제약폐기 은행잎중에 함유된 식물생육억제물질 분리동정 (Isolation and Chemical Structure Identification of Allelopathic Substances from the Ginkgo(Ginkgo biloba L.) Leaf Waste Produced by Phamaceutical Process)

  • 제약폐기은행잎의 유기물자원화 및 식물생장억제제 개발을 위한 기초자료로 활용코자 은행잎 제약폐기물중에 함유된 식물생육 억제물질을 분리 동정하고 이들물질을 처리하여 무와 벼의 발아 및 초기생육을 검토한 결과는 다음과 같다. 1) 폐기은행잎 조추출물 10% 용액은 무 종자 발아율을 저하시키고 유묘의 생육을 억제하였으나, 1% 농도에서는 뿌리 길이가 약간 짧아졌고, 0.1% 농도는 무처리와 비슷하였다. 벼 발아율은 조추출물 10% 농도에서 31%이상 억제되었다. 2) 지베렐린 $A_3(3.3{\times}10^{-6}M)$와 조추출액(10%)을 벼 유묘와 함께 처리하면 지베렐린 $A_3(3.3{\times}10^{-6}M)$ 단독처리보다 제2엽초장이 짧았다. 3) 조추출액의 산도를 달리한 3종의 액액분리물은 (EtOAc $(pH9)A_1$, EtOAc $(pH3)A_2$, BuOH $A_3$) 모두 1% 농도에서 무 종자의 발아를 완전히 억제하였다. 4) 조추출액을 액액분리, column chromatography, HPLC에 의한 분리정제를 통하여 활성성분을 분리하고, GC/MS, NMR 분석 결과 phenol류인 catechol로 판명되었으며, 폐기은행잎 29kg으로부터 분리한 catechol량은 0.76g이었다.

에탄올 농도에 따른 미나리 추출물의 영양성분 변화 (Changes in Nutritional Composition of Dropwort (Oenanthe javanica) Ethanol Extracts)

  • 본 연구는 미나리의 최적 추출 조건을 확립하기 위해 다양한 에탄올추출 농도가 미나리의 영양학적 성분 변화에 미치는 정도를 관찰함으로써 진행되었다. 본 실험에서는 열수추출물, 에탄올추출물(50%, 80%, 95%)로 추출 농도를 다르게 하여 미나리를 추출하였다. 수율을 측정한 결과 50% 에탄올 추출물에서 44.67%로 가장 높게 나타났다. 일반성분 분석 결과, 50% 에탄올추출물에서 단백질 함량이 6.76%, 탄수화물 함량이 19.60%로 가장 높게 나타났다. 지방 함량은 열수추출물과 비교하여 에탄올추출물에서 유의적으로 증가하였으며(P<0.05), 에탄올추출 농도에 따라 유의차가 있었다 (P<0.05). 식이섬유를 측정한 결과, 50% 에탄올추출물을 제외한 에탄올추출물(80%, 95%)에서 열수추출물과 유의차가 나타났으며(P<0.05), 두 실험군 간에도 유의적 차이가 있었다(P<0.05). 총당 함량을 측정한 결과, 열수추출물에 대해 에탄올추출물은 통계적 유의차가 있었고(P<0.05), 에탄올 농도에 따라서도 유의차를 나타냈으며(P<0.05), 95% 에탄올추출 시 5.35%로 가장 낮게 측정되었다. 비타민 A 함량은 열수추출물에 대해 에탄올추출물이 유의하게 증가하였으며(P<0.05), 에탄올추출 농도에 따라 유의차가 있었다(P<0.05). 무기성분 측정 결과, 나트륨, 칼슘, 철의 경우 열수추출물에 대해 에탄올추출물이 유의하게 감소하였고(P<0.05), 에탄올추출 농도가 증가함에 따라 유의적으로 감소하였다(P<0.05). 따라서 에탄올추출 방법을 사용 목적에 따라 적용함으로써 미나리를 이용한 우수한 가공식품 개발 가능성과 이용률 향상에 도움이 될 것으로 판단된다.

