, and SG were 102.12, 89.52, and $102.25\;{\mu}m$, respectively, using the pin mill pulverizer. The sizes were 9.43, 9.52, and $10.18\;{\mu}m$, respectively, using the ultra fine pulverizer. There was no difference in moisture, crude fat, ash, or protein content between the two different pulverizing methods. The total ${\gamma}$-oryzanol content of the FM using the pin mill and ultra fine pulverizers was 116.07 and $145.30\;{\mu}g$/g, respectively. The total polyphenol content of the SG using the pin mill and ultra fine pulverizers was 14.58 and 15.03 mg/g extract residue, respectively. There was no difference in total flavonoid or tannin content of the methanol extracts between the two different pulverizing methods. The major phenolic compounds in FM were pyrogallol, gallic acid, (+)-catechin, salicylic acid, ferulic acid, and rutin; in PM, they were pyrogallol, rutin, gallic acid, kaempfrol, and salicylic acid; in SG, they were (+)-catechin, salicylic acid, pyrogallol, myricetin, hesperidin and chlorogenic acid. SG had a higher radical scavenging activity than FM or PM extracts. The DPPH and ABTS radical scavenging activities of the SG extracts using the ultra fine pulverizer were 178.10 and 251.56 mg TE/g extract residue, respectively. We noted a significant correlation between free radical scavenging activity and polyphenolic compound.

  • To study the effects of substances in paeonia seeds (Paeonia lactiflora Pall.) on lipid metabolism, crude methanol extract and secondary ether-soluble fraction out of defatted methanol extract and trans-resveratrol were prepared from the seeds and added to 0.5% (w/w) cholesterol diets for rats. Male Sprague-Dawley rats weighing 120$\pm$11 g were divided into six experimental groups: control group with no extra supplement, 0.1% (MP1) and 0.2% methanol extract (MP2) supplemented groups, 0.05% (EP1) and 0.1% ether-soluble fraction (EP2) supplemented groups and 0.02% resveratrol supplemented group. Experimental diets were fed ad libitum to the rats for 3 weeks. Body weight gains and food efficiencies were not different among the six experimental groups. Relative liver weights were lower in EP2 group compared to those in control group, but serum GOT and GPT levels of paeonia seed groups including trans-resveratrol group were not different from those of the control group. Serum total cholesterol levels reduced in EP2 and resveratrol groups but HDL-/total cholesterol ratios significantly increased in the four paeonia groups except EP1 group compared with the control group and serum triglyceride level lowered only in EP2 group than that of the control. However, liver cholesterol levels lowered in the five paeonia groups but triglyceride level lowered in MP2, EP1, EP2 groups than that of the control group. Fecal cholesterol excretion significantly increased in MP2, EP1, and EP2 groups than that of the control group, but bile acid excretions were not changed except that a reduction in EP2 group. These results suggest that paeonia seeds contain substances improving serum lipid status mostly via HDL pathway and resveratrol as monomer is one of the effective components but others including resveratrol oligmer are involved in the lipid improving effect.

  • The extracts from natural and fermented products such as Artemisia plants, Rhodiola Salientness, fermented soybeans and soybean paste were used to investigate the whitening effect. 10 g of Artemisia plant were added to 300 mL of ethanol and extracted by sonification at room temperature for 3 h. The extract was further partitioned by the equal volume percent in the order of the n-hexane, chloroform and ethyl acetate. 5 g of Rhodiola salientness was also added to 150 mL of methanol and extracted at the room temperature for 12 h. The effluents from a chromatographic column ($3.9{\times}250mm$, $C_{18}$, $15{\mu}m$) were collected and concentrated in two parts. The extraction of fermented soybeans and soybean paste were done by 60% ethanol. In this work, tyrosinase inhibitory activity and melanin inhibitory effect were measured to confirm the whitening effect. The water layer of Artemisia princeps Pampan showed the good inhibitory of antioxidant, while the hexane layer of Artemisia iwayomogi Kitamura and the chloroform layer of Artemisia princeps Pampan had the excellent melanin inhibitory effect. The Rhodiola salientness had the superior whitening effect to the arbutin in in-vivo melanin production ratio assay. However, the fermented soybeans and soybean paste did not show any whitening effect.

