생약재중 천연유래 이산화황 함유량 및 그 출처 (Naturally Occurring of Sulfur Dioxide in Medicinal Herbs(Crude Drug Materials) and Its Origin)

  • 표백제 규제 대상인 한약재 28종 101건을 재배 현장에서 채취하여 수세, 건조 후 천연유래 이산화황 함유량을 모니어-월리암스 변법과 산증류-이온 크로마토그래피법으로 조사하고 그 출처를 규명하였다. 모니어-윌리암스 변법으로 측정한 한약재의 천연유래의 이산화황 함유량은 0.6-4.7ppm으로 의이인(0.6ppm)이 가장 낮았으며 황기(4.7ppm)가 가장 높았으나 산증류-이온 크로마토그래피법에 의해 측정한 천연유래 이산화황 함유량은 0.31-3.20ppm으로 택사(0.31ppm)가 가장 낮고 고본(3.3ppm)이 가장 높은 함유량을 보였다. 시험법에 따른 측정값의 차이 및 이산화황의 출처를 규명하기 위하여 GC-MS로 휘발성분을 분석한 결과, 측정값의 차이는 모니어-윌리암스 변법에서 휘발성 유기화합물에 의한 false positive response로 일어났으며, 한약재에서 측정되는 천연유래 이산화황은 한약재가 함유하고 있는 휘발성 황화합물에 기인함을 확인하였다.

  • With the Sunshine policy, exchange of materials and cultures inter Koreas has been broadened and expectancy of reunification is getting higher. Especially, medical supplies and medicines are one of the biggest parts in the exchange goods. So, preparing an unified official drug standard preparing new medical administration system is required. We compared the Korean pharmacopoeia with North Korean Pharmacopoeia. Two pharmacopoeias have been developed in different direction and have many differences in the nomenclature and format. In this study, we compared general notices, general rules for preparations and crude drugs, monographs, general tests, processes and apparatus.

Antinociceptive and Antiinflammatory Effect of a Diterpene Isolated from the Aerial Part of Siegesbeckia pubescens

  • The aerial part of Siegesbeckia pubescens (Compositae) has been used to treat rheumatoid arthritis and hypertension in the Oriental medicine. This crude drug has been used without process (SP-0) or with three times-process of steaming and drying (SP-3) or the nine times of that process (SP-9). To search for the antinociceptive anti-inflammatory components from this crude drug, activity-directed fractionation was performed on this crude drug. Since the $CHCl_3$ extract was shown to have a more potent effect than other extracts, it was subjected to silica gel & ODS column chromatography to yield two diterpene compounds (1). Compound 1 was structurally identified as ent-16 (H, 17-hydroxykauran-19-oic acid, which were tentatively named siegeskaurolic acid A. A main diterpene, siegeskaurolic acid A was tested for the antiiflammatory antinociceptive effects using both hot plate- and writhing anti-nociceptive assays and carrageenan-induced anti-inflammatory assays in mice and rats. Pretreatment with siegeskaurolic acid A (20 and 30mg/kg) significantly reduced the stretching episodes, action time of mice and carrageenan-induced edema. These results support that siegeskaurolic acid is a main diterpene responsible for antinociceptive and antiiflammatory action of S. pubescens. In addition, the assays on SP-0, SP-3 and SP-9 produced the experimental results that SP-9 had more significant effects than other two crude drugs. These results suggest that the processing on the original plant may lead to the higher pharmacological effect.

청쾌원의 현미감정 연구 (Microscopic Identification of the 'Chung Kwei Won')

  • 'Chung Kwei Won(淸快園)' is a Chinese patent medicine, which is used for stomachic disorder, acute and chronic indigestion, dispepsia, vomiting and diarrhea in Korea. This preparation consists of 16 kinds of powdered crude drugs. For the identification of individual crude drugs in such powdery mixtures, microscopic method may be used conviniently as it requires only a small amount of specimens. In this paper, the effectiveness of this method is exemplified by the identification of the ingredients in 'Chung Kwei Won' which contains 16 powder crude drug ingredients.

  • Geo Pung Ji Bo Dan (祛風至寶丹) is a Chinese patent medicine, which has been used for various purposes in Korea. Geo Pung Ji Bo Dan is composed 26 kinds of powdered crude drugs and has been used for palsy and mental disorder. For the identification of individual crude drugs in such powdery mixtures, microscope method may advantageously be used as it require only a small amount of specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 26 crude drug ingredients in Geo Pung Ji Bo Dan.

  • Bangpoong (防風) is a popular crude drug used to expel wind from the body surface (祛風解表) to remove dampness (勝濕). And to relieve pain (正痛) and spasm (正痙). In China and Japan, roots of Saposhnikovia divaricata (Turcz.) Schischk. Is used as Bangpoong. However, the roots of Peucedanum japonicum Thunb. Or Glehnia littoralis (A. Gray) Fr. Schmidt ex Miquel, being called Sikbangpoong (植防風) and Wonbangpoong (元防風) respectively are used instead of Bangpoong in Korea. The ITS regions of nuclear ribosomal DNA were analyzed to determine original plants and to design a molecular identification method for the crude drugs used as Bangpoong in Korea and China. It is demonstrated that RFLP analysis via PCR has the great potential as a novel tool to test crude drugs for the quality control and standardization.

  • An Chung Hwan(安中丸) is a Chinesε patent medicine, which has been used for various purposes in Korea. An Chung Hwan is composed of 15 kinds of powdered crude drugs and has been used for stomachic disorder, acute and chronic indigestion, dyspepsia, and diarrhea. For the identification of individual crude drugs in such powdery mixtures, microscopic method may advantageously be used as it requires only a small amount of specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 15 crude drug ingredients in An Chung Hwan.

  • 'Chung Shin Whan(靑神丸)' is a Chinese patent medicine, which has been used for stomachic disorder, acute and chronic indigestion, dyspepsia, vomiting and diarrhea in Korea. This medicine consists of 14 kinds of powdery mixtures, microscopic method be used advantageously as it requires only a small amount of specimens. In this paper, the effectiveness of this method is exemplified by the identification of the ingredients in "Chung Shin Whan' which contains 14 powdered crude drug ingredients.

  • Chinese crude drug 'JeonHo (前胡)' has been used mainly cough and dyspnea in respiratory infections. To clarify the botanical origin of 'JeonHo' in Korean market, the anatomical characterization of Anthriscus sylvestris, Angelica decursiva and Peucedanum praeruptorum were studied. As a result, it was clarified that 'JeonHo' in Korean market was the root of Peucedanum praeruptorum.

  • 'Hae Cheong Whan' is a Chinese patent preparation, which has been used for cough in Korea. This preparation consists of 18 powder crude drugs. The microscopic method is very useful to identify individual gradients in powdery mixture because it requires only a small amount of specimens. In this study, it is demonstrated that the microscopic method is very effective for the identification of 18 crude drug ingredients including in 'Hae Chung Hwan'.