약용식물로부터 오이흰가루병에 대한 항균성물질 탐색 및 동정 (Screening and Identification of Fungicidal Compounds Derived from Medicinal Plants against Cucumber Powdery Mildew)

  • 50종의 약용식물추출액을 공시하여 흰가루병에 대한 방제효과를 검정하고 생리활성 물질을 분리, 동정하였다. 공시약용식물중에서 대황의 물추출액은 200배의 희석농도에서도 100%의 포자발아억제 효과를 나타내서 가장 효과가 있었고, 대황의 물추출액 50배, 알콜추출액 100배, 조물질 500배, 표준품 1000배 희석농도까지도 60%이상의 발병억제효과가 있었으며, 조물질 500배액에서 2회 이상 살포하면 100% 발병억제 효과가 있었다. 그리고 조물질 100배 액에서는 약해가 있었으나 500배액에서는 약해가 없었다. 대황에 함유되어 있는 생리활성물질은 anthraquinone유도체인 1,8-dihydroxy-3-methyl-9,10-anthracenedione과 1,8-dihydroxy-3-methoxy-6-methyl-9,10-anthracenedione으로 잠정 동정되었다.

Antinociceptive and gastro-protective effect of the ethanolic extract of the flowering top of Anthocephalus Cadamba Roxb

  • The effect of alcoholic extract of Anthocephalus (A.) Cadamba Roxb. was evaluated in experimental models of pain and ulcer. Hot tail flick test, hot plate test and acetic acid induced writhing test were employed for evaluating the peripheral as well as central analgesic mechanism exerted by the extracts. Gastroprotective activity was examined by HCl and ethanol induced gastric damage test. Test group received crude extract 500 mg/kg showed maximum time needed for the response against thermal stimuli (6.26 ${\pm}$ 0.439 s) which is comparable to diclofenac sodium (6.56 ${\pm}$ 0.381 s) in hot tail flick method. These experimental results also followed the experimental results of hot plate test where crude extract 500 mg/kg showed maximum time needed for the response against thermal stimuli (4.74 ${\pm}$ 0.234 s) which is comparable to diclofenac sodium (5.58 ${\pm}$ 0.585 s). The crude extract at 500 and 250 mg/kg showed significant reduction in acetic acid induced writhing in mice with a maximum effect of 68.026% reduction at 500 mg/kg dose which is comparable to standard diclofenac sodium (79.93%). In gastroprotective study the extract of A. Cadamba (250 and 500 mg/kg) significantly inhibited ulceration induced by both HCl and ethanol dose dependently. Results of the study suggest that the extract possesses both analgesic and gastroprotective activity on mice.

다시마 추출액의 항산화 및 항염증 활성에 대한 효모 발효의 영향 (Effect of Yeast Fermentation on the Antioxidant and Anti-inflammatory Activity of Sea Tangle Water Extract)

  • To examine the effective use of seaweeds, sea tangle (Laminaria japonica) was extracted with water and the resultant extracts were fermented with Saccharomyces cerevisiae. Four strains of S. cerevisiae were cultured in aqueous extracts from sea tangle. S. cerevisiae SC-2, which was isolated from a traditional Korean fermented food (Meju), was selected for further study based on the results of a sensory evaluation. No significant differences in proximate compositions, such as moisture, crude protein, crude fat, and crude ash, of the sea tangle extracts before and after fermentation were observed. The reducing sugar decreased as the fermentation period increased, and the contents of some free amino acids were also affected by S. cerevisiae SC-2 fermentation. However, the content of glutamic acid, which is a major taste compound in sea tangle extract, was not affected by fermentation for up to 36 hr by the SC-2 strain. To determine the antioxidant activity of fermented sea tangle extract (fermented for 36 hr by SC-2 strain), the radical scavenging activities of 1,1-diphenyl-2-picrylhydrazyl (DPPH), superoxide, and nitric oxide were investigated and xanthin oxidase inhibition assay was performed. The antioxidant activity increased by 8 to 35%. The greatest enhancement of antioxidant activity was seen in the superoxide radical scavenging assay with $100\;{\mu}g/mL$ of raw and fermented sea tangle extract. The anti-inflammatory activity of fermented sea tangle extract was also enhanced. The fermented sea tangle extract showed 34.2% inhibitory activity against nitric oxide synthesis versus 11.9% for raw sea tangle extract at $100\;{\mu}g/mL$ concentration. These results suggest that fermented aqueous extracts from sea tangle are a useful resources.