  • Gibberellin(GA) 3-$\beta$ hydroxylation is very important for the shoot elogation in the higher plants, since only 3$\beta$-hydryoxylated GAs promote shoot elogation in several plants. Fluctuation of 3$\beta$-hydryoxylase activity was examined during seed maturation using two cultivars of , P. vulgaris, Kentucky Wonder (normal) and Masterpiece (dwarf). Very immature seeds of both cultivars contain high level of 3$\beta$-hydroxylase activity (per mg protein). Both cultivars showed maximum of enzyme activity (per seed) in the middle of their maturation process. Gibberellin 3$\beta$-hydroxylase catalyzing the hydroxylation of GA20 to GA1 was purified 313-fold from very early immature seeds of P. vulgaris. Crude soluble enzyme extracts were purified by 15% methanol precipitation, hydrophobic interaction chromatogrphy, DEAE ion exchange column chromatography and gel filtration HPLC. The 3$\beta$-hydroxylase activity was unstable and lost much of its activity duting the purification. The molecular weight of purified enzyme was extimated to be 42, 000 by gel filtration HPLC and SDS-PAGE. The enzyme exhibited maximum activity at pH 7.7. The Km values for [2.3-3H] GA20 and [2.3-3H]GA9 were 0.29 $\mu$M and 0.33 $\mu$M, respectively. The enzyme requires 2-oxoglutarate as a cosubstrate; the Km value for 2-oxoglutarate was 250 $\mu$M using 3H GA20 as a substrate. Fe2+ and ascorbate significantly activated the enzyme at all purification steps, while catalase and BSA activated the purified enzyme only. The enzyme was inhibited by divalent cations Mn2+, Co2+, Ni2+, Cu2+, Zn2+, Cd2+ and Hg2+. Effects of several GAs and GA anaogues on the putrified 3$\beta$-hydroxylase were examined using [3H]GA9 and GA20 as a substrates. Among them, GA5, GA9, GA15, GA20 and GA44 inhibited the enzyme activity. [13C, 3H] GA20 was converted by the partially purified enzyme preparation to [13C, 3H]GA1, GA5 and GA6, which were identified by GC-MS, GA9 was converted only GA4, GA15 and GA44 were converted to GA37 and GA38, respectively. GA5 was epoxidized to GA6 by the preparation. This suggests that 3$\beta$-hydroxylation of GA20 and epoxidation of GA5 are catalyzed by the same enzyme in P, vulgaris.

  • Chemical fungicides have reduced Fusarium head blight (FHB) severity. However, by the effects of fungicide residues, they can only be used up to 30 days before time of harvest. Therefore, the development of new biofungicides that are applicable until harvest is required. In order to select plant extracts having antifungal activity against Fusarium graminearum for the control of FHB, we investigated the inhibitory effects of 225 medicinal plant extracts on spore germination of F. graminearum. Of these plant extracts, the methanol extract of Cirsium japonicum (CJ) roots showed the strongest antifungal activity. Through solvent partitioning, repeated column chromatography, and spore germination bioassay, two chemicals were purified and then their chemical structures were identified as ciryneol C (CC) and 1-heptadecene-11,13-diyne-8,9,10-triol (HD-ol) which are polyacetylene substances. Two active compounds effectively inhibited the germination of F. graminearum macroconidia; HD-ol ($IC_{50}$ of $3.17{\mu}g/ml$) showed stronger spore germination inhibitory activity than that of CC ($IC_{50}$ of $28.14{\mu}g/ml$). In addition, the wettable powder type formulation of ethyl acetate extract of CJ roots suppressed the development of FHB in dose-dependent manner, with control values of 78.92% and 31.56% at 250- and 500-fold dilutions, respectively. Combining these findings suggest that the crude extract of CJ roots containing polyacetylene compounds could be used as botanical fungicide for the control of FHB.