유산균 발효에 의한 톳(Hizikia fusiforme) 추출액의 이화학적 및 관능적 특성 변화 (Changes in Physicochemical and Sensory Properties of Hizikia fusiforme Water Extract by Fermentation of Lactic Acid Bacteria)

  • This study was conducted to improve the food functional and sensory food quality of Hizikia fusiforme by the fermentation of lactic acid bacteria. Seven strains of lactic acid bacteria isolated from traditional Korean fermented food were inoculated and cultivated in H. fusiforme water extract. Among them, Lactobacillus brevis LB-20, isolated from Kimchi, was selected for further study by considering the results of bacterial growth, DPPH radical scavenging activity, and sensory evaluation. No significant differences in proximate compositions (moisture, crude protein, crude fat, and crude ash) were observed by the fermentation of L. brevis LB-20. The most dramatical change was the conversion from glutamate to ${\gamma}$-aminobutyric acid (GABA) in H. fusiforme water extract fermented by L. brevis LB-20. The GABA content increased approximately 60-fold after 48 hr of fermentation. The bacterial fermentation also resulted in low-molecularization of the extract. The particle size of the fermented extract became approximately 4-fold smaller than that of the law extract. In addition, the analysis of volatile flavor compounds using GC/MS revealed that the bacterial fermentation dramatically removed off-flavors such as acetaldehyde, haxanal, diallyl disulphide and 1-penten-2-ol in the H. fusiforme extract.

Proximate, Phytochemical, and In Vitro Antimicrobial Properties of Dried Leaves from Ocimum gratissimum

  • Ocimum gratissimum is a common plant in the tropics and has been used in food and medicine. Its usage in food and medicine could be attributed to its phtyochemical and antimicrobial properties. In this study we investigated the proximate, phytochemical, and antimicrobial attributes of air dried leaves of O. gratissimum. The aqueous extract was found to contain phtyochemicals with alkaloid and saponin present in appreciable amounts. The proximate analysis (crude protein and crude fibre content were 15.075% and 17.365%, respectively) showed that the leaf could be a good source of protein and fibre. The aqueous ethanolic extract of the leaf exhibited activity against a wider range of organisms when compared to the aqueous extract at the investigated concentrations. Aqueous ethanolic extracts of O. gratissimum leaf was active against Escherichia coli, Pseudomonas aeruginosa, Staphylococcus aureus, and Bacillus cereus and the aqueous extract of the leaf was active against P. aeruginosa.

Antinociceptive and sedative effects of the bark of Cerbera odollam Gaertn.

  • The crude methanolic extract of the bark of Cerbera odollam Gaertn. was evaluated for its possible antinociceptive and neuropharmacological activities in animal models. At the dose of 250 and 500 mg/kg body weight, the extract showed a significant antinociceptive effect in acetic acid induced writhing in mice comparable to that produced by aspirin, used as standard drug (P<0.001). The extract significantly reduced the time of onset of sleep (P<0.01) and potentiated the pentobarbital induced sleeping time in mice at the dose of 400 mg/kg of body weight significantly (P<0.001). It also decreased the open field score in open field test significantly at the dose of 400 mg/kg of body weight (P < 0.05). The obtained results tend to suggest the probable antinociceptive and neuropharmacological activities of the crude extract.

홍삼의 알콜 농도별 추출 엑스의 수율과 사포닌 함량 (Extract Yields and Saponin Contents of Red Ginseng Extracts prepared with various Concentrations of Ethanol)

  • Red ginseng extracts were prepared with various concentrations of ethanol. Extract yields were examined and saponins in the extracts were identified and determined by TLC and HPLC, respectively. Yields of the extracts, $19.7{\sim]50.3%$, were the highest in water extract and showed significant decrease with the increase of ethanol concentration used for extraction. Contrary to the extract yields, saponin yields from red ginseng were conspicuously increased with the increase of ethanol concentration and were $3.47{\sim}5.13%$ of crude saponins and $1.28{\sim}1.93%$ of six major ginsenosides. Saponin contents in the red ginseng extracts were $6.9{\sim}24.2%$, of crude saponin and $2.57{\sim}9.22%$, of six major ginsenosides.

Insecticidal Activity of Extracts Isolated from Syzygium Aromaticum

  • This study separated the crude extract (70% ethanol) of and its three fractions (hexane, chloroform and ethyl acetate extracts) on the basis of polarity indexes, and examined for their insecticidal activities against aphid (Uroleucon lactucicola). For crude extraction, the 70% ethanol extract showed the best extract yield (58.0%) and insecticidal activity (69.0%) among the various concentrations tested (water, 30% ethanol, 50% ethanol, 70% ethanol and 95% ethanol). The major chemical compounds of different fractions (hexane, chloroform and ethyl acetate extracts) were identified as eugenol by head space-GC-MS analysis. The hexane extract showed the highest eugenol content (43.7%) and insecticidal activity (80.0%). The insecticidal activity is accordingly believed to be attributable to the eugenol component. This may provide a useful starting point for the development of bio-pesticides.
